Identifying biomarkers for arsenic exposure-induced nonalcoholic fatty liver disease: An integrated proteomics, metabolomics, and machine learning approach.

Arsenic, a toxic metalloid prevalent in the environment, poses significant health risks to humans primarily via the contamination of water. Arsenic exposure has been demonstrated to cause nonalcoholic fatty liver disease (NAFLD); however, the biomarkers used for the identification of its NAFLD risk remain inadequately explored. Based on our previous finding that arsenic induces NAFLD (characterized by hepatic lipid accumulation) in rats, this study aimed to identify potential biomarkers for the risk assessment of environmentally relevant levels of arsenic-induced NAFLD using an integrated mult

Transcriptional and biochemical changes in mouse liver following exposure to a metal/drug cocktail. Attenuating effect of a selenium-enriched diet.

Real-life pollution usually involves simultaneous co-exposure to different chemicals. Metals and drugs are frequently and abundantly released into the environment, where they interact and bioaccumulate. Few studies analyze potential interactions between metals and pharmaceuticals in these mixtures, although their joint effects cannot be inferred from their individual properties. We have previously demonstrated that the mixture (PC) of the metals Cd and Hg, the metalloid As and the pharmaceuticals diclofenac (DCF) and flumequine (FLQ) impairs hepatic proteostasis. To gain a deeper vision of how

Epicatechin ameliorates glucose intolerance and hepatotoxicity in sodium arsenite-treated mice.

Arsenic is a metalloid found in the environment that causes toxic effects in different organs, mainly the liver. This study aimed to investigate the protective effects of epicatechin (EC), a natural flavonol, on glucose intolerance (GI) and liver toxicity caused by sodium arsenite (SA) in mice. Our findings showed that SA exposure led to the development of GI. Liver tissue damage and decreased pancreatic Langerhans islet size were also observed in this study. Mice exposed to SA exhibited hepatic oxidative damage, indicated by reduced antioxidant markers (such as superoxide dismutase, catalase,

Hepatoprotective and anti-hyperglycemic effects of ferulic acid in arsenic-exposed mice.

Arsenic is a toxic metalloid that increases the risk of hepatotoxicity and hyperglycemia. The objective of the present study was to assess the effect of ferulic acid (FA) in mitigating glucose intolerance and hepatotoxicity caused by sodium arsenite (SA). A total of six groups including control, FA 100 mg/kg, SA 10 mg/kg, and groups that received different doses of FA (10, 30, and 100 mg/kg), respectively just before SA (10 mg/kg) for 28 days were examined. Fasting blood sugar (FBS) and glucose tolerance tests were conducted on the 29th day. On day 30, mice were sacrificed and blood and tissue

The Design of Experiment as a Tool to Model Plant Trace-Metal Bioindication Abilities.

Bioindicator plants are species that have the capacity to linearly uptake some elements (metal and metalloids) from the growing substrate, thus reflecting their concentration in the soil. Many factors can influence the uptake of these elements by plants, among which is the simultaneous presence of several metals, a common situation in contaminated or natural soils. A novel approach that can be used to validate the bioindication ability of a species growing on a polymetallic substrate is the design of experiment (DoE) approach. The aim of the present study was to apply the DoE in full factorial

ATF3 inhibits arsenic-induced malignant transformation of human bronchial epithelial cells by attenuating inflammation.

Arsenic is a naturally occurring metalloid strongly associated with the incidence of lung cancer. Understanding the mechanisms of arsenic-induced carcinogenesis favors the development of effective interventions to reduce the incidence and mortality of lung cancer. In this study, we investigated the role of activating transcription factor 3 (ATF3) in arsenic-induced transformation of human bronchial epithelial cells. ATF3 was upregulated during chronic exposure to 0.25 μM arsenic, and loss of ATF3 promoted arsenic-induced transformation. Moreover, arsenic-transformed ATF3 knockout (ATF3 KO-AsT)

Sex-specific neurotoxic effects of heavy metal pollutants: Epidemiological, experimental evidence and candidate mechanisms.

The heavy metals lead (Pb), mercury (Hg), and cadmium (Cd) are ubiquitous environmental pollutants and are known to exert severe adverse impacts on the nervous system even at low concentrations. In contrast, the heavy metal manganese (Mn) is first and foremost an essential nutrient, but it becomes neurotoxic at high levels. Neurotoxic metals also include the less prevalent metalloid arsenic (As) which is found in excessive concentrations in drinking water and food sources in many regions of the world. Males and females often differ in how they respond to environmental exposures and adverse eff

Arsenic and Human Health: Genotoxicity, Epigenomic Effects, and Cancer Signaling.

Arsenic is a well-known element because of its toxicity. Humans as well as plants and animals are negatively affected by its exposure. Some countries suffer from high levels of arsenic in their tap water and soils, which is considered a primary arsenic-linked risk factor for living beings. Humans generally get exposed to arsenic by contaminated drinking waters, resulting in many health problems, ranging from cancer to skin diseases. On the other hand, the FDA-certified drug arsenic trioxide provides solutions for various diseases, including several types of cancers. This issue emphasizes the i

Neurotoxic Mechanism of Arsenic: Synergistic Effect of Mitochondrial Instability, Oxidative Stress, and Hormonal-Neurotransmitter Impairment.

Arsenic toxicity which is now a global concern is predicted to affect more than 200 million people. Chronic arsenic exposure conduce carcinogenicity, hepatotoxicity, and neurotoxicity. Here we have reviewed numerous epidemiological and experimental reports related to arsenic toxicity to explore its neurotoxicity mechanism. Penetrability of this metalloid through blood-brain barrier makes it a potent neuro-toxicant by inducing mitochondrial membrane instability and calorie exhaustion. It directly affects the cortex, cerebellum region, and specially microglial cells by the induction of a variety

Exogenous spermidine, arsenic and beta-aminobutyric acid modulate tobacco resistance to tobacco mosaic virus, and affect local and systemic glucosylsalicylic acid levels and arginine decarboxylase gene expression in tobacco leaves.

The polyamine spermidine and the metalloid arsenic increased resistance responses in the well-known pathosystem NN tobacco/tobacco mosaic virus (TMV). Both the hypersensitive response to TMV in a leaf disk model system (inoculated disks floating in the 0.1mM treatments) and systemic acquired resistance (SAR) in whole plants were significantly affected. In the latter case, 1mM foliar sprays of spermidine and arsenic were as effective as TMV and dl-beta-aminobutyric acid (BABA), both taken as positive controls, in improving the plant's response to subsequent challenge inoculation with TMV. Physiological and toxicological changes in the skin resulting from the action and interaction of metal ions.

The human environment contains more than 50 metal or metalloid elements. At least 15 are recognized as trace elements, with zinc, calcium, copper, magnesium, and iron having specific roles in skin morphogenesis and function. The present review focuses on the presumed role of metal ions in the skin, their competition for carrier proteins, and membrane receptors. Evidence presented shows that the balance of trace metal ions is critical for normal skin and repair mechanisms following injury. Xenobiotic ions can impair this balance, leading to pathological change. The skin acts as an organ of elim

Disruption of Antioxidant Defense Systems in Honey Bees and Wild Bees Under Environmental Xenobiotic Pressure.

Honey bee colonies play a vital role in ecosystem stability and global food security. Together with bumble bees and other wild bee species, they form a diverse pollinator community that is particularly vulnerable to environmental pollution. Among stressors, environmental xenobiotics including heavy metals, metalloids, pesticides, polycyclic aromatic hydrocarbons, per- and polyfluoroalkyl substances, and emerging contaminants such as microplastics pose a growing concern due to their persistence, bioaccumulation potential and capacity to trigger oxidative stress and interact with pathogens, nutr

Prevalent and diverse new plasmid-encoded heavy metal and antimicrobial resistance in Klebsiella strains isolated from hospital wastewater.

The rise of antibiotic-resistant Klebsiella pneumoniae poses a significant global health threat. Plasmids, as mobile genetic elements, play a critical role in bacterial adaptation by facilitating the spread of resistance genes. To analyze plasmid-mediated antibiotic and heavy metal resistance in clinical Klebsiella strains, 33 Klebsiella strains isolated from wastewater were subjected to third-generation nanopore sequencing to obtain high-quality whole-genome assemblies. The presence and diversity of plasmids associated with antibiotic and heavy metal resistance were analyzed, and phenotypic a

Novel multimodal approach of LA-ICP-MS exploring innovative Ru-tetrazene: In situ tracking and bioimaging-guided cancer treatment.

In this study, we employed LA-ICP-MS to trace a novel, unique Ru-based complex in a lung cancer cell line. This work highlights the importance of the time synchronization of all instrumentation, including the Aerosol Rapid Introduction System. It introduces bulk in situ LA-ICP-MS for rapid screening of chemotherapeutic penetration into the cell and the use of high-resolution 2D single cell imaging, which is used for both large-scale mapping for the monitoring of large cellular variability and for imaging single cells for the precise localization of ruthenium tetrazene accumulation. Glucocorticoid and mineralocorticoid receptors and corticosteroid homeostasis are potential targets for endocrine-disrupting chemicals.

Endocrine-disrupting chemicals (EDCs) have received significant concern, since they ubiquitously exist in the environment and are able to induce adverse health effects on human and wildlife. Increasing evidence shows that the glucocorticoid receptor (GR) and the mineralocorticoid receptor (MR), members of the steroid receptor subfamily, are potential targets for EDCs. GR and MR mediate the actions of glucocorticoids and mineralocorticoids, respectively, which are two main classes of corticosteroids involved in many physiological processes. The effects of EDCs on the homeostasis of these two cl

Geochemical conditions conducive for retention of trace elements and radionuclides during shale-fluid interactions.

Produced water generated during unconventional oil and gas extractions contains a complex milieu of natural and anthropogenic potentially toxic chemical constituents including arsenic (As), chromium (Cr), and cadmium (Cd), naturally occurring radioactive materials (NORMs) including U and Ra, and a myriad of organic compounds. The human-ecological health risks and challenges associated with the disposal of produced water may be alleviated by understanding geochemical controls on processes responsible for the solubilization of potentially hazardous natural shale constituents to produced water. Laser ablation inductively coupled plasma mass spectrometry imaging of metals in experimental and clinical Wilson's disease.

Wilson's disease is an autosomal recessive disorder in which the liver does not properly release copper into bile, resulting in prominent copper accumulation in various tissues. Affected patients suffer from hepatic disorders and severe neurological defects. Experimental studies in mutant mice in which the copper-transporting ATPase gene (Atp7b) is disrupted revealed a drastic, time-dependent accumulation of hepatic copper that is accompanied by formation of regenerative nodes resembling cirrhosis. Therefore, these mice represent an excellent exploratory model for Wilson's disease. However, th
