This study investigated the regulatory effects of triptolide on PTEN-induced putative kinase 1 (PINK1)/Parkin-mediated mitophagy and NOD-like receptor family pyrin domain containing 3 (NLRP3)-mediated pyroptosis. Furthermore, it aimed to elucidate the potential relationship between mitophagy and pyroptosis in triptolide-induced hepatotoxicity. We established in vitro and in vivo models using a human normal hepatic cell line, HL7702, and C57BL/6J mice treated with triptolide. Furthermore, mitophagy inhibition experiments were performed using cyclosporin A, chloroquine, or PINK1 knockdown. The r

Silver nanoparticles(AgNPs)have been widely used in biomedicine and industry. Growing studies have shown that AgNPs can induce sperm motility decrease and spermiogenesis disorders. In this study, animal experiments were used to investigate the role of mitophagy and pyroptosis caused by AgNPs (25.93 nm) in testicular injury. Results showed that AgNPs induced the production of NLRP3, IL-1β and IL-18, activated Caspase-1, increased the expression of GSDMD protein, and activated the PINK1/Parkin signaling pathway, which induced in mitophagy in mice testicle tissue. In summary, AgNPs induced mitoph

Pyroptosis, a lytic inflammatory cell death mechanism, plays dual roles in tumorigenesis, but its clinical relevance in acute myeloid leukemia (AML) remains poorly understood. Through an integrative analysis of 40 pyroptosis-related genes in newly diagnosed AML patients (TCGA, n = 151) and healthy controls (GTEx, n = 386), we identified 32 genes with aberrant expression. Among these, 9 genes were found to be significant prognostic markers, including ELANE (protective), and CASP1, CHMP4B, BAK1, and CHMP2A (risk), which retained their prognostic significance after adjusting for age and gender. Therapeutic resistance and recurrent metastasis continue to pose major obstacles in the treatment of malignant tumors worldwide. Immunogenic cell death (ICD), characterized by its ability to both eliminate cancer cells and stimulate antitumor immune responses, has emerged as a promising strategy in the field of cancer immunotherapy. As key subtypes of ICD, pyroptosis and necroptosis contribute significantly to remodeling the tumor microenvironment (TME) and modulating immune responses through their distinct death-immunity coupling mechanisms. Characterized by plasma membrane pore formation and

Cell death and inflammation are key innate immune responses, but excessive activation can cause tissue damage. The NLRP3 inflammasome is a promising target for reducing inflammation and promoting recovery. Immunometabolism regulates NLRP3 responses in neurological and inflammatory diseases through cyclic nucleotide signaling. Targeting phosphodiesterases (PDEs), which hydrolyze cAMP and cGMP, offer a novel approach to mitigate inflammation. While 14 PDE inhibitors are FDA-approved, PDE10A's role in NLRP3 inflammasome activation remains unclear. This study investigates the effects of PDE10A inh

As an inevitable end of an organism, cell death plays a crucial role in human metabolism. In recent years, inappropriate cell death, such as cellular apoptosis, pyroptosis and ferroptosis, has been shown to be involved in a variety of metabolic diseases. The placenta is an extremely important endocrine organ during pregnancy, which significantly influences fetal health by providing material exchange. Cell death has been found to be associated with several placenta-associated diseases, including gestational diabetes mellitus, preeclampsia and others. Inflammation is closely linked to the direct

Ischemic stroke is one of the diseases which pose a significant threat to human health. Recent studies have suggested that programmed cell death plays an important role in brain tissues affected by ischemia and hypoxia. Pyroptosis, which is characterized by both apoptosis and necrosis, is mediated by activation of inflammasomes, such as NOD-like receptor family, pyrin domain containing 3 (NLRP3). Pyroptosis, which depends on caspase-1 activation and the release of pro-inflammatory cytokines, such as Interleukin 1β and Interleukin 18, plays a vital role in regulating cell survival and death fol

Retinal diseases encompass various conditions associated with sight-threatening immune responses and are leading causes of blindness worldwide. These diseases include age-related macular degeneration, diabetic retinopathy, glaucoma and uveitis. Emerging evidence underscores the vital role of the innate immune response in retinal diseases, beyond the previously emphasized T-cell-driven processes of the adaptive immune system. In particular, pyroptosis, a newly discovered programmed cell death process involving inflammasome formation, has been implicated in the loss of membrane integrity and the

BACKGROUND: In contrast to childhood minimal change disease (MCD), adult-onset MCD frequently recurs and requires prolonged immunosuppressive therapy. Accordingly, an investigation of the pathogenesis of adult MCD is required. MCD is usually accompanied by severe dyslipidaemia. Oxidized low-density lipoprotein (ox-LDL) is known to function in a damage-associated molecular pattern (DAMP) through CD36, triggering the NOD-like receptor thermal protein domain-associated protein 3 (NLRP3) inflammasome and programmed cell death called pyroptosis. However, the relationship between MCD pathogenesis an

This manuscript explores the role of pyroptosis, an inflammatory programmed cell death, in the pathogenesis of two chronic dermatoses, psoriasis and hidradenitis suppurativa (HS). The diseases, though clinically diverse, share common pathogenetic pathways involving the unbalanced interaction between the adaptive and innate immune systems. This review focuses on the molecular changes in psoriatic and HS skin, emphasizing the activation of dendritic cells, secretion of interleukins (IL-17, IL-22, and TNF-α), and the involvement of inflammasomes, particularly NLRP3. This manuscript discusses the

Sepsis is a life-threatening systemic inflammatory response syndrome caused by the host imbalanced response to infection. Lung injury is the most common complication of sepsis and one of the leading causes of patient death. Pyroptosis is a specific programmed cell death characterized by the release of inflammatory cytokines. Appropriate pyroptosis can reduce tissue damage and exert a protective effect against infection during sepsis. However, overactivated pyroptosis results in massive cell death, leading to septic shock, multiple organ dysfunction syndrome, and even an increased risk of secon

Gasdermin D (GSDMD)-mediated pyroptosis has a significant pro-inflammation characteristic due to dramatic secretion of pro-inflammatory substances. However, its role remains unclear in psoriasis as one chronic inflammatory skin disorder with high prevalence. We found that N-terminal GSDMD (N-GSDMD) was aberrantly expressed in epidermis of skin lesion in psoriasis patients and imiquimod-induced psoriasis-like dermatitis (IIPLD) mice. In epidermis of IIPLD mice and M5 (simulating psoriatic inflammatory challenge)-treated keratinocytes cultured in vitro, cleavage products of caspase-1, GSDMD and

The incidence of Chlamydia trachomatis respiratory infection is increasing, and its pathogenesis is still unclear. Pyroptosis, as a mode of inflammatory cell death, plays a vital role in the occurrence and development of Chlamydia trachomatis respiratory infection. In this study, the potential pyroptosis-related genes involved in Chlamydia trachomatis respiratory infection were identified by constructing a mouse model of C. muridarum infection combined with bioinformatics analysis. Through in-depth analysis of the RNA sequencing data, 13 differentially expressed pyroptosis-related genes were s

Inflammasome activation is a critical defense mechanism against bacterial infection. Previous studies suggest that inflammasome activation protects against Salmonella oral infection. Here we find inflammasome activation plays a critical role in the pathogenesis of Salmonella systemic infection. We show that in a systemic infection model by i.p. injection of Salmonella, deficiency of caspase-1 or gasdermin-D prolonged survival time, reduced plasma concentrations of the proinflammatory cytokines IL-1β, IL-6 and TNFα. These deficiencies also protected against coagulopathy during Salmonella infect

Sepsis, a life-threatening disease caused by dysregulated host response to infection, is a major public health problem with a high mortality and morbidity rate. Pyroptosis is a new type of programmed cell death discovered in recent years, which has been proved to play an important role in sepsis. Nevertheless, there is no comprehensive report, which can help researchers get a quick overview and find research hotspots. Thus, we aimed to identify the study status and knowledge structures of pyroptosis in sepsis and summarize the key mechanism of pyroptosis in sepsis. The data were retrieved and
