Effects of chronic treatment with selective and nonselective antagonists on the subtypes of dopamine receptors.

The effects of chronic blockade of D-1, D-2, or both subtypes of dopamine receptors on the densities and properties of the D-1 and D-2 subtypes of dopamine receptors were measured in rat brain. Animals were treated with 14 daily injections (i.p.) of the D-1-selective antagonist SCH-23390, the D-2-selective antagonist sulpiride, the nonselective antagonist fluphenazine, or vehicle. Serial 32-microns horizontal sections that included the caudate putamen were cut and alternately assigned to assays for D-1 or D-2 receptors. D-1 receptors were labeled with 3H-SKF-83566 or 3H-SCH-23390, and D-2 rece

Long-term consequences of antagonism by neuroleptics of behavioural events occurring during mesolimbic dopamine infusion.

Rats selected as low-activity responders to peripherally administered (-)N-n-propylnorapomorphine [(-)NPA] were subjected to a 13-day continuous infusion of dopamine bilaterally into the nucleus accumbens (0.48 microliter/hr, 25 micrograms in 24 hr). This caused biphasic increases in spontaneous levels of hyperactivity with peaks occurring between days 2-5 and 8-12 of infusion. Two weeks after the infusion was withdrawn the low-activity status of the animals, to (-)NPA challenge, converted to high-activity and remained at this changed level for 18 to 20 weeks. The effects of sulpiride (2.5 mg/

Dopamine D1 and D2 receptors mediate neuropeptide S-induced antinociception in the mouse formalin test.

Neuropeptide S (NPS) is the endogenous ligand of a G-protein coupled receptor named NPS receptor. The NPS system controls several biological functions, including anxiety, wakefulness, locomotor activity, food intake, and pain transmission. A growing body of evidence supports facilitatory effects for NPS over dopaminergic neurotransmission. The present study was aimed to investigate the role of dopamine receptors signaling in the antinociceptive effects of NPS in the mouse formalin test. The following dopamine receptor antagonists were employed: SCH 23390 (selective dopamine D1 antagonist, 0.05

Drug-induced movement disorders.

Parkinsonism, tremor, chorea-ballismus, dystonia, tardive dyskinesia, myoclonus, tics and akathisia can be induced by many drugs. The drugs that are most frequently implicated in movement disorders are antipsychotics, calcium antagonists, orthopramides and substituted benzamides (e.g. metoclopramide, sulpiride, clebopride, domperidone), CNS stimulants, antidepressants, anticonvulsants, antiparkinsonian drugs and lithium. It is possible for a single drug to induce 2 or more types of movement disorders in the same patient. Movement disorders are not always reversible after drug withdrawal.

[Disorders of the functional relations of the caudate nucleus and putamen as a possible cause of torsion muscular dystonia].

In cats electrostimulation of nucleus caudatus and putamen produced contralateral rotation of the head which was used to simulate the extrapyramidal torsion disorders. Single or repeated injections of cataleptogenic neuroleptic drugs (haloperidol, metoclopramide) caused nonuniform changes of the thresholds of rotation induced by putamen stimulation. On the contrary, atypical neuroleptics (clozapine, sulpiride, tioridazine) facilitated equally both types of striatum-induced responses. The authors infer that acute muscular dystonia can result from disorder in intrastriatal relations between nucl

Single-dose methamphetamine administration impairs ORM retrieval in mice via excessive DA-mediated inhibition of PrLGlu activity.

Methamphetamine (METH), an abused psychostimulant, impairs cognition through prolonged or even single-dose exposure, but animal experiments have shown contradictory effects on memory deficits. In this study we investigated the effects and underlying mechanisms of single-dose METH administration on the retrieval of object recognition memory (ORM) in mice. We showed that single-dose METH administration (2 mg/kg, i.p.) significantly impaired ORM retrieval in mice. Fiber photometry recording in METH-treated mice revealed that the activity of prelimbic cortex glutamatergic neurons (PrLGlu) was sign

Involvement of dopaminergic system in the dentate gyrus of the hippocampus in modulating the orofacial pain-related behaviors in the rats.

Chemical stimulation of the lateral hypothalamus (LH) induces analgesia by forming neural circuitries with multiple brain regions. The involvement of hippocampal dopaminergic receptors in the LH stimulation-induced antinociception in specific pain models in animals has been documented. However, because the neural circuitries involved in the mediation of orofacial pain are not the same as those that mediate the other types of pain, the present study aims to detect the role of dopamine receptors within the dentate gyrus (DG) in the antinociceptive responses induced by LH stimulation in an animal

Detecting drug-drug interactions that increase the incidence of long QT syndrome using a spontaneous reporting system.

WHAT IS KNOWN AND OBJECTIVE: Drug-induced long QT syndrome (diLQTS) is a rare but serious adverse drug reaction. Drug-drug interaction (DDI) is one of the risk factors for the development of diLQTS. However, the combinations of drugs that increase the risk of diLQTS have not been extensively investigated. This study was performed to analyse the potential DDIs that elevate the incidence of diLQTS using a spontaneous reporting system. METHODS: The Japanese Adverse Drug Event Report database from April 2004 to January 2020 was used to assess adverse event reports. We calculated the reporting odd

Role of D1-like and D2-like dopamine receptors within the ventral tegmental area in stress-induced and drug priming-induced reinstatement of morphine seeking in rats.

The ventral tegmental area (VTA) as a major source of dopamine neurons projecting to cortical and limbic regions has a crucial role in reward as well as stress processes. Dopamine is a predominant neurotransmitter in the reward system, which plays an important role in both drug priming-induced and cue-induced reinstatement of cocaine and heroin seeking. It has been shown that this neurotransmitter has a role in stress-induced relapse to drug seeking. Therefore, the present study aims to evaluate the effects of intra-VTA administration of SCH-23390, as a dopamine D1-like receptor antagonist, an

Effects of metoclopramide on mRNA levels of steroid 5α-reductase isozymes in prostate of adult rats.

The rising incidence of prostate cancer and benign prostatic hypertrophy in the Western world is a cause of increasing public health concern. The most active androgen in the prostate is 5α-dihydrotestosterone obtained from testosterone (T) by the enzyme 5α-reductase (5α-R), expressed in the prostate as two isozymes, 5α-R1 and 5α-R2. These isozymes are involved in the growth and development of normal prostate and in the onset and progression of prostate disease. Besides androgens, prolactin (PRL) may also play a role, although it is not clear whether its effects on the prostate are in synergism

[Significance of CRF and dopamine receptors in amygdala for reinforcing effects of opiates and opioids on self-stimulation of lateral hypothalamus in rats].

Bipolar electrodes were implanted in the lateral hypothalamus in a group of 44 Wistar male rats in order to study self-stimulation reaction in the Skinner box. Simultaneously, microcanules were implanted into the central nucleus of the amygdala to inject the drugs (1 microl per injection). The blockade of corticoliberin (CRF) receptors (astressin, 1 microg) or Na+influx currents (xycaine or lidocain 1 microg) by the intrastructural administration of drugs into the amygdala decreased self-stimulation reaction of the lateral hypothalamus in rats by 29-55%. Cloning, polymorphism, and inhibition of beta-carbonic anhydrase of Helicobacter pylori.

BACKGROUND: Carbonic anhydrase (CA) catalyzes the reversible hydration of CO(2) to bicarbonate and a proton, and alpha-class CA has been reported to facilitate the acid acclimation of Helicobacter pylori (hpalphaCA). The purpose of this study was to characterize the beta-class CA of H. pylori (hpbetaCA) and elucidate the role of this enzyme as a possible drug target for eradication therapy. METHODS: We isolated DNA clones of independent H. pylori strains obtained from patients with gastritis (n = 15), gastric ulcer (n = 6), or gastric cancer (n = 16), and then studied genetic polymorphisms. Modulation of dopaminergic terminal excitability by D1 selective agents.

The effect of the active R (+) enantiomer of the dopaminergic selective D1 agonist 1-Phenyl-2,3,4,5-tetrahydrol-(1H)-3-benzazepine-7,8-diol HCL (R-SKF 38393) was examined on the excitability of antidromically identified nigro-striatal dopaminergic neurons. Striatal infusions of R-SKF 38393 produced a decrease in terminal excitability, which was reversed by subsequent infusion of the Dopaminergic D1 selective antagonist R-(+)-8-chloro-2,3,4,5-tetrahydro-3-methyl-5-phenyl-1H-3-benzazepine-7-o l-HCL (SCH 23390). A nicotinic-dopaminergic hybrid probe enhances D2-like autoreceptor-mediated inhibition of dopamine release.

β2-containing nicotinic acetylcholine receptors (nAChRs) and dopamine D2 receptors (D2Rs) cooperate to shape striatal dopamine (DA) output, yet the mechanisms by which nicotinic-dopaminergic crosstalk influences D2-like autoreceptor-mediated inhibition of DA release remain unclear. Here we use NiCh8, a covalently linked nicotinic-dopaminergic hybrid, as a molecular probe to test whether single-molecule co-engagement can enhance D2-like autoreceptor-mediated inhibition of DA release. NiCh8 binds native α4β2* and α6β2* nAChRs as well as D2Rs, and behaves as a very low-efficacy partial agonist wi

We reported a case of neuroleptic malignant syndrome (NMS) associated with the syndrome of inappropriate secretion of antidiuretic hormone (SIADH). A 71-year-old woman, who had been diagnosed as hypertension and multiple cerebral infarction, was given sulpiride 150 mg daily for depressive state. Three days after started sulpiride, she developed fever, sweating, difficulty of movement and was admitted to the hospital. The white blood cell count rose to 16,300/mm3 and serum creatine kinase (CK) to 3,063 IU/L. Two days later CK rose to 20,050 IU/L regardless of stopping the drug, so she was trans

Inhibition in carotid body chemoreceptors mediated by D-2 dopaminoceptors: antagonism by benzamides.

Inhibition of chemosensory nerve impulses in the cat is evoked by dopamine (DA) applied to carotid body chemoreceptors. Pharmacological characterization of the dopaminoceptors involved in this action was determined through their blockade with benzamides, selective antagonists of D-2 receptors. Both metoclopramide and sulpiride were effective blockers of DA-induced chemosensory inhibition. Furthermore, both drugs induced an immediate increase in the frequency of carotid nerve chemosensory impulses, suggesting the presence of previous tonic inhibition of chemoreceptor discharges by endogenous DA

[Analysis of mechanisms whereby pharmacologically-induced hyperprolactinemia leads to altered gonadotropin secretion (author's transl)].

Chronic hyperprolactinaemia was induced in 10 women since the onset of a cycle for 2 or 3 consecutive cycles by administration of sulpiride (3 X 50 mg/day) or tiapride (100-200 mg/day). The resulting perturbations of the menstrual cycle included short luteal phase cycles, anovulatory cycles and amenorrhoea. The analysis of the hormonal profiles under treatment, as compared to the control cycles, indicate an impact of hyperprolactinaemia both preferentially at the hypothalamic (alteration of the secretion or endogenous LRH) and accessorily at the ovarian (lack of progesterone secretion by the n

Echinatin as a multimodal modulator of monoaminergic system: Preclinical evidence for antidepressant-like activity.

Mounting evidence highlights flavonoids as multitarget candidates for central nervous system disorders because they can cross the blood-brain barrier and modulate diverse neurotransmitter pathways. Echinatin, a chalcone-type flavonoid from Glycyrrhiza species, has not previously been evaluated in behavioral models relevant to mood disorders. Here, we examined whether echinatin exerts antidepressant-like effects in validated rodent tests and explored the underlying monoaminergic mechanisms using pharmacological manipulations. Male BALB/c mice received echinatin (20 or 30 mg/kg), fluoxetine (10

Modulatory role of intra-accumbal dopamine receptors in the restraint stress-induced antinociceptive responses.

Stress contributes to pain sensation by affecting several neural pathways, including mesolimbic-cortical dopamine neurons. Nucleus accumbens, an essential element of the mesolimbic dopaminergic pathway, plays a fundamental role in modulating pain and is differentially influenced by stressful events. Since we previously demonstrated the marked association of intra-NAc dopamine receptors with forced swim stress-evoked analgesia in acute pain state, this research was conducted to consider the contribution of intra-accumbal D1- and D2-like dopamine receptors to modulating effects of exposure to re
