A river runs through it: intraspecific variability in hotter-drought-related physiological traits across the Mississippi divide in Pinus taeda L.

Human activities are intensifying heatwaves and droughts, threatening forest ecosystems worldwide. However, understanding intraspecific variation in physiological traits that confer resistance to hotter drought is critical for predicting tree responses to climate change. In Pinus taeda L., long-term geographic isolation across the Mississippi River Valley after the last glacial maximum has resulted in distinct population genetic structures, likely reflecting historical adaptation to contrasting climatic conditions in western and eastern refugia. However, it remains unclear whether contemporary

A tracking work on how Sm4CL2 re-directed the biosynthesis of salvianolic acids and tanshinones in Salvia miltiorrhiza hairy roots.

Overexpression and antisense expression of Sm4CL2 re-directed the biosynthesis of salvianolic acids and tanshinones in Salvia miltiorrhiza hairy roots. Danshen (Salvia miltiorrhiza Bunge) is a widely used traditional Chinese medicine and its main active ingredients are water-soluble phenolic acids and lipophilic diterpenoids which are produced through the phenylpropanoid pathway and terpenoid pathway, respectively. 4-Coumaric acid: Coenzyme A ligase (4CL) is a key enzyme in the phenylpropanoid metabolism. We had obtained Sm4CL2-overexpressing (Sm4CL2-OE) and antisense Sm4CL2-expressing (anti-S

Ammonium-nitrogen addition at the seedling stage does not reduce grain cadmium concentration in two common wheat (Triticum aestivum L.) cultivars.

High cadmium (Cd) concentration in common wheat (Triticum aestivum L.) grains poses potential health risks. Several management strategies have been used to reduce grain Cd concentration. However, limited information is available on the use of ammonium-nitrogen (NH4+-N) as a strategy to manage Cd concentration in wheat grains. In this study, NH4+-N addition at the seedling stage unchanged the grain Cd concentration in the high-Cd accumulator, Zhoumai 18 (ZM18), but dramatically increased that in the low-Cd accumulator, Yunmai 51 (YM51). Further analysis revealed that the effects of NH4+-N addit

Spatial gene expression analysis in tomato hypocotyls suggests cysteine as key precursor of vascular sulfur accumulation implicated in Verticillium dahliae defense.

Verticillium dahliae is a prominent generator of plant vascular wilting disease and sulfur (S)-enhanced defense (SED) mechanisms contribute to its in-planta elimination. The accumulation of S-containing defense compounds (SDCs) including elemental S (S(0) ) has been described based on the comparison of two near-isogenic tomato (Solanum lycopersicum) lines differing in fungal susceptibility. To better understand the effect of S nutrition on V. dahliae resistance both lines were supplied with low, optimal or supraoptimal sulfate-S. An absolute quantification demonstrated a most effective fungal

Assessing the Likelihood of Transmission of Candidatus Liberibacter solanacearum to Carrot by Potato Psyllid, Bactericera cockerelli (Hemiptera: Triozidae).

'Candidatus Liberibacter solanacearum' (Lso) is a phloem-limited bacterium that severely affects important Solanaceae and Apiaceae crops, including potato, tomato, pepper, tobacco, carrot and celery. This bacterium is transmitted to solanaceous species by potato psyllid, Bactericera cockerelli, and to Apiaceae by carrot psyllids, including Trioza apicalis and Bactericera trigonica. Five haplotypes of Lso have so far been described, two are associated with solanaceous species and potato psyllids, whereas the other three are associated with carrot and celery crops and carrot psyllids. Little is

Aggregation and toxicity in bacteria of Rep-WH1 prion-like proteins from the phytopathogen Xylella fastidiosa.

Xylella fastidiosa is a global bacterial phytopathogen that colonizes the xylem vessels, leading to their occlusion and thus to plant death. The expression of the amyloidogenic, prion-like WH1 domain of the replication protein RepA from the Pseudomonas savastanoi plasmid pPS10 severely damages Escherichia coli cells. Here, we characterize the WH1 domains in the Rep proteins from the X. fastidiosa plasmids pXF51 and pXF64. The expression of these Rep-WH1 proteins, as happens with the archetypical RepA-WH1 protein from pPS10, decreased the culturability of E. coli in vitro, promoting dormancy. E

Characterization of physiochemical and anatomical features associated with enhanced phytostabilization of copper in Bruguiera cylindrica (L.) Blume.

Copper is an essential micronutrient for normal plant metabolism and it is involved in number of physiological processes in plants but at the same time, at concentrations above threshold level, it acts as a potential stress factor. In this study, the phytoremediation potential of Bruguiera cylindrica (L.) Blume with respect to Cu was evaluated for the first time. Various physiochemical and anatomical parameters were analyzed in three-month-old healthy plantlets of B. cylindrica on exposure to different concentrations of CuSO4 (0, 0.05, 0.15, and 0.25 mM)for 20 d. Higher uptake and accumulation

Physiological and Metabolic Changes Induced by Fullerene C60 Derivatives in Zinc-Stressed Cucumber.

Zinc (Zn) in excess is very toxic for plants and can limit agriculture. Carbon-based engineered nanomaterials with high electron mobility and electron-accepting capability may be essential for mitigating heavy metal stress. In the present study, the protective role of some fullerene C60 derivatives (fullerenol [C60(OH)22-24] and the arginine C60 [C60(C6H13N4O2)8H8]) were tested for the first time against Zn toxicity in Cucumis sativus L. (cucumber). Plants were grown hydroponically at three concentrations of fullerenes (0, 2, and 10 mg L-1) without or with 40 µM Zn for 17 days. Plant growth, l

Chitosan and copper nanoparticles in vase solutions elevate the quality and longevity of cut tulips, setting a new standard for sustainability in floriculture.

Nanoparticles present innovative solutions for postharvest applications, allowing for the development of compounds that effectively extend the vase life of cut flowers by reducing ethylene production and preventing bacterial growth. As a leading choice among cut flowers, Tulips encounter common issues such as neck bending and a limited display life, which can severely impact their marketability. This experiment aims to evaluate the effectiveness of nano chitosan (CHS-NPs) at 3.5 and 7.0 mg L− 1, nano copper (Cu-NPs) at 15 and 30 mg L− 1, and Cymbopogon flexuosus essential oil (LG) at 150 and 3

Transcriptome Analyses in Adult Olive Trees Indicate Acetaldehyde Release and Cyanide-Mediated Respiration Traits as Critical for Tolerance against Xylella fastidiosa and Suggest AOX Gene Family as Marker for Multiple-Resilience.

Xylella fastidiosa (Xf) is a global bacterial threat for a diversity of plants, including olive trees. However, current understanding of host responses upon Xf-infection is limited to allow early disease prediction, diagnosis, and sustainable strategies for breeding on plant tolerance. Recently, we identified a major complex trait for early de novo programming, named CoV-MAC-TED, by comparing early transcriptome data during plant cell survival with SARS-CoV-2-infected human cells. This trait linked ROS/RNS balancing during first hours of stress perception with increased aerobic fermentation co

Seasonal shifts in water-source partitioning and fine-root overlap in a Populus simonii-maize shelterbelt system.

INTRODUCTION: Farmland shelterbelts are critical for agricultural production in arid and semiarid regions, but they may compete with crops for limited soil water. This study aimed to quantify seasonal water-source use, soil moisture dynamics, and fine-root distribution in a Populus simonii-maize shelterbelt system. METHODS: The study was conducted in Zhangbei County, northwestern Hebei Province, China. Three land-use conditions were compared: a pure P. simonii shelterbelt (P), a P. simonii-maize intercropping system (PC), and a maize monoculture reference (C). Synchronous sampling was conduct

Exogenous calcium modulates oxalic acid metabolism via repression of LcAAE3 and mediates calcium accumulation in litchi pericarp and fruit pedicel.

Calcium plays a crucial role in fruit development and quality formation, yet the regulatory mechanisms underlying calcium distribution and oxalic acid metabolism in litchi remain poorly understood. In this study, the effects of exogenous calcium on calcium accumulation, fruit quality, and oxalic acid metabolism in litchi were investigated through stem infusion experiments using calcium chloride (CaCl2) and ethylene glycol-bis(2-aminoethylether)-N,N,N',N'-tetraacetic acid (EGTA) treatments. The results demonstrated that exogenous CaCl2 application significantly enhanced calcium accumulation in

5-Aminolevulinic acid improves strawberry salt tolerance through a NO-H2O2 signaling circuit regulated by FaWRKY70 and FaWRKY40.

INTRODUCTION: 5-Aminolevulinic acid (ALA) is an essential biosynthetic precursor of tetrapyrrole compounds, naturally occurring in all living organisms. It has also been suggested as a new plant growth regulator. Treatment with ALA promotes strawberry Na+ homeostasis under salt stress. Regulation of this process requires the signaling molecules nitric oxide (NO) and hydrogen peroxide (H2O2), but the specific signaling cascade and transcriptional regulatory mechanism have not previously been characterized. OBJECTIVES: Our work focused on the dissection of the NO and H2O2 signaling cascade and

1H-NMR Metabolite Fingerprinting Analysis Reveals a Disease Biomarker and a Field Treatment Response in Xylella fastidiosa subsp. pauca-Infected Olive Trees.

Xylella fastidiosa subsp. pauca is a xylem-limited bacterial phytopathogen currently found associated on many hectares with the "olive quick decline syndrome" in the Apulia region (Southern Italy), and the cultivars Ogliarola salentina and Cellina di Nardò result in being particularly sensitive to the disease. In order to find compounds showing the capability of reducing the population cell density of the pathogen within the leaves, we tested, in some olive orchards naturally-infected by the bacterium, a zinc-copper-citric acid biocomplex, namely Dentamet®, by spraying it to the crown, once pe

Phloem small RNAs, nutrient stress responses, and systemic mobility.

BACKGROUND: Nutrient availabilities and needs have to be tightly coordinated between organs to ensure a balance between uptake and consumption for metabolism, growth, and defense reactions. Since plants often have to grow in environments with sub-optimal nutrient availability, a fine tuning is vital. To achieve this, information has to flow cell-to-cell and over long-distance via xylem and phloem. Recently, specific miRNAs emerged as a new type of regulating molecules during stress and nutrient deficiency responses, and miR399 was suggested to be a phloem-mobile long-distance signal involved i

Spatiotemporal reprogramming of auxin signaling and resistance outputs across model plants and solanaceae crops under Ralstonia solanacearum infection.

Bacterial wilt is a devastating vascular disease caused by the soil-borne bacterium Ralstonia solanacearum, which invades and blocks the xylem of susceptible plants, especially economically important solanaceous crops. In addition to classical plant defense hormones, recent research has focused on understanding the role of auxin as a key and continuous regulator of infection and development. Auxin connects changes during infection to root development and the progression of bacterial wilt disease. The auxin related shift in plant hormone patterns associated with root development has been observ

Physiological disruption, structural deformation and low grain yield induced by neonicotinoid insecticides in chickpea: A long term phytotoxicity investigation.

Arbitrary use of insecticides in agricultural practices cause severe environmental hazard that adversely affects the growth and productivity of edible crops. Considering theses, the aim of the present study was to evaluate the toxicological potential of two neonicotinoid insecticides, imidacloprid (IMID) and thiamethoxam (THIA) using chickpea as a test crop. Application of insecticides at three varying doses revealed a gradual decrease in biological performance of chickpea plants which however, varied noticeably among insecticides and their doses. Significant (P ≤ 0.05) decline in germination

Spatial metabolomics reveals brassinosteroid-mediated compartmentalization of saikosaponin biosynthesis in Bupleurum chinense DC. under abiotic stress.

Although the biosynthesis of active ingredients in medicinal plants is co-regulated by diverse environmental stimuli, the endocrine mechanism by which brassinosteroids (BRs) coordinate physiological resilience with the in situ spatial distribution of saikosaponins in Bupleurum chinense DC. under combined drought and salinity remains largely elusive. We hypothesized that exogenous brassinosteroids (BRs) application alleviates oxidative impairment provoked by single and combined osmotic/ionic stresses while orchestrating the tissue-level topological routing of saikosaponin biosynthesis. Comparative genomics reveal signatures of ecological specialization in the striped ambrosia beetle Trypodendron lineatum.

BACKGROUND: Beetles (Coleoptera) display exceptional dietary diversity and occupy a wide range of ecological niches, often involving close associations with plants and microbes. Ambrosia beetles (Curculionidae; Scolytinae and Platypodinae) exemplify ecological specialization by cultivating mutualistic fungi within galleries excavated in their host trees' xylem, with the fungi serving as their main food source. The striped ambrosia beetle Trypodendron lineatum is a pest of conifers, relying on its nutritional mutualist Phialophoropsis ferruginea for survival. This fungiculture-based lifestyle p

Morpho-anatomical changes and physio-biochemical responses of Carex siderosticta under water stress.

Carex siderosticta Hance is an advantageous understory and lawn greening resource due to its strong environmental adaptability and ornamental value, but extreme climates render soil moisture a key growth-limiting factor. Thus, this study explored the response strategies of C. siderosticta to drought and flooding via pot water control and double-pot flooding simulations. Results showed that all the water stresses significantly inhibited the growth of C. siderosticta with increasing stress intensity; leaf and root biomass, water content and root activity decreased gradually, and root architectur
