Exposure to Aerosolized Algal Toxins in South Florida Increases Short- and Long-Term Health Risk in Drosophila Model of Aging.

Harmful algal blooms (HABs) are a rising health and environmental concern in the United States, particularly in South Florida. Skin contact and the ingestion of contaminated water or fish and other seafood have been proven to have severe toxicity to humans in some cases. However, the impact of aerosolized HAB toxins is poorly understood. In particular, knowledge regarding either the immediate or long-term effects of exposure to aerosolized cyanotoxins produced by freshwater blue-green algae does not exist. The aim of this study was to probe the toxicity of aerosolized cyanobacterial blooms usi

Interactions between Microcystis aeruginosa and coexisting bisphenol A at different phosphorus levels.

Microcystis aeruginosa is known as the main contributor to cyanobacterial bloom, which is prevalent globally and degrades freshwater systems worldwide. The argument that the introduction of anthropogenic contaminants in fresh water stimulates cyanobacterial growth and microcystin production has attracted widespread attention. Bisphenol A (BPA), one of the most abundant endocrine-disrupting compounds, is often detected in various water bodies due to its notably high annual levels of production and use. Research on the combined effects of endocrine-disrupting compounds and environmental factors

Toxicity evaluation of freshwater cyanobacterium Microcystis aeruginosa PCC 7806: II. Nephrotoxicity in rats.

Nephrotoxic potential of laboratory cultures of freshwater cyanobacterium (blue-green alga) Microcystis aeruginosa PCC 7806 (Pasteur Institute) was assessed in male rats. The animals were injected intraperitoneally with 0.5, 1.0 and 2.0 LD50 doses of lyophilized cell extract. Elevated plasma urea and creatinine levels were accompanied by decrease in protein and albumin levels, followed by hematuria, proteinuria and bilirubinuria. Also decrease in kidney lactate dehydrogenase and glutamic oxaloacetic transaminase indicated possible nephrotoxic potential of the cyanobacteria. The extract also pr

Mosquito inoculation: an alternative bioassay for toxins.

Mosquitoes were evaluated as a bioassay host for several classes of biological toxins. Mosquitoes were sensitive to snake toxic or neurotoxic phospholipase A2 enzymes (but not to nontoxic phospholipase A2 enzymes), cobrotoxin, saxitoxin, microcystin and the scorpion insect sodium channel toxin. Mosquitoes were not sensitive to ricin, diphtheria toxin, anthrax toxin, botulinum toxin, tetanus toxin, conotoxin G or a scorpion sodium channel toxin toxic to mammals. Specific antisera neutralization tests with mosquitoes gave comparable results to those of a mouse assay. The mosquito is a suitable b

Challenges of using blooms of Microcystis spp. in animal feeds: A comprehensive review of nutritional, toxicological and microbial health evaluation.

Microcystis spp., are Gram-negative, oxygenic, photosynthetic prokaryotes which use solar energy to convert carbon dioxide (CO2) and minerals into organic compounds and biomass. Eutrophication, rising CO2 concentrations and global warming are increasing Microcystis blooms globally. Due to its high availability and protein content, Microcystis biomass has been suggested as a protein source for animal feeds. This would reduce dependency on soybean and other agricultural crops and could make use of "waste" biomass when Microcystis scums and blooms are harvested. Besides proteins, Microcystis cont

In vivo assessment of the hepatotoxicity of a new Nostoc isolate from the Nile River: Nostoc sp. strain NRI.

Nostoc sp. is one of the most widely distributed cyanobacterial genera that produce potentially protein phosphatase (PP) inhibitor; microcystins (MCs). MCs have posed a worldwide concern due to predominant hepatotoxicity to human health. We have previously isolated a Nostoc strain (NR1) from the Nile River (the main water supply in Egypt) and this strain exerted production of rare and highly toxic MC; demethylated microcystin-LR. There is no data concerning risk factors of liver diseases for human and animal exposure to NR1-contaminated drinking water yet. It is thus important to evaluate acut

Cyanobacteria, cyanotoxins and lipopolysaccharides in aerosols from inland freshwater bodies and their effects on human bronchial cells.

Components of cyanobacterial water blooms were quantified in aerosols above agitated water surfaces of five freshwater bodies. The thoracic and respirable aerosol fraction (0.1-10 µm) was sampled using a high-volume sampler. Cyanotoxins microcystins were detected by LC-MS/MS at levels 0.3-13.5 ng/mL (water) and < 35-415 fg/m3 (aerosol). Lipopolysaccharides (endotoxins) were quantified by Pyrogene rFC assay at levels < 10-119 EU/mL (water) and 0.13-0.64 EU/m3 (aerosol). Cyanobacterial DNA was detected by qPCR at concentrations corresponding to 104-105 cells eq./mL (water) and 101-103 cells eq./

Chronic and Acute Toxicities of Aflatoxins: Mechanisms of Action.

There are presently more than 18 known aflatoxins most of which have been insufficiently studied for their incidence, health-risk, and mechanisms of toxicity to allow effective intervention and control means that would significantly and sustainably reduce their incidence and adverse effects on health and economy. Among these, aflatoxin B1 (AFB1) has been by far the most studied; yet, many aspects of the range and mechanisms of the diseases it causes remain to be elucidated. Its mutagenicity, tumorigenicity, and carcinogenicity-which are the best known-still suffer from limitations regarding th

Nutrition and metabolism in hepatocellular carcinoma.

Hepatocellular carcinoma is the fifth most common human cancer worldwide, with an overall 5-year survival in the range of 10%. In addition to the very substantial role of chronic viral hepatitis in causing hepatocellular carcinoma, nutritional status and specific nutritional factors appear to influence disease risk. This is apparent in the increased risk associated with non-alcoholic hepatic cirrhosis occurring in the context of obesity, the metabolic syndrome, and type 2 diabetes. Specific nutrients and ingested toxins, including ethanol, aflatoxin, microcystins, iron, and possibly components

Lesser-Known Cyanotoxins: A Comprehensive Review of Their Health and Environmental Impacts.

Cyanobacteria, also known as blue-green algae, are a diverse phylum of photosynthetic, Gram-negative bacteria and one of the largest microbial taxa. These organisms produce cyanotoxins, which are secondary metabolites that can have significant impacts on both human health and the environment. While toxins like Microcystins and Cylindrospermopsins are well-documented and have been extensively studied, other cyanotoxins, including those produced by Lyngbya and Nostoc, remain underexplored. These lesser-known toxins can cause various health issues in humans, including neurotoxicity, hepatotoxicit

Effects of UVC/H2O2 on dissolved organic matter derived from a cyanobacterial bloom.

Cyanobacterial blooms release complex metabolite mixtures, yet their degradation during water treatment remains poorly understood. This study investigates the degradation of cyanometabolite-rich natural bloom extracts using UVC/H2O2 to generate hydroxyl radicals (HO•). By applying LC-HR-MS/MS combined with computational metabolomics, we identified nine microcystins (MCs), four anabaenopeptins (APs), and sixteen microginins (MGs), proposing structures for five new congeners and two novel transformation products (TPs). UVC/H2O2 accelerated cyanopeptide degradation compared to individual control

Female reproductive impacts of emerging cyanobacterial harmful algal bloom toxins.

Harmful algal blooms (HABs), denoted by the excessive growth or 'bloom' of cyanobacteria, have increased in frequency, duration, and intensity worldwide over the past decades, primarily due to the rise in global temperature and nutrient enrichment. The blooms of cyanobacteria release various HAB toxins, with the cyclic heptapeptide microcystin-LR (MC-LR) as the most prevalent and well-characterized. Evidence suggests that exposure to HAB toxins poses significant risks to female reproductive health. Herein, we summarized current knowledge on the female reproductive impacts of MC-LR and other HA

Hawthorn fruit extract protect against MC-LR-induced hepatotoxicity by attenuating oxidative stress and apoptosis.

Microcystins (MCs) is a class of cyclic heptapeptide compounds with biological activity. There is no effective treatment for liver injury caused by MCs. Hawthorn is a medicinal and edible plant traditional Chinese medicine with hypolipidemic, reducing inflammation and oxidative stress in the liver. This study discussed the protective effect of hawthorn fruit extract (HFE) on liver damage caused by MC-LR and the underlying molecular mechanism. After MC-LR exposure, pathological changes were observed and hepatic activity of ALT, AST and ALP were increased obviously, but they were remarkably rest

Okara and okara modified and functionalized with iron oxide nanoparticles for the removal of Microcystis aeruginosa and cyanotoxin.

Eutrophicating compounds promote the growth of cyanobacteria, which has the potential of releasing toxic compounds. Alternative raw materials, such as residues, have been used in efficient adsorption systems in water treatment. The aim of the present study was to apply the residue Okara in its original form and modified by hydrolysis with immobilization of magnetic nanoparticles as an adsorbent. For the removal, the cyanobacteria Microcystis aeruginosa was chosen, as well as its secondary metabolites, L-amino acids leucine and arginine (MC-LR microcystin), from aqueous solutions. The adsorbent

Viral community assembly during cyanobacterial blooms reveals ecosystem-specific regulation and nutrient cycling in freshwater lakes.

Cyanobacterial blooms pose escalating threats to freshwater ecosystems and public health, yet the ecological roles of viral communities in bloom development, succession and toxin dynamics remain poorly understood. We characterized bacterial and viral community dynamics during bloom seasons in two chronically affected lakes with contrasting ecological properties: Lake Erie, a large Great Lake, and Buckeye Lake, a shallow inland reservoir. Buckeye Lake exhibited substantially higher bloom severity, with fivefold higher microcystin concentrations (mean 24.40 vs. 5.30 μg L-¹), all samples exceedin
