Selective ligand activity at Nur/retinoid X receptor complexes revealed by dimer-specific bioluminescence resonance energy transfer-based sensors.

Retinoid X receptors (RXRs) play a role as master regulators because of their capacity to form heterodimers with other nuclear receptors (NRs). Accordingly, retinoid signaling is involved in multiple biologic processes, including development, cell differentiation, metabolism, and cell death. However, the role and function of RXRs in different heterodimer complexes remain unidentified, mainly because most RXR drugs (called rexinoids) are not selective of specific heterodimer complexes. The lack of selectivity strongly limits the use of rexinoids for specific therapeutic approaches. To better ch

Highly selective detection of microRNA based on distance-dependent electrochemiluminescence resonance energy transfer between CdTe nanocrystals and Au nanoclusters.

A distance-dependent electrochemiluminescence resonance energy transfer (ERET) system based on CdTe nanocrystals and Au nanoclusters (Au NCs) was designed with the aid of ligase for highly selective detection of microRNA (miRNA). First, Au NCs functionalized hairpin DNA was synthesized via Au-S chemistry, and characterized with transmission electron microscopy and dynamic light scattering. The resulting hairpin DNA-Au NCs composite can be bound to the carboxylated CdTe nanocrystals via amide reaction on glass carbon electrode. The strong interaction between CdTe nanocrystals and AuNCs led to t

Sensitive detection of polycyclic aromatic hydrocarbons using CdTe quantum dot-modified TiO₂ nanotube array through fluorescence resonance energy transfer.

CdTe quantum dots (QDs) are prepared on TiO(2) nanotubes (TiO(2) NTs), for the first time, with pulse electrodeposition. A novel single-drop optical sensor is prepared with the CdTe QDs-modified TiO(2) NTs, and applied for the detection of polycyclic aromatic hydrocarbons (PAHs) based on fluorescence resonance energy transfer (FRET). Excited at 270 nm, the sensor shows fluorescence emission at around 370 nm. As PAHs are with absorption/fluorescence emission at around 364/410 nm, FRET happens between the CdTe QDs and PAHs with the CdTe QDs as donors and PAHs as receptors. The sensitivity is dep

Spatially pathogenic forms of tau detected in Alzheimer's disease brain tissue by fluorescence lifetime-based Förster resonance energy transfer.

In tauopathies including Alzheimer's disease (AD) tau molecules have lost their normal spatial distance to each other and appear in oligomeric or aggregated forms. Conventional immunostaining methods allow detection of abnormally phosphorylated or conformationally altered aggregated tau proteins, but fail to visualize oligomeric forms of tau. Here we show that tau molecules that lost their normal spatial localization can be detected on a subcellular level in postmortem central nervous system (CNS) tissue sections of AD patients by fluorescence lifetime-based Förster resonance energy transfer (

Conformational substates of calmodulin revealed by single-pair fluorescence resonance energy transfer: influence of solution conditions and oxidative modification.

A calmodulin (CaM) mutant (T34,110C-CaM) doubly labeled with fluorescence probes AlexaFluor 488 and Texas Red in opposing domains (CaM-DA) has been used to examine conformational heterogeneity in CaM by single-pair fluorescence resonance energy transfer (spFRET). Burst-integrated FRET efficiencies of freely diffusing CaM-DA single molecules yielded distributions of distance between domains of CaM-DA. We recently reported distinct conformational substates of Ca(2+)-CaM-DA and apoCaM-DA, with peaks in the distance distributions centered at approximately 28 A, 34-38 A, and 55 A [Slaughter et al.

Use of fluorescence resonance energy transfer hybridization probes to evaluate quantitative real-time PCR for diagnosis of ocular toxoplasmosis.

Toxoplasma gondii infection is an important cause of chorioretinitis in Europe and the United States. Ophthalmological examination and a good clinical response to adequate therapy mainly support ocular toxoplasmosis diagnosis. However, clinical diagnostic may be difficult in some atypical cases. In these cases, laboratory confirmation, based on detection of local specific antibodies and parasite DNA by conventional PCR, is therefore important to confirm the disease etiology. More recently, real-time PCR has been developed to improve prenatal congenital toxoplasmosis diagnosis. We therefore exa

Rapid detection and differentiation of human pathogenic orthopox viruses by a fluorescence resonance energy transfer real-time PCR assay.

BACKGROUND: The orthopox viruses that are pathogenic for humans include variola major virus (VAR), monkeypox virus (MPV), cowpox virus (CPV), and to a lesser extent, camelpox virus (CML) and vaccinia virus (VAC). PCR is a powerful tool to detect and differentiate orthopox viruses, and real-time PCR has the further advantages of rapid turnaround time, low risk of contamination, capability of strain differentiation, and use of multiplexed probes. METHODS: We used real-time PCR with fluorescence resonance energy transfer technology to simultaneously detect and differentiate VAR, MPV, CPV/VAC, an

Detection, differentiation, and quantitation of pathogenic leishmania organisms by a fluorescence resonance energy transfer-based real-time PCR assay.

Real-time technology eliminates many of the pitfalls of diagnostic PCR, but this method has not been applied to differentiation of Leishmania organisms so far. We have developed a real-time PCR that simultaneously detects, quantitates, and categorizes Leishmania organisms into three relevant groups causing distinct clinical pictures. The analytical sensitivity (detection rate of >or=95% at 94.1 parasites/ml of blood) was within a range that has been determined previously to facilitate the confirmation of visceral leishmaniasis from peripheral blood. Parasites were successfully detected in 12 d

Laser induced resonance energy transfer--a novel approach towards achieving high sensitivity in capillary electrophoresis. I. Clinical diagnostic application.

Most of the procedures currently performed by capillary zone electrophoresis (CZE) with laser induced fluorescence detection requires prior derivatization. This increases cost, the turn-around-time and chances of extraneous contaminations. CZE with laser induced resonance energy transfer is demonstrated as a viable alternative for detecting non-fluorescent compounds with no prior derivatization. The feasibility of this approach is demonstrated by separating and directly detecting salicylic acid (2,4-dihydroxybenzoic acid), gentisic acid (o-methoxybenzoic acid), salicyluric acid (o-hydroxyhippu

Two-wave mixing and energy transfer in Bi(12) SiO(20) crystals: application to image amplification and vibration analysis.

The energy transfer between two beams (signal and reference, respectively) writing a dynamic-volume hologram in photorefractive BSO crystals is applied to the image amplification of a diffuse object. The image intensity transmitted by the crystal is amplified 10x in the presence of the pump reference beam. The crystal is used in the drift recording mode (applied electric field, E(0) = 10 kV cm(-1); fringe spacing, Lambda = 3 microm), and beam coupling is induced by the nonlocal response of the crystal that is due to the fringe displacement at a constant speed. We have applied this two-wave mix

UV-Absorbing Energy Transfer and Morphology Regulation by D-A-Type Integrated Additives for Stable and Efficient Organic Solar Cells.

Organic solar cells (OSCs) face persistent challenges in maintaining both high efficiency and operational stability. Here, we report a rational design of D-A-type additives (UV326-DBOPA and UV327-DBOPA) that modulate morphology and transfer ultraviolet (UV) photons into utilizable photoexcitations through an energy-transfer process in the active layer, thereby simultaneously enhancing device performance and suppressing photodegradation. In PM6: Y6 and PM6: L8-BO systems, the power conversion efficiencies (PCE) reach 16.1% and 17.5%, respectively, accompanied by markedly prolonged lifetimes und

A Hydrogen Bond-Mediated Förster Resonance Energy Transfer (FRET) Glue Strategy for Long-Lived Room-Temperature Phosphorescence From Silver Clusters.

Luminescent coinage metal clusters (CMCs) hold great promise for bioimaging and optical applications, but long-lived room-temperature phosphorescence (RTP) with high quantum yield remains challenging. Herein, a Förster resonance energy transfer (FRET) glue strategy using Ag+ to bridge donor-acceptor (D/A) pairs is adopted to synthesize a series of clusters in CH3CN (Ag4), CH3OH [Ag4(CH3OH)2], C2H5OH [Ag4(C2H5OH)], and i-C3H7OH [Ag2(i-C3H7OH)2] realizing near-unity FRET efficiency. Among them, Ag4(CH3OH)2 delivers superior thermally activated delayed phosphorescence with a high photoluminescenc

Rapid and Dense Immobilization of Nucleic Acids on Lanthanide-Doped Nanoparticles for Enhanced-Sensitivity Förster Resonance Energy Transfer-Based Diagnostics.

Rapid and dense DNA functionalization of upconversion nanoparticles (UCNPs) remains a critical bottleneck, as conventional covalent and electrostatic methods suffer from low labeling efficiency and time-consuming processing. We report a microwave-assisted rapid dehydration method for efficient DNA immobilization on UCNPs within 3 min, drastically outperforming conventional strategies. This rapid dehydration method demonstrates broad applicability across diverse UCNP compositions, morphologies, and different DNA chain lengths. Molecular dynamics simulations confirm the underlying mechanism: str

Association Between Linear Energy Transfer Distribution and Local Recurrence in Recurrent Nasopharyngeal Carcinoma Reirradiated With Intensity-Modulated Carbon Ion Therapy.

PURPOSE: Local recurrence (LR) after reirradiation with intensity-modulated carbon ion therapy (IMCT) in recurrent nasopharyngeal carcinoma (rNPC) may relate to insufficient dose-averaged linear energy transfer (LETd) within the tumor. This study evaluated the prognostic value of LETd metrics for local control (LC) in rNPC. METHODS AND MATERIALS: This retrospective study enrolled 76 patients with rNPC who underwent salvage IMCT reirradiation at Shanghai Proton and Heavy Ion Center (SPHIC) from 2015 to 2019. Clinically, dosimetric and LETd variables were evaluated for association with LC using

Mapping the Energy Transfer Network in the Diatom Cyclotella meneghiniana by Picosecond Time-Resolved Fluorescence Spectroscopy down to Cryogenic Temperatures.

Diatoms are unicellular photosynthetic microalgae that contribute nearly a quarter of the global primary production. Knowledge of the mechanisms, networks, and time scales of the excitation transfer is crucial for a deeper understanding of their photosynthetic machinery. Unfortunately, the short excited-state lifetime of photosystem I at room temperature and research focus on isolated structures have limited the development of a complete excitation transfer scheme in intact cells. Here, we address this gap through picosecond time-resolved fluorescence measurements on intact Cyclotella meneghin

Hot-Exciton-Involved Dual-Channel Stepwise Energy Transfer Enabling Efficient and Stable Blue OLEDs with Narrow Emission and High Luminance.

Marching toward next-generation ultrahigh-definition and high-resolution displays, the development of high-performance blue organic light-emitting diodes (OLEDs) with narrow emission and high luminance is essential and requires conceptual advancements in both molecular and device design. Herein, a blue organic emitter is reported that exhibits hot-exciton and aggregation-induced emission characteristics, and use it as a sensitizer in the proposed triplet-triplet annihilation (TTA)-assisted hot-exciton-sensitized fluorescence (HSF) device, abbreviated THSF. In Vivo Monitoring of Hydrogen Polysulfide via a NIR-Excitable Reversible Fluorescent Probe Based on Upconversion Luminescence Resonance Energy Transfer.

Hydrogen polysulfide (H2Sn), derived from hydrogen sulfide (H2S), has attracted increasing attention, which is suggested to be the actual signal molecule instead of H2S in physiological and pathological processes. Reversible detection of H2Sn through a NIR-excitable fluorescence probe is an effective means to understand its functions but is quite challenging. Herein, we reported a NIR-excitable ratiometric nanoprobe for the reversible detection of H2Sn based on luminescence resonance energy transfer principle with upconversion nanoparticles as the energy donor and an organic molecule, SiR1, as

Self-Catalyzed Surface Reaction-Induced Fluorescence Resonance Energy Transfer on Cysteine-Stabilized MnO2 Quantum Dots for Selective Detection of Dopamine.

A simple one-step ultrasonic method was developed for the synthesis of luminescent MnO2 quantum dots (MnO2 QDs) in the presence of cysteine, in which cysteine acted as the exfoliating agent and stabilization ligand. The cysteine-stabilized MnO2 QDs (Cys-MnO2 QDs) possess a fluorescence quantum yield of 4.7%, and the fluorescence intensity of Cys-MnO2 QDs is sensitive to dopamine (DA). The mechanism by which the Cys-MnO2 QDs catalyzed the self-polymerization of DA to form polydopamine nanoparticles (PDA NPs) and caused the fluorescence resonance energy transfer (FRET) between MnO2 QDs and PDA N
