Integration of hydrogen peroxide sensing and detoxification by the C. elegans nervous system.

Hydrogen peroxide (H2O2) is the most common reactive chemical threat organisms face. Here, we map the neural circuit that drives chemotactic escape from environmental H2O2 in the nematode C. elegans. Twenty-four neuron classes with sensory endings at the mouth and nose respond to H2O2, with dynamics encoding stimulus intensity and exposure history. Sensing begins when H2O2 oxidizes the peroxidatic and resolving cysteines of the cytosolic peroxiredoxin PRDX-2, which relays this oxidative signal to cysteines on the LITE-1 and GUR-3 ion channels, triggering calcium influx in sensory neurons that

Predictive Modeling of Bacterial Inactivation With Hydrogen Peroxide Over a Cobalt Ferrite Catalyst.

Hydrogen peroxide activated by a cobalt ferrite catalyst generates hydroxyl radicals that effectively inactivate bacteria. A continuous-flow packed-bed reactor containing cobalt ferrite was developed and tested for bacterial disinfection. The inactivation kinetics were highly nonlinear. The classical Weibull model accurately described the initial decline in bacterial counts at low H2O2 concentration (3 mM) but lost accuracy at longer contact times and higher concentrations (6-10 mM). To improve prediction, 10 machine learning algorithms were evaluated. Gradient boosting (GB) and random forest

MOFs-Derived Integrated Hierarchical Carbon Electrode for Hydrogen Peroxide Synthesis in Acidic Media.

Current research on electrosynthesis of hydrogen peroxide mainly focuses on optimizing intrinsic active sites, while overlooking the constraints imposed by mass transport limitations and weak oxygen adsorption capacity during the reaction. Herein, we developed an integrated hierarchically porous carbon electrode composed of MOF-derived porous carbon sheets arrayed on carbon cloth and epitaxially grown carbon nanotube networks, with highly active Co-Nx sites generated in situ. During the synthesis, pure water was initially employed as the solvent to promote the ordered growth of sheet-like MOFs

High-quality hydrogen peroxide production through electrochemical reduction of anthraquinone in ambient environment.

At present, the traditional anthraquinone method is the main method for producing high-quality and high-concentration H2O2, but it involves the use of H2, resulting in high carbon emissions and certain risks. The double-electron oxygen reduction reaction has become a more environmentally friendly alternative method for producing H2O2. However, at high overpotentials, the production and conversion rates of H2O2 are still limited. Here, we report an electrochemical method that can convert H2O and formaldehyde into high-concentration H2O2 and high-value formate salts with high product purity unde

A selective near-infrared fluorescent probe for detecting hydrogen peroxide in Alzheimer's disease.

Alzheimer's disease (AD) is a progressive neurodegenerative disorder with growing incidence among younger populations, highlighting the urgent need for early and accurate detection of AD-related biomarkers. Excessive production of reactive oxygen species (ROS), particularly hydrogen peroxide (H2O2), induces oxidative stress, leading to neuronal damage and death, which implicates H2O2 as a potential biomarker for AD. In this study, leveraging the advantages of fluorescence imaging, we developed a novel near-infrared (NIR) fluorescent probe, LDMD-B, capable of selectively detecting H2O2 through

Impact of bleaching on white spot lesions: hydrogen peroxide permeability and color alteration.

OBJECTIVES: To assess the penetration of hydrogen peroxide (HP) into the pulp chamber and the bleaching efficacy on teeth with white spot lesions (WSLs) compared to sound teeth, when subjected to both in-office and at-home bleaching treatments. MATERIALS AND METHODS: Forty premolars were allocated into four groups (n = 10 each): sound teeth and teeth with artificially induced WSLs, both subjected to at-home bleaching (16% carbamide peroxide) and in-office bleaching (35% hydrogen peroxide). WSLs were induced in half of the specimens using a 14-day pH-cycling protocol alternating between demine

Electrochemical sensing of hydrogen peroxide using SnO2-SA nanoparticles with enhanced catalytic activity.

Accurate quantification of hydrogen peroxide (H2O2) is essential for biomedical analysis and process monitoring, yet metal-oxide sensors often suffer from limited active-site exposure and sluggish charge transport. Here we report a sodium-alginate (SA)-templated route to porous SnO2 nanoparticles (SnO2-SA) that integrates high crystallinity with abundant surface oxygen vacancies and a permeable mesoporous network. When drop-cast on a glassy carbon electrode (GCE), the SnO2-SA enables non-enzymatic H2O2 detection with a wide linear range of 0.02-2.8 mM, high sensitivity of 381.12 μA mM-1 cm-2,

Glycerol metabolism contributes to competition by oral streptococci through production of hydrogen peroxide.

UNLABELLED: As a biological byproduct from both humans and microbes, glycerol's contribution to microbial homeostasis in the oral cavity remains understudied. In this study, we examined glycerol metabolism by Streptococcus sanguinis, a commensal associated with oral health. Genetic mutants of glucose-PTS enzyme II (manL), glycerol metabolism (glp and dha pathways), and transcriptional regulators were characterized with regard to glycerol catabolism, growth, production of hydrogen peroxide (H2O2), transcription, and competition with Streptococcus mutans. Biochemical assays identified the glp pa

Scinderin Promotes Hydrogen Peroxide-induced Lens Epithelial Cell Injury in Age-related Cataract.

BACKGROUND: Scinderin (SCIN) is a calcium-dependent protein implicated in cell growth and apoptosis by regulating actin cleavage and capping. In this study, we investigated the role of SCIN in hydrogen peroxide-induced lens epithelial cell (LEC) injury related to age-related cataract (ARC). METHODS: Anterior lens capsules from ARC patients were collected to examine SCIN expression levels. Immortalized human LEC cell line SRA01/04 and lens capsules freshly isolated from mice were induced by H2O2 to mimic the oxidative stress in ARC. The role of SCIN was investigated by gain-of-function (overex

Evidence for endogenous hydrogen peroxide production by E. coli fatty acyl-CoA dehydrogenase.

Aerobic organisms continuously generate internal superoxide and hydrogen peroxide, which can damage enzymes and impair growth. To avoid this problem cells maintain high levels of superoxide dismutases, catalases, and peroxidases. Surprisingly, we do not know the primary sources of these reactive oxygen species (ROS) in living cells. However, in vitro studies have shown that flavoenzymes can inadvertently transfer electrons to oxygen. Therefore, it seems plausible that substantial ROS may be generated when large metabolic fluxes flow through flavoproteins. Such a situation may arise during the

Hydrogen peroxide is not generated intracellularly in human neural spheroids during ischemia-reperfusion.

Reactive oxygen species (ROS) are considered a primary source of damage during ischemic stroke. However, the precise timing of ROS production (during hypoxia or reperfusion) remains unclear. Cellular 3D spheroids are often proposed as an optimal alternative to both 2D cell cultures and animal models in modeling disease conditions. Here we report live imaging of hydrogen peroxide dynamics during the acute phase of hypoxia and reperfusion in human iPSC-derived neural spheroids, stably expressing fluorescent biosensor HyPer7. Contrary to previous reports, we did not observe a hydrogen peroxide pr

Hydrogen peroxide attenuates rhinovirus-induced anti-viral interferon secretion in sinonasal epithelial cells.

BACKGROUND: Altered innate defense mechanisms, including an imbalance between oxidants and antioxidants release, have been implicated in the pathogenesis of chronic rhinosinusitis (CRS). The aim of this study is to investigate whether oxidative stress may attenuate the secretion of anti-viral interferons in human sinonasal mucosa. METHODS: The levels of H2O2 in nasal secretion were increased in patients with CRS with nasal polyps, compared with that of CRS patients without nasal polyps and control subjects. Normal sinonasal epithelial cells derived from healthy subjects were cultured under an

Hydrogen Peroxide-Triggered Disassembly of Boronic Ester-Cross-Linked Brush-Arm Star Polymers.

The concentrations of reactive oxygen species (ROS), e.g., H2O2, are often elevated in diseased tissue microenvironments. Therefore, the selective detection of ROS could enable new diagnostic methods or tools for chemical biology. Here, we report the synthesis of boronic ester-bis-norbornene core-cross-linked brush-arm star polymers (BASPs) with polyethylene glycol (PEG) or PEG-branch-spirocyclohexyl nitroxide (chex) shells. Size exclusion chromatography (SEC) and dynamic light scattering (DLS) showed that these BASPs have narrowly dispersed molar masses and average hydrodynamic diameters of 2

Endophytic Fungus Phomopsis liquidambaris Enhances Fe Absorption in Peanuts by Reducing Hydrogen Peroxide.

Iron (Fe) deficiency in alkaline calcium soil is a problem that needs to be solved urgently as Fe is an essential and commonly limiting nutrient for plants. Endophytic fungus, Phomopsis liquidambaris (P. liquidambaris), has been reported to promote Fe absorption in peanuts (Arachis hypogaea L.), however, the mechanisms remain unclear. Under prolonged Fe deficiency, an increase in hydrogen peroxide (H2O2) often triggers a series of signaling events and leads to the inhibition of Fe acquisition. The main purpose of this study was to explore whether and how the endophytic fungus P. liquidambaris

Hydrogen peroxide responsive covalent cyclodextrin framework for targeted therapy of inflammatory bowel disease.

The incidence and prevalence of inflammatory bowel disease (IBD) are steadily rising worldwide, where the pathogenesis and progression are intrinsically connected to oxidative stress and irregular high levels of reactive oxygen species. To achieve effective therapy for IBD, using hydrogen peroxide (H2O2)-activatable antioxidant prodrug (BRAP) as a linker to crosslink cyclodextrin metal-organic framework, a novel H2O2 responsive covalent cyclodextrin framework (defined as BCOF) was formulated. BCOF featured excellent ROS-responsive hydrolysis and therapeutic p-hydroxybenzyl alcohol released in

Protective effects of GHRH antagonists against hydrogen peroxide-induced lung endothelial barrier disruption.

PURPOSE: Growth hormone-releasing hormone (GHRH) is a hypothalamic hormone, which regulates growth hormone release from the anterior pituitary gland. GHRH antagonists (GHRHAnt) are anticancer agents, which also exert robust anti-inflammatory activities in malignancies. GHRHAnt exhibit anti-oxidative and anti-inflammatory effects in vascular endothelial cells, indicating their potential use against disorders related to barrier dysfunction (e.g. sepsis). Herein, we aim to investigate the effects of GHRHAnt against lung endothelial hyperpermeability. METHODS: The in vitro effects of GHRHAnt in H

Determination of hydrogen peroxide on N95 masks after sanitization using a colorimetric method.

Hydrogen peroxide is commonly used as a sterilizing agent for medical devices and its use has recently been extended to N95 masks during PPE shortages as a result of the COVID-19 pandemic. The hydrogen peroxide remaining on the masks after sterilization could potentially pose a health hazard to the mask users. In the present study a colorimetric method was optimized for the determination of hydrogen peroxide on N95 masks following chemical sanitizations. The developed analytical method demonstrated an overall recovery of 98% ± 7%. The limit of detection ranged from 0.16 to 0.25 mg/mask, depend

Polythiophene-Doped Resorcinol-Formaldehyde Resin Photocatalysts for Solar-to-Hydrogen Peroxide Energy Conversion.

The generation of hydrogen peroxide (H2O2) from water and dioxygen by sunlight-driven heterogeneous photocatalysis is a promising method for the artificial photosynthesis of a liquid solar fuel. We previously found that resorcinol-formaldehyde (RF) resin powders prepared by high-temperature hydrothermal synthesis act as highly active semiconductor photocatalysts for H2O2 generation. Herein, we report that RF resin powders doped with poly(3-hexylthiophene-2,5-diyl) (RF/P3HT) exhibit enhanced photocatalytic activities. The highly dispersed P3HT within the resin particles created charge transfer

Nanospatial Charge Modulation of Monodispersed Polymeric Microsphere Photocatalysts for Exceptional Hydrogen Peroxide Production.

Photocatalysis offers a sustainable strategy for hydrogen peroxide (H2 O2 ) production, which is an essential oxidant and emerging energy carrier in modern chemical industry. The development of polymer-based photocatalysts to produce H2 O2 has great potential but is limited by lower efficiency due to the limitation of light utilization and the low charge separation efficiency. Herein, a series of monodispersed mesoporous resorcinol-formaldehyde resin spheres (MRFS) are reported with a rational designed spatial charge distribution, exhibiting wide light absorption with a solar-to-chemical conve
