Zebrafish Zic2a and Zic2b regulate neural crest and craniofacial development.

Holoprosencephaly (HPE), the most common malformation of the human forebrain, is associated with defects of the craniofacial skeleton. ZIC2, a zinc-finger transcription factor, is strongly linked to HPE and to a characteristic set of dysmorphic facial features in humans. We have previously identified important functions for zebrafish Zic2 in the developing forebrain. Here, we demonstrate that ZIC2 orthologs zic2a and zic2b also regulate the forming zebrafish craniofacial skeleton, including the jaw and neurocranial cartilages, and use the zebrafish to study Zic2-regulated processes that may co

Morphology of experimental spina bifida in the chick embryo.

Open malformations of the central nervous system may involve the brain or spinal cord, or both. Preliminary experiments in which a window was cut in the shell overlying early chick embryos (with removal of 2 ml of albumen) produced a range of neural and non-neural malformations. Exposure of Stage 5--10 embryos at 26 hours of incubation produced open brain and cord defects. Embryos were recovered at 11--12 days for gross examination. Open cord defects in 12 day experimental embryos could be divided morphologically into 2 types. One group showed an everted symmetrical plaque of neural tissue. In

Simple generation of neurons from human embryonic stem cells using agarose multiwell dishes.

Human embryonic stem cells (hESC) are potentially an unlimited source of neurons for study and therapy for human disease. Directed differentiation of hESC has been performed using many different methods, often via neural precursor intermediates generated from aggregates of hESC. We describe here a protocol based on commercially available reusable silicone micromolds and two small molecule growth factor inhibitors to simply and reproducibly generate human neurons from hESC. Hundreds of neurospheres were generated with a single pipettation of hESC into agarose multiwell plates made with the micr

Degenerative spondylolisthesis and degenerative scoliosis treated with a combination segmental rod-plate and transpedicular screw instrumentation system: a preliminary report.

Fourteen patients with degenerative spondylolisthesis and three patients with degenerative scoliosis, all of whom experienced low-back pain, lumbar radiculopathy, and/or intermittent claudication were treated with posterolateral fusion and correction of deformities using a new instrumentation system. This new spinal fixation system combines the advantages of a rod for scoliotic deformities and a plate for sagittal plane disorders. The combination rod-plates can apply multiple forces to facilitate correction of complex deformities as well as enable indirect neurologic decompression. The system

Multiple roles for Hedgehog signaling in zebrafish pituitary development.

The endocrine-secreting lobe of the pituitary gland, or adenohypophysis, forms from cells at the anterior margin of the neural plate through inductive interactions involving secreted morphogens of the Hedgehog (Hh), fibroblast growth factor (FGF), and bone morphogenetic protein (BMP) families. To better understand when and where Hh signaling influences pituitary development, we have analyzed the effects of blocking Hh signaling both pharmacologically (cyclopamine treatments) and genetically (zebrafish Hh pathway mutants). While current models state that Shh signaling from the oral ectoderm pat

Animal models for human craniofacial malformations.

Holoprosencephaly malformations, of which the fetal alcohol syndrome appears to be a mild form, can result from medial anterior neural plate deficiencies as demonstrated in an ethanol treated animal model. These malformations are associated with more medial positioning of the nasal placodes and resulting underdevelopment or absence of the medial nasal prominences (MNPs) and their derivatives. Malformations seen in the human retinoic acid syndrome (RAS) can be produced by administration of the drug 13-cis-retinoic acid in animals. Primary effects on neural crest cells account for most of these

Primary culture of neural cells isolated from the cerebellum of newborn and adult mucopolysaccharidosis type IIIA mice.

In order to evaluate the mechanisms leading to neuropathology in Mucopolysaccharidosis type IIIA (MPS-IIIA, Sanfilippo syndrome), we have harvested and cultured primary neural cells isolated from the cerebellum of newborn and adult MPS-IIIA and unaffected mice. Cell viability and plating efficiency were comparable for brain tissue obtained from either newborn or adult MPS-IIIA and unaffected mice. Cultures (newborn and adult) comprised a mixed brain cell population including astrocytes, oligodendrocytes, and neurons. Newborn MPS-IIIA cells contained inclusions and vacuoles consistent with the

Generation of Spontaneously Beating Neuromyocardial Organoids with Neural Crest-Derived Proprioceptive-Biased Neurons and Real-Time Respirometry.

The neural crest gives rise to diverse cell types, including the proprioceptive sensory neurons that relay limb and organ position to the central nervous system. Studying how these neurons interact with their peripheral targets in a human context has been constrained by the lack of integrated in vitro models that combine neural crest derivatives with functional effector tissue. Here, we present a protocol for generating spontaneously beating neuromyocardial organoids from human pluripotent stem cells (hPSCs), incorporating neural crest-derived proprioceptive-biased neurons. Differentiation pro

Morphogenesis of the cranial segments and distribution of neural crest in the embryos of the snapping turtle, Chelydra serpentina.

Recent studies of the heads of vertebrates have shown a primitive pattern of segmentation in the mesoderm and neural plate not previously recognized. The role of this pattern in the subsequent distribution of cranial crest and the development of branchial arches and cranial nerves, may resolve century-old arguments about the evolution of vertebrate segmentation. In this study, we examine the early embryonic development of the cranium of a primitive amniote, the snapping turtle, with the SEM. We show that the paraxial mesoderm cranial to the first-formed somites is segmented and that this patte

Machine learning-based analysis of heat and mass transfer in casson hybrid nanofluid flow over a variable porous riga plate.

This study uses an artificial neural network (ANN) architecture in combination with the Levenberg-Marquardt backpropagation (LMBP) method to investigate the impact of heat radiation on the hybrid nanofluid flow through a variable porous medium beneath a stretchy Riga plate. The purpose of this work is driven by the increasing demand for accurately modeling complex nonlinear thermal systems involving electromagnetic control, porous media, and nanofluids, which are not sufficiently covered in the existing literature. Single-walled carbon nanotubes (SWCNTs) and multiwalled carbon nanotubes (MWCNT

Anterior decompression and plate fixation in fracture dislocations of the lower cervical spine.

Surgical treatment of unstable traumatic injuries of the cervical spine can be carried out by a posterior or anterior approach, with different advantages and disadvantages. Twenty patients were treated with anterior decompression, interbody fusion with autogenous iliac bone graft, and osteosynthesis with a Louis anterior plate. The screws were inserted in the vertebral body without reaching the posterior vertebral wall. There were 18 male and 2 female patients, aged between 18 and 66 years (average 36 years). The osteoarticular lesion was in 8 cases a tear-drop fracture and in 12 a fracture-di

Specific and redundant functions of Gli2 and Gli3 zinc finger genes in skeletal patterning and development.

The correct patterning of vertebrate skeletal elements is controlled by inductive interactions. Two vertebrate hedgehog proteins, Sonic hedgehog and Indian hedgehog, have been implicated in skeletal development. During somite differentiation and limb development, Sonic hedgehog functions as an inductive signal from the notochord, floor plate and zone of polarizing activity. Later in skeletogenesis, Indian hedgehog functions as a regulator of chondrogenesis during endochondral ossification. The vertebrate Gli zinc finger proteins are putative transcription factors that respond to Hedgehog signa

Whole-brain input architecture of primary and secondary somatosensory cortices in mice.

The primary and secondary somatosensory cortices (S1 and S2) play crucial roles in processing sensory inputs from various body regions, encompassing tactile, pressure, thermal, and nociceptive stimuli. These cortices are anatomically distinct, with S1 primarily involved in mechanical and cold stimulus discrimination and S2 in the interpretation of mechanical and thermal inputs, particularly in pain perception. However, the upstream innervation patterns of the somatosensory system remain less explored. In this study, we employed a modified rabies virus (RV)-mediated transsynaptic retrograde tra

A nanofibrous polycaprolactone/collagen neural guidance channel filled with sciatic allogeneic schwann cells and platelet-rich plasma for sciatic nerve repair.

Sciatic nerve damage, a common condition affecting approximately 2.8% of the US population, can lead to significant disability due to impaired nerve signal transmission, resulting in loss of sensation and motor function in the lower extremities. In this study, a neural guidance channel was developed by rolling a nanofibrous scaffold produced via electrospinning. The scaffold's microstructure, biocompatibility, biodegradation rate, porosity, mechanical properties, and hemocompatibility were evaluated. Platelet-rich plasma (PRP) activated with 30,000 allogeneic Schwann cells (SCs) was injected i

Regulation of autoimmune-mediated neuroinflammation by endothelial cells.

The CNS has traditionally been considered an immune-privileged organ, but recent studies have identified a plethora of immune cells in the choroid plexus, meninges, perivascular spaces, and cribriform plate. Although those immune cells are crucial for the maintenance of CNS homeostasis and for neural protection against infections, they can lead to neuroinflammation in some circumstances. The blood and the lymphatic vasculatures exhibit distinct structural and molecular features depending on their location in the CNS, greatly influencing the compartmentalization and the nature of CNS immune res

The issue of the multipotency of the neural crest cells.

In the neural primordium of vertebrate embryos, the neural crest (NC) displays a unique character: the capacity of its component cells to leave the neural primordium, migrate along definite (and, for long, not identified) routes in the developing embryo and invade virtually all tissues and organs, while producing a large array of differentiated cell types. The most striking diversity of the NC derivatives is found in its cephalic domain that produces, not only melanocytes and peripheral nerves and ganglia, but also various mesenchymal derivatives (connective tissues, bones, cartilages…) which,

[Development of the isocortex of the human brain during the embryonic and fetal periods up to the 24th week of gestation].

From 98 specimens (Pr YAKOVLEV's Collection) we have been able to define the cytoarchitectonic criteria of the different stages of the development of the human Isocortex, and to stress some important features of the organogenesis of the human brain. Most of those findings are in agreement with G.I. POLIAKOV's publications on the cerebral development. Schematically it is possible to individualise three periods made of several stages: - The first one, from the 4th to the 6th week of Gestation during which the primitive neural epithelium, with its four layers (layers of His), is built up. - The s

Quantitative Analysis of Cell and Tissue Shape During Mouse Cranial Neural Tube Closure.

Neural tube closure is a critical process that transforms the neural plate, an open epithelial tissue, into the closed tube that serves as the structural basis of the central nervous system. Defects in this process are among the most common and severe developmental diseases in the human population, with failures in cranial closure accounting for approximately one-third of total defects. However, the cell and tissue mechanisms that drive cranial closure remain opaque relative to the better studied process of spinal closure, in large part due to the unique challenges in characterizing cranial ti
