Pleural macrophages differentially alter pleural mesothelial cell glycosaminoglycan production.

Glycosaminoglycans are produced in abundance by the pleural mesothelium and likely participate in the inflammatory response to pleural injury. Because intrapleural tetracycline (TCN) results in pleural macrophage influx and pleural fibrosis, this study attempted to define the role of pleural macrophage products on mesothelial glycosaminoglycan (GAG) production. Pleural macrophages were isolated 72 h after intrapleural TCN or intrapleural carrageenan (CAR), a substance that recruits pleural macrophages without producing pleural fibrosis. Macrophage cultured for 24 h produced a conditioned mediu

Deciphering the Interference of the Anti-Integrin α4β7 Vedolizumab With Cell Crossmatch.

In organ transplantation, immunological risk assessment uses cell crossmatch (XM) results, which can be altered by several drugs. We observed two recipients awaiting kidney and liver transplantation with positive flow cytometry XM (FCXM) on T-cells in the absence of Donor Specific Antibodies (DSA). Both were treated with vedolizumab (VDZ) for an inflammatory bowel disease (IBD). VDZ is an IgG1 directed against integrin α4β7, expressed on T and B cell subsets, we therefore suspected that VDZ interfered with XM. We tested with Luminex screening and single antigen assays and with auto- and allo-F

A distinctive patchy osteomalacia characterises Phospho1-deficient mice.

The phosphatase PHOSPHO1 is involved in the initiation of biomineralisation. Bones in Phospho1 knockout (KO) mice show histological osteomalacia with frequent bowing of long bones and spontaneous fractures: they contain less mineral, with smaller mineral crystals. However, the consequences of Phospho1 ablation on the microscale structure of bone are not yet fully elucidated. Tibias and femurs obtained from wild-type and Phospho1 null (KO) mice (25-32 weeks old) were embedded in PMMA, cut and polished to produce near longitudinal sections. Block surfaces were studied using 20 kV backscattered-e

Effects of image enhancement on reliability of landmark identification in digital cephalometry.

INTRODUCTION: Although digital cephalometric radiography is gaining popularity in orthodontic practice, the most important source of error in its tracing is uncertainty in landmark identification. Therefore, efforts to improve accuracy in landmark identification were directed primarily toward the improvement in image quality. One of the more useful techniques of this process involves digital image enhancement which can increase overall visual quality of image, but this does not necessarily mean a better identification of landmarks. The purpose of this study was to evaluate the effectiveness of

Eradication of Helicobacter pylori: are rifaximin-based regimens effective?

Rifaximin is a non-absorbed semisynthetic rifamycin derivative with a broad spectrum of antibacterial activity including Gram-positive and Gram-negative bacteria, both aerobes and anaerobes. Although originally developed for the treatment of infectious diarrhea, the appreciation of the pathogenic role of gut bacteria in several organic and functional gastrointestinal diseases has increasingly broadened its clinical use. Being the antibiotic active against Helicobacter pylori, even towards clarithromycin-resistant strain, and being the primary resistance very rare, several investigations explor

Diagnosis of fungal infections: current status.

Diagnosing infections remains a problem in the management of fungal diseases, particularly in the immunocompromised host. Signs and symptoms are non-specific, colonization is difficult to distinguish from invasive disease, blood cultures are commonly negative and patients are often unable to undergo invasive diagnostic procedures. This situation has led to the strategy of initiating empirical therapy in the high-risk patient. A variety of tests has been applied to several body fluids. At the simplest level, the clinician must be familiar with the appearance of various fungi in tissue. Non-cult

Functional characterization of protease-treated Bacillus anthracis protective antigen.

Characterization of the functional domains of Bacillus anthracis protective antigen (PA, 83-kDa), the common cellular binding molecule for both anthrax edema toxin and anthrax lethal toxin, is important for understanding the mechanism of entry and action of the anthrax toxins. In this study, we generated both biologically active (facilitates killing of J774A.1 cells in combination with lethal factor, LF) and inactive preparations of PA by protease treatment. Limited proteolytic digestion of PA in vitro with trypsin generated a 20-kDa fragment and a biologically active 63-kDa fragment. In contr

Normal human B cells express endogenous sheep erythrocyte (E) receptor after appropriate in vitro culture.

When normal human B cells from blood, tonsil or spleen are cultured with 20-30% autologous or allogeneic T cells and mitogen, the majority of cells rosette with sheep erythrocytes (E) after 3-5 days in culture. These E+ cells are of transformed appearance and several points indicate that many are derived from B cells. Some simultaneously possess surface immunoglobulin (sIg) or a B cell antigen [detected with BA1 monoclonal antibody (mAb)] and E receptor. Many do not stain with anti-T cell mAb (UCHT1, OKT4, OKT8), while cultured T cells continue to express these antigens. Furthermore, many E+ c

Liquiritigenin attenuates inflammatory periapical bone loss by stabilizing TRIM31 and restraining NLRP3 inflammasome and cGAS-STING inflammatory signaling.

Apical periodontitis (AP) is characterized by persistent inflammation and progressive periapical bone resorption, processes increasingly associated with NLRP3 inflammasome activation and cGAS-STING-related inflammatory signaling. However, pharmacological approaches targeting these inflammatory pathways and their upstream regulatory mechanisms remain limited. Here, we investigated whether liquiritigenin, a natural flavonoid derived from licorice, attenuates inflammatory injury and periapical bone loss through TRIM31-associated regulation of NLRP3 and modulation of cGAS-STING-related signaling.

Human epidermal growth factor receptor 2 as a target of 1,2,3,4,6-penta-O-galloyl-β-d-glucose in colon cancer.

OBJECTIVES: Although HER2 overexpression is observed in only 2%-3% of colorectal cancer (CRC) patients, its amplification can function as a compensatory mechanism that drives CRC resistance to targeted therapy. 1,2,3,4,6-Penta-O-galloyl-β-d-glucose (PGG) has been reported to possess anti-tumor and anti-metastatic activities in CRC. Nevertheless, the molecular targets through which PGG exerts its effects in CRC remain unclear. This study aimed to evaluate the role of HER2 as a potential molecular target of PGG in CRC. METHODS: Ultra performance liquid chromatography-mass spectrometer (UPLC-MS)

Continuous release of secretory granule proteoglycans from a cell strain derived from the bone marrow of a patient with diffuse cutaneous mastocytosis.

A human cell strain (designated HBM-M) that was derived from the bone marrow of a child with diffuse cutaneous mastocytosis was previously found to possess features that suggested it belonged in the mast cell/monocyte lineage. HBM-M cells synthesized approximately 150-Kd Pronase-resistant proteoglycans that were recognized by an antihuman secretory granule proteoglycan peptide core antibody. These cells also contained in relatively high abundance the same sized mRNA transcript that encodes the peptide core of proteoglycans that are normally localized to secretory granules of hematopoietic cell

Finding the etiology of membranoproliferative glomerulonephritis.

Membranoproliferative glomerulonephritis (MPGN) has undergone a change in classification that is focused on the etiology of MPGN. Thus, MPGN is classified into immune-complex (IC)-mediated MPGN and complement-mediated MPGN (C3 glomerulopathy) based on immunofluorescence microscopy studies that show positive staining for Igs and/or C3. IC-MPGN typically results from an infection or an autoimmune disease. Deposition of monotypic/monoclonal Ig also results in an MPGN. On the other hand, MPGN with bright C3 and minimal/no Ig is classified as C3 glomerulopathy. On the basis of the absence of any cl

Biochemical characterization of an anti-Candida factor produced by Enterococcus faecalis.

BACKGROUND: Because Candida albicans is resistant to several antifungal antibiotics, there is a need to identify other less toxic natural products, particularly antimicrobial proteins, peptides or bacteriocin like inhibitory substances. An attempt has been made to purify and characterise an anti-Candida compound produced by Enterococcus faecalis. RESULTS: An anti-Candida protein (ACP) produced by E. faecalis active against 8 C. albicans strains was characterised and partially purified. The ACP showed a broad-spectrum activity against multidrug resistant C. albicans MTCC 183, MTCC 7315, MTCC 3

Phosphopeptides in highly purified calf thymus DNA.

Highly purified DNA obtained from calf thymus nuclei was found to cleave after reaction with a chelating agent and subsequent dialysis against 0.01 M phosphate. During the cleavage release of proteineous material into the dialysate was observed. By means of anion exchange resin column chromatography, this material was separated into 9 main fractions. Two of these fractions P1 and P5) were found to contain the amino acids phosphoserine, asp, thr, ser, glu, gly, ala, val, ile, leu, and arg, as well as metal ion complexes of phosphoserine. The complexes were dissociated by Chelex 100 treatment. T

Comparative study on the composition of the organic matrix of the long bones of the "normal" rat and of the mutant Op/Orl.

The organic matrix of the long bones of the "normal" rat strain and of its mutant Op/Orl were investigated. This latter strain exhibits some anomalles similar to Albers-Schönberg disease as well as tooth retentions. Bones (tibia, femur) were incubated with 14C-glucose and also submitted to a fractional extraction procedure (EDTA, urea) to separate soluble and insoluble fractions. The proteins, hexoses, hexosamines, hydroxyproline content of these fractions was determined as well as the radioactivity of the soluble extracts Glycosaminoglycans were also studied by cellogel electrophoresis after

Triiodothyronine, thyroxine, and iodine in purified thyroglobulin from patients with Graves' disease.

Previous studies have suggested that there is an overproduction of triiodothyronine (T(3)) relative to thyroxine (T(4)) in patients with thyrotoxicosis associated with Graves' disease. To evaluate whether or not an increased ratio of T(3) to T(4) in thyroidal secretion could be contributing to this relative T(3) hyperproduction, T(3), T(4), and iodine were measured in thyroglobulin (Tg) from controls and patients with Graves' disease who had been treated either with propranolol only or with antithyroid drugs plus iodide before surgery. To avoid possible artifacts associated with pulse labeling

A role for phosphorylation in the maintenance of resurgent sodium current in cerebellar purkinje neurons.

Cerebellar Purkinje neurons express voltage-gated, tetrodotoxin (TTX)-sensitive sodium channels that not only open and inactivate rapidly during depolarization but also reopen during repolarization, carrying an unusual "resurgent" sodium current. Expression of Na(V)1.6 alpha subunits appears necessary but not sufficient to generate this component of current; Purkinje cells without Na(V)1.6 lack resurgent current, but resurgent current is absent from many other Na(V)1.6-expressing neurons. These observations raise the question of how modulation or modification of the Na(V)1.6 subunit may lead t
