[Changes in GABAA/benzodiazepine receptor complex function in the pentobarbital-dependent rat].

Changes in GABAA receptor function were studied in pentobarbital (PB)-dependent rats. Physical dependence on PB in male Lewis rats was induced by the drug-admixed food method. The 36Cl- influx into cerebral cortical synaptoneurosomes induced by 10 microM GABA in the PB-dependent rat was significantly decreased compared with the control. The enhancement of GABA-dependent 36Cl- influx by the addition of PB, ethanol (EtOH) and flunitrazepam (FZ) was not recognized in the PB-dependent group. The addition of picrotoxin and bicuculline had no effect on GABA-dependent 36Cl- influx in PB-dependent rat

Benzodiazepines and the mammalian retina. I. Autoradiographic localisation of receptor sites and the lack of effect on the electroretinogram.

The majority of specific benzodiazepine binding sites were found to be restricted to the innerplexiform layer of the rat retina, although there were minor amounts of binding in the inner nuclear and ganglion cell layers. Relatively high levels of non-specific benzodiazepine binding sites were, on the other hand, found in the pigment epithelium and ciliary body, as well as in the corneal epithelium. The specific binding was enhanced by the GABA-A agonist, muscimol. In both rats and cats, neither acute nor chronic administration of benzodiazepines or their antagonists altered the retinal functio

Pneumonia Risk Associated with the Use of Individual Benzodiazepines and Benzodiazepine Related Drugs among the Elderly with Parkinson's Disease.

Most patients with Parkinson's disease (PD) gradually develop oropharyngeal dysphagia which is often associated with pneumonia risk. The possible association of benzodiazepine (BZD) and benzodiazepine related drugs (BZRD) use with pneumonia risk has received increasing attention but remains controversial. We investigated pneumonia risk associated with the use of BZDs and BZRDs in older adult patients with PD. This case-control study analyzed data of 551,975 older adult patients with PD between 2001 and 2018 in Taiwan. To minimize potential confounding, we used 1:4 propensity score matching to

Conversion factors for assessment of driving impairment after exposure to multiple benzodiazepines/z-hypnotics or opioids.

AIMS: Norway has introduced legal concentration limits in blood for 28 non-alcohol drugs in driving under the influence cases. As of 2016 this legislation also regulates the assessment of combined effects of multiple benzodiazepines and opioids. We herein describe the employed methodology for the equivalence tables for concentrations of benzodiazepines/z-hypnotics and opioids implemented in the Norwegian Road Traffic Act. METHODS: Legislative limits corresponding to impairment at blood alcohol concentrations (BAC) of 0.02%, 0.05% and 0.12% were established for 15 different benzodiazepines and

Endoscopic treatment for esophageal varices complicated by Isaacs' syndrome involving difficulty with conventional sedation.

A 54-year-old male consulted a local doctor with a chief complaint of systemic convulsions and muscle stiffness and was diagnosed with Isaacs' syndrome based on positive findings for antibodies against voltage-gated potassium channels in 2009. He subsequently experienced repeated hematemesis in 2013, at which time he was taken to our hospital by ambulance. Emergent endoscopy revealed esophageal varices with spurting bleeding. The bleeding was stopped with urgent endoscopic variceal ligation. Three days later, the patient developed sudden dyspnea with stridor during inspiration under sedation w

Modafinil prevents the MPTP-induced increase in GABAA receptor binding in the internal globus pallidus of MPTP-treated common marmosets.

The psychostimulant drug modafinil induces a reversal of motor deficits in MPTP treated primates and prevents MPTP toxicity to substantia nigra but its mechanism of action is not clear. In common marmosets acutely treated with MPTP in the presence or absence of modafinil, we have studied changes in GABA(A) receptor binding in the basal ganglia. MPTP treatment had no effect on [(3)H]-flunitrazepam (FNZ) binding density in the striatum or external globus pallidus (GPe) but increased [(3)H]-FNZ binding density in the internal globus pallidus (GPi). Administration of modafinil (10-100 mg/kg) with

Barbiturate tolerance: effects on GABA-operated chloride channel function.

Male ICR mice were fed powdered laboratory chow containing phenobarbital for 7 days to induce tolerance. Mice were sacrificed and brains assayed for changes in GABA-mediated chloride flux into brain membrane vesicles (microsacs). Concentration-dependent stimulation of chloride flux by GABA alone was not affected by the development of tolerance to phenobarbital. Phenobarbital potentiation of GABA-mediated chloride flux was significantly attenuated in the membranes prepared from phenobarbital-tolerant mice compared with those from pair-fed control mice. Similarly, stimulation of GABA-mediated fl

Simultaneous screening and quantitation of alpidem, zolpidem, buspirone and benzodiazepines by dual-channel gas chromatography using electron-capture and nitrogen-phosphorus detection after solid-phase extraction.

A rapid twin-column gas chromatographic (GC) method for simultaneous screening and determination of commonly prescribed benzodiazepines and other new anxiolytics from plasma is described. Identical fused-silica Ultra 2 (5% phenyl methyl silicone) columns were connected to nitrogen-phosphorus and electron-capture detectors. The drugs were isolated from 1 ml of plasma by solid-phase extraction (SPE) onto a C8 reversed-phase sorbent and recovered with 0.5% acetic acid in methanol. The eluate was reconstituted with isopropanol which was found suitable for on-column injection. Prazepam was used as

Peripheral-type benzodiazepine receptors are highly concentrated in mitochondrial membranes of rat testicular interstitial cells.

The binding of 3H-RO 5-4864 to the peripheral-type benzodiazepine receptors (PBZDR) in rat testicular interstitial cells (TIC) was characterized. The binding was saturable, reversible and showed a single high-affinity (Kd = 5.02 +/- 0.86 nM) class of binding sites. The maximal binding capacity (Bmax) in crude mitochondrial fractions (77.6 +/- 9.1 pmol/mg protein) represents the highest density of PBZDR in tissues thus far studied. In comparison with the crude mitochondrial fraction the subcellular fractionation of TIC revealed a 2-fold enrichment of 3H-RO 5-4864 binding sites to the purified m

Responses to GABA by isolated insect neuronal somata: pharmacology and modulation by a benzodiazepine and a barbiturate.

Mechanically dissociated neuronal somata from the thoracic ganglia of Locusta migratoria and Schistocerca gregaria were viable in vitro for hours and were current- and voltage-clamped to record the responses evoked by brief pressure applications of gamma-aminobutyric acid (GABA) in the presence of various modulators. The application of GABA and muscimol, but not baclofen, produced a hyperpolarization and concurrent increase in the membrane conductance. The current underlying this response reversed at -65 mV, was evoked in all cells tested and showed outward rectification. In 6 of 74 Locusta ne

[Changes in GABAA/benzodiazepine receptor complex function in the pentobarbital-dependent rat. II: Strain differences between Lewis and Wistar-Kyoto rats].

We studied the differences in alterations of GABAAergic receptor function between pentobarbital (PB)-dependent female Lewis (LEW) and Wistar-Kyoto (WKY) rats. The 36Cl- influx induced by 10 microM GABA in the PB-dependent WKY was significantly lower than that in the control, while there was no significant 36Cl- influx change in both PB-dependent and control LEW. The additions of PB, flunitrazepam (FZ) and ethanol (EtOH) enhanced the GABA-dependent 36Cl- influx in control rats of both strains. However, the enhancements of 36Cl- influx by PB, FZ, EtOH were not recognized in PB-dependent WKY. On

Benzodiazepines enhance the muscimol-dependent activation of phospholipase A2 in glioma C6 cells.

Glioma C6 cells were incubated with [14C]arachidonate to label membrane phospholipids. Muscimol, a selective gamma-aminobutyric acid A receptor agonist, but not (-)-baclofen, a selective gamma-aminobutyric acid B receptor agonist, stimulates [14C]arachidonate release from C6 cells as a result of hydrolysis of a small pool of phosphatidylcholine and phosphatidylethanolamine by phospholipase A2. This release is facilitated by diazepam and a number of other benzodiazepines such as flunitrazepam, medazepam and midazolam (but very little by clonazepam), although these benzodiazepines per se are ina

Binding and neuropharmacological profile of zaleplon, a novel nonbenzodiazepine sedative/hypnotic.

The binding properties of CL284,846 (zaleplon), a novel nonbenzodiazepine sedative/hypnotic, at benzodiazepine receptor subtypes were evaluated. Zaleplon was 14.3 times more potent at inhibiting [3H]flunitrazepam binding to membrane preparations of the cerebellum than to membrane preparations of the spinal cord. The gamma-aminobutyric acid (GABA) ratio of zaleplon was 2.07. Zaleplon produced significant increases in muscimol binding similar to those of diazepam, and it was antagonized by flumazenil. Furthermore, zaleplon showed little affinity for other receptors. Spectral analysis of the elec

Discrepancy of benzodiazepine receptor occupancy between 3H-flumazenil and 125I-iomazenil in intact mouse brain.

The in vivo benzodiazepine (BZ) receptor occupancy in mouse brain was measured by employing 3H-flumazenil (FMZ) in comparison with 125I-iomazenil (IMZ), in order to obtain fundamental data for PET and SPECT studies, respectively. Mice were pretreated with various doses of flunitrazepam (FNP) 40 min prior to the tracer injection. At 20 min after the tracer injection, mice were killed by decapitation and receptor occupancy in cerebral cortex, hippocampus, cerebellum and pons-medulla were determined by the modified method reported by Goeders and Kuhar (1985). A multifactorial critical appraisal of substances found in drug facilitated sexual assault cases.

Drug-facilitated sexual assault (DFSA) is a sexual act in which the victim is unable to give or rescind consent due to intoxication with alcohol and/or drugs that have been self-administered (opportunistic DFSA) or covertly administered by the perpetrator (predatory DFSA). The drugs that are most commonly associated with DFSA are flunitrazepam and gamma-hydroxybutyric acid (GHB). They cause sedation and amnesia, are readily dissolved in beverages and are rapidly eliminated from the system. However, drugs such as amphetamine and cocaine, which are central nervous system (CNS) stimulants, have a

Expression of functional receptors by the human gamma-aminobutyric acid A gamma 2 subunit.

gamma-Aminobutyric acid A (GABA(A)) receptors are heteromeric membrane proteins formed mainly by various combinations of alpha, beta, and gamma subunits; and it is commonly thought that the gamma 2 subunit alone does not form functional receptors. In contrast, we found that cDNA encoding the gamma 2L subunit of the human GABA(A) receptor, injected alone into Xenopus oocytes, expressed functional GABA receptors whose properties were investigated by using the two-microelectrode voltage-clamp technique. GABA elicited desensitizing membrane currents that recovered after a few minutes' wash. Repeti
