The proximodistal gradient of enterokinase activity was studied in mucosal homogenates of the duodenum, jejunum, ileum, caecum (or megacaecum) and sigmoid flexure of various mammals. The study was carried out in groups of 5 monkeys, guinea pigs, dogs and rats and also in gnotobiological rats--conventional (n = 5), germ-free (n = 10) and monocontaminated (n = 5) with Escherichia coli 0 86. Enterokinase activity was likewise determined in duodenal mucosal homogenates prepared from material resected at operation from adult humans (n = 5) and from a group of monkeys (n = 5), with and without the a

Five gnotobiotic lambs were fed on sterile diets until they were killed at 13 to 21 weeks of age. They were dosed orally with different combinations of 11 species of rumen bacteria. The biochemical reactions of each of the bacteria inoculated had been determined in pure culture in vitro, and they were chosen to perform the main reactions known to be associated with digestion in the normal mature rumen. Two of the bacteria could not be reisolated, but the remainder had established readily in the rumen, forming stable, mixed, defined populations. The total numbers of bacteria in the rumen, and t

Early cellular and vascular changes in response to invasion of lamina propria by Entamoeba histolytica were studied sequentially, at the ultrastructural level, in germfree guinea pigs inoculated intracecally with amebae and enteric flora derived from patients with acute amebic colitis. Approximately one week post-inoculation the animals developed acute colitis with mucosal invasion by trophic amebae. Although epithelial cells at the sites of amebic invasion showed progressive cytoplasmic changes and desquamation resulting in microerosions, most mesenchymal elements in the lamina propria appear

Mitochondrial DNA (mtDNA) that codes protein subunits essential for the maintenance of mitochondrial ATP synthesis system acquires mutations at a much higher rate than that in nuclear DNA. Recent study has revealed that somatically acquired mutations such as deletions in mtDNA are caused mainly by oxygen free-radical damage. Cumulative accumulation of these somatic mutations during the life of an individual causes bioenergetic deficit leading to cell death and normal ageing. The base-sequencing of the entire mtDNA from 48 individuals revealed that germ-line point mutations accelerate extensive

Fever is a common adverse reaction to vaccination, contributing to vaccine hesitancy and reduced uptake. To understand variation in fever risk, we longitudinally profiled fecal microbiota, oral temperature, and serological markers in 171 healthy adults receiving Severe acute respiratory syndrome coronavirus 2 mRNA vaccines. Fever risk correlated with low-grade intestinal inflammation, increased abundance of flagellated Lachnospiraceae bacteria, and increased flagellin expression prevaccine. Microbiomes from fever-high donors triggered stronger inflammation in human intestinal organoids and dro

The gut microbiota is increasingly recognized as a regulator of host metabolism and bone physiology. However, how microbial colonization integrates systemic metabolic cues with skeletal remodeling under metabolic stress remains unclear. We used germ-free (GF) and gnotobiotic C57BL/6J mice colonized with the defined 12-member Oligo-Mouse-Microbiota (Oligo-MM12) to dissect microbiota-dependent bone adaptation during high-fat diet (HFD)-induced obesity. Micro-CT analysis revealed that only colonized mice exhibited structural adaptations, namely increased cortical thickness and trabecular area, in

BACKGROUND: Periodontitis is increasingly linked to systemic conditions such as diabetes, yet the underlying mechanisms remain unclear. This study aimed to determine whether gut microbiota mediates the impact of periodontitis on glucose homeostasis.
METHODS: Germ-free (GF) mice were obtained and maintained in sterile isolators (GemPharmatech, China) and colonized with microbiota derived from donor mice with ligature-induced periodontitis (GF-LIG) or healthy controls (GF-CON). Donor faecal samples were collected from donor mice with ligature-induced periodontitis or healthy controls. Fasting b

The gastrointestinal tract harbors a dynamic microbial ecosystem that interfaces with the intestinal epithelium. Among this community, Akkermansia muciniphila, a mucin-degrading microbe, has garnered attention for its impact on gut health. While well studied in the colon, its influence on the small intestine remains underexplored. To examine direct effects of A. muciniphila, we used gnotobiotic mice. Germ-free mice were inoculated with Brain Heart Infusion (BHI) media or 10⁹ viable A. muciniphila in BHI. After 21 days, small intestinal tissue was collected. Fluorescence in situ hybridization c

The fungal species Candida albicans and the bacterium Enterococcus faecalis are members of the human gut microbiome. To explore the range of interactions between these two species, we utilized dual RNA-sequencing to transcriptionally profile both C. albicans and E. faecalis during coculture (compared with monoculture controls) under two conditions: 1) an in vitro setting that mimics certain features of the gut environment and 2) a gnotobiotic mouse gut model. RNA-seq analysis revealed a large number of gene expression changes induced by one species in the presence of the other. More specifical

Crosstalk between the immune system and the microbiome is critical for maintaining immune homeostasis. Here, we examined this communication and the impact of immune-suppressive IL-10 signaling on pulmonary homeostasis. We found that IL-10 sensing by interstitial macrophages (IMs) is required to prevent spontaneous lung inflammation. Loss of IL-10 signaling in IMs initiated an inflammatory cascade through the activation of classical monocytes and CD4+ T cell subsets, leading to chronic lung inflammation with age. Analyses of antibiotic-treated and germ-free mice established that lung inflammati

Disturbances in gut microbiota are prevalent in inflammatory bowel disease (IBD), which includes ulcerative colitis (UC). However, whether these disturbances contribute to development of the disease or are a result of the disease is unclear. In pairs of human twins discordant for IBD, the healthy twin has a higher risk of developing IBD and a gut microbiota that is more similar to that of IBD patients as compared with healthy individuals. Furthermore, appropriate medical treatment may mitigate these disturbances. To study the correlation between microbiota and IBD, we transferred stool samples

Tryptophan is catabolized by gut microorganisms resulting in a wide range of metabolites implicated in both beneficial and adverse host effects. How gut microbial tryptophan metabolism is directed towards indole, associated with chronic kidney disease, or towards protective indolelactic acid (ILA) and indolepropionic acid (IPA) is unclear. Here we used in vitro culturing and animal experiments to assess gut microbial competition for tryptophan and the resulting metabolites in a controlled three-species defined community and in complex undefined human faecal communities. The generation of speci

To study the possible influence of intestinal micro-organisms on the course of strongyloidiasis in mice, a method was developed to obtain axenic infective larvae of Strongyloides venezuelensis. Cultured larvae from conventional mice were treated with sodium hypochlorite 0.25% for 10 min, washed in distilled water and then exposed to various combinations of antibiotics for 30 or 60 min. Success was achieved with a combination of penicillin 180 mg/L and ceftazidime 1 mg/ml. Decontamination of the larvae was determined by aerobic and anaerobic culture and by inoculation into gnotobiotic mice. Via

The enteric pathogenicity of the ovine C. psittaci serotype 1 isolate S26/3 was assessed using a litter of gnotobiotic piglets. In one group, eight piglets were inoculated at 3 days of age; at 10 days, two of these were re-inoculated. In a second group, six animals were mock-inoculated at 3 days of age as negative controls; subsequently, at 10 days, three of these piglets were inoculated with C. psittaci. The animals were observed for clinical signs, killed and necropsied sequentially between 4 and 17 days of age. At necropsy, specimens were collected for histopathology, immunohistochemistry a

Barrier-raised transforming growth factor beta 1 (TGF beta 1)-deficient mice consistently die before 35 days of age of a severe multiorgan inflammatory disease that can affect the skeletal muscle, heart, liver, pancreas, salivary gland, lung, oesophagus and stomach. The underlying cause of this disease is not known. To determine whether abnormal responsiveness of the immune system to the presence of enteric flora plays a causative role, a colony of TGF beta 1-deficient and wild-type mice were raised in a sterile environment. Seven germ-free TGF beta 1-deficient and 5 germ-free TGF beta 1 wild-

A formalin fixative and a formalin-free fixative were used to study mast cells in the small intestine of conventional, gnotobiotic and parasitized pigs. Many more mast cells were identified after basic lead acetate fixation ('mucosal mast cells', MMC) than after routine formalin fixation ('connective tissue mast cells'). The MMC were preferentially localized in the lamina propria. There were no differences between conventional and gnotobiotic pigs. However, in parasitized animals, the number of mast cells was several times higher, mainly because there were more MMC. The heterogeneity of intest
