Species-specific divergence in Phytophthora infestans resistance between Nicotiana benthamiana and potato mediated by 1-aminocyclopropane-1-carboxylic acid pretreatment.

BACKGROUND: Late blight, caused by Phytophthora infestans, is a devastating oomycete disease threatening global potato (Solanum tuberosum) production. 1-Aminocyclopropane-1-carboxylic acid (ACC), has also been proposed to act as a bioactive signal in plant stress responses, but its contribution to plant immunity remains elusive. RESULTS: Here, we show that ACC pretreatment exerts opposite effects on P. infestans resistance in Nicotiana benthamiana and the potato cultivar 'Favorita', revealing strong species-specific transcriptional reprogramming. In N. benthamiana, differentially expressed ge

AFM-YOLOv8s: An Accurate, Fast, and Highly Robust Model for Detection of Sporangia of Plasmopara viticola with Various Morphological Variants.

Monitoring spores is crucial for predicting and preventing fungal- or oomycete-induced diseases like grapevine downy mildew. However, manual spore or sporangium detection using microscopes is time-consuming and labor-intensive, often resulting in low accuracy and slow processing speed. Emerging deep learning models like YOLOv8 aim to rapidly detect objects accurately but struggle with efficiency and accuracy when identifying various sporangia formations amidst complex backgrounds. To address these challenges, we developed an enhanced YOLOv8s, namely, AFM-YOLOv8s, by introducing an Adaptive Cro

Breaking-Down Barriers: Proposal of Using Cellulose Biosynthesis Inhibitors and Cellulase Enzyme as a Novel Treatment Modality for Vision Threatening Pythium Insidiosum Keratitis.

Pythium insidiosum, an Oomycete, causes severe keratitis that endangers vision. Its clinical, morphological, and microbiological characteristics are often indistinguishable from those of fungal keratitis, earning it the moniker "parafungus". Distinctive clinical hallmarks that set it apart from other forms of keratitis include radial keratoneuritis, tentacles, marginal infiltration, and a propensity for rapid limbal spread. The therapeutic approach to Pythium keratitis (PK) has long been a subject of debate, and topical and systemic antifungals and antibacterials have been tried with limited s

Comparison of systemic and local interactions between the arbuscular mycorrhizal fungus Funneliformis mosseae and the root pathogen Aphanomyces euteiches in Medicago truncatula.

It has been shown in a number of pathosystems that arbuscular mycorrhizal (AM) fungi confer resistance against root pathogens, including in interactions between Medicago truncatula and the root rot-causing oomycete Aphanomyces euteiches. For the current study of these interactions, a split root system was established for plant marker gene analysis in order to study systemic defense responses and to compare them with local interactions in conventional pot cultures. It turned out, however, that split root systems and pot cultures were in different physiological stages. Genes for pathogenesis-rel

Abscisic acid has a key role in modulating diverse plant-pathogen interactions.

We isolated an activation-tagged Arabidopsis (Arabidopsis thaliana) line, constitutive disease susceptibility2-1D (cds2-1D), that showed enhanced bacterial growth when challenged with various Pseudomonas syringae strains. Systemic acquired resistance and systemic PATHOGENESIS-RELATED GENE1 induction were also compromised in cds2-1D. The T-DNA insertion adjacent to NINE-CIS-EPOXYCAROTENOID DIOXYGENASE5 (NCED5), one of six genes encoding the abscisic acid (ABA) biosynthetic enzyme NCED, caused a massive increase in transcript level and enhanced ABA levels >2-fold. Overexpression of NCED genes re

Overexpression of Arabidopsis MAP kinase kinase 7 leads to activation of plant basal and systemic acquired resistance.

There is a growing body of evidence indicating that mitogen-activated protein kinase (MAPK) cascades are involved in plant defense responses. Analysis of the completed Arabidopsis thaliana genome sequence has revealed the existence of 20 MAPKs, 10 MAPKKs and 60 MAPKKKs, implying a high level of complexity in MAPK signaling pathways, and making the assignment of gene functions difficult. The MAP kinase kinase 7 (MKK7) gene of Arabidopsis has previously been shown to negatively regulate polar auxin transport. Here we provide evidence that MKK7 positively regulates plant basal and systemic acquir

Altering expression of benzoic acid/salicylic acid carboxyl methyltransferase 1 compromises systemic acquired resistance and PAMP-triggered immunity in arabidopsis.

Methyl salicylate (MeSA), which is synthesized in plants from salicylic acid (SA) by methyltransferases, has roles in defense against microbial and insect pests. Most of the MeSA that accumulates after pathogen attack is synthesized by benzoic acid/SA carboxyl methyltransferase 1 (AtBSMT1). To investigate the role of AtBSMT1 in plant defense, transgenic Arabidopsis with altered AtBSMT1 function or expression were assessed for their ability to resist pathogen infection. A knockout mutant (Atbsmt1) failed to accumulate MeSA following pathogen infection; these plants also failed to accumulate SA

A novel Phytophthora-specific putative lipid-binding protein triggers plant immunity.

Plants recognize conserved pathogen-derived molecules to activate pattern-triggered immunity (PTI), yet many secreted proteins from oomycete pathogens remain functionally uncharacterized. Here, we identified PcCF16, a previously uncharacterized secreted protein from Phytophthora capsici, through activity-guided fractionation of the pathogen secretome. PcCF16 induced cell death and PTI-associated gene expression in Nicotiana benthamiana, and purified recombinant PcCF16 triggered reactive oxygen species production across multiple plant species. PcCF16-induced cell death was independent of the co

Effects of Phytophthora species on seed germination and seedling survival and performance in five sweet chestnut varieties.

The oomycete genus Phytophthora includes numerous pathogens, some causing severe tree diseases. In forestry, their impact is usually assessed on adult trees, while their effects on seed germination and seedling development-crucial phases in forest regeneration-remain poorly investigated. Here, we tested the effects of the two main causal agents of ink disease in sweet chestnut, P. cinnamomi and P. x cambivora, along with three other Phytophthora species commonly found in European sweet chestnut forests (P. plurivora, P. cactorum, P. cryptogea), on seed germination and seedling development of f

Plant essential oils against woody plant fungal pathogens: chemical composition, antifungal mechanisms, and translational challenges.

Woody plants are threatened by fungal and oomycete pathogens that cause root and wood rots, cankers, vascular wilts, dieback, fruit decay, and other diseases. Meanwhile, intensive production systems and repeated fungicide use have raised concerns about resistance, environmental persistence, and non-target effects. Essential oils (EOs) have emerged as promising natural antimicrobial resources, yet their effectiveness depends strongly on chemical composition, pathogen susceptibility, concentration, formulation, and application strategy. This review synthesizes current evidence on EO-based manage

Molecular Targets and Trait Innovation in CRISPR-Edited Vegetable Crops: An Up-to-Date Review (2020-2026).

Vegetable crops are of fundamental nutritional and economic importance worldwide, yet their improvement through conventional breeding remains time consuming and constrained by genomic complexity. For that reason, over the period 2020-2026, CRISPR/Cas-mediated genome editing, a new genomic technique (NGT), has emerged as the leading tool for precise, rapid and targeted modification of vegetable crop genomes. In this review, we summarize selected peer-reviewed studies on CRISPR/Cas applications across a broad range of vegetable species. Tomato (Solanum lycopersicum) and potato (Solanum tuberosum

High variation of virulence in Aphanomyces astaci strains lacks association with pathogenic traits and mtDNA haplogroups.

Introduced into Europe from North America 150 years ago alongside its native crayfish hosts, the invasive pathogen Aphanomyces astaci is considered one of the main causes of European crayfish population decline. For the past two centuries, this oomycete pathogen has been extensively studied, with the more recent efforts focused on containing and monitoring its spread across the continent. However, after the recent introduction of new strains, the newly-discovered diversity of A. astaci in North America and several years of coevolution with its European host, a new assessment of the traits link

Linking Comparative Genomics of Nine Potato-Associated Pseudomonas Isolates With Their Differing Biocontrol Potential Against Late Blight.

For plants, the advantages of associating with beneficial bacteria include plant growth promotion, reduction of abiotic and biotic stresses and enhanced protection against various pests and diseases. Beneficial bacteria rightly equipped for successful plant colonization and showing antagonistic activity toward plant pathogens seem to be actively recruited by plants. To gain more insights into the genetic determinants responsible for plant colonization and antagonistic activities, we first sequenced and de novo assembled the complete genomes of nine Pseudomonas strains that had exhibited varyin

Contribution of Hydrogen Cyanide to the Antagonistic Activity of Pseudomonas Strains Against Phytophthora infestans.

Plants face many biotic and abiotic challenges in nature; one of them is attack by disease-causing microbes. Phytophthora infestans, the causal agent of late blight is one of the most prominent pathogens of the potato responsible for multi-billion-dollar losses every year. We have previously reported that potato-associated Pseudomonas strains inhibited P. infestans at various developmental stages. A comparative genomics approach identified several factors putatively involved in this anti-oomycete activity, among which was the production of hydrogen cyanide (HCN). Here, we report the relative c

Deep RNA-Seq profile reveals biodiversity, plant-microbe interactions and a large family of NBS-LRR resistance genes in walnut (Juglans regia) tissues.

Deep RNA-Seq profiling, a revolutionary method used for quantifying transcriptional levels, often includes non-specific transcripts from other co-existing organisms in spite of stringent protocols. Using the recently published walnut genome sequence as a filter, we present a broad analysis of the RNA-Seq derived transcriptome profiles obtained from twenty different tissues to extract the biodiversity and possible plant-microbe interactions in the walnut ecosystem in California. Since the residual nature of the transcripts being analyzed does not provide sufficient information to identify the e

Induced systemic resistance in Arabidopsis thaliana in response to root inoculation with Pseudomonas fluorescens CHA0.

Root inoculation of Arabidopsis thaliana ecotype Columbia with Pseudomonas fluorescens CHA0r partially protected leaves from the oomycete Peronospora parasitica. The molecular determinants of Pseudomonas fluorescens CHA0r for this induced systemic resistance (ISR) were investigated, using mutants derived from strain CHA0: CHA400 (pyoverdine deficient), CHA805 (exoprotease deficient), CHA77 (HCN deficient), CHA660 (pyoluteorin deficient), CHA631 (2,4-diacetylphloroglucinol [DAPG] deficient), and CHA89 (HCN, DAPG- and pyoluteorin deficient). Only mutations interfering with DAPG production led to

The sulfated laminarin triggers a stress transcriptome before priming the SA- and ROS-dependent defenses during grapevine's induced resistance against Plasmopara viticola.

Grapevine (Vitis vinifera) is susceptible to many pathogens which cause significant losses to viticulture worldwide. Chemical control is available, but agro-ecological concerns have raised interest in alternative methods, especially in triggering plant immunity by elicitor treatments. The β-glucan laminarin (Lam) and its sulfated derivative (PS3) have been previously demonstrated to induce resistance in grapevine against downy mildew (Plasmopara viticola). However, if Lam elicits classical grapevine defenses such as oxidative burst, pathogenesis-related (PR)-proteins and phytoalexin production

A mutation in a coproporphyrinogen III oxidase gene confers growth inhibition, enhanced powdery mildew resistance and powdery mildew-induced cell death in Arabidopsis.

A gene encoding a coproporphyrinogen III oxidase mediates disease resistance in plants by the salicylic acid pathway. A number of genes that regulate powdery mildew resistance have been identified in Arabidopsis, such as ENHANCED DISEASE RESISTANCE 1 to 3 (EDR1 to 3). To further study the molecular interactions between the powdery mildew pathogen and Arabidopsis, we isolated and characterized a mutant that exhibited enhanced resistance to powdery mildew. The mutant also showed dramatic powdery mildew-induced cell death as well as growth defects and early senescence in the absence of pathogens.

The expression of the t-SNARE AtSNAP33 is induced by pathogens and mechanical stimulation.

The fusion of vesicles in the secretory pathway involves the interaction of t-soluble N-ethylmaleimide-sensitive factor attachment protein receptors (t-SNAREs) on the target membrane and v-SNAREs on the vesicle membrane. AtSNAP33 is an Arabidopsis homolog of the neuronal t-SNARE SNAP-25 involved in exocytosis and is localized at the cell plate and at the plasma membrane. In this paper, the expression of AtSNAP33 was analyzed after different biotic and abiotic stresses. The expression of AtSNAP33 increased after inoculation with the pathogens Plectosporium tabacinum and virulent and avirulent f
