Optimization and validation of liquid chromatography and headspace-gas chromatography based methods for the quantitative determination of capsaicinoids, salicylic acid, glycol monosalicylate, methyl salicylate, ethyl salicylate, camphor and l-menthol in a topical formulation.

Capsaicinoids, salicylic acid, methyl and ethyl salicylate, glycol monosalicylate, camphor and l-menthol are widely used in topical formulations to relieve local pain. For each separate compound or simple mixtures, quantitative analysis methods are reported. However, for a mixture containing all above mentioned active compounds, no assay methods were found. Due to the differing physicochemical characteristics, two methods were developed and optimized simultaneously. The non-volatile capsaicinoids, salicylic acid and glycol monosalicylate were analyzed with liquid chromatography following liqui

Reduction of the olfactory cognitive ability in horses during preslaughter: stress-related hormones evaluation.

As horses may perceive several odour signals of danger at slaughter, application of mentholated ointment to their nostrils may limit their perception of danger. To assess the effect of the application of a mentholated ointment to horse nostrils on the stress response during pre-slaughter handling, plasma levels were evaluated for cortisol, beta-endorphin, epinephrine and norepinephrine prior to and after stunning. Twenty draught-type horses were divided into control (n=10) and treated (n=10) groups and a mentholated ointment applied to the nostrils of the treated horses following blood samplin

The action of selected monoterpenes as biofilm control agents and antibiotic resistance modifiers.

Multidrug-resistant bacteria in biofilms are a growing public health threat, due to their resistance to conventional antibiotics. Phytochemicals are attractive candidates because of their structural diversity and ability to potentiate antimicrobial activity. This study investigated the antibiofilm and resistance-modifying effects of two monoterpenes, menthol and linalool, alone and in combination with ten antibiotics, against Escherichia coli and Staphylococcus epidermidis. Menthol exhibited MIC and MBC of 800 µg/mL against E. coli and the same MIC against S. epidermidis, while linalool showed

TRPM8 voltage sensor mutants reveal a mechanism for integrating thermal and chemical stimuli.

TRPM8, a member of the transient receptor potential (TRP) channel superfamily, is expressed in thermosensitive neurons, in which it functions as a cold and menthol sensor. TRPM8 and most other temperature-sensitive TRP channels (thermoTRPs) are voltage gated; temperature and ligands regulate channel opening by shifting the voltage dependence of activation. The mechanisms and structures underlying gating of thermoTRPs are currently poorly understood. Here we show that charge-neutralizing mutations in transmembrane segment 4 (S4) and the S4-S5 linker of human TRPM8 reduce the channel's gating ch

The roles of iPLA2, TRPM8 and TRPA1 in chemically induced cold hypersensitivity.

BACKGROUND: The cooling agents menthol and icilin act as agonists at TRPM8 and TRPA1. In vitro, activation of TRPM8 by icilin and cold, but not menthol, is dependent on the activity of a sub-type of phospholipase A2, iPLA2. Lysophospholipids (e.g. LPC) produced by PLA2 activity can also activate TRPM8. The role of TRPA1 as a primary cold sensor in vitro is controversial, although there is evidence that TRPA1 plays a role in behavioural responses to noxious cold stimuli. In this study, we have investigated the roles of TRPM8 and TRPA1 and the influence of iPLA2 on noxious cold sensitivities in

Study on the mechanism of penetration of particles of air pollution into the brain through the nasal cavity and the potential neurological health risks.

The presence of nanopollutants in the atmosphere poses significant risks to human health. However, research on their adverse effects on the central nervous system (CNS) remains limited. To address this gap, we constructed an in vitro model utilizing mesoporous silica nanoparticles (MSNs) to simulate airborne solid particulates and menthol/curcumol to represent adsorbed liquid microparticles. This model was employed to investigate the impact of nanopollutants on the CNS, focusing on their entry pathways into the CNS and the mechanisms underlying their cellular transport and uptake. The results

Viable, Multi-Drug-Resistant Bacteria Recovered from E-Liquids Used with Commercial Electronic Cigarettes.

The use of electronic cigarettes has increased in the U.S. with menthol and mint flavors showing notably higher sales. While research on the bacterial microbiome of traditional tobacco products is growing, particularly regarding menthol and nicotine effects, data regarding potential microbial contaminants within electronic liquids (e-liquids) remain limited. Additionally, the potential antibacterial properties of e-liquids remain sparse. To address these gaps, we evaluated the prevalence of viable bacteria in e-liquids; characterized their antimicrobial susceptibility patterns; and tested the

Prospects for prostate cancer imaging and therapy using high-affinity TRPM8 activators.

One of the best-studied temperature-gated channels is transient receptor potential melastatin 8 (TRPM8), which is activated by cold and cooling agents, such as menthol. Besides inducing a cooling sensation in sensory neurons, TRPM8 channel activation also plays a major role in physiopathology. Indeed, TRPMP8 expression increases in early stages of prostate cancer and its involvement in prostate cell apoptosis has recently been demonstrated. Thus, as TRPM8 is a tumor marker with significant potential use in diagnosis, as well as a target for cancer therapy, there is a need for new TRPM8-specifi
