Implementing the cities readiness initiative: lessons learned from Boston.

The federally funded Cities Readiness Initiative (CRI) requires seamless federal, state, and local public health coordination to provide antibiotics to an entire city population within 48 hours of an aerosolized release of anthrax. We document practical lessons learned from the development and implementation of the Boston CRI plan. Key themes center on heightened emphasis on security, a new mass protection model of dispensing, neighborhood-centric clinic site selection, online training of Medical Reserve Corps volunteers, and the testing of operations through drills and exercises. Sharing such

Improving data participation for the development of artificial intelligence in dermatology.

Artificial intelligence (AI) has the potential to significantly impact many aspects of dermatology. The visual nature of dermatology lends itself to innovations in this space. The robustness of AI algorithms depends on the quality, quantity, and variety of data on which it is trained and tested. Image collections can suffer from inconsistencies in image quality, underrepresentation of various anatomic sites and skin tones, and lack of benign counterparts leading to underperformance of algorithms in contexts other than one in which it is developed. Access to care, trust, rights, control, and tr

Bayesian Federated Inference for regression models based on non-shared medical center data.

To estimate accurately the parameters of a regression model, the sample size must be large enough relative to the number of possible predictors for the model. In practice, sufficient data is often lacking, which can lead to overfitting of the model and, as a consequence, unreliable predictions of the outcome of new patients. Pooling data from different data sets collected in different (medical) centers would alleviate this problem, but is often not feasible due to privacy regulation or logistic problems. An alternative route would be to analyze the local data in the centers separately and comb

Graph neural networks for multi-scale gene-drug interaction modeling in cancer: applications, challenges, and therapeutic opportunities for breast cancer drug resistance.

Breast cancer drug resistance remains a major clinical challenge driven by complex genetic, signaling, and microenvironmental interactions. Conventional machine learning and deep learning represent genes, drugs, and patients as independent feature vectors, limiting their ability to capture biological relationships governing therapeutic response. Graph neural networks have emerged as a powerful paradigm by modelling biological systems as interconnected networks rather than isolated entities. This review synthesizes recent advances in graph neural network based multi-scale modelling of gene-drug

Artificial intelligence and personalised medicine in liver cancer.

Primary liver cancers, including hepatocellular carcinoma and cholangiocarcinoma, represent a growing global health burden marked by rising incidence and high mortality. Clinical management requires integration of tumour stage, liver function, and patient-related factors to guide treatment decisions, yet current frameworks capture only a fraction of underlying disease complexity. In this setting, artificial intelligence (AI) is emerging as a key enabler of precision medicine in liver oncology. AI encompasses machine learning and deep learning approaches capable of identifying complex, non-line

Artificial intelligence-enabled multi-omics biomarkers for immune checkpoint blockade: mechanisms, predictive modeling, and clinical translation.

Immune checkpoint inhibitors (ICIs) have transformed oncology, yet durable benefit remains confined to a minority of patients, revealing the limitations of single biomarkers such as PD-L1 expression, tumor mutational burden, and microsatellite instability. Multi-omics profiling, spanning genomics, transcriptomics, epigenomics, proteomics, metabolomics, microbiomics, and imaging-derived radiomics/pathomics, enables a systems-level interrogation of tumor-immune interactions. It captures lineage plasticity, antigen-presentation defects, metabolic and epigenetic suppression, stromal remodeling, an

Biological Foundation Models for Complex Disease Research and Clinical Translation.

Complex diseases, including cancer, rare genetic disorders, neurodevelopmental and psychiatric conditions, and neurodegenerative diseases, arise from interactions among genetic variation, gene regulation, and cellular states that are difficult to capture using a single data type or biological scale. Biological foundation models address this challenge by treating nucleotides and genes as tokens and learning representations that can be transferred to downstream biomedical and clinical tasks. In this review, we examine two major model classes, genomic sequence foundation models and cell foundatio

Artificial intelligence and digital solutions for myopia.

Myopia as an uncorrected visual impairment is recognized as a global public health issue with an increasing burden on health-care systems. Moreover, high myopia increases one's risk of developing pathologic myopia, which can lead to irreversible visual impairment. Thus, increased resources are needed for the early identification of complications, timely intervention to prevent myopia progression, and treatment of complications. Emerging artificial intelligence (AI) and digital technologies may have the potential to tackle these unmet needs through automated detection for screening and risk str

Evolutionary multi-agent reinforcement learning for crisis-aware demographic policy optimization.

Demographic systems face unprecedented challenges from simultaneous crises. Conventional statistical demography techniques and agent-based models often struggle to capture nonlinear inter-regional interactions during periods of severe socio-economic disruption. To address this, we propose MADDPG-EVO-DGM, a hybrid algorithm that integrates multi-agent deep reinforcement learning with evolutionary optimisation and meta-learning principles to model regional demographic processes under multiple crisis scenarios. Each region is treated as an autonomous agent learning to steer demographic policy lev
