A redox switch allows binding of Fe(II) and Fe(III) ions in the cyanobacterial iron-binding protein FutA from Prochlorococcus.

The marine cyanobacterium Prochlorococcus is a main contributor to global photosynthesis, whilst being limited by iron availability. Cyanobacterial genomes generally encode two different types of FutA iron-binding proteins: periplasmic FutA2 ABC transporter subunits bind Fe(III), while cytosolic FutA1 binds Fe(II). Owing to their small size and their economized genome Prochlorococcus ecotypes typically possess a single futA gene. How the encoded FutA protein might bind different Fe oxidation states was previously unknown. Here, we use structural biology techniques at room temperature to probe

Vibrio cholerae O1 El Tor strains linked to global cholera show region-specific patterns by pulsed-field gel electrophoresis.

Vibrio cholerae O1 El Tor, causative agent of the ongoing seventh cholera pandemic, is native to the aquatic environment of the Ganges Delta, Bay of Bengal (GDBB). Recent studies traced pandemic strains to the GDBB and proposed global spread of cholera had occurred via intercontinental transmission. In the research presented here, NotI-digested genomic DNA extracted from V. cholerae O1 clinical and environmental strains isolated in Bangladesh during 20042014 was analyzed by pulsed-field gel electrophoresis (PFGE). Results of cluster analysis showed 94.67% of the V. cholerae strains belonged to

The rice/maize pathogen Cochliobolus spp. infect and reproduce on Arabidopsis revealing differences in defensive phytohormone function between monocots and dicots.

The fungal genus Cochliobolus describes necrotrophic pathogens that give rise to significant losses on rice, wheat, and maize. Revealing plant mechanisms of non-host resistance (NHR) against Cochliobolus will help to uncover strategies that can be exploited in engineered cereals. Therefore, we developed a heterogeneous pathosystem and studied the ability of Cochliobolus to infect dicotyledons. We report here that C. miyabeanus and C. heterostrophus infect Arabidopsis accessions and produce functional conidia, thereby demonstrating the ability to accept Brassica spp. as host plants. Some ecotyp

Complexities of gene expression patterns in natural populations of an extremophile fish (Poecilia mexicana, Poeciliidae).

Variation in gene expression can provide insights into organismal responses to environmental stress and physiological mechanisms mediating adaptation to habitats with contrasting environmental conditions. We performed an RNA-sequencing experiment to quantify gene expression patterns in fish adapted to habitats with different combinations of environmental stressors, including the presence of toxic hydrogen sulphide (H2 S) and the absence of light in caves. We specifically asked how gene expression varies among populations living in different habitats, whether population differences were consist

Evidence for cross-tolerance to nutrient deficiency in three disjunct populations of Arabidopsis lyrata ssp. lyrata in response to substrate calcium to magnesium ratio.

Species with widespread distributions that grow in varied habitats may consist of ecotypes adapted to a particular habitat, or may exhibit cross-tolerance that enables them to exploit a variety of habitats. Populations of Arabidopsis lyrata ssp. lyrata (L.) O'Kane & Al-Shehbaz grow in a wide variety of edaphic settings including serpentine soil, limestone sand, and alluvial flood plains. While all three of these environments share some stressors, a crucial difference among these environments is soil calcium to magnesium ratio, which ranges from 25:1 in the limestone sand to 0.2:1 in serpentine

Contrasting egg-laying, adult abundance and genetics of the dengue vector Aedes aegypti in two ecologically distinct regions in Kenya.

BACKGROUND: Aedes aegypti is widely distributed in the tropics and sub-tropics and is a major vector of several arboviruses, such as dengue and chikungunya viruses. Mapping the behavioral and genetic divergence among the mosquito populations can enhance our understanding of spatiotemporal variation in disease transmission. METHODS: Cross-sectional surveys were carried out between 2022 and 2023 to investigate egg-laying abundance in ovitraps as well as adult abundance in BG Sentinel traps baited with carbon dioxide. Both trap types were deployed in outdoor environments in two peri-urban areas

The genomic architecture of local adaptation in two connected populations of three-spined stickleback.

Populations often adapt to their local environments despite the homogenizing effects of gene flow, but the genomic mechanisms enabling this process remain unclear. Theory predicts that adaptive divergence under high connectivity is favored when beneficial alleles cluster in regions of reduced recombination, a pattern that can be reinforced by structural variants (SVs). We investigated this in three-spined sticklebacks (Gasterosteus aculeatus) from the St. Lawrence Estuary, where distinct freshwater and marine ecotypes meet and interbreed along a short ecological gradient. Using long- and short

Genetic and Ecological Divergence Between Northwest Atlantic Killer Whale Populations.

Killer whales (Orcinus orca) exhibit substantial genetic and ecological variation across their global distribution. Differentiation between neighboring or sympatric populations is thought to be driven by foraging specialization and social organization, which can lead to reproductive isolation and facilitate the emergence of distinct ecotypes or morphotypes. Here, we use whole-genome resequencing and compound-specific stable isotope analysis of amino acids to investigate links between genetic and ecological differentiation in two genetically distinct killer whale populations in the northwest At

Functional mutations in the thyroid-stimulating hormone receptor in natural stickleback populations at sites identical to human disease-causing mutations.

BACKGROUND: Thyroid hormones regulate multiple physiological functions, including metabolism, reproduction, and metamorphosis. Although there are variations in thyroid hormone signaling between populations and species, the causative mutations underlying these variations have rarely been identified. Here, we investigated whether information regarding the causative genes and mutations responsible for human thyroid diseases could assist with the identification of functional mutations in natural stickleback populations, which vary in thyroid hormone signaling between marine and stream-resident eco

Different Molecular Phenotypes of Progression in BRAF- and RAS-Like Papillary Thyroid Carcinoma.

BACKGRUOUND: Papillary thyroid carcinoma (PTC) can be classified into two distinct molecular subtypes, BRAF-like (BL) and RASlike (RL). However, the molecular characteristics of each subtype according to clinicopathological factors have not yet been determined. We aimed to investigate the gene signatures and tumor microenvironment according to clinicopathological factors, and to identify the mechanism of progression in BL-PTCs and RL-PTCs. METHODS: We analyzed RNA sequencing data and corresponding clinicopathological information of 503 patients with PTC from The Cancer Genome Atlas database.

Ringed seal (Pusa hispida) seasonal movements, diving, and haul-out behavior in the Beaufort, Chukchi, and Bering Seas (2011-2017).

Continued Arctic warming and sea-ice loss will have important implications for the conservation of ringed seals, a highly ice-dependent species. A better understanding of their spatial ecology will help characterize emerging ecological trends and inform management decisions. We deployed satellite transmitters on ringed seals in the summers of 2011, 2014, and 2016 near Utqiaġvik (formerly Barrow), Alaska, to monitor their movements, diving, and haul-out behavior. We present analyses of tracking and dive data provided by 17 seals that were tracked until at least January of the following year. Se

The epigenetic memory of temperature during embryogenesis modifies the expression of bud burst-related genes in Norway spruce epitypes.

Epigenetic memory affects the timing of bud burst phenology and the expression of bud burst-related genes in genetically identical Norway spruce epitypes in a manner usually associated with ecotypes. In Norway spruce, a temperature-dependent epigenetic memory established during embryogenesis affects the timing of bud burst and bud set in a reproducible and predictable manner. We hypothesize that the clinal variation in these phenological traits, which is associated with adaptation to growth under frost-free conditions, has an epigenetic component. In Norway spruce, dehydrins (DHNs) have been a

Eco-genetic structure of Bacillus cereus sensu lato populations from different environments in northeastern Poland.

The Bacillus cereus group, which includes entomopathogens and etiologic agents of foodborne illness or anthrax, persists in various environments. The basis of their ecological diversification remains largely undescribed. Here we present the genetic structure and phylogeny of 273 soil B. cereus s.l. isolates from diverse habitats in northeastern Poland, with samplings acquired from the last European natural forest (Białowieża National Park), the largest marshes in Europe (Biebrza National Park), and a farm. In multi-locus sequence typing (MLST), despite negative selection in seven housekeeping

A complex eIF4E locus impacts the durability of va resistance to Potato virus Y in tobacco.

Many recessive resistances against potyviruses are mediated by eukaryotic translation initiation factor 4E (eIF4E). In tobacco, the va resistance gene commonly used to control Potato virus Y (PVY) corresponds to a large deletion affecting the eIF4E-1 gene on chromosome 21. Here, we compared the resistance durability conferred by various types of mutations affecting eIF4E-1 (deletions of various sizes, frameshift or nonsense mutations). The 'large deletion' genotypes displayed the broadest and most durable resistance, whereas frameshift and nonsense mutants displayed a less durable resistance,

Assessment of genetic diversity of landraces of Dendrocalamus hamiltonii using AFLP markers and association with biochemical traits.

Fermented bamboo shoots are popular traditional food items of various ethnic groups of the northeastern India, especially in Manipur State. Dendrocalamus hamiltonii is an economically important bamboo species used to produce fermented bamboo shoots. We studied genetic variability of this bamboo species in Chandel and Imphal-East (commercial production districts), using AFLP molecular markers. Each of the selected primers detected polymorphisms and 1614 (95.8%) were found to be polymorphic. Cluster analysis based on Dice similarity coefficients using UPGMA differentiated the populations into tw

Concentrations of essential and non-essential elements in eastern North Pacific killer whales (Orcinus orca).

Essential and non-essential elements can harm marine wildlife and impact ecosystems. Using liver (n = 35) and kidney (n = 17) samples from 35 animals from three distinct killer whale (Orcinus orca) ecotypes (fish-eating residents, mammal-eating transients, and offshore shark-eating specialists) stranded from California to Alaska and Hawaii, we determined elemental concentrations (arsenic (As), barium (Ba), beryllium (Be), calcium (Ca), cadmium (Cd), cobalt (Co), copper (Cu), chromium (Cr), iron (Fe), mercury (Hg), methylmercury (MeHg), manganese (Mn), molybdenum (Mo), magnesium (Mg), nickel (N

Phytostabilization potential and microbial response to the reclamation of native Cynodon dactylon in spoil heaps from a multiple-metal mining site in Southwest China.

Phytocapping offers a sustainable approach for managing exposed tailings by mitigating pollutant spread and enhancing phytoremediation. This study investigates the potential of Bermudagrass (Cynodon dactylon) as a pioneering plant for rehabilitating tailings from an open-pit lead-zinc mine in Southwest China. Our findings demonstrate that Bermudagrass significantly improved soil quality and multifunctionality compared to adjacent bare tailings. Soil improvements included increases in organic matter (107%), total and available nitrogen (50% and 110%, respectively), available phosphorus (170%),

Local ancestry analysis reveals genomic convergence in extremophile fishes.

The molecular basis of convergent phenotypes is often unknown. However, convergence at a genomic level is predicted when there are large population sizes, gene flow among diverging lineages or strong genetic constraints. We used whole-genome resequencing to investigate genomic convergence in fishes ( Poecilia spp.) that have repeatedly colonized hydrogen sulfide (H2S)-rich environments in Mexico. We identified genomic similarities in both single nucleotide polymorphisms (SNPs) and structural variants (SVs) among independently derived sulfide spring populations, with approximately 1.2% of the g

Genotypic characterization and prevalence of virulence factors among Canadian Escherichia coli O157:H7 strains.

In this study, the association between genotypic and selected phenotypic characteristics was examined in a collection of Canadian Escherichia coli O157:H7 strains isolated from humans and cattle in the provinces of Alberta, Ontario, Saskatchewan, and Quebec. In a subset of 69 strains selected on the basis of specific phage types (PTs), a strong correlation between the lineage-specific polymorphism assay (LSPA6) genotype and PT was observed with all strains of PTs 4, 14, 21, 31, 33, and 87 belonging to the LSPA6 lineage I (LSPA6-LI) genotype, while those of PTs 23, 45, 67, and 74 belonged to LS
