The thiamin, riboflavin and pyridoxine status of patients on emergency admission to hospital.

The aim of this study was to assess the prevalence of thiamin, riboflavin and pyridoxine deficiencies at admission to an acute hospital. One hundred and twenty adult patients were selected at random from those admitted via the Accident and Emergency department over 3 days. Comparisons were made with a group of 80 healthy blood donors sequentially attending a local transfusion centre. The alcohol intake of 500 patients admitted sequentially via the same Accident and Emergency department was also assessed. Erythrocyte transketolase (ETK), glutathione reductase (EGR) and aspartate aminotransferas

Intrauterine growth retardation induced by thiamine deficiency and pyrithiamine during pregnancy in the rat.

The role of nutritional factors in the development of prenatal and postnatal growth retardation is not well understood. We tested if thiamine deficiency may cause intrauterine growth retardation in rats. From the second day of gestation Sprague-Dawley rats were freely fed either a nutritionally complete or a thiamine-deficient diet. A similar group of rats was pair-fed with a complete or a thiamine-deficient diet and daily pyrithiamine injections (50 micrograms/100 gm of body weight) were given to precipitate thiamine deficiency during the short gestation of the rat. Maternal thiamine levels i

Thiamin status of a Melanesian population.

A semiautomated method is described which uses the Abbott ABA-100 bichromatic analyzer to measure the stimulation of erythrocyte transketolase by thiamin pyrophosphate (the thiamin pyrophosphate effect). The method was accurate with a throughput of 30 samples per hour and is suitable for the study of the thiamin status of large populations. A significant proportion (21%) of a total of 1920 individuals of a predominantly Melanesian population were found to have an elevated thiamin pyrophosphate effect (greater than 15% activation). This same proportion with an elevated thiamin pyrophosphate eff

Prevalence of thiamin deficiency in anorexia nervosa.

OBJECTIVE: Deficiency of thiamin (vitamin B1) causes a range of neuropsychiatric symptoms that resemble those reported in patients with anorexia nervosa (AN) but the prevalence of thiamin deficiency in AN has not been reliably established. This study was designed to investigate the prevalence of thiamin deficiency in AN. METHOD: Thirty-seven patients attending a specialist eating disorders unit and meeting all or some of the DSM-IV criteria for AN were compared with 50 blood donor controls. All subjects underwent measurement of erythrocyte transketolase activation following the addition of th

[An elderly patient with beriberi].

An 82-year-old man was admitted to the hospital in the summer of 1995 due to dyspnea, peripheral edema, and a tingling sensation and muscle weakness in all extremities. Physical examination showed heart failure and polyneuropathy. Laboratory data showed a low concentration of vitamin B1 (10 ng/ml normal 23.8-45.9 ng/ml) and a low level of erythrocyte transketolase activity (0.58 IU/gHb normal 0.75-1.30 IU/gHb). A chest X-ray film revealed cardiomegaly (cardiothoracic ratio 57.1%) and ultrasonic cardiography revealed increased motion of the left ventricle. A diagnosis of beriberi was made. The

Recurrent febrile lymphadenopathy treated with large doses of vitamin B1: report of two cases.

The 2 children whose cases are reported here both had recurrent episodes of fever and cervical lymphadenopathy. The conventional approach had been unsuccessful in identifying the cause or therapy. In neither case was there an infectious agent demonstrated, and biopsy of a pathologically enlarged lymph gland revealed only reactive hyperplasia in each case. Abnormal metabolism was revealed in the first patient by detecting a substance in urine which is reported to be diagnostic for a form of subacute necrotizing encephalomyelopathy. In the second case, red cell transketolase indicated thiamine p

Specific irreversible inhibition of enzymes concomitant to the oxidation of carbanionic enzyme-substrate intermediates by hexacyanoferrate (III).

Four different enzymes, class I fructose-1,6-biphosphate aldolase from rabbit muscle, class II fructose-1,6-bisphosphate aldolase from yeast, transaldolase, and transketolase, are inactivated progressively in the presence of their specific substrates and hexacyanoferrate(III). The inactivation is strictly linked to the oxidation of the carbanionic enzyme-substrate intermediates of these enzymes reported previously [Healy, M. J. and Christen, P. (1973) Biochemistry, 12, 35]. However, the loss of activity is not due to the products of this oxidation, i.e. to hexacyanoferrate(II), or to the oxida

Neocortical neurogenesis in development and evolution-Human-specific features.

In this review, we focus on human-specific features of neocortical neurogenesis in development and evolution. Two distinct topics will be addressed. In the first section, we discuss the expansion of the neocortex during human evolution and concentrate on the human-specific gene ARHGAP11B. We review the ability of ARHGAP11B to amplify basal progenitors and to expand a primate neocortex. We discuss the contribution of ARHGAP11B to neocortex expansion during human evolution and its potential implications for neurodevelopmental disorders and brain tumors. We then review the action of ARHGAP11B in

[Lactic acidosis treated with thiamine. 3 cases].

Of 3 alcoholic patients with severe lactic acidosis, one had shoshin beriberi; the second--a beer drinker--presented with convulsions associated with hyponatraemia and complicated by rhabdomyolysis and was not thiamine-deficient; the third patient had convulsions associated with Korsakoff's syndrome and was thiamine-deficient. In all three patients treatment with thiamine administered alone corrected the lactic acidosis within less than 4 hours. In patient 1, this result was obtained after symptomatic treatment of shock and lactic acidosis had failed and more than 24 hours before the haemodyna

Nonoxidative pentose phosphate pathway regulates CD8+ T cell immunity by maintaining NADPH homeostasis.

NADPH is essential for cellular biosynthesis and redox balance in CD8+ T cells. Here, we demonstrate that the nonoxidative pentose phosphate pathway (non-oxPPP), mediated by transketolase (TKT) and transaldolase (TALDO1), is critical for CD8+ T cell activation, proliferation, and memory formation by maintaining NADPH homeostasis. Metabolomic profiling and isotopic tracing revealed upregulated non-oxPPP flux in effector (Teff) and memory (Tm) CD8+ T cells, enabling a pentose cycle that amplifies NADPH yield and sustains metabolic fitness for T cell immunity. Genetic knockdown or pharmacological

Mechanism of the synergistic action of oxythiamine and ketoconazole against the yeast Malassezia pachydermatis.

This article explains the biochemical basis of the synergistic effect of oxythiamine (OT) and ketoconazole (KTC) against Malassezia pachydermatis yeast, which was isolated from dogs exhibiting clinical signs of otitis externa. All strains were incubated on MLNA medium supplemented with OT, KTC, or a mixture of both compounds. We found that the ergosterol content was reduced by the compounds tested, both separately (20%-50%) and in combination (80%). OT alone and in combination with KTC reduced NADPH levels. However, we found no differences in acetyl-CoA levels under the influence of the compou

Andrographolide and Its 14-Aryloxy Analogues Inhibit Zika and Dengue Virus Infection.

Andrographolide is a labdene diterpenoid with potential applications against a number of viruses, including the mosquito-transmitted dengue virus (DENV). In this study, we evaluated the anti-viral activity of three 14-aryloxy analogues (ZAD-1 to ZAD-3) of andrographolide against Zika virus (ZIKV) and DENV. Interestingly, one analogue, ZAD-1, showed better activity against both ZIKV and DENV than the parental andrographolide. A two-dimension (2D) proteomic analysis of human A549 cells treated with ZAD-1 compared to cells treated with andrographolide identified four differentially expressed prot

Thiamine deficiency in vivo produces fiber cell degeneration in mouse lenses.

Thiamine (Vitamin B1) is a co-factor for enzymes key in bridging aerobic and anaerobic metabolism. One such enzyme, transketolase, catalyzes two of three reactions for entry into the pentose-phosphate pathway, a major source of chemical reducing power. Thus, thiamine deprivation (TD) is considered a classic model of systemic oxidative stress and is linked with degenerative diseases. TD in mice and rats produces neurodegeneration with Alzheimer's disease characteristics. Age-related disease of the lens, commonly cataract, is also linked with thiamine and oxidative stress. To test the effects of

[Cognitive deficits, vitamin status and controlled thiamine substitution in alcohol dependent patients in withdrawal treatment].

Vitamin status and cognitive performance in a test battery were analyzed in 45 alcohol dependent men after admission to and after two weeks of withdrawal treatment in two psychiatric clinics. To examine the effect of thiamin substitution versus alcohol withdrawal and hospital diet alone, 23 patients received 300 mg thiamin daily, 22 patients no vitamin substitution. Thiamin substitution alone ameliorated selectively and significantly the mean marginal thiamin status. Multiple correlations between measurements of the thiamin parameter transketolase activity and test performances were found in t

Can heatstable antithiamin factors inactivate thiamin in vitro and/or in vivo?

Aqueous solutions of thiamin hydrochloride were incubated with catechol for 24 hr, pH 7.4 at 50 degrees C. Additional solutions of thiamin and catechol were incubated separately. After incubation the remaining thiamin was analyzed by the thiochrome procedure which showed that the thiamin in the thiamin-catechol mixture was reduced by 82.5-90% and the thiamin incubated alone by 18-20%. The solutions were fed to groups of rats after they had been on a thiamin free diet for 17 days. Body weight gains, erythrocyte transketolase (ETKA), thiamin pyrophosphate effect (TPPE) and liver and urine thiami

Neurologic deficit following gastric partitioning: possible role of thiamine.

Gastric partitioning has been proposed as an alternative to jejunoileal bypass for the control of morbid obesity because of the relative ease of performance and the lack of metabolic complications. From our series of 250 patients who underwent gastric partitioning, one of them developed a neurologic syndrome consistent with Wernicke's disease. The clinical findings were characterized by ataxia, confusion, impaired extra-ocular movements with diplopia, and weakness. A second patient with a similar condition was referred to us for evaluation and treatment. Pretreatment thiamine levels were norma

Vitamin B2 status in athletes and the influence of riboflavin administration on neuromuscular irritability.

In order to obtain information on the vitamin B2 status in athletes in training, the following aspects were investigated: in one group of athletes, riboflavinuria at rest, during the autumn and winter months (mean values 23.8 and 9.45 mug/h, respectively); in the second group (n = 18), the activity of the nicotinamide-adenine-dinucleotide-phosphate-dependent red blood cell glutathione reductase (EC 1.6.4.2) and its activation by added flavin adenine dinucleotide. In 8 athletes, activation coefficients higher than 1.25 were found, indicating a biochemical deficiency of riboflavin. Erythrocyte t

A non-cofactor role of thiamine derivatives in excitable cells?

Thiamine diphosphate (TDP) is an important cofactor of pyruvate (PDH) and alpha-ketoglutarate (KGDH) dehydrogenases and transketolase. Thiamine deficiency leads to reversible and irreversible brain lesions due to impaired oxidative metabolism. A specific non-cofactor role for thiamine has also been proposed in excitable cells and thiamine triphosphate (TTP) might be involved in the regulation of ion channels. Thiamine is taken up by neuroblastoma cells through a high affinity transporter. Inside the cells, it is rapidly phosphorylated to TDP. This high turnover TDP pool is the precursor for TT
