To what extent are genetics clinically useful?

The volume of research undertaken on the genetic susceptibility of inflammatory bowel diseases has been tremendous. International collaborative efforts which initiated in 1997 and have not stopped since, led to the identification at present of more than 100 IBD risk loci. Yet, only 25% of the genetic variance is explained. It is hypothesized that rare variants, other forms of genetic variation (copy number variation), as well as gene-gene and geneenvironment interactions, explain the missing heritability. From all genes identified, the pattern recognition receptor NOD2/CARD15 is still the most

Plant immune co-receptors: bridging gaps toward next-generation crop defense.

Plant innate immunity is a crucial, multi-layered defense system that protects crops from a wide range of bacterial, fungal, and viral pathogens, all of which pose a significant threat to global food security. Co-receptors function as central molecular modulators that amplify and fine-tune immune signaling. This signaling is initiated by cell-surface pattern-recognition receptors (PRRs) and intracellular nucleotide-binding leucine-rich repeat (NLR) proteins. Critical co-receptors for strong immune signal propagation include the SOMATIC EMBRYOGENESIS RECEPTOR KINASE (SERK) protein family, repre

Role of Innate Immune Receptors in Cardiac Damage Linked to Metabolic Disorders.

This chapter underscores the pivotal role of immune receptors, particularly Toll-like receptors (TLRs) and NOD-like receptors (NLRs), in the intricate interplay between metabolic diseases and their associated cardiovascular comorbidities. Their involvement extends beyond mere initiation, profoundly influencing the progression of these complex conditions. The recognition of TLRs and NLRs as promising therapeutic targets is a significant advancement, offering potential avenues for intervention in both metabolic disorders and their cardiovascular sequels. The significance of these pattern recognit

Therapeutic targeting of NOD1 receptors.

The nucleotide-binding oligomerization domain 1 (NOD1) protein is an intracellular receptor for breakdown products of peptidoglycan (PGN), an essential bacterial cell wall component. NOD1 responds to γ-D-glutamyl-meso-diaminopimelic acid, which is an epitope unique to PGN structures from all Gram-negative bacteria and certain Gram-positive bacteria. Upon ligand recognition, NOD1 undergoes conformational changes and self-oligomerization mediated by the nucleotide-binding NACHT domains, followed by the recruitment and activation of the serine threonine kinase receptor-interacting protein 2 leadi

A G-protein-coupled chemoattractant receptor recognizes lipopolysaccharide for bacterial phagocytosis.

Phagocytes locate microorganisms via chemotaxis and then consume them using phagocytosis. Dictyostelium amoebas are stereotypical phagocytes that prey on diverse bacteria using both processes. However, as typical phagocytic receptors, such as complement receptors or Fcγ receptors, have not been found in Dictyostelium, it remains mysterious how these cells recognize bacteria. Here, we show that a single G-protein-coupled receptor (GPCR), folic acid receptor 1 (fAR1), simultaneously recognizes the chemoattractant folate and the phagocytic cue lipopolysaccharide (LPS), a major component of bacter

A genome-wide survey of expansive NLR-C subfamily in miiuy croaker and characterization of the NLR-B30.2 genes.

NOD-like receptors (NLRs) are essential intracellular pattern-recognition receptors that respond to pathogens and regulate innate immunity. NLRs include three distinct subfamilies: NLR-A, NLR-B and NLR-C, thereinto, NLR-C as a large subfamily is unique to bony fish and little research about it has been done. In the current study, we identified the members of NLR-B and NLR-C subfamilies containing 2 and 48 genes respectively in miiuy croaker. Compared with other teleosts except for zebrafish, NLR-C subfamily genes occurred expansion in miiuy croaker. The gene expansions of NLR-C subfamily may i

Propofol directly binds to and inhibits TLR7.

Sedatives/anesthetics are important medical tools to facilitate medical care and increase patients' comfort. Increasingly, there is recognition that sedatives/anesthetics can modulate immune functions. Toll-like receptors (TLRs) are major pattern recognition receptors involved in the recognition of microbial components. TLR7 recognizes single-strand RNA virus such as influenza and SARS-CoV2 viruses and initiates interferon (IFN) responses. IFN production triggered by TLR7 stimulation is a critical anti-viral response. For example, patients with TLR7 variants including loss-of- function variant

UuCTL6, a novel C-type lectin of Urechis unicinctus, agglutinates various bacteria and promotes immune defense against bacterial infection.

C-type lectins (CTLs) serve as pivotal pattern recognition receptors (PRRs) within the innate immune system of invertebrates. In this study, we identified and characterized a novel CTL from the echiuran worm (Urechis unicinctus), termed UuCTL6, to elucidate its function in antimicrobial defense. Sequence analysis revealed that the open reading frame (ORF) of UuCTL6 comprises 1074 bp, encoding a polypeptide of 358-amino acid, which includes a conserved carbohydrate recognition domain (CRD) stabilized by two disulfide bonds. Sequence identity analysis showed that UuCTL6 shares 26.7%-31.6% amino

Targeting innate immune sensors for intranasal vaccine adjuvant design.

While injectable vaccines can prevent respiratory pathogens from causing severe disease, their ability to elicit protective local immunity in the respiratory mucosa is more limited. For viral pathogens, infection outcome is often determined at the site of entry, where innate immune sensing within the airway mucosa precedes and conditions adaptive immune responses. At these surfaces, epithelial cells and antigen-presenting cells express a wide repertoire of pattern recognition receptors (PRRs), positioning innate immune activation as a central determinant of vaccine efficacy and durability of m

Japanese Encephalitis Virus and Host Innate Immunity: Insights Into TLR Signaling, PRR Crosstalk, and Viral Immune Evasion.

Japanese encephalitis virus (JEV), a neurotropic flavivirus and a major cause of viral encephalitis, poses a significant global health threat due to its neuroinvasive potential. Host innate immune responses, particularly those mediated by pattern-recognition receptors such as Toll-like receptors (TLRs) and RIG-I-like receptors, play critical roles in detecting JEV infection in neurons and glial cells, triggering antiviral defenses through induction of type I interferons (IFNs), inflammatory cytokines, and interferon-stimulated genes. However, dysregulated inflammatory responses may contribute

Counting the Toll of Inflammation on Schizophrenia-A Potential Role for Toll-like Receptors.

Toll-like receptors (TLRs) are a family of pattern recognition receptors (PRRs) that are ubiquitously expressed in the human body. They protect the brain and central nervous system from self and foreign antigens/pathogens. The immune response elicited by these receptors culminates in the release of cytokines, chemokines, and interferons causing an inflammatory response, which can be both beneficial and harmful to neurodevelopment. In addition, the detrimental effects of TLR activation have been implicated in multiple neurodegenerative diseases such as Alzheimer's, multiple sclerosis, etc. Many

Application and prospect of targeting innate immune sensors in the treatment of autoimmune diseases.

Dysregulation of auto-reactive T cells and autoantibody-producing B cells and excessive inflammation are responsible for the occurrence and development of autoimmune diseases. The suppression of autoreactive T cell activation and autoantibody production, as well as inhibition of inflammatory cytokine production have been utilized to ameliorate autoimmune disease symptoms. However, the existing treatment strategies are not sufficient to cure autoimmune diseases since patients can quickly suffer a relapse following the end of treatments. Pattern recognition receptors (PRRs), including Toll-like

Regulation of platelet function by class B scavenger receptors in hyperlipidemia.

Platelets constitutively express class B scavenger receptors CD36 and SR-BI, 2 closely related pattern recognition receptors best known for their roles in lipoprotein and lipid metabolism. The biological role of scavenger receptors in platelets is poorly understood. However, in vitro and in vivo data suggest that class B scavenger receptors modulate platelet function and contribute significantly to thrombosis by sensing pathological or physiological ligands, inducing prothrombotic signaling, and increasing platelet reactivity. Platelet CD36 recognizes a novel family of endogenous oxidized chol

Zika virus and host innate immunity: mechanisms of activation and evasion.

Zika virus (ZIKV) is an Orthoflavivirus that poses a significant public health threat due to its unique capacity to cause congenital malformations, neurological disorders, and reproductive complications. Upon infection, host cells sense viral RNA and danger signals through pattern recognition receptors, including Toll-like receptors, RIG-I-like receptors, inflammasomes and the cGAS-STING pathway, leading to the production of type I interferons and the subsequent expression of interferon-stimulated genes. Additional intrinsic restriction mechanisms, such as apoptosis, autophagy, RNA interferenc

Comparative innate immune responses across major RNA and DNA viral infections: Mechanisms, immunopathology, and therapeutic perspectives.

Innate immunity is the first defense against viral infections, limiting viral replication and initiating adaptive immunity. Viral pathogens are recognized by pattern recognition receptors (PRRs), including Toll-like receptors (TLRs), (RIG-I-like receptors) RLRs, and the (cyclic GMP-AMP synthase) cGAS-STING pathway, which trigger interferon production and antiviral responses. This review compares innate immune responses to major RNA viruses (influenza, SARS-CoV-2, HIV) and DNA viruses (HSV, HBV, CMV). While these viruses activate similar pathways, they differ in interferon dynamics, inflammator

C-type lectin receptors differentially induce th17 cells and vaccine immunity to the endemic mycosis of North America.

Vaccine immunity to the endemic mycoses of North America requires Th17 cells, but the pattern recognition receptors and signaling pathways that drive these protective responses have not been defined. We show that C-type lectin receptors exert divergent contributions to the development of antifungal Th17 cells and vaccine resistance against Blastomyces dermatitidis, Histoplasma capsulatum, and Coccidioides posadasii. Acquired immunity to B. dermatitidis requires Dectin-2, whereas vaccination against H. capsulatum and C. posadasii infection depends on innate sensing by Dectin-1 and Dectin-2, but

Toll-like receptors in fish: evolution, structure, immune signaling, and translational prospects for adjuvant-based vaccine design in aquaculture.

Toll-like receptors (TLRs) are central to innate immunity and the most studied class of pattern recognition receptors (PRRs). Upon recognition of pathogen-associated molecular patterns (PAMPs) from bacteria, viruses, fungi, and parasites, strong immune responses that bridge innate and adaptive immunity are initiated. Their ability to sense various ligands has made TLR agonists promising candidates for adjuvant development. Unlike traditional adjuvants, TLR ligands directly activate immune pathways, leading to strong, specific, and lasting protection. In aquaculture, there is a critical need fo

Role of scavenger receptors in innate immunity in crustaceans.

Crustaceans are indispensable aquaculture species worldwide, and the scale of crustacean cultivation has been expanding continuously over recent years. However, pathogen infections have caused substantial economic losses in the global crustacean aquaculture industry. As invertebrates, crustaceans lack acquired immunity and entirely depend on innate immunity to resist pathogen invasion. Scavenger receptors (SRs) are an important type of pattern-recognition receptors (PRRs) in crustaceans that can specifically recognize pathogen-associated molecular patterns (PAMPs) and play a crucial role in in

Biomarkers for sepsis: more than just fever and leukocytosis-a narrative review.

A biomarker describes a measurable indicator of a patient's clinical condition that can be measured accurately and reproducibly. Biomarkers offer utility for diagnosis, prognosis, early disease recognition, risk stratification, appropriate treatment (theranostics), and trial enrichment for patients with sepsis or suspected sepsis. In this narrative review, we aim to answer the question, "Do biomarkers in patients with sepsis or septic shock predict mortality, multiple organ dysfunction syndrome (MODS), or organ dysfunction?" We also discuss the role of pro- and anti-inflammatory biomarkers and

The CARD9-Associated C-Type Lectin, Mincle, Recognizes La Crosse Virus (LACV) but Plays a Limited Role in Early Antiviral Responses against LACV.

La Crosse virus (LACV) is a mosquito-transmitted arbovirus and the main cause of virus-mediated neurological diseases in children. To date, little is known about the role of C-type lectin receptors (CLRs)-an important class of pattern recognition receptors-in LACV recognition. DC-SIGN remains the only well-described CLR that recognizes LACV. In this study, we investigated the role of additional CLR/LACV interactions. To this end, we applied a flow-through chromatography method for the purification of LACV to perform an unbiased high-throughput screening of LACV with a CLR-hFc fusion protein li
