A caspase-like decoy molecule enhances the activity of a paralogous caspase in the yellow fever mosquito, Aedes aegypti.

Caspases are cysteine proteases that play critical roles in apoptosis and other key cellular processes. A mechanism of caspase regulation that has been described in mammals and nematodes involves caspase-like decoy molecules, enzymatically inactive caspase homologs that have arisen by gene duplication and acquired the ability to regulate other caspases. Caspase-like decoy molecules are not found in Drosophila melanogaster, raising the question of whether this type of caspase regulation exists in insects. Phylogenomic analysis of caspase genes from twelve Drosophila and three mosquito species r

A salivary elicitor in Laedelphax stiatellus positively regulates plant defenses on optimal and suboptimal hosts.

While herbivorous insects often secrete effector proteins to suppress plant immunity, alternative adaptation strategies remain less explored. Here, we report a counterintuitive approach in the small brown planthopper, Laodelphax striatellus. SBPH downregulated a salivary gland-specific elicitor gene, LsG40, when transferred from the optimal host rice to the suboptimal host corn. When expressed in plants, including the model organism tobacco and the two host plants corn and rice, LsG40 acts as a defense elicitor triggering calcium-dependent reactive oxygen species burst, cell death, callose dep

Cellular remodeling of ovarian follicular epithelial cells transmits an obligate nutritional endosymbiont in a scale insect.

Many insects show complex associations with vertically transmitted endosymbionts. Here, we describe unique cellular remodeling of the oocyte's follicular epithelial cells into endosymbiont-bearing tubular structures in the ensign scale insect Insignorthezia insignis (Hemiptera: Ortheziidae). Vitellogenic oocytes develop a bulge-like distension at the posterior pole. Here, follicular epithelial cells undergo extensive cellular reorganization, remodeling their actin cytoskeleton and plasma membrane to produce cellular protrusions. These tubular protrusions, which are densely packed with the endo

Identification of candidate chemosensory receptors based on transcriptome analysis in the monophagous insect Protegira songi (Lepidoptera, Noctuidae).

BACKGROUND: Insects perceive chemical cues from conspecifics and hosts through their sensitive chemosensory system. The chemosensory receptors, which are mainly represented by odorant receptors (ORs), ionotropic receptors (IRs), and gustatory receptors (GRs), are highly enriched in the chemosensory tissues and are crucial for perceiving and transducing chemical signals. RESULTS: In this study, we performed a transcriptome analysis of different tissues of Protegira songi, a notorious defoliator that specifically infests the medicinal and rubber plant Eucommia ulmoides and identified 77 candidat

Cyanogenic millipede genome illuminates convergent evolution of cyanogenesis-related enzymes.

Hydrogen cyanide (HCN) is a highly toxic biogenic compound. Unlike most natural defensive chemicals, which are typically lineage-specific, the biosynthesis and liberation of HCN, called "cyanogenesis", occur sporadically among arthropod and plant lineages. This suggests that cyanogenesis has evolved independently numerous times in the animal and plant kingdoms. Although cyanogenesis was identified in millipedes 140 years ago, the cyanogenesis-related enzymes in these arthropods have not yet been fully identified. Here, we report a complete set of cyanogenesis-related enzymes in the millipede C

De novo sequencing and transcriptome analysis of female venom glands of ectoparasitoid Bracon hebetor (Say.) (Hymenoptera: Braconidae).

Venom is a key-factor in the regulation of host physiology by parasitic Hymenoptera and a potentially rich source of novel bioactive substances for biotechnological applications. The limited study of venom from the ectoparasitoid Bracon hebetor, a tiny wasp that attacks larval pest insects of field and stored products and is thus a potential insect control agent, has not described the full complement and composition of these biomolecules. To have a comprehensive picture of genes expressed in the venom glands of B. hebetor, a venom gland transcriptome was assembled by using next generation sequ

Viral paratransgenesis in the malaria vector Anopheles gambiae.

Paratransgenesis, the genetic manipulation of insect symbiotic microorganisms, is being considered as a potential method to control vector-borne diseases such as malaria. The feasibility of paratransgenic malaria control has been hampered by the lack of candidate symbiotic microorganisms for the major vector Anopheles gambiae. In other systems, densonucleosis viruses (DNVs) are attractive agents for viral paratransgenesis because they infect important vector insects, can be genetically manipulated and are transmitted to subsequent generations. However, An. gambiae has been shown to be refracto

Transcriptome profiling to decipher molecular mechanisms of diapause in Chilo partellus (Swinhoe) (Lepidoptera: Crambidae).

Diapause is a critical adaptive strategy in insects to survive unfavourable environmental conditions. The present study investigated molecular mechanisms underlying diapause in Chilo partellus, using high-throughput RNA sequencing and de novo transcriptome assembly. Six transcriptome libraries from diapause and non-diapause larvae yielded over 50 million reads per sample, with >98% mapping efficiency. The Evidential Gene-refined assembly produced 67,116 high-quality transcripts, and BUSCO analysis confirmed its completeness. DEG analysis identified 1204 DEGs, including 660 upregulated and 544

Identification of candidate host-manipulating effector genes in Aphis gossypii (Hemiptera: Aphididae) using a combination of transcriptome, genome, and differential gene expression data.

The cotton aphid, Aphis gossypii Glover, is an important plant disease vector and a highly polyphagous agricultural pest that feeds on a broad range of host plants. During feeding, its salivary glands serve as a route for the transmission of circulative plant viruses and produce a range of secretory proteins, called effectors, to modulate host cellular processes. To understand the molecular mechanisms underlying aphid-plant interactions, we developed a bioinformatics pipeline that incorporated the salivary gland transcriptome, genome, and head vs abdomen differential gene expression data to pr

Aquaporin expression in the alimentary canal of the honey bee Apis mellifera L. (Hymenoptera: Apidae) and functional characterization of Am_Eglp 1.

Aquaporins (AQP) are a family of plasma membrane proteins responsible for water transport through cell membranes. They are differentially expressed in different parts of the alimentary canal of insects where they regulate water transport. These proteins have been studied in detail in some insects, but few data are available for aquaporins of the honey bee, Apis mellifera. We used quantitative PCR to study the expression of six putative aquaporin genes in forager honey bees. We found differential expression of all putative AQP genes in crop, midgut, ileum, rectum and Malpighian tubules. We foun

RNAi of Mannosidase-Ia in the Colorado potato beetle and changes in the midgut and peritrophic membrane.

BACKGROUND: In addition to its role in the digestive system, the peritrophic membrane (PM) provides a physical barrier protecting the intestine from abrasion and against pathogens. Because of its sensitivity to RNA interference (RNAi), the notorious pest insect, the Colorado potato beetle (CPB, Leptinotarsa decemlineata), has become a model insect for functional studies. Previously, RNAi-mediated silencing of Mannosidase-Ia (ManIa), a key enzyme in the transition from high-mannose glycan moieties to paucimannose N-glycans, was shown to disrupt the transition from larva to pupa and the metamorp

Joint Transcriptional Control of Virulence and Resistance to Antibiotic and Environmental Stress in Acinetobacter baumannii.

UNLABELLED: The increasing emergence of antibiotic-resistant bacterial pathogens represents a serious risk to human health and the entire health care system. Many currently circulating strains of Acinetobacter baumannii exhibit resistance to multiple antibiotics. A key limitation in combating A. baumannii is that our understanding of the molecular mechanisms underlying the pathogenesis of A. baumannii is lacking. To identify potential virulence determinants of a contemporary multidrug-resistant isolate of A. baumannii, we used transposon insertion sequencing (TnSeq) of strain AB5075. A collect

Autophagy genes AMBRA1 and ATG8 play key roles in midgut remodeling of the yellow fever mosquito, Aedes aegypti.

The function of two autophagy genes, an activating molecule BECN1 regulated autophagy (AMBRA1) and autophagy-related gene 8 (ATG8) in the midgut remodeling of Aedes aegypti was investigated. Real-time quantitative polymerase chain reaction (RT-qPCR) analysis of RNA samples collected from the last instar larvae and pupae showed that these two genes are predominantly expressed during the last 12 h and first 24 h of the last larval and pupal stages, respectively. Stable ecdysteroid analog induced and juvenile hormone (JH) analog suppressed these genes. RNA interference (RNAi) studies showed that

Molecular characterization and elucidation of the function of Hap38 MAPK in the response of Helicoverpa armigera (Hübner) to UV-A stress.

The cotton bollworm Helicoverpa armigera (Hübner) (Lepidoptera: Noctuidae), an important pest of cotton, is detrimental to cotton production. Light from UV-A ultraviolet lamps is regarded as a form of environmental stress for insects. In order to investigate the response of H. armigera exposed to UV-A, we explored Hap38 MAPK expression and functions. We hope that the findings of this study will lay the foundation for future investigations into the insect's phototaxis mechanism. A p38 MAPK was cloned and named Hap38 MAPK. A phylogenetic tree showed that Hap38 MAPK was highly conserved. The gene

Amino acid synthesis loss in parasitoid wasps and other hymenopterans.

Источник пометил публикацию как отозванную

Insects utilize diverse food resources which can affect the evolution of their genomic repertoire, including leading to gene losses in different nutrient pathways. Here, we investigate gene loss in amino acid synthesis pathways, with special attention to hymenopterans and parasitoid wasps. Using comparative genomics, we find that synthesis capability for tryptophan, phenylalanine, tyrosine, and histidine was lost in holometabolous insects prior to hymenopteran divergence, while valine, leucine, and isoleucine were lost in the common ancestor of Hymenoptera. Subsequently, multiple loss events o

The complete mitochondrial genome of the scuttle fly, Megaselia scalaris (Diptera: Phoridae).

More than 1400 scuttle flies species in worldwide comprise the Megaselia genus, the largest genus in the family Phoridae. The complete mitochondrial genome of Megaselia scalaris, a medically important entomology was sequenced for the first time. The 15,599 bp circular genome contains the 37 genes found in a typical Metazoan genome: 13 protein-coding genes, 22 transfer RNA genes and 2 ribosomal RNA genes. The mitochondrial genome also contains one non-coding A + T-rich region. The arrangement of the genes was identical with other insect. Each of the base composition on heavy strand was as follo

Gb3 encodes a unique kinase fusion protein conferring greenbug resistance in wheat.

Understanding plant resistance to insects has lagged owing to a scarcity of cloned resistance genes. Here we report cloning of the greenbug resistance gene Gb3 in wheat (Triticum aestivum L.), initially introgressed into cultivars such as 'TAM112' from Aegilops tauschii. Gb3 encodes an intracellular kinase-pseudokinase protein, featuring a unique MAP3K-like domain. This gene probably originated through fusion, duplication and indel events in the Triticeae tribe, retaining its functionality in several Aegilops species. Transgenic Gb3 expression in the susceptible Fielder wheat enhanced resistan

Identification and characterization of two arginine kinases from the parasitic insect Ctenocephalides felis.

Arginine kinase (ATP:l-arginine omega-N-phosphotransferase, EC2.7.3.3.; AK) is an enzyme crucial for the energy metabolism of insects and other invertebrates, that has known allergenic potential in humans and that has been proposed as a pesticidal drug target. Here we report the identification, cDNA cloning, genomic gene structure and functional expression of AK genes from Ctenocephalides (C.) felis (cat flea). In contrast to other insect species investigated so far, C. felis possesses two AK genes, cfak1 and cfak2, encoding the functional enzymes CfAK1 and CfAK2 that can be distinguished by t

The sandfly Lutzomyia longipalpis LL5 embryonic cell line has active Toll and Imd pathways and shows immune responses to bacteria, yeast and Leishmania.

BACKGROUND: Lutzomyia longipalpis is the main vector of visceral leishmaniasis in Latin America. Sandfly immune responses are poorly understood. In previous work we showed that these vector insects respond to bacterial infections by modulating a defensin gene expression and activate the Imd pathway in response to Leishmania infection. Aspects of innate immune pathways in insects (including mosquito vectors of human diseases) have been revealed by studying insect cell lines, and we have previously demonstrated antiviral responses in the L. longipalpis embryonic cell line LL5. Gene construction, expression and functional testing of an inotropic peptide from the venom of the black scorpion Hottentotta judaicus.

Anti-insect depressant toxins represent a subfamily of scorpion venom-derived β-toxins that are polypeptides composed of 61-65 amino acid residues stabilized by four disulfide bridges. These toxins affect the activation of voltage-sensitive sodium channels (NaScTx) and exhibit the preferential ability to induce flaccid paralysis in insect larvae. Here we demonstrate the recombinant expression of the novel cardiac inotropic peptide (Bj-IP) that was classified as an anti-insect depressant βNaScTx isolated from the venom of Hottentotta judaicus. By using "splicing by overlap extension" (SOE)-PCR,
