Demyelination induced in mice by avirulent Semliki Forest virus. I. Virology and effects on optic nerve.

Swiss A2G mice were infected intraperitoneally with an avirulent strain of Semliki Forest virus. The virus titres in the optic nerves, retina and brain were estimated on post-inoculation days 3 to 8. High titres of virus were obtained in the optic nerve, retina and brain. The optic nerves, retina and brain were studied by light microscopy up to post-inoculation day 67. Electron microscope studies wee carried out on the optic nerve on post-inoculation days 11 and 14. Inflammatory, microcystic changes and demyelination seen in the optic nerve were similar to those found in he central nervous sys

Durability of the virological response after lamivudine discontinuation in lamivudine-resistant patients with a complete virological response after lamivudine and adefovir combination therapy.

We investigated the durability of virological response after lamivudine (LAM) discontinuation in LAM-resistant chronic hepatitis B (CHB) patients with complete virological response after LAM-adefovir (ADV) combination therapy. We enrolled 58 patients switched to ADV monotherapy with undetectable viral loads (<12 IU/ml) and normal alanine aminotransferase levels after ADV add-on combination treatment for at least 6 months in LAM-resistant CHB patients. Virologic relapse was defined as HBV DNA detection at more than 20 IU/ml by quantitative polymerase chain reaction determined on two consecutive

Virological response after short-term CCR5 antagonist exposure in HIV-infected patients: frequency of subjects with virological response and associated factors.

The virological response after an 8-day maraviroc monotherapy has been proposed to be an alternative method to determine whether an CCR5 antagonist should be prescribed to HIV-infected patients. The frequency of patients eligible for a combined antiretroviral therapy which includes maraviroc on the basis of the result of this clinical test is not well-known at the moment. In the same way, clinical and immunovirological factors associated with the virological response after antagonist exposure need to be determined. Ninety consecutive HIV-infected patients were exposed to an 8-day maraviroc mon

Virologic suppression in response to antiretroviral therapy despite extensive resistance within HIV-1 reverse transcriptase after the first virologic failure.

BACKGROUND: Incomplete virologic suppression results in mutations associated with resistance and is a major obstacle to disease control. We analyzed the genotypic profiles of HIV-1 patients at the time of the first virologic failure and the response to a salvage regimen after 48 weeks. METHODS: This work was a cross-sectional, retrospective, analytical study based on data collected from medical records and genotyping tests between 2006 and 2016. The sample consisted of data on individuals living with HIV (PLWH) from three major reference centers. RESULTS: A total of 184 patients were include

Nasopharyngeal Carcinoma at the Virology Precision Oncology Nexus: Decoding Molecular Alterations for Early Intervention and Therapeutic Innovation.

Nasopharyngeal carcinoma (NPC) represents an Epstein-Barr virus (EBV)-associated malignancy showing elevated incidence in East and Southeast Asia. Early detection remains vital, as molecular abnormalities precede visible histological changes during tumor development. This review summarizes recent progress in decoding NPC's molecular profile, including genetic mutations, epigenetic alterations, non-coding RNA networks and proteomic alterations. Importantly, these molecular discoveries are increasingly informing clinical approaches to disease management. Modern diagnostic integration of histopat

Gain-of-function virology as dual-use research of concern: Variations of securitization in the United States.

Ever since the advent of recombinant DNA technology in the early 1970s concerns have been expressed about the misuse potential of this and subsequent biotechnology breakthroughs. This article focuses on the securitization of gain-of-function (GOF) virology research in the United States, utilizing an updated theoretical framework that distinguishes between "riskification" and "threatification." The paper examines three distinct cases, two historical, one ongoing. It argues that early attempts to govern GOF research primarily employed a riskification approach, characterized by self-governance by

Hantavirus: A Comprehensive Contemporary Review of Virology, Global Epidemiology, Clinical Variants, Diagnosis, Treatment, and Intensive Care Management.

BACKGROUND: Hantaviruses are negative-sense, single-stranded RNA viruses of the family Hantaviridae that cause two potentially lethal zoonotic syndromes: Hemorrhagic fever with renal syndrome (HFRS) and hantavirus cardiopulmonary syndrome (HCPS). With approximately 2,00,000 reported cases of HFRS annually worldwide and case-fatality rates reaching 36-40% in severe HCPS, hantaviruses constitute a significant global public health burden. Rodent excreta are the principal source of human infection through inhalation of infectious aerosols. MATERIALS AND METHODS: A comprehensive literature search

Use of a generative AI tool to design RNA-based antiviral therapeutics for undergraduate virology laboratory teaching.

RNA medicines have taken the drug development world by storm since the introduction of mRNA vaccines post-pandemic. As this field is rapidly evolving at an unprecedented speed, it is crucial that higher education institutions keep up with this at all levels of teaching, including at the undergraduate level. In parallel, the necessity of embedding the fast-changing artificial intelligence (AI) landscape in undergraduate teaching and learning is also crucial. Here, I have developed a succinct but informative, in silico-based laboratory activity using a generative AI tool called Biomod AI (https:

Pathology and virology of natural highly pathogenic avian influenza H5N8 infection in wild Common buzzards (Buteo buteo).

Highly pathogenic avian influenza (HPAI) in wild birds is a major emerging disease, and a cause of increased mortality during outbreaks. The Common buzzard (Buteo buteo) has a considerable chance of acquiring the infection and therefore may function as bio-sentinel for the presence of virus in wildlife. This study aimed to determine the virus distribution and associated pathological changes in the tissues of Common buzzards that died with HPAI H5 virus infection during the 2020-2021 epizootic. Eleven freshly dead, HPAI H5 virus-positive Common buzzards were necropsied. Based on RT-PCR, all bir

Hepatitis D virus in 2021: virology, immunology and new treatment approaches for a difficult-to-treat disease.

Approximately 5% of individuals infected with hepatitis B virus (HBV) are coinfected with hepatitis D virus (HDV). Chronic HBV/HDV coinfection is associated with an unfavourable outcome, with many patients developing liver cirrhosis, liver failure and eventually hepatocellular carcinoma within 5-10 years. The identification of the HBV/HDV receptor and the development of novel in vitro and animal infection models allowed a more detailed study of the HDV life cycle in recent years, facilitating the development of specific antiviral drugs. The characterisation of HDV-specific CD4+ and CD8+T cell

Cyprinid herpesvirus-2 causing mass mortality in goldfish: applying electron microscopy to histological samples for diagnostic virology.

In June 2013, a major fish kill of adult goldfish Carassius auratus occurred in Runnemede Lake, New Jersey, USA: an estimated 3000 to 5000 fish died within ~5 d. Necropsy of 4 moribund fish revealed severely pale gills, and histopathology showed type I and II fusion of the gills, diffuse necrosis of hematopoietic tissue in anterior and posterior kidney, and multifocal necrosis of the spleen. Within necrotic areas, pyknosis and enlarged nuclei with marginalized chromatin were observed. Cyprinid herpesvirus-2, the etiological agent for herpesviral hematopoietic necrosis disease, was confirmed in

A study of the safety, immunology, virology, and microbiology of adjunctive etanercept in HIV-1-associated tuberculosis.

OBJECTIVE: Tumor necrosis factor (TNF), an important inflammatory mediator in tuberculosis, has been implicated in causing accelerated HIV disease progression in HIV-associated tuberculosis. However, TNF blockade, particularly by monoclonal antibody, has been associated with the reactivation of latent Mycobacterium tuberculosis infection by the impairment of mycobacterial immunity. This phase 1 study examined the safety, microbiology, immunology, and virology of TNF blockade using etanercept (soluble TNF receptor, Enbrel) during the initial treatment of HIV-associated tuberculosis. The epidemiology and virology of C-type virus-associated hematological cancers and related diseases in wild mice.

In several different populations of wild mice, observed over a 35-month period in laboratory geriatric colonies, a direct correlation was found between the prevalence and titer of spleen complement-fixing gs (p30) antigen and C-type particles in newly trapped healthy mice and a predilection to lymphoma and a hind leg paralytic disease upon aging. Other studies have established the indigenous C-type virus as the essential etiological determinant of both diseases in wild mice. An increased incidence of breast carcinomas, hepatomas, and pulmonary adenomas associated with C-type virus also occurre

Virological Markers in Epstein-Barr Virus-Associated Diseases.

Epstein-Barr virus (EBV) is an oncogenic virus infecting more than 95% of the world's population. After primary infection-responsible for infectious mononucleosis in young adults-the virus persists lifelong in the infected host, especially in memory B cells. Viral persistence is usually without clinical consequences, although it can lead to EBV-associated cancers such as lymphoma or carcinoma. Recent reports also suggest a link between EBV infection and multiple sclerosis. In the absence of vaccines, research efforts have focused on virological markers applicable in clinical practice for the m

[Treatment of chronic hepatitis B: virological and pharmacological aspects].

Tenofovir is a nucleotidic analog inhibitor used in monotherapy as first line treatment of chronic Hepatitis B virus (HBV) infection. This drug requires a two-step phosphorylation by cellular kinases. The active triphosphate form inhibits viral DNA polymerase. Tenofovir has a very low oral bioavailability following oral administration. Hence, its oral administration requires the use of prodrugs: tenofovir disoproxil fumararate (TDF) or tenofovir alafénamide (TAF). TAF demonstrated a lower kidney and bone toxicity than TDF. TAF and TDF treatments allow prevention of chronic hepatitis B complica

Deciphering SARS-CoV-2 Virologic and Immunologic Features.

Severe acute respiratory syndrome coronavirus (SARS-CoV)-2 and its associated pathology, COVID-19, have been of particular concerns these last months due to the worldwide burden they represent. The number of cases requiring intensive care being the critical point in this epidemic, a better understanding of the pathophysiology leading to these severe cases is urgently needed. Tissue lesions can be caused by the pathogen or can be driven by an overwhelmed immune response. Focusing on SARS-CoV-2, we and others have observed that this virus can trigger indeed an immune response that can be dysregu

Mapping of the US Domestic Influenza Virologic Surveillance Landscape.

Influenza virologic surveillance is critical each season for tracking influenza circulation, following trends in antiviral drug resistance, detecting novel influenza infections in humans, and selecting viruses for use in annual seasonal vaccine production. We developed a framework and process map for characterizing the landscape of US influenza virologic surveillance into 5 tiers of influenza testing: outpatient settings (tier 1), inpatient settings and commercial laboratories (tier 2), state public health laboratories (tier 3), National Influenza Reference Center laboratories (tier 4), and Ce
