Spatial and temporal patterns in dynamic-contrast enhanced intraoperative fluorescence imaging enable classification of bone perfusion in patients undergoing leg amputation.

Dynamic contrast-enhanced fluorescence imaging (DCE-FI) classification of tissue viability in twelve adult patients undergoing below knee leg amputation is presented. During amputation and with the distal bone exposed, indocyanine green contrast-enhanced images were acquired sequentially during baseline, following transverse osteotomy and following periosteal stripping, offering a uniquely well-controlled fluorescence dataset. An unsupervised classification machine leveraging 21 different spatiotemporal features was trained and evaluated by cross-validation in 3.5 million regions-of-interest o

How Realistic Are Idealized Copper Surfaces? A Machine Learning Study of Rough Copper-Water Interfaces.

Copper is a highly promising catalyst for the electrochemical CO2 reduction reaction (CO2RR) since it is the only pure metal that can form highly added-value products such as ethylene and ethanol. Since the CO2RR takes place in aqueous solution, the detailed atomic structure of the water-copper interface is essential for unraveling the key reaction mechanisms. In this study, we investigate copper-water interfaces exhibiting nanometer-scale roughnesses. We introduce two molecular dynamics protocols to create rough copper surfaces, which are subsequently brought into contact with water. From the

Identification of CYP2B6 as a Novel Biomarker of HRD in Colon Adenocarcinoma through WGCNA and Machine Learning.

The potential role of homologous recombination deficiency (HRD) in the diagnosis and treatment of colon adenocarcinoma (COAD) remains incompletely explored. Differential gene expression analysis was conducted using Limma to identify genes with altered expression levels. Key genes associated with HRD were identified through the integration of WGCNA and machine learning techniques. For the unsupervised grouping of samples, ConsensusClusterPlus was applied. To quantify gene expression and protein abundance in clinical tissues and cell lines, RT-qPCR and Western Blotting (WB) assays were performed

Cocaine Use Prediction With Tensor-Based Machine Learning on Multimodal MRI Connectome Data.

This letter considers the use of machine learning algorithms for predicting cocaine use based on magnetic resonance imaging (MRI) connectomic data. The study used functional MRI (fMRI) and diffusion MRI (dMRI) data collected from 275 individuals, which was then parcellated into 246 regions of interest (ROIs) using the Brainnetome atlas. After data preprocessing, the data sets were transformed into tensor form. We developed a tensor-based unsupervised machine learning algorithm to reduce the size of the data tensor from 275 (individuals) × 2 (fMRI and dMRI) × 246 (ROIs) × 246 (ROIs) to 275 (ind

Inferring fungal cis-regulatory networks from genome sequences via unsupervised and interpretable representation learning.

Gene expression patterns are determined to a large extent by transcription factor (TF) binding to noncoding regulatory regions in the genome. However, gene expression cannot yet be systematically predicted from genome sequences, in part because nonfunctional matches to the sequence patterns (motifs) recognized by TFs occur frequently throughout the genome. Large-scale functional genomics data for many TFs has enabled characterization of regulatory networks in experimentally accessible cells such as budding yeast. Beyond yeast, fungi are important industrial organisms and pathogens, but large-s

Quantifying the spatial information loss of Dose-Volume Histograms in Gamma Knife radiosurgery via unsupervised 3D dose geometry embedding.

Dose-Volume Histograms (DVH) discard spatial information by summarizing 3D distributions into 1D curves. This study quantifies this loss in Gamma Knife radiosurgery and introduces TopoGK, a novel unsupervised deep learning framework capturing full 3D dose geometry. Ninety-six vestibular schwannoma plans were analyzed. Dose grids were tumor-centered, resampled to [Formula: see text] voxels, and normalized to [Formula: see text]. A 3D Convolutional Variational Autoencoder compressed each dose-mask volume into a 64dimensional spatial embedding. Spatial information loss was quantified by comparing

ISGAN: Unsupervised Domain Adaptation With Improved Symmetric GAN for Cross-Modality Multi-Organ Segmentation.

The differences between cross-modality medical images are significant, so several studies are working on unsupervised domain adaptation (UDA) segmentation, which aims to adapt a segmentation model trained on a labeled source domain to an unlabeled target domain. The conventional UDA segmentation strategy aims to integrate image generation and segmentation. However, conventional image generation modules only consider information from a single domain (source or target), resulting in visual inconsistencies. The image generation module may also lack anatomical constraints, leading to incorrect pse

An unsupervised learning approach for clustering joint trajectories of Alzheimer's disease biomarkers: An application to ADNI Data.

INTRODUCTION: Current models of Alzheimer's disease (AD) progression assume a common pattern and pathology, oversimplifying the heterogeneity of clinical AD. METHODS: We define a syndrome as a unique biomarker progression pattern and develop a lag measure to cluster pre-dementia individuals, reflecting their pathology's multi-dimensionality. The technique uses the time-ordering of events to group individuals based on their position along the disease process and the relative positions of their markers. RESULTS: An application using Alzheimer's Disease Neuroimaging Initiative (ADNI) data highl

Rhythm profiling using COFE reveals multi-omic circadian rhythms in human cancers in vivo.

The study of ubiquitous circadian rhythms in human physiology requires regular measurements across time. Repeated sampling of the different internal tissues that house circadian clocks is both practically and ethically infeasible. Here, we present a novel unsupervised machine learning approach (COFE) that can use single high-throughput omics samples (without time labels) from individuals to reconstruct circadian rhythms across cohorts. COFE can simultaneously assign time labels to samples and identify rhythmic data features used for temporal reconstruction, while also detecting invalid orderin

Toddlers' Active Gaze Behavior Supports Self-Supervised Object Learning.

Toddlers learn to recognize objects from different viewpoints with almost no supervision. During this learning, they execute frequent eye and head movements that shape their visual experience. It is presently unclear if and how these behaviors contribute to toddlers' emerging object recognition abilities. To answer this question, we here combine head-mounted eye tracking during dyadic play with unsupervised machine learning. We approximate toddlers' central visual field experience by cropping image regions from a head-mounted camera centered on the current gaze location estimated via eye track

Unsupervised Brain MRI Anomaly Detection via Inter-Realization Channels.

Accurate anomaly detection in brain Magnetic Resonance Imaging (MRI) is crucial for early diagnosis of neurological disorders, yet remains a significant challenge due to the high heterogeneity of brain abnormalities and the scarcity of annotated data. Traditional one-class classification models require extensive training on normal samples, limiting their adaptability to diverse clinical cases. In this work, we introduce MadIRC, an unsupervised anomaly detection framework that leverages Inter-Realization Channels (IRC) to construct a robust nominal model without any reliance on labeled data. We

Structure preservation constraints for unsupervised domain adaptation intracranial vessel segmentation.

Unsupervised domain adaptation (UDA) has received interest as a means to alleviate the burden of data annotation. Nevertheless, existing UDA segmentation methods exhibit performance degradation in fine intracranial vessel segmentation tasks due to the problem of structure mismatch in the image synthesis procedure. To improve the image synthesis quality and the segmentation performance, a novel UDA segmentation method with structure preservation approaches, named StruP-Net, is proposed. The StruP-Net employs adversarial learning for image synthesis and utilizes two domain-specific segmentation
