Activity-dependent metabolic vulnerability in the APPPS1 Alzheimer's disease mouse model during isoflurane anesthesia.

General anesthesia is an essential component of modern surgical and diagnostic care. Although it is overall safe in younger and healthy individuals, the situation is markedly different in patients with underlying neurological conditions like Alzheimer´s disease (AD), where perioperative neurological complications are common. However, the underlying mechanisms and tissue-level interaction between AD-related pathology, cerebral energy metabolism, and anesthetic exposure remain incompletely understood. We investigated acute entorhinal cortex slices from wild-type (WT) and AD-like APPPS1 transgeni

Transcranial ultrasound neuromodulation facilitates isoflurane-induced general anesthesia recovery and improves cognition in mice.

Delayed arousal and cognitive dysfunction are common, especially in older patients after general anesthesia (GA). Elevating central nervous system serotonin (5-HT) levels can promote recovery from GA and increase synaptic plasticity to improve cognition. Ultrasound neuromodulation has become a noninvasive physical intervention therapy with high spatial resolution and penetration depth, which can modulate neuronal excitability to treat psychiatric and neurodegenerative diseases. This study aims to use ultrasound to noninvasively modulate the brain 5-HT levels of mice to promote recovery from GA

Exposure to emulsified isoflurane and sevoflurane protects canine primary hepatocytes against hypoxia-induced apoptosis.

OBJECTIVE: Emulsified isoflurane and sevoflurane have immunomodulating and anti-inflammatory effects in vital organs such as the brain, myocardium, and kidneys subjected to ischemia-reperfusion injury. This study aims to investigate the cellular protective effects of both emulsified anesthetics in cultured canine hepatocytes. PROCEDURES: We analyzed the apoptosis and viability responses of cultured primary canine hepatocytes exposed to 1% O2 for 30 versus 120 minutes after being treated with emulsified isoflurane or sevoflurane in 10% lipid, or 10% lipid alone or no-treatment control at 24 ho

Isoflurane Preconditioning Protects the Myocardium Against Ischemia and Reperfusion Injury by Upregulating GRM1 Expression.

BACKGROUND: Reduction in myocardial I/R injury has become the key to the therapy of ischemic cardiovascular disease. Isoflurane (ISO) preconditioning can mimic the major potent protective mechanisms and attenuate ischemia injury. Nevertheless, the mechanisms involved in the cardioprotective effects afforded by isoflurane preconditioning have never been evaluated systematically. METHODS: Mice were randomly divided into an ISO preconditioning group and control group. The size of the infarcted region was measured, and comparisons between ISO preconditioning and control animals were made. The met

Xenon pretreatment may prevent early memory decline after isoflurane anesthesia and surgery in mice.

Postoperative cognitive decline (POCD) is a common complication following surgery, but its aetiology remains unclear. We hypothesized that xenon pretreatment prevents POCD by suppressing the systemic inflammatory response or through an associated protective signaling pathway involving heat shock protein 72 (Hsp72) and PI3-kinase. Twenty-four hours after establishing long-term memory using fear conditioning training, C57BL/6 adult male mice (n = 12/group) received one of the following treatments: 1) no treatment group (control); 2) 1.8% isoflurane anesthesia; 3) 70% xenon anesthesia; 4) 1.8% is

The common inhalational anesthetic isoflurane induces apoptosis via activation of inositol 1,4,5-trisphosphate receptors.

BACKGROUND: Isoflurane induces cell apoptosis by an unknown mechanism. The authors hypothesized that isoflurane activates inositol 1,4,5-trisphosphate (IP3) receptors on the endoplasmic reticulum (ER) membrane, causing excessive calcium release, triggering apoptosis. METHODS: The authors determined isoflurane-induced cytotoxicity by measuring caspase-3 activity, lactate dehydrogenase release, MTS (3-(4,5-dimethylthiazol-2-yl)-5-(3carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium, inner salt) reduction, and imaging analysis of cell damage markers (annexin V and propidium iodide staining)

Molecular understanding of Abeta peptide interaction with isoflurane, propofol, and thiopental: NMR spectroscopic study.

Источник пометил публикацию как отозванную

Abeta peptide is the major component of senile plaques (SP), which accumulate in the brain of a patient with Alzheimer's disease (AD). A recent report indicated that isoflurane enhanced Abeta oligomerization (micro-aggregation) and subsequent cytotoxicity of the Abeta peptide. A separate study showed that a clinically relevant concentration of isoflurane induces apoptosis and increases Abeta production in a human neuroglioma cell line. In vitro studies have indicated that halothane interacts specifically with Abeta peptide to induce oligomerization and that Abeta42 oligomerizes faster than Abe

Isoflurane preconditioning induces neuroprotection that is inducible nitric oxide synthase-dependent in neonatal rats.

BACKGROUND: Perinatal stroke is a common human disease. Neonatal brains are immature and engaged in active synaptogenesis. Preconditioning adult rats with the volatile anesthetic isoflurane induces neuroprotection. Whether isoflurane preconditioning induces neuroprotection in neonates is not known. METHODS: Seven-day-old Sprague-Dawley rats had left common carotid arterial ligation followed by hypoxia with 8% oxygen for 1, 2, or 2.5 h at 37 degrees C. Isoflurane preconditioning with 1 or 1.5% isoflurane for 30 min was performed at 24 h before the brain hypoxia/ischemia. The inducible nitric o

Cross-sensitivity between isoflurane and diazepam: evidence from a bidirectional tolerance study in mice.

We examined in mice the effect of chronic diazepam treatment on the sensitivity to isoflurane, and that of repeated isoflurane exposure on the sensitivity to diazepam. Mice were divided into four groups: group 1, treated with diazepam, 10 mg/kg i.p. twice daily; group 2, vehicle-treated controls; group 3, exposed to 3% isoflurane for 25 min twice daily; and group 4, untreated controls. After 14 days the effect of the treatment was assessed. Twenty-four hours after the last 10 mg/kg diazepam treatment, groups 1 and 2 received diazepam, 5 mg/kg i.p., and were subjected to the horizontal wire tes

Isoflurane and sevoflurane induce vasodilation of cerebral vessels via ATP-sensitive K+ channel activation.

BACKGROUND: Activation of adenosine triphosphate-sensitive K+ channels causes cerebral vasodilation. To assess their contribution to volatile anesthetic-induced cerebral vasodilation, the effects of glibenclamide, an adenosine triphosphate-sensitive K+ channel blocker, on the cerebral vasodilation induced by isoflurane and sevoflurane were studied. METHODS: Pentobarbital-anesthetized dogs (n = 24) assigned to one of two groups were prepared for measurement of pial vessel diameter using a cranial window preparation. Each dog received three minimum alveolar concentrations (MAC; 0.5, 1, and 1.5

Slow ventilatory dynamics after isocapnic hypoxia and voluntary hyperventilation in humans: effects of isoflurane.

Short-term potentiation (STP) of breathing refers to respiratory activity that persists at termination of a primary stimulus and is not related just to the dynamics of chemoreceptors. In humans, STP is activated by brief episodes of hypoxia and voluntary hyperventilation (VHV). STP exerts a stabilizing influence on breathing pattern. To investigate the effects of a subanaesthetic concentration of isoflurane on STP, we studied recovery from mild and moderate hypoxic hyperpnoea and VHV. Experiments were performed in eight healthy volunteers. If necessary, subjects were aroused to maintain a stat

[Cognitive and psychomotor performance following isoflurane, midazolam/alfentanil and propofol anesthesia. A comparative study].

Mental and psychomotor abilities are impaired to varying degrees after general anaesthesia. This has important implications for the time over which patients are monitored in the recovery room and for the discharge of outpatients after day surgery. The present study was undertaken to compare recovery and mental and psychomotor skills in the first 60 min following general anaesthesia with isoflurane, midazolam/alfentanil and propofol. METHODS. A total of 45 patients undergoing microsurgical lumbar nucleotomy were randomized to three study groups. Group 1 (n = 15): anaesthesia was induced with th

Port of Choice: Induction Chamber Inlet Position Affects Speed of Isoflurane Induction in Mice.

Various factors affect inhalant anesthetic induction speed in mice. We investigated the effects of different inhalant anesthetic inlet port positions on isoflurane induction speed in 3 mouse strains. We considered 3 anesthetic inlet port positions: conventional (port located in a traditional location approximately halfway between the induction box lid and base), top (port incorporated into the induction box lid), and bottom (port located adjacent to the base, but on the side wall). We hypothesized that positioning the anesthetic inlet port at the bottom of the induction chamber would result in

COMPARISON OF THREE SEDATION COMBINATIONS IN ISOFLURANE-ANESTHETIZED GARDEN DORMICE (ELIOMYS QUERCINUS) UNDERGOING LAPAROTOMY.

The garden dormouse (Eliomys quercinus) is commonly used as model species for studies on hibernation, which may involve surgery. Similar to laboratory rodents, inhalational anesthesia, which does not provide analgesia, is often performed for surgical procedures. We retrospectively compared cardiorespiratory effects between ketamine-butorphanol-medetomidine (KBMed), ketamine-butorphanol-midazolam (KBMid) and butorphanol-medetomidine-midazolam (BMM) administered SC in 48 garden dormice undergoing laparotomy for bio-logger implantation plus tissue (liver, brown fat) biopsy surgery (n = 48/48 [28

Capture of isoflurane from anaesthetised dogs and cats following methadone, medetomidine and ketamine administration.

BACKGROUND: Volatile anaesthetic agents contribute to climate change, and reducing this impact is a commendable ambition. We evaluated the efficiency of a veterinary capture device for isoflurane (VET-can/VET-dock, SageTech Veterinary) in anaesthetised cats and dogs. METHODS: Purposeful sampling was used to recruit enough animals undergoing isoflurane general anaesthesia to fill one VET-can to at least 95% capacity. The vaporiser and VET-can were weighed pre- and post-anaesthesia to determine in vivo mass transfer (MT) of isoflurane and water. The primary outcomes were capture efficiency (iso
