Postherpetic neuralgia: irritable nociceptors and deafferentation.

Postherpetic neuralgia (PHN) is a common and often devastatingly painful condition. It is also one of the most extensively investigated of the neuropathic pains. Patients with PHN have been studied using quantitative testing of primary afferent function, skin biopsies, and controlled treatment trials. Together with insights drawn from an extensive and growing literature on experimental models of neuropathic pain these patient studies have provided a preliminary glimpse of the pain-generating mechanisms in PHN. It is clear that both peripheral and central pathophysiological mechanisms contribut

[Afferent projections to the uvula in the cat. I. Climbing fiber projections].

The inferior olive afferents to the cerebellar uvula were studied by means of retrograde axonal transport of horseradish peroxidase (HRP) in the cat. Following large and small injections of HRP into the uvula, the distributions of labeled cells in the inferior olive were investigated. As to the climbing fiber projection, it is revealed that there are six longitudinal parasagittal zone extending throughout the dorsal and ventral uvula. That is, the caudal aspect of the nucleus beta projects to a most medially oriented zone (caudal beta zone), the rostral aspect of the nucleus beta projects to a

Dopamine-induced ventilatory depression in the rat, mediated by carotid nerve afferents.

In pentobarbitone-anesthetized rats, i.v. injections of dopamine (DA) at (10-8)-(10-5 g/kg led to transient ventilatory depression, usually not associated with changes in systemic arterial pressure. DA-induced ventilatory depression consisted of decreases in tidal volume and respiratory frequency in 12 rats, and in bradypnea without changes in tidal volume in 5 rats. After bilateral section of the carotid nerves, ventilatory responses to DA were abolished or greatly diminished. It is concluded that DA inhibits carotid body chemosensory discharges in the rat. The possibility that chemosensory a

Cortico-prefrontal afferents in the guinea pig.

The prefrontal cortex of the guinea pig was investigated in a search for afferents from cortical areas outside the frontal lobe using the horseradish peroxidase retrograde tracing technique. Based on afferents from the medial, the lateral, and from both parts of the thalamic mediodorsal nucleus, three prefrontal subregions were defined, namely the sulcal (within and above the rhinal sulcus), the medial (anterior and superior to the genu of the corpus callosum), and the fronto-polar subfield. Only the fronto-polar region was found to receive afferents from insular, cingulate, retrosplenial, par

TSH secretion following acute heat exposure, in intact and in hypothalamic deafferentated male rats.

Male rats, intact (N) or with complete, anterior, or posterior hypothalamic deafferentations (CHD, AHD, or PHD, respectively), were acutely exposed to an environmental temperature of 36 degrees C, and serum TSH concentrations were determined by RIA. In intact animals, 30 min of exposure to elevated environmental temperature resulted in decreased serum TSH levels. In both CHD and AHD animals, basal serum TSH concentrations were reduced, and heat exposure produced further decreases. In the PHD group, no significant difference in resting serum TSH concentrations was observed, as compared to intac

Neck muscle afferents influence oromotor and cardiorespiratory brainstem neural circuits.

Sensory information arising from the upper neck is important in the reflex control of posture and eye position. It has also been linked to the autonomic control of the cardiovascular and respiratory systems. Whiplash associated disorders (WAD) and cervical dystonia, which involve disturbance to the neck region, can often present with abnormalities to the oromotor, respiratory and cardiovascular systems. We investigated the potential neural pathways underlying such symptoms. Simulating neck afferent activity by electrical stimulation of the second cervical nerve in a working heart brainstem pre

Primate models of postural disorders.

Dorsal column lesions in the high cervical region of the monkey result in severe defects of movements projected into space and contactual orienting reactions of the forelimbs. The hindlimbs are less affected provided a pathway through the lateral columns, Morin's tract, remains intact. Interruption of this pathway results in a defect of hindlimb function similar to that of the forelimbs. Cerebellar ablations in monkeys result in postural and movement disorders, including hypotonia of limb extensor muscles. An important mechanism underlying the hypotonia is a depression of the responses to musc

Long-Term Depression Induced by Optogenetically Driven Nociceptive Inputs to Trigeminal Nucleus Caudalis or Headache Triggers.

The peripheral trigeminovascular pathway mediates orofacial and craniofacial pain and projects centrally to the brainstem trigeminal nucleus caudalis (TNc). Sensitization of this pathway is involved in many pain conditions, but little is known about synaptic plasticity at its first central synapse. We have taken advantage of optogenetics to investigate plasticity selectively evoked at synapses of nociceptive primary afferents onto TNc neurons. Based on immunolabeling in the trigeminal ganglia, TRPV1-lineage neurons comprise primarily peptidergic and nonpeptidergic nociceptors. Optical stimulat

Intermittent phrenic afferent activation induces phrenic motor plasticity.

Approaches such as electrical stimulation of the phrenic neuromuscular system are currently used in humans to manage breathing impairments. We recently showed that electrical stimulation of the diaphragm muscle induces phrenic afferent-dependent plasticity. These findings highlight the need to better understand the mechanisms of afferent-mediated plasticity to guide and improve the development of novel therapies using electrical stimulation. Here, we tested the hypothesis that intermittent activation of large-diameter phrenic afferents induces phrenic motor plasticity, which, like other forms

Possible neural network mediating jaw opening during prey-catching behavior of the frog.

The prey-catching behavior of the frog is a complex, well-timed sequence of stimulus response chain of movements. After visual analysis of the prey, a size dependent program is selected in the motor pattern generator of the brainstem. Besides this predetermined feeding program, various direct and indirect sensory inputs provide flexible adjustment for the optimal contraction of the executive muscles. The aim of the present study was to investigate whether trigeminal primary afferents establish direct contacts with the jaw opening motoneurons innervated by the facial nerve. The experiments were

Vagal afferent innervation of the proximal gastrointestinal tract mucosa: chemoreceptor and mechanoreceptor architecture.

The vagus nerve supplies low-threshold chemo- and mechanosensitive afferents to the mucosa of the proximal gastrointestinal (GI) tract. The absence of a full characterization of the morphology and distributions of these projections has hampered comprehensive functional analyses. In the present experiment, dextran (10K) conjugated with tetramethylrhodamine and biotin was injected into the nodose ganglion and used to label the terminal arbors of individual vagal afferents of both rats and mice. Series of serial 100-μm thick sections of the initial segment of the duodenum as well as the pyloric a

Neural inputs of the hypothalamic "aggression area" in the rat.

A part of the mediobasal hypothalamus (known as hypothalamic attack area) plays a central role in the control of aggressive behavior as its electrical stimulation reliably and rapidly elicits biting attacks in cats and rodents. The efferent connections of this brain region were described in rats, but afferent pathways were not investigated so far. We injected the retrograde tracer cholera toxin B subunit into the mediobasal hypothalamus of male Wistar rats and studied the distribution of labeled cells by immunohistochemical method. The retrograde labeling outlined three continuous, distinct af

Encoding of the cough reflex.

Coughing can be both voluntarily induced and involuntarily initiated by activation of vagal afferent nerves innervating the airways and lungs. Centrally, cough is regulated at the level of the brainstem through integration of vagal afferent nerve input by relay neurones in the nucleus tractus solitarius (nTS). Projections to and from the nTS add further complexity to cough regulation, as do the profound influences of psychological and social factors known to regulate cough. Peripherally, both neuronal and non-neuronal elements in the airways regulate the excitability of the vagal afferent nerv

A neurologic basis for the overactive bladder.

OBJECTIVES: The functions of the lower urinary tract (LUT) to store and periodically release urine are dependent on neural circuits in the brain and spinal cord. This paper reviews the central neural control of micturition and how disruption of this control can lead to bladder overactivity and incontinence. METHODS: Neuroanatomic, electrophysiologic, and pharmacologic techniques have provided information about the neural circuitry and the neurotransmitters involved in the central nervous control of voiding. Experimental models of neural injury, including spinal cord transection, cerebral infa

Magnetic stimulation of the sacral roots.

Magnetic stimulation using an external surface coil induces an electrodynamic field that penetrates various tissues and stimulates peripheral nerves in a similar fashion to conventional electrical stimulation. An 83 mm magnetic surface coil was used to stimulate 11 spinal cord injury (SCI) patients, during which time detrusor activity and evoked potentials of the striated urinary sphincter motor pathways were evaluated. All patients had urodynamic studies and conventional sacral evoked potentials prior to magnetic stimulation. The mean bladder capacity was 337 ml (range 109-590), mean leak poi

Neural mechanisms of the reflex inhibition and excitation of gastric motility elicited by acupuncture-like stimulation in anesthetized rats.

The effects of acupuncture-like stimulation of the various segmental areas on gastric motility were examined in anesthetized rats. An acupuncture needle (diameter 340 microns) was inserted into the skin and underlying muscles at a depth of 4-5 mm and was twisted right and left once every second for 60 s. Gastric motility in the pyloric region was measured with the balloon method. Gastric motility was inhibited by acupuncture-like stimulation applied to the abdomen and lower chest region, and was often excited when the limbs were stimulated, in all cases in which stimuli were delivered to the s

Sensory signals mediating high blood pressure via sympathetic activation: role of adipose afferent reflex.

Blood pressure regulation in health and disease involves a balance between afferent and efferent signals from multiple organs and tissues. Although there are numerous reviews focused on the role of sympathetic nerves in different models of hypertension, few have revised the contribution of afferent nerves innervating adipose tissue and their role in the development of obesity-induced hypertension. Both clinical and basic research support the beneficial effects of bilateral renal denervation in lowering blood pressure. However, recent studies revealed that afferent signals from adipose tissue,
