Nutritional Modulation of the Gut-Kidney Axis.

BACKGROUND: Chronic kidney disease (CKD) represents a state of persistent, sterile low-grade inflammation in which sustained innate immune activation accelerates renal decline and cardiovascular complications. Diet-induced gut dysbiosis and intestinal barrier dysfunction lower mucosal immune tolerance, promote metabolic endotoxemia, and position the gut as an upstream modulator of systemic inflammatory signaling along the gut-kidney axis. SCOPE: Most studies address microbiota-derived metabolites, food-derived bioactive peptides, or omega-3 fatty acids separately. This review integrates evide

Adiposity associates with lower plasma resolvin E1 (Rve1): a population study.

BACKGROUND: Inadequate inflammation resolution may contribute to persistent low-grade inflammation that accompanies many chronic conditions. Resolution of inflammation is an active process driven by Specialized Pro-resolving Mediators (SPM) that derive from long chain n-3 and n-6 fatty acids. This study examined plasma SPM in relation to sex differences, lifestyle and a broad range cardiovascular disease (CVD) risk factors in 978, 27-year olds from the Australian Raine Study. METHODS: Plasma SPM pathway intermediates (18-HEPE, 17-HDHA and 14-HDHA), and SPM (E- and D-series resolvins, PD1, MaR

The role of acetylated cyclooxygenase-2 in the biosynthesis of resolvin precursors derived from eicosapentaenoic acid.

Specialized pro-resolving lipid mediators (SPMs) are natural bioactive agents actively involved in inflammation resolution. SPMs act when uncontrolled inflammatory processes are developed, for instance, in patients of COVID-19 or other diseases. The so-called resolution pharmacology aims at developing new treatments based on the use of SPMs as agonists, which promote inflammation resolution without unwanted side effects. It has been shown that the biosynthesis of SPMs called eicosapentaenoic acid (EPA)-derived E-series resolvins is initiated by aspirin-acetylated COX-2 from EPA, leading to 18-

Fatty Acids and Oxylipins in Osteoarthritis and Rheumatoid Arthritis-a Complex Field with Significant Potential for Future Treatments.

PURPOSE OF REVIEW: Osteoarthritis (OA) and rheumatoid arthritis (RA) are characterized by abnormal lipid metabolism manifested as altered fatty acid (FA) profiles of synovial fluid and tissues and in the way dietary FA supplements can influence the symptoms of especially RA. In addition to classic eicosanoids, the potential roles of polyunsaturated FA (PUFA)-derived specialized pro-resolving lipid mediators (SPM) have become the focus of intensive research. Here, we summarize the current state of knowledge of the roles of FA and oxylipins in the degradation or protection of synovial joints. R

Granulation tissue enriched by aspirin and omega-3 fatty acids in healing experimental periodontal lesion.

AIMS: Granulation tissue (GT) and specialized pro‑resolving mediators such as lipoxins and resolvins are key elements in the successful resolution of periodontitis. Aspirin‑triggered lipoxins and resolvins are even more powerful than their natural analogues. Their biosynthesis can be accelerated by omega-3 fatty acids. The aim of this study was to evaluate the use of GT enriched by aspirin and omega-3 fatty acids during the surgical treatment of periodontitis in an experimental animal model (rabbit). METHODS: In each of 24 rabbits, two experimental periodontal defects were created. In total,

Resolvin D1 Halts Remote Neuroinflammation and Improves Functional Recovery after Focal Brain Damage Via ALX/FPR2 Receptor-Regulated MicroRNAs.

Remote damage is a secondary phenomenon that usually occurs after a primary brain damage in regions that are distant, yet functionally connected, and that is critical for determining the outcomes of several CNS pathologies, including traumatic brain and spinal cord injuries. The understanding of remote damage-associated mechanisms has been mostly achieved in several models of focal brain injury such as the hemicerebellectomy (HCb) experimental paradigm, which helped to identify the involvement of many key players, such as inflammation, oxidative stress, apoptosis and autophagy. Currently, few

Long-chain Polyunsaturated Fatty Acids Mitigate In Vitro Skeletal Muscle Wasting Induced by Colorectal Carcinoma Cells via a 15-LOX-dependent Pathway.

Up to 50% of adults with colorectal cancer (CRC) are at risk of progressive involuntary loss of skeletal muscle mass and function known as cachexia. Available options to prevent and treat cachexia in cancer survivors are currently limited. Long-chain polyunsaturated fatty acids (LC-PUFAs) and their bioactive metabolites, termed specialized pro-resolving lipid mediators (SPMs), promote the resolution of inflammation and support muscle growth and repair. However, prior studies of cachexia have mainly focused on fish oil supplements, and it is not fully understood how different individual omega-3

Lipid emulsion rich in n-3 polyunsaturated fatty acids elicits a pro-resolution lipid mediator profile in mouse tissues and in human immune cells.

BACKGROUND: Lipid emulsions are a key component of total parenteral nutrition (TPN) and are administered to patients who are unable to ingest their daily required calories orally. Lipid emulsions rich with n-6 (ω-6) PUFAs are known to cause parenteral nutrition-associated liver disease and have inflammatory side effects, whereas n-3 PUFA-rich emulsions have favourable clinical outcomes. OBJECTIVES: The present study used targeted lipid mediator analysis to investigate the metabolism of a n-3 PUFA-rich lipid emulsion and a n-6 PUFA-rich lipid emulsion in a mouse model of TPN and in primary hum

Periodontal disease and endocrine imbalance: a molecular pathways perspective.

Periodontitis is a chronic multifactorial inflammatory disease whose pathological consequences extend well beyond the oral cavity to affect systemic health through the dissemination of pro-inflammatory mediators, bacteremic episodes, and the amplification of low-grade systemic inflammation. Concurrently, the endocrine system functions as a central regulator of periodontal tissue homeostasis, with hormone receptors for estrogens, androgens, glucocorticoids, insulin, thyroid hormones, and vitamin D expressed across the principal cell populations of the periodontium, including gingival fibroblast

Blueberry intake elevates post-exercise anti-inflammatory oxylipins: a randomized trial.

This study determined if 18 days of supplementation with blueberries (BL) compared to placebo (PL) could mitigate muscle soreness and damage and improve inflammation resolution in untrained adults (n = 49, ages 18-50 years) after engaging in a 90-min bout of "weekend warrior" eccentric exercise. The BL freeze dried supplement provided 1 cup of fresh blueberries per day equivalent with 805 mg/day total phenolics and 280 mg/day anthocyanins. Urine levels of eight BL gut-derived phenolics increased after 14- and 18-days supplementation with 83% higher concentrations in BL vs. PL (p < 0.001). The

Unsaturated fatty acids as immunometabolic modulators in high-risk pregnancies: mechanisms, clinical evidence, and translational perspectives.

High-risk pregnancies (HRPs) are characterized by the convergence of metabolic dysfunction, chronic low-grade inflammation, and placental maladaptation, leading to increased maternal and perinatal morbidity and mortality. Emerging evidence indicates that these conditions are driven by a tightly integrated immunometabolic axis, in which metabolic stress activates innate immune pathways, amplifying systemic and placental inflammation. Unsaturated fatty acids (UFAs), including monounsaturated and polyunsaturated fatty acids (PUFAs), have gained attention as key modulators of this axis. Omega-3 PU

The Therapeutic Effect of Phosphopeptide P140 Attenuates Inflammation Induced by Uric Acid Crystals in Gout Arthritis Mouse Model.

Gout is a painful form of inflammatory arthritis characterized by the deposition of monosodium urate (MSU) crystals in the joints. The aim of this study was to investigate the effect of peptide P140 on the inflammatory responses in crystal-induced mouse models of gout and cell models including MSU-treated human cells. Injection of MSU crystals into the knee joint of mice induced neutrophil influx and inflammatory hypernociception. Injection of MSU crystals subcutaneously into the hind paw induced edema and increased pro-inflammatory cytokines levels. Treatment with P140 effectively reduced hyp

Pathogenesis of bronchopulmonary dysplasia: an immunological perspective.

Bronchopulmonary dysplasia (BPD), the most common chronic lung disease of prematurity, is at its mechanistic core an immune-developmental disease in which inflammatory injury disrupts normal lung maturation. In the United States, BPD remains among the most common morbidities of very preterm birth, affecting a substantial proportion of infants born before 32 weeks' gestation. These infants are born before alveolarization is complete, into an inflammatory environment that can impair ongoing lung development. This review advances a single thesis: the heterogeneous prenatal and postnatal insults t

Bioactive Anti-Inflammatory Compounds and Therapeutic Strategies for Promoting Resolution.

Inflammation plays a crucial role in defending the body against harmful stimuli and maintaining physiological balance; however, when it becomes chronic, it contributes to the pathogenesis of several long-term diseases, including autoimmune conditions, cardiovascular and neurodegenerative disorders, and various cancers. Although conventional anti-inflammatory drugs provide symptomatic relief, their long-term use is often associated with adverse side effects. This limitation has shifted scientific attention toward naturally occurring bioactive molecules with potent, safer anti-inflammatory activ

Lipoxin A4/FPR2 Signaling Mitigates Ferroptosis of Alveolar Epithelial Cells via NRF2-Dependent Pathway During Lung Ischemia-Reperfusion Injury.

Post-lung transplant (LTx) injury can involve sterile inflammation due to ischemia-reperfusion injury (IRI) that contributes to allograft dysfunction. In this study, we investigated the cell-specific role of ferroptosis (excessive iron-mediated cell death) in mediating lung IRI and investigated if specialized pro-resolving mediators such as Lipoxin A4 (LxA4) can protect against ferroptosis in lung IRI. Single-cell RNA sequencing analysis of lung tissue from post-LTx patients was performed, and lung IRI was evaluated in C57BL/6 (WT), formyl peptide receptor 2 knockout (Fpr2-/-) and nuclear fact

Resolution of post-lung transplant ischemia-reperfusion injury is modulated via Resolvin D1-FPR2 and Maresin 1-LGR6 signaling.

BACKGROUND: Dysregulation of inflammation-resolution pathways leads to postlung transplant (LTx) ischemia-reperfusion (IR) injury and allograft dysfunction. Our hypothesis is that combined treatment with specialized pro-resolving lipid mediators, that is, Resolvin D1 (RvD1) and Maresin-1 (MaR1), enhances inflammation-resolution of lung IR injury. METHODS: Expression of RvD1 and MaR1 was analyzed in bronchoalveolar lavage (BAL) fluid of patients on days 0, 1, and 7 post-LTx. Lung IR injury was evaluated in C57BL/6 (WT), FPR2-/-, and LGR6 siRNA treated mice using a hilar-ligation model with or

Blood pro-resolving mediators are linked with synovial pathology and are predictive of DMARD responsiveness in rheumatoid arthritis.

Biomarkers are needed for predicting the effectiveness of disease modifying antirheumatic drugs (DMARDs). Here, using functional lipid mediator profiling and deeply phenotyped patients with early rheumatoid arthritis (RA), we observe that peripheral blood  specialized pro-resolving mediator (SPM) concentrations are linked with both DMARD responsiveness and disease pathotype. Machine learning analysis demonstrates that baseline plasma concentrations of resolvin D4, 10S, 17S-dihydroxy-docosapentaenoic acid, 15R-Lipoxin (LX)A4 and n-3 docosapentaenoic-derived Maresin 1 are predictive of DMARD res

Protectins and maresins: New pro-resolving families of mediators in acute inflammation and resolution bioactive metabolome.

Acute inflammatory responses are protective, yet without timely resolution can lead to chronic inflammation and organ fibrosis. A systems approach to investigate self-limited (self-resolving) inflammatory exudates in mice and structural elucidation uncovered novel resolution phase mediators in vivo that stimulate endogenous resolution mechanisms in inflammation. Resolving inflammatory exudates and human leukocytes utilize DHA and other n-3 EFA to produce three structurally distinct families of potent di- and trihydroxy-containing products, with several stereospecific potent mediators in each f

Impact of exercise on immune cell infiltration in muscle tissue: implications for muscle repair and chronic disease.

Exercise has long been recognized for its systemic health benefits, including modulation of the immune system. Contemporary scientific inquiry has increasingly turned toward understanding the regulatory effects of exercise on immune cell dynamics within muscle tissue, highlighting their potential role in facilitating tissue repair and modulating chronic disease pathways. Following acute bouts of exercise, especially those involving eccentric or high-intensity contractions, muscle fibers experience micro-damage that triggers a well-orchestrated immune response. This phenomenon entails a coordin
