An in vitro model to study effects of airborne pollutants on human ciliary activity.

A method to study the effects of airborne pollutants on ciliary activity of isolated human respiratory cells is presented. Human respiratory cells were scraped from nasal cavities of 25 healthy volunteers and washed in Ringer's solution. The cells were placed on polycarbonate membranes (3 microns pore size) and kept in macroplate holders swimming on Ringer's solution. Cells were thus kept humid and were supplied with nutrients through the pores of the membrane by capillary forces, while their surface was exposed to the gaseous environment. Isolated respiratory cells were exposed to SO2, NO2, a

Efficacy of pharmacologic glucagon in acute experimental pancreatitis.

Microcirculatory derangements in the pancreas associated with acute pancreatitis may contribute to a low-flow state and lead to pancreatic necrosis. This study investigated the effects of glucagon, a selective mesenteric arterial dilator, on pancreatic ischemia in canine bile-trypsin-induced pancreatitis (BTP). Measurements of cardiac Index (CI), total pancreatic blood flow (QP), pancreatic oxygen consumption (O2CP), and pancreatic arteriovenous shunt flow (QAVS) were obtained prior to and after inducing BTP. Bile-trypsin-induced pancreatitis was induced in 18 dogs. Nine received lactated Ring

Fluid management for dengue in children.

Dengue is a serious public health problem worldwide. Dengue shock syndrome (DSS), the severe form of dengue fever, can cause death within 12-24 hours if appropriate treatment is not promptly administered. For patients with DSS and the 30% of non-shocked dengue patients who require intravenous fluid therapy, a range of solutions is available for plasma volume support. Crystalloid solutions, such as normal 0·9% saline or Ringer's lactate, are the ones most commonly used. In severe cases, colloid solutions may be administered for their greater osmotic effect, although they carry a greater risk of

Effects of Joint Lavage with Dimethylsulfoxide on LPS-Induced Synovitis in Horses-Clinical and Laboratorial Aspects.

Several studies in human and equine medicine have produced controversial results regarding the role of dimethylsulfoxide (DMSO) as a therapeutic agent. This study aimed to evaluate the effect of joint lavage with different DMSO concentrations on biomarkers of synovial fluid inflammation and cartilage degradation in joints with lipopolysaccharide (LPS)-induced synovitis. Twenty-six tibiotarsal joints of 13 horses were randomly distributed into four groups (lactated Ringer's solution; 5% DMSO in lactated Ringer's; 10% DMSO in lactated Ringer's; and sham). All animals were evaluated for the prese

Plasma volume expansion with 5% albumin compared to Ringer's acetate during normal and increased microvascular permeability in the rat.

BACKGROUND: It is believed that the effectiveness of colloids as plasma volume expanders is dependent on the endothelial permeability for macromolecules. The objective of this study was to test the hypothesis that the plasma volume expanding effect of 5% albumin relative to that of a crystalloid solution is reduced if microvascular permeability is increased. METHODS: A control group was resuscitated with either 5% albumin (8 ml/kg) or Ringer's acetate (36 ml/kg) immediately after a hemorrhage of 8 ml/kg (n = 29). In a second group, permeability was increased by inducing sepsis through cecal l

Objective approach to the management of shock.

The objective in managing shock is to prevent anaerobic metabolism in the tissue. In the resuscitation of the acutely hypovolemic patient, volume resuscitation is the key. Drugs are seldom needed to accomplish this goal. Only when shock state results from an increase in the size of the container or from a decrease in the output of the pump, are drugs necessary. The objective management of shock is an understanding of the interrelationship of the pump, the container and the fluid, the mechanisms of monitoring their functions, and the use of the appropriate methods to return them to normal.

Monitoring and Evaluation of Corrosion at the Interface of Zirconium Alloy Biomaterials Under Simulated Oxidative Biological Environment.

The present work investigates the electrochemical behavior of the Zr2.5Nb alloy in a biomedical context, emphasizing the influence of electrochemical oxidation treatment on its stability in simulated physiological environments. The alloy samples were oxidized in 1 M H2SO4 under controlled voltages (200-275 V) and times (1 min), identifying 200 V-1 min as the optimal condition for obtaining a uniform porous oxide layer with an average pore diameter of ~90 nm. The corrosion resistance was evaluated using open circuit potential (OCP) and electrochemical impedance spectroscopy (EIS) in Ringer's so

Pressure-volume ratio in human cadaver hip joints.

In postmortem examinations 26 hip joints of 23 adult individuals were filled with Ringer's solution in order to furnish data on the relationship between intra-articular pressure, capsular content, and joint position. A tube system was connected to an 18-gauge needle and to a pressure transducer. After instillation of 2.5, 5.0, 7.5, and 10.0 ml of Ringer's solution, intra-articular pressure was recorded in six different joint positions (extension, 45 degrees and 90 degrees flexion, 40 degrees internal and 40 degrees external rotation, and 45 degrees abduction in extension). Intra-articular pres

The significance of gastric mucosal blood flow and acid secretion in erosive gastritis following hemorrhagic shock in cats.

Gastric acid secretion was stimulated in 21 anesthetized cats by infusion of pentagastrin for 45 min. Thirty minutes later the arterial blood pressure in two groups of the cats was lowered by bleeding to 75 mmHg for two hours, during which time one group was infused with cimetidine and the other with Ringer's solution. The tapped blood was then reinfused and normotension was maintained for two hours. In the third group the cats received pentagastrin and Ringer's solution but were kept normotensive throughout the experimental period. Blood flow was determined with the microsphere distribution t

Effect of amiloride and bumetanide on ionic currents in the epithelium of caecum from starved rabbits.

The aim of this study was to determine the effect of starvation on the transport of sodium and chloride ions in the epithelium of rabbit caecum. The experiment consisted in measuring transepithelial electrical potential (PD in mV) and the transepithelial electrical potential difference (dPD in mV) of an isolated fragment of rabbit caecum, before and after 4-day-long starvation. The studied tissue was incubated in Ringer solution and subsequently ion transport was modified through incubation in the Ringer solution supplemented with amiloride or/and bumetanide. It was demonstrated that the value

Membrane potential stabilization in amphibian skeletal muscle fibres in hypertonic solutions.

This study investigated membrane transport mechanisms influencing relative changes in cell volume (V) and resting membrane potential (E(m)) following osmotic challenge in amphibian skeletal muscle fibres. It demonstrated a stabilization of E(m) despite cell shrinkage, which was attributable to elevation of intracellular [Cl(-)] above electrochemical equilibrium through Na(+)-Cl(-) and Na(+)-K(+)-2Cl(-) cotransporter action following exposures to extracellular hypertonicity. Fibre volumes (V) determined by confocal microscope x z - scanning of cutaneous pectoris muscle fibres varied linearly wi

Amiloride and carotid body chemoreception of hypercapnia and hypoxia.

The sodium-proton (Na(+)-H+) antiporter has been found in virtually every tissue where its presence has been investigated. Its principal physiological role is to regulate intracellular pH (pHi). Amiloride (10(-3)-10(-4) M) is a known blocker of the antiporter when Na is present in normal physiological concentrations (130-140 x 10(-3) M). In order to determine if the Na(+)-H+ antiporter participated in the chemoreception of hypercapnia or hypoxia anesthetized, paralyzed, artificially ventilated cats were fitted with a loop in the right common carotid artery for the selective perfusion of the ca

[Replacement solutions and hemodilution].

By definition, haemodilution involves the use of filling solutions, but in this technique the role of different solutions has yet to be clearly defined. Amongst the criteria of choice used at the present time, two would seem to be essential: good tolerance, limiting the risks of complication and side effects to an absolute minimum; and a sufficiently long intravascular half-life ensuring good control of blood volume. On this basis, amongst the crystalloids Ringer lactate offers definite advantages. It is almost always used in association with a colloidal solution, very often human albumin or a
