An optically accessible two-chamber perfusion bioreactor for label-free metabolic imaging of three-dimensional tissue models under dynamic flow conditions.

Bioreactors play an important role in tissue engineering as they tackle challenges that arise when developing three-dimensional (3D) tissue models. Bioreactor design is a complex task due to the combination of technology-specific and tissue-specific requirements, with general requirements like biocompatibility and sterility. Here we present a novel design for a two-chamber perfusion bioreactor that provides optical accessibility on both the apical and basal surfaces of 3D tissue models based on small intestine submucosa (SIS) scaffolds, while also enabling tissue culture under dynamic flow con

Investigation of new flow modifying endovascular image-guided interventional (EIGI) techniques in patient-specific aneurysm phantoms (PSAPs) using optical imaging.

Effective minimally invasive treatment of cerebral bifurcation aneurysms is challenging due to the complex and remote vessel morphology. An evaluation of endovascular treatment in a phantom involving image-guided deployment of new asymmetric stents consisting of polyurethane patches placed to modify blood flow into the aneurysm is reported. The 3D lumen-geometry of a patient-specific basilar-artery bifurcation aneurysm was derived from a segmented computed-tomography dataset. This was used in a stereolithographic rapid-prototyping process to generate a mold which was then used to create any nu

Comparison of Stent Expansion Guided by Optical Coherence Tomography Versus Intravascular Ultrasound: The ILUMIEN II Study (Observational Study of Optical Coherence Tomography [OCT] in Patients Undergoing Fractional Flow Reserve [FFR] and Percutaneous Coronary Intervention).

OBJECTIVES: The present study sought to determine whether optical coherence tomography (OCT) guidance results in a degree of stent expansion comparable to that with intravascular ultrasound (IVUS) guidance. BACKGROUND: The most important predictor of adverse outcomes (thrombosis and restenosis) after stent implantation with IVUS guidance is the degree of stent expansion achieved. METHODS: We compared the relative degree of stent expansion (defined as the minimal stent area divided by the mean of the proximal and distal reference lumen areas) after OCT-guided stenting in patients in the ILUMI

KCl-induced cortical spreading depression waves more heterogeneously propagate than optogenetically-induced waves in lissencephalic brain: an analysis with optical flow tools.

Although cortical spreading depolarizations (CSD) were originally assumed to be homogeneously and concentrically propagating waves, evidence obtained first in gyrencephalic brains and later in lissencephalic brains suggested a rather non-uniform propagation, shaped heterogeneously by factors like cortical region differences, vascular anatomy, wave recurrences and refractory periods. Understanding this heterogeneity is important to better evaluate the experimental models on the mechanistics of CSD and to make appropriate clinical estimations on neurological disorders like migraine, stroke, and

High-speed optical imaging of afferent flow through rat olfactory bulb slices: voltage-sensitive dye signals reveal periglomerular cell activity.

Fast, multiple-site optical recording and video imaging techniques were combined to visualize the olfactory processing stream as it flowed through rat olfactory bulb slices stained with the voltage-sensitive dye RH155. A 464 element photodiode detector array was used to record the voltage-sensitive dye signals. Focal electrical stimulation of the olfactory nerve layer evoked relatively large optical responses in the olfactory nerve and glomerular layers but only small responses within the external plexiform layer. With paired-pulse stimulation, glomerular attenuation was evident in signals rec

Optical coherence tomography: Translation from 3D-printed vascular models of the anterior cerebral circulation to the first human images of implanted surface modified flow diverters.

BACKGROUND: The new generation of flow diverters includes a surface modification with a synthetic biocompatible polymer, which makes the device more biocompatible and less thrombogenic. Optical coherence tomography (OCT) can be used to visualize perforators, stent wall apposition, and intra-stent thrombus. Unfortunately real world application of this technology has been limited because of the limited navigability of these devices in the intracranial vessels. In this report, we share our experience of using 3D-printed neurovascular anatomy models to simulate and test the navigability of a comme

Impact of ophthalmic draping on end-tidal CO₂, cerebral oximetry, and optic nerve sheath diameter in cataract surgery: an observational comparison by COVID-19 pneumonia history with randomized oxygen flow allocation.

BACKGROUND: Ophthalmic draping during cataract surgery creates an isolated surgical field but may lead to carbon dioxide (CO₂) accumulation beneath the drape. Elevated CO₂ can cause hypercapnia, cerebral vasodilation, and increased intracranial pressure, potentially reflected by changes in cerebral oximetry and optic nerve sheath diameter (ONSD). Patients with a history of COVID-19 pneumonia may exhibit residual pulmonary dysfunction that influences these parameters during surgery. This study aimed to investigate the effects of ophthalmic draping on end-tidal CO₂, cerebral oximetry, and ONSD i

Fractional flow reserve-guided coronary angioplasty using paclitaxel-coated balloons without stent implantation: feasibility, safety and 6-month results by angiography and optical coherence tomography.

BACKGROUND: Percutaneous coronary interventions (PCI) with drug-coated balloons (DCB) might be a promising trade-off between balloon angioplasty and drug-eluting stents, since DCB inhibit neointimal proliferation and limit duration of dual antiplatelet therapy. We investigated the safety, feasibility, and 6-month results of fractional flow reserve (FFR)-guided use of the paclitaxel-coated SeQuent Please® balloon without stenting for elective PCI of de novo lesions. Impact of thin-cap fibroatheroma on predicting deteriorated coronary flow during interventional procedures in acute as well as stable coronary syndromes: insights from optical coherence tomography analysis.

The occurrence of deteriorated coronary flow associated with distal embolization during percutaneous coronary intervention results in impaired myocardial perfusion and worsens the clinical prognosis. This study aimed to examine the impact of optical coherence tomography (OCT)-determined coronary plaque morphology on the prediction of deteriorated coronary flow after stent implantation in acute as well as stable coronary syndromes (ACS and SAP, respectively). We studied 126 patients who underwent OCT during stenting for ACS (n = 44) and SAP (n = 82) with a de novo lesion. Angiographic deteriora

Overestimation of Automated Platelet Counts by Blast Fragments in Acute Hypercellular Leukemias: A Retrospective Study Comparing Impedance, Optical (PLT-O), Fluorescent (PLT-F), and CD41/CD61 Flow Cytometry Methods.

INTRODUCTION: Acute leukemias with hyperleukocytosis (> 100 × 109/L) can produce cytoplasmic blast fragments that interfere with platelet counts, notably in impedance-based methods, potentially masking severe thrombocytopenia and increasing hemorrhagic risk. While fluorescent platelet counting (PLT-F) is promoted as platelet-specific, its accuracy in the presence of blast fragments remains uncertain. METHODS: This retrospective study analyzed 269 blood samples from 87 patients with hypercellular acute leukemia. Blast fragments were identified on blood smears. Platelet counts by impedance were
