Studies on chicken liver xanthine dehydrogenase with reference to the problem of non-equivalence of FAD moieties.

1. Reduction of chicken liver xanthine dehydrogenase (xanthine: NAD+ oxidoreductase, EC 1.2.1.37) by xanthine under anaerobic condition proceeded in two phases. This biphasicity may be due to functional and non-functional enzymes in the enzyme preparation. 2. Cyanolysis of a persulfide group of chicken liver enzyme resulted in an inactivation of the enzyme. The non-functional enzyme in the standard enzyme preparation was found to lack persulfide groups at the active sites. 3. The remaining NADH-Methylene Blue oxidoreductase activity, after KI treatment of the xanthine-reduced enzyme of a high

Distribution of reducing equivalents on xanthine oxidase molecules and the rates of the intramolecular electron-transfer reactions.

When xanthine oxidase turns over 1-methylxanthine aerobically at pH8.2, the time-sequence in development of its electron-paramagnetic-resonance signals is not primarily due to slow intramolecular reactions among its centres. It derives instead from gross differences in electron distribution within enzyme molecules reduced by the substrate in comparison with those that have subsequently been partly reoxidized by one-electron reaction with oxygen.

Turkey liver xanthine dehydrogenase: properties of the enzyme dependent on the content of functional active sites.

Turkey liver xanthine dehydrogenase containing the full complement of molybdenum, flavin and iron-sulphur prosthetic groups is, as normally isolated, a mixture of functional and non-functional enzyme. The latter apparently lacks the cyanolysable persulphide groups essential to the oxidation of xanthine and to interaction with arsenite. These groups are not required for the oxidation of NADH by Methylene Blue. That KI treatment effects a differential release of flavin from xanthine-prereduced and NADH-prereduced enzyme merely reflects the degree of functionality of the preparations used and may

Design, synthesis, and evaluation of N-substituted indolyl-diazine derivatives as potent xanthine oxidase inhibitors.

Xanthine oxidase (XO) is a key enzyme in purine metabolism and a well-established target for treating hyperuricemia and gout. However, clinically used XO inhibitors are limited by suboptimal efficacy and adverse effects, underscoring the necessity for novel scaffolds. N-substituted indolyl-heterocycle scaffold has emerged as an effective and promising framework for XO inhibition, with isoxazole incorporation in our previous study shown to yield derivatives exhibiting favorable activity in vitro. In this study, we further investigated this scaffold by systematically introducing a series of six-

Effects of Smilax China L. extracts on Hyperuricemia chicken model via inhibiting xanthine oxidase activity.

Hyperuricemia (HUA) is a metabolic disorder caused by excessive production of uric acid (UA) or impaired uric acid metabolism. Smilax China L. has a wide range of pharmacological activities such as immunomodulatory, anti-inflammatory, and antioxidant. Its roots and rhizomes have been widely used for the treatment of HUA. However, its mechanisms for treating HUA and reducing renal impairment have not been fully elucidated. In the present study, we evaluated the effect of Smilax China L. extract (SC) on UA metabolism and further explored its mechanism of action by feeding a high-calcium and high

Synthesis, Crystal Structures, and Biological Activity Evaluation of Novel Xanthine Derivatives Containing a Pyrethroid Moiety.

On the basis of the structures of natural xanthines and pyrethroid insecticides, a series of novel xanthine derivatives Ia-Is containing pyrethroid motifs were synthesized and identified by means of melting points, 1H NMR, 13C NMR, and HRMS. The single crystals of compounds In and Iq were obtained, which further confirmed the structures and configurations of this type of compounds. The biological tests showed that some of them exhibited favorable insecticidal activities toward Mythimna separata Walker and Plutella xylostella L. and were superior to the natural methylxanthine compound caffeine

Xanthine Oxidase and Transforming Growth Factor Beta-activated Kinase 1: Potential Targets for Gout Intervention.

BACKGROUND: Gout, inflammatory arthritis caused by the deposition of monosodium urate crystals into affected joints and other tissues, has become one of the major health problems of today's world. The main risk factor for gout is hyperuricemia, which may be caused by excessive or insufficient excretion of uric acid. The incidence is usually in the age group of 30- 50 years, commonly in males. In developed countries, the incidence of gout ranges from 1 to 4%. Despite effective treatments, there has been an increase in the number of cases over the past few decades. OBJECTIVE: In recent years, t

Codeine exerts cardiorenal injury via upregulation of adenine deaminase/xanthine oxidase and caspase 3 signaling.

AIMS: Codeine treatment has been shown to be associated with glucolipid deregulation, though data reporting this are inconsistent and the mechanisms are not well understood. Perturbation of glutathione-dependent antioxidant defense and adenosine deaminase (ADA)/xanthine oxidase (XO) signaling has been implicated in the pathogenesis of cardiometabolic disorders. We thus, hypothesized that depletion of glutathione contents and upregulation of ADA/XO are involved in codeine-induced glucolipid deregulation. The present study also investigated whether or not codeine administration would induce geno

Xanthine oxidase inhibitory peptides derived from tuna protein: virtual screening, inhibitory activity, and molecular mechanisms.

BACKGROUND: There has been growing interest in the use of xanthine oxidase (XO) as a therapeutic agent to prevent gout and hyperuricemia. In the present study, XO inhibitory peptides were identified from tuna protein by virtual screening, and molecular docking was used to elicit the interaction mechanism between XO and peptides. RESULTS: A novel tetrapeptide, EEAK, exhibited high XO inhibitory activity with an IC50 of 173.00 ± 0.06 μM. Molecular docking analysis revealed that EEAK bound with the pivotal residues of XO's active sites (i.e., Glu802, Arg880, Glu1261) through two conventional hyd

Acute lung injury by endotoxin-induced mediators: prevention by HWA 138, a new xanthine derivative.

We attempted to mimic septic conditions in vitro by using a model of isolated perfused rabbit lung (IPRL) and evaluated the effects of endotoxin or endotoxin-induced mediators (or both) on it. Moreover, we determined the salutary effects of HWA 138, a new xanthine derivative, against endotoxin-related lung injury. To study this, heparinized human blood was centrifuged, following which the plasma complement was inactivated by heat treatment and the isolated and washed buffy coat cells were then added to it. This was followed by incubation of aliquot suspension with and without endotoxin (lipopo

Exploring the relationship between XDH gene variants and serum urate and xanthine dehydrogenase levels in gout.

BACKGROUND: Gout is a type of arthritis caused by high levels of urate in the body, leading to inflammation and joint pain. Xanthine dehydrogenase (XDH) is a key enzyme in urate biosynthesis, and genetic variations in its XDH gene can influence the amount or activity of the enzyme, as well as the levels of urate produced. OBJECTIVES: This study aimed to explore the association of two single nucleotide variants (SNVs) in the XDH gene with gout susceptibility in the Mexican population, as well as their correlation with urate and XDH levels. METHODS: Genotyping of rs17323225 and rs17011368 XDH

Xanthine oxidase inhibitor allopurinol preserves cardiac function after experimental malocclusion induced by occlusal disharmony in mice.

Oxidative stress caused by poor oral condition is associated with systemic diseases, including cardiovascular disease. In this work, therefore, we examined the effect of allopurinol, an inhibitor of the reactive oxygen species (ROS)-producing enzyme xanthine oxidase, on cardiac dysfunction in our bite-opening (BO) mouse model, in which a suitable appliance is cemented onto the mandibular incisior. After two weeks, we confirmed that cardiac function was significantly decreased in the BO group compared to the control, while allopurinol ameliorated the dysfunction. The impairment of cardiac funct

Febuxostat facilitates neovasculogenesis in chronic kidney disease through xanthine oxidase/NADPH oxidase/c-Jun signaling pathways.

The accumulation of uremic toxins in circulation contributes to the cardiovascular diseases that result from chronic kidney disease (CKD). Indoxyl sulfate (IS), which is a protein-bound uremic toxin, promotes cardiovascular diseases with impaired neovascularization by increasing the reactive oxygen species (ROS). This study aimed to investigate febuxostat, a potent xanthine oxidase (XO) inhibitor, for its potential effects on the mechanisms of neovasculogenesis in CKD. CKD mice were generated by 5/6 subtotal nephrectomy and orally administered with febuxostat. Human aortic endothelial cells (H

Xanthine Oxidase-Dependent Activation of NLPR3 Inflammasome in Epithelial Cells Sustains Inflammation in Inflammatory Bowel Disease.

IMPORTANCE AND OBJECTIVE: Xanthine oxidase (XO) plays a key role in purine metabolism, catalyzing the oxidation of hypoxanthine to xanthine, and xanthine to uric acid (UA) with the production of superoxide anions. The accumulation of UA has been shown to initiate the inflammatory process through NLRP3 inflammasome, and the production of reactive oxygen species (ROS) contributes to inflammation-related tissue damage in different organs. This study aimed to investigate the role of XO in the pathogenesis of Inflammatory Bowel Disease (IBD) and the anti-inflammatory potential of XO inhibition. DE

Discovery of 4-(isopentyloxy)-3-nitrobenzamide derivatives as xanthine oxidase inhibitors through a non-anthraquinone exploration.

In our previous study, we reported a series of N-(9,10-anthraquinone-2-carbonyl) amino acid derivatives as novel inhibitors of xanthine oxidase (XO). Recognizing the suboptimal drug-like properties associated with the anthraquinone moiety, we embarked on a nonanthraquinone medicinal chemistry exploration in the current investigation. Through systematic structure-activity relationship (SAR) studies, we identified a series of 4-(isopentyloxy)-3-nitrobenzamide derivatives exhibiting excellent in vitro potency against XO. The optimized compound, 4-isopentyloxy-N-(1H-pyrazol-3-yl)-3-nitrobenzamide

Antiplatelet Effects of Selected Xanthine-Based Adenosine A2A and A2B Receptor Antagonists Determined in Rat Blood.

The platelet aggregation inhibitory activity of selected xanthine-based adenosine A2A and A2B receptor antagonists was investigated, and attempts were made to explain the observed effects. The selective A2B receptor antagonist PSB-603 and the A2A receptor antagonist TB-42 inhibited platelet aggregation induced by collagen or ADP. In addition to adenosine receptor blockade, the compounds were found to act as moderately potent non-selective inhibitors of phosphodiesterases (PDEs). TB-42 showed the highest inhibitory activity against PDE3A along with moderate activity against PDE2A and PDE5A. The

Flavonoid extract of saffron by-product alleviates hyperuricemia via inhibiting xanthine oxidase and modulating gut microbiota.

Hyperuricemia was associated with the overproduction or inadequate excretion of uric acid, while its association with gut microbiota has emerged although few studies were focused on it. Previously, we have reported a flavonoid extract from saffron floral bio-residues lowered uric acid in potassium oxonate-induced hyperuricemic mice. In this study, the impacts of the flavonoid extract on potassium oxonate-induced hyperuricemic rats were evaluated through its effects on serum, renal, intestinal uric acid, and xanthine oxidase activity. At the same time, the microbial and metabolic features of th
