Taurine (2-aminoethanesulfonic acid) is the most abundant free amino acid in mammalian cells. It plays a significant role in cell development, nutrition, and survival, such as in the regulation of ion transport and osmoregulation. Cysteine sulfinate decarboxylase (CSD) is the rate-limiting biosynthetic enzyme of taurine. Recently, the synthesis of taurine has been observed in the central nervous system, kidney, liver, and muscle. However, the synthesis of taurine in the salivary glands has still not been described in detail. We have detected CSD expression in the major salivary glands of adult

Serratia marcescens is a Gram-negative enterobacterium that has become an important opportunistic pathogen, largely due to its high degree of natural antibiotic resistance. One factor contributing to this natural antibiotic resistance is reduced outer membrane permeability, which is controlled in part by OmpC and OmpF porin proteins. OmpF expression is regulated by micF, an RNA transcript encoded upstream of the ompC gene, which hybridizes with the ompF transcript to inhibit its translation. Regulation of S. marcescens porin gene expression, as well as that of micF, was investigated using beta

Many organisms, including higher plants, accumulate free proline (Pro) in response to osmotic stress. Although various studies have focused on the ability of Pro as a compatible osmolyte involved in osmotolerance, its specific role throughout plant growth is still unclear. It has been reported that Pro is synthesized from Glu catalyzed by a key enzyme, delta 1-pyrroline-5-carboxylate synthetase (P5CS), in plants. To elucidate essential roles of Pro, we generated antisense transgenic Arabidopsis plants with a P5CS cDNA. Several transgenics accumulated Pro at a significantly lower level than wil

OBJECTIVE: To provide an overview of diagnostic and treatment strategies in pregnant patients with diabetes insipidus (DI).
METHODS: We review the changes in osmoregulation during normal pregnancy, characterize the various types of DI that can occur during pregnancy, and discuss the recommended management.
RESULTS: The incidence of DI is 2 to 4 cases per 100,000 gestations. Central DI can precede pregnancy or manifest initially during gestation. With preexistent central DI, pregnancy usually aggravates the disorder, and the requirements for antidiuretic hormone (ADH) usually increase. Such a

We have examined the occurrence of acetylcholinesterase (AChE)-positive cells in the pineal organ of different developmental stages of the Pacific coho salmon. Large numbers of AChE cells were present in freshwater living alevins, in all stages of presmolts (n = 307-544), and in adult spawners (n = 696-1774), whereas seawater-living postmolts displayed a total lack of labeled cells. The AChE-reactive cells were evenly distributed within the pineal end-vesicle and stalk of the presmolts and adults. However, the AChE-positive cells that occurred in the pineal stalk were of a smaller type and mor

Saline-alkaline fishes maintain internal homeostasis through effective intestinal ion and acid-base regulation. However, the specific intestinal epithelial cell populations mediating these functions remain poorly characterized. Here, we performed single-cell RNA sequencing on 23,909 mid-intestinal cells from juvenile Gymnocypris przewalskii, a fish native to the saline-alkaline Lake Qinghai, to generate a cell atlas of the mid-intestine. Several major cell classes were identified, including epithelial cells (ECs), enteroendocrine cells (EECs), goblet cells (GCs), and leukocytes (LCs), each exh

Cyclic AMP (cAMP) signaling is crucial for environmental sensing and response to stress conditions in trypanosomatids. However, the mechanisms driving the specificity of cAMP signals remain poorly understood in these protozoan parasites. We recently identified two putative cAMP microdomains in Trypanosoma cruzi, the causative agent of Chagas disease. Here, considering the localization of three phosphodiesterases, PDEC at the contractile vacuole complex (CVC), and PDEB1 and PDEB2 along the flagellum, we modulated their expression to functionally characterize the flagellar tip (FT) and the CVC a

Sharks and rays present an osmoregulatory mechanism essentially exercised by a rectal salt gland. Histological assessments of this gland, however, are notoriously lacking. In this sense, histological assessments of the rectal gland of the Brazilian guitarfish, Pseudobatos horkelii obtained from off the coast of Rio de Janeiro, Brazil, were carried out herein. Rectal gland samples were histologically processed with hematoxylin/eosin and special Masson's Trichrome stain. Three main regions were identified: the capsule, secretory parenchyma and central duct. Highly vascularized connective tissue

Measurement of adrenal hormones in amphibians is important in studies on stress, development, osmoregulation, endocrine disruption, and conservation to help reveal mechanisms within amphibians and evolution among vertebrates. Corticosteroids measured in frogs are typically corticosterone and/or aldosterone, but steroid intermediates that may activate hormone receptors are thus far not quantified. Also, steroidogenesis in frogs has been examined in vitro but little work has been done using tadpoles with mutations affecting steroidogenesis. To advance such studies, we developed a comprehensive l

Polyamines, γ-aminobutyric acid (GABA), proline, and glycine betaine (GB) are important plant stress metabolites that are interconnected in common metabolic cycles. These compounds have a multifunctional protective effect on plant cells under stress conditions, acting as osmoregulators, antioxidants, chaperones, and intracellular pH stabilizers, among other roles. The key role of these compounds, however, lies in their involvement in the signaling network of plant cells. Stress metabolites, in particular, engage in a complex functional interaction with reactive oxygen species (ROS) and nitric

The gastroenteropancreatic system in elasmobranchs presents a number of firsts in the evolutionary history of vertebrates. This group of fishes were the first to have evolved an acid-pepsin-secreting stomach and demonstrate a glandular pancreas with exocrine and endocrine cells. They additionally were the first to have evolved a well-developed spiral or scroll-like intestine in comparison to the precursor typhlosole - an infolding of the intestinal wall - found in lampreys. Coupled with these features, the unusual metabolic strategy of elasmobranchs is characterized by a lack of reliance on fa

Dry eye disease (DED) is a multifactorial disorder of the lacrimal system and ocular surface, characterized by a deficiency in the quality and/or quantity of the tear fluid. The multifactorial nature of DED encompasses a number of interconnected underlying pathologies, including loss of homeostasis, instability and hyperosmolarity of the tears, and the induction and propagation of detrimental inflammatory responses in the eyes, which finally results in the development of neurosensory dysfunction and visual disruption. Dryness, grittiness, scratchiness, discomfort, inflammation, burning, wateri

Serum albumin (SA) exists in relatively high concentrations, in close contact with most cells. However, in the adult brain, except for cerebrospinal fluid (CSF), SA concentration is relatively low. It is mainly produced in the liver to serve as the main protein of the blood plasma. In the plasma, it functions as a carrier, chaperon, antioxidant, source of amino acids, osmoregulator, etc. As a carrier, it facilitates the stable presence and transport of the hydrophobic and hydrophilic molecules, including free fatty acids, steroid hormones, medicines, and metal ions. As a chaperon, SA binds to

Combined pituitary hormone deficiency (CPHD) is a genetically heterogeneous disorder caused by mutations in over 30 genes. The loss-of-function mutations in many of these genes, including orthodenticle homeobox 2 (OTX2), can present with a broad range of clinical symptoms, which provides a challenge for predicting phenotype from genotype. Another challenge in human genetics is functional evaluation of rare genetic variants that are predicted to be deleterious. Zebrafish are an excellent vertebrate model for evaluating gene function and disease pathogenesis, especially because large numbers of

High osmolarity, bound water, and hydrostatic pressure contribute to notochord mechanics and its morphogenesis into the nucleus pulposus (NP) compartment of the intervertebral disc. Indeed, the osmoadaptive transcription factor, nuclear factor of activated T-cells 5 (NFAT5 aka TonEBP), is robustly expressed by resident cells of the notochord and NP. Nevertheless, the molecular mechanisms that drive notochord osmoregulation and the functions of NFAT5 in disc embryogenesis remain largely unexplored. In this study, we show that deletion of NFAT5 in mice results in delayed vertebral column develop

Select species of the bacterial genus Leptospira are causative agents of leptospirosis, an emerging global zoonosis affecting nearly one million people worldwide annually. We examined two Leptospira pathogens, Leptospira interrogans serovar Lai str. 56601 and Leptospira borgpetersenii serovar Hardjo-bovis str. L550, as well as the free-living leptospiral saprophyte, Leptospira biflexa serovar Patoc str. 'Patoc 1 (Ames)'. The transport proteins of these leptospires were identified and compared using bioinformatics to gain an appreciation for which proteins may be related to pathogenesis and sap

Aldose reductase enzyme (ALR2) of the polyol metabolic pathway, apart from its role as detoxifying enzyme towards toxic aldehydes, osmoregulator in the kidney and regulator of sperm maturation, was first found to be implicated in the etiology of the long term diabetic complications. However, to date, emerging reports have suggested that under normal glucose concentration, ALR2 may be up-regulated by factors other than hyperglycemia and therefore be involved also in other pathological processes that have become major threats to human health in the 21(st) century. Such pathologies are a number o

Individual and topographical variation in the metabolic profiles of multiple human gastrointestinal tract (GIT) biopsies have been characterized using high-resolution magic-angle spinning (HRMAS) 1H NMR spectroscopy and pattern recognition. Samples from antrum, duodenum, jejunum, ileum, and transverse colon were obtained from 8 male and 8 female participants. Each gut region generated a highly characteristic metabolic profile consistent with the varying structural and functional properties of the tissue at different longitudinal levels of the gut. The antral (stomach) mucosa contained higher l

The pufferfishes Sphoeroides testudineus and Sphoeroides greeleyi are estuarine species that osmoregulate efficiently, but S. testudineus tolerates seawater dilution to a much higher degree than S. greeleyi. This study aimed at testing whether NKCC is involved with their differential tolerance of seawater dilution, through the analysis of in vivo furosemide (NKCC inhibitor) injection both on hypo-regulation (in 35 per thousand salinity) and hyper-regulation (in 5 per thousand salinity). After exposure for 6 h or 5 days to both salinities, blood samples were obtained for determination of plasma

Atrial natriuretic peptide-(1-28) (ANP), brain natriuretic peptide-(1-32) (BNP) and C-Type natriuretic polypeptide (CNP) occur in the brain, are concentrated in the anteroventral area of the third cerebral ventricle and participate in the regulation of body fluid homeostasis. The ventricles of the mammalian brain are lined by a continuous monolayered epithelium of polyciliated ependymal cells. In the adult rat, the ependymocytes continue to express the intermediate filament vimentin, but do not contain glial fibrillary acidic protein. Ependymal functions are poorly understood, but may extend t
