Therapeutic hypothermia reduces cortical inflammation associated with utah array implants.

OBJECTIVE: Neuroprosthetics hold tremendous promise to restore function through brain-computer interfaced devices. However, clinical applications of implantable microelectrodes remain limited given the challenges of maintaining neuronal signals for extended periods of time and with multiple biological mechanisms negatively affecting electrode performance. Acute and chronic inflammation, oxidative stress, and blood brain barrier disruption contribute to inconsistent electrode performance. We hypothesized that therapeutic hypothermia (TH) applied at the microelectrode insertion site will positiv

Insertion mechanics of amorphous SiC ultra-micro scale neural probes.

Objective. Trauma induced by the insertion of microelectrodes into cortical neural tissue is a significant problem. Further, micromotion and mechanical mismatch between microelectrode probes and neural tissue is implicated in an adverse foreign body response (FBR). Hence, intracortical ultra-microelectrode probes have been proposed as alternatives that minimize this FBR. However, significant challenges in implanting these flexible probes remain. We investigated the insertion mechanics of amorphous silicon carbide (a-SiC) probes with a view to defining probe geometries that can be inserted into

Investigation of neurotrophic factor concentrations with a novel in vitro concept for peripheral nerve regeneration.

The regeneration of nerves of the peripheral nervous system after injuries is a complex process. This study presents a novel in vitro neurite regeneration concept to investigate the regeneration of neurons and their processes with different concentrations of neurotrophic factors. The core part of the concept is a transparent microfluidic neurite isolation (NI) device affixed on top of a microelectrode array (MEA), providing a fast and easy way to assess both the growth and the electrical activity of neurites. The NI-MEA isolates neurites from the culture with microchannels that serve as guidan

Improvement of the embryonic stem cell test endpoint analysis by use of field potential detection.

The embryonic stem cell test (EST) is a validated in vitro method to assess the embryotoxic potential of compounds and is a promising tool for drug screening. EST requires microscopic observation of beating cardiomyocytes differentiated from embryonic stem cells as a toxicological endpoint. However, this process is time-consuming and lacks throughput performance. To improve the analysis, we introduced an electrophysiological method with a microelectrode array system for the evaluation of differentiated cardiomyocytes. Embryotoxic (valproic acid, verapamil, and 5-fluorouracil) and non-embryotox

Scanning Photoelectrochemical Microscopy for the Investigation of Local Photocatalytic H2 Evolution in Matrixed Langmuir Films.

Molecule-in-matrix systems such as Langmuir films with photosensitizers and hydrogen evolution reaction (HER) catalysts may exhibit heterogeneity in photocatalytic activity across large-area films. Here, we introduce a confined illumination approach and simultaneous localized detection of light-driven hydrogen (H2) evolution to directly correlate information on structural organization with catalytic performance of Langmuir-Blodgett and Langmuir-Schaefer films with incorporated [Mo3S13]2- catalyst and a Ru-based photosensitizer. Derived from a coating of optical fibers used in scanning near-fie

Studies on Square Wave and Cyclic Voltammetric Behavior of 1,2- and 1,4-Dihydroxybenzenes and Their Derivatives in Acetic Acid, Ethyl Acetate and Mixtures of the Two.

An electrochemical investigation of 1,2- and 1,4-dihydroxybenzenes was carried out with platinum macro- and microelectrodes using square wave and cyclic voltammetry techniques. Furthermore, the effect of the two solvents-acetic acid and ethyl acetate-was compared. When using square wave voltammetry, signals only appeared at lower frequencies and only when the supporting electrolyte was in excess, as expected due to the relatively low permittivity of the used solvents. The behavior of hydroquinone and catechol did not differ significantly from that of their derivatives (dihydroxybenzaldehydes,

Advanced age is not a barrier to chronic intracortical single-unit recording in rat cortex.

INTRODUCTION: Available evidence suggests that as we age, our brain and immune system undergo changes that increase our susceptibility to injury, inflammation, and neurodegeneration. Since a significant portion of the potential patients treated with a microelectrode-based implant may be older, it is important to understand the recording performance of such devices in an aged population. METHODS: We studied the chronic recording performance and the foreign body response (FBR) to a clinically used microelectrode array implanted in the cortex of 18-month-old Sprague Dawley rats. RESULTS AND DIS

Array Focal Cortical Stimulation Enhances Motor Function Recovery and Brain Remodeling in a Rat Model of Ischemia.

OBJECTIVE: Using a new microelectrode array implanted into the cranial window employing a new stimulation protocol, we investigated the effects of the implanted electrode arrays on both motor map plasticity and neural regeneration in a rodent model of stroke. MATERIALS AND METHODS: Rats were pretrained on single-pellet retrieval task, then received focal ischemic infarction and microelectrode arrays implantation. Rats in the cortical stimulation (CS) group received daily electrical stimulation (1 hour each day) for 14 days whereas animals in the no stimulation (NS) group did not receive elect

Spatiotemporal Changes of Neuronal Responses in the Primary Somatosensory Cortex to Noxious Tail Stimulation in Awake and Pentobarbital-Anesthetized Rats.

Primary somatosensory cortex (SI) is a key area in the processing of nociceptor inputs to our consciousness. To clarify the columnar and laminar organization of SI for pain processing, we compared spatiotemporal changes in neuronal activities of the primary sensorimotor cortex (SmI) of the rat in response to noxious laser heat stimulation applied to the mid-tail. Longitudinal and vertical array microelectrodes were chronically implanted in the cerebral cortex. Evoked neuronal activities, including intracortical local field potentials (LFP) and ensemble single-unit activity (SU) around SmI were

Four women whose pioneering contributions to science have been largely overlooked.

Ethel Browne Harvey and Hilde Pröscholdt Mangold did pioneering research in embryology, Ida Henrietta Hyde helped develop the first microelectrodes for the stimulation of single cells, and Marthe Gautier had a vital role in discovering that Down syndrome is caused by an extra copy of chromosome 21. So why are their names so little known by the scientific community at large?

[The action of the venom of the spider Eresus niger on the neuromuscular synapses of the frog].

The effect of Eresus niger spider venom on frog neuromuscular preparations has been studied using the intracellular microelectrodes. The spider venom has been found to block both spontaneous and elicited transmitter release and possesses the phospholipase activity. It has been suggested that the venom blocking synaptic transmission results from its phospholipase activity by analogy with the action of snake presynaptic neurotoxins.

A potential difference across mouse ovarian follicle.

A small potential difference (antrum positive) has been measured with fine-tipped glass microelectrodes across the epithelial cell layers of the mouse ovarian follicle wall. As ovulation approached the potential in the antrum became more positive compared to the outside. Metabolic inhibitors and locally active hormones also altered the potential difference. The ionic basis and the significance of the potential difference are unknown.

An Optically Controlled Virtual Microsensor for Biomarker Detection In Vivo.

Current technologies for the real-time analysis of biomarkers in vivo, such as needle-type microelectrodes and molecular imaging methods based on exogenous contrast agents, are still facing great challenges in either invasive detection or lack of active control of the imaging probes. In this study, by combining the design concepts of needle-type microelectrodes and the fluorescence imaging method, a new technique is developed for detecting biomarkers in vivo, named as "optically controlled virtual microsensor" (OCViM). OCViM is established by the organic integration of a specially shaped laser

Electrochemical Nitric Oxide Microsensors Based on a Fluorinated Xerogel Screening Layer for in Vivo Brain Monitoring.

Nitric oxide (NO) is an important free radical synthesized and released by brain cells. At low (nanomolar) levels, it modulates synaptic transmission and neuronal activity, but at much higher levels mediates neuronal injury through oxidative stress. However, the precise concentrations at which these biological actions are exerted are still poorly defined. Electrochemical detection of NO in vivo requires rigorous exclusion of endogenous redox molecules such as ascorbate or nitrite. A fluorinated xerogel composed of trimethoxymethylsilane and heptadecafluoro-1,1,2,2-tetrahydrodecyl silane has be

Chronic monitoring of lower urinary tract activity via a sacral dorsal root ganglia interface.

OBJECTIVE: Our goal is to develop an interface that integrates chronic monitoring of lower urinary tract (LUT) activity with stimulation of peripheral pathways. APPROACH: Penetrating microelectrodes were implanted in sacral dorsal root ganglia (DRG) of adult male felines. Peripheral electrodes were placed on or in the pudendal nerve, bladder neck and near the external urethral sphincter. Supra-pubic bladder catheters were implanted for saline infusion and pressure monitoring. Electrode and catheter leads were enclosed in an external housing on the back. Neural signals from microelectrodes and

A feasibility study of optical coherence tomography for guiding deep brain probes.

Deep brain simulation (DBS) is effective for the treatment of various diseases including Parkinson's disease and essential tremor. However, anatomical targeting combined with microelectrode mapping of the region requires significant surgical time. Also, the fine-tipped microelectrode imposes a risk of hemorrhage in the event that the trajectory intersects subcortical vessels. To reduce the operation time and the risk of hemorrhage, we propose to use optical coherence tomography (OCT) to guide the insertion of the DBS probe. We conducted in vitro experiments in the rat brain to study the feasib

Interpretation of oxygen disappearance curves measured in blood perfused tissues.

We have developed a two compartment (tissue and blood) lumped parameter model to interpret oxygen disappearance curves (O2 DCs) measured in vivo with PO2 microelectrodes in tissues which are perfused with blood. To include the properties of the oxyhemoglobin equilibrium curve (HEC), we used an algorithm we have recently developed for both standard and nonstandard conditions. The new blood and tissue model is more useful than a previous analysis using the Hill equation for blood and constant oxygen consumption (VO2). The model can be adapted for constant (zero-order) consumption, Michelis-Mente

FRET Modulated Signaling: A Versatile Strategy to Construct Photoelectrochemical Microsensors for In Vivo Analysis.

Microelectrode-based electrochemical (EC) and photoelectrochemical (PEC) sensors are promising candidates for in vivo analysis of biologically important chemicals. However, limited selectivity in complicated biological systems and poor adaptability to electrochemically non-active species restrained their applications. Herein, we propose the concept of modulating the PEC output by a fluorescence resonance energy transfer (FRET) process. The emission of energy donor was dependent on the concentration of target SO2 , which in turn served as the modulator of the photocurrent signal of the photoact

A combined dielectrophoresis, traveling wave dielectrophoresis and electrorotation microchip for the manipulation and characterization of human malignant cells.

The study of the dielectric properties of micrometer- or nanometer-scale particles is of particular interest in present-day applications of biomedical engineering. Electrokinetics utilises electrically energised microelectrode structures within microfluidic chambers to noninvasively probe the physiological structure of live cancer cells. A system is described that combines the three complementary techniques of dielectrophoresis (DEP), travelling wave dielectrophoresis (TWD) and electrorotation (ROT) for the first time on a single, integrated chip (3 x 6 mm). The chip employs planar microelectr
