The many pathways driving liver inflammation in MASH.

Metabolic dysfunction-associated steatotic liver disease (MASLD) is the most prevalent chronic liver disease worldwide, affecting one-third of the global population. Most patients exhibit simple steatosis, whereas up to 20% develop metabolic dysfunction-associated steatohepatitis (MASH), potentially culminating in liver cirrhosis and hepatocellular carcinoma. Diverse parallel mechanisms contribute to the development of MASH, which are fueled by hepatic lipotoxicity, intestinal dysbiosis, and pro-inflammatory diets shaping innate and adaptive immune responses. Moreover, adipose tissue is drivin

Effects of mixed traffic flow and time pressure on violated driving behaviors of bus drivers.

This study aimed to examine the effects of mixed traffic flow (MTF) environments with time pressure (TP) on violated driving behaviours (VDBs) of bus drivers. A questionnaire-based survey was conducted among bus drivers. Factor analyses, partial correlation and linear regression analyses were utilised to explore the associations among VDB, MTF and TP. Our results proposed two novel measurements of MTF and TP and one modified VDB scale. Bus drivers who perceived a higher presence of motorcycles and more dangerous interactions among vehicles were less likely to participate in VDB. Those who perc

High endogenous CCL2 expression promotes the aggressive phenotype of human inflammatory breast cancer.

Inflammatory Breast Cancer (IBC) is a highly aggressive malignancy with distinct clinical and histopathological features whose molecular basis is unresolved. Here we describe a human IBC cell line, A3250, that recapitulates key IBC features in a mouse xenograft model, including skin erythema, diffuse tumor growth, dermal lymphatic invasion, and extensive metastases. A3250 cells express very high levels of the CCL2 chemokine and induce tumors enriched in macrophages. CCL2 knockdown leads to a striking reduction in macrophage densities, tumor proliferation, skin erythema, and metastasis. These r

Multifocal cohort analysis unveils cell types associated with regional lymph node seeding in prostate cancer.

BACKGROUND: Understanding the molecular features that underlie metastatic prostate cancer (PCa) is essential to develop prognostic markers and improve treatment decisions. However, such studies are hampered by substantial intratumor and interpatient heterogeneity. METHODS: To cope with this heterogeneity, we propose a unique study design that leverages the statistical power of multifocal bulk transcriptome profiling with the resolution of a single-cell analysis to identify processes and cell types associated with regional metastatic lymph node seeding in PCa. RESULTS: Elaborate analysis of the

Formation of post-traumatic and non-traumatic knee joint contracture: Role of macrophage polarization in joint capsule and skeletal muscle.

PURPOSE: This study investigated how differential macrophage polarization dynamics drive the severity of post-traumatic joint contracture (PTJC) compared to non-traumatic joint contracture (NTJC). METHODS: We first performed a bioinformatic analysis intersecting the human dataset GSE135854 with macrophage polarization gene sets to identify core pathogenic targets. Subsequently, rat models of surgically-induced post-traumatic contracture and immobilization-only non-traumatic contracture were established. This study assessed temporal changes in joint range of motion, histopathological fibrosis,

NG2-ITGA4 axis regulates Rho GTPases and leukemic aggressiveness in KMT2A-r B-ALL and is targetable with natalizumab.

KMT2A-rearranged B-cell acute lymphoblastic leukemia (KMT2A-r B-ALL) is an aggressive subtype of leukemia, characterized by high relapse rates, therapy resistance, and poor prognosis. Although CD19-targeted immunotherapies have significantly benefited patients with relapsed/refractory (R/R) disease, relapses remain common and long-term survival is especially poor in KMT2A-r B-ALL patients. We recently identified the membrane-bound proteoglycan NG2 (CSPG4) as direct transcriptional target of KMT2A fusions, with its expression associated with poor prognosis, early relapse, and glucocorticoid res

IL-8 secretion in primary cultures of prostate cells is associated with prostate cancer aggressiveness.

BACKGROUND: Chronic inflammation is believed to be a major factor in prostate cancer initiation and promotion and has been studied using prostate cancer cells and immortalized cell lines. However, little is known about the contribution of normal cells to the prostatic microenvironment and inflammation. We aim to study the contribution of normal prostate epithelial cells to prostate inflammation and to link the inflammatory status of normal cells to prostate cancer aggressiveness. MATERIALS AND METHODS: Short-term primary cell cultures of normal epithelial prostate cells were derived from pros

Loss of NCOA6 protein causes aggressive prostate cancer with EGFR overexpression and dependency.

Loss of tumor suppressors can drive carcinogenesis while simultaneously creating actionable therapeutic vulnerabilities. Epidermal growth factor receptor (EGFR) is frequently overexpressed in prostate cancer (PCa), yet EGFR inhibitors have shown limited clinical benefit, and the basis for this variability remains unclear. Here, analysis of 1,719 human PCa specimens reveals frequent downregulation of the nuclear receptor coactivator 6 (NCOA6), particularly in metastatic disease. Genetic loss of NCOA6 accelerates tumor growth in human PCa xenografts, induces prostate tumorigenesis in mice, and c

A peritumoral microenvironment engaged by Reg-EXTL3 axis fosters nerve-cancer interactions in pancreatic ductal adenocarcinoma.

Tumor innervation (TIN) and perineural invasion (PNI) are well-established pathological features of pancreatic ductal adenocarcinoma (PDAC) that drive its aggressiveness and associated pain. Here, we reveal that regenerating islet-derived (Reg) proteins, secreted by peritumoral exocrine acinar cells, facilitate TIN and PNI through two paracrine mechanisms. In PDAC cells, Reg proteins drive cancer invasiveness along nerves via autocrine transforming growth factor β (TGF-β) signaling. In neurons, Reg proteins are neurotrophic and potentiate neuronal excitability, resulting in hyperinnervation an

CCNE1: A Cell Cycle Regulator That Influences Tumor Progression.

Cyclin E1 (CCNE1), a pivotal member of the cyclin family, governs the G1/S phase transition of the cell cycle by binding to and activating cyclin-dependent kinase 2 (CDK2), thereby initiating DNA replication and driving S-phase entry. In a broad spectrum of human malignancies-including breast, ovarian, gastric, and non-small cell lung cancers-amplification or overexpression of CCNE1 disrupts the orderly regulation of cell cycle progression, leading to uncontrolled proliferation, impaired DNA damage repair, and genomic instability. These oncogenic consequences are intimately associated with inc

The Role and Function of Non-Coding RNAs in Cholangiocarcinoma Invasiveness.

Cholangiocarcinoma (CCA) is an aggressive tumor that originates from the epithelial cells of the bile duct and has the ability to metastasize to the liver or lymph nodes at an early stage. CCA metastasis represents a complex, multi-stage cascade process. Among these stages, the acquisition of invasiveness by CCA cells is a critical prerequisite for metastatic progression. Elucidating the molecular mechanisms driving CCA cell invasiveness is critical for advancing our knowledge in this field. Emerging evidence highlights the critical role of non-coding RNAs (ncRNAs), including microRNAs (miRNAs

Oncolytic virotherapy induced CSDE1 neo-antigenesis restricts VSV replication but can be targeted by immunotherapy.

In our clinical trials of oncolytic vesicular stomatitis virus expressing interferon beta (VSV-IFNβ), several patients achieved initial responses followed by aggressive relapse. We show here that VSV-IFNβ-escape tumors predictably express a point-mutated CSDE1P5S form of the RNA-binding Cold Shock Domain-containing E1 protein, which promotes escape as an inhibitor of VSV replication by disrupting viral transcription. Given time, VSV-IFNβ evolves a compensatory mutation in the P/M Inter-Genic Region which rescues replication in CSDE1P5S cells. These data show that CSDE1 is a major cellular co-f

Magnetic fields mimic the behavioral effects of REM sleep deprivation in humans.

The discovery of rapid eye movement (REM) sleep by Aserinsky and Kleitman in 1953 initiated the impetus for sleep research and specifically the investigations of the effects of REM sleep deprivation (RSD) on animal and human behavior. The behavioral effects of RSD include the enhancement of motivational and "drive"-related behaviors. In laboratory animals, RSD has been reported to increase appetite, sexual behavior, aggressiveness, and locomotor activity. Moreover, RSD reportedly improves mood in patients with endogenous depression and heightens appetite and sexual interest in normal subjects.

Clathrin recruitment and assembly of phosphorylated B cell receptor is impaired in human diffuse large B cell lymphoma.

The B cell receptor (BCR) drives the differentiation of naive B cells into activated plasma cells that produce antibodies. The BCR consists of a plasma membrane-bound surface immunoglobulin (sIg) that binds antigens, and two coreceptor subunits CD79A and CD79B. The intracellular domains of CD79A and CD79B initiate immune signaling and endocytosis of the receptor in healthy B cells. In diffuse large B cell lymphoma (DLBCL), an aggressive form of human blood cancer, the activated B cell like (ABC) subtype exhibits constitutive signaling that drives survival and proliferation. Nuclear factor I/B: Duality in action in cancer pathophysiology.

The nuclear factor I (NFI) family of transcription factors plays a decisive role in organ development and maturation. Their deregulation has been linked with various diseases, most notably cancer. NFIB stands apart from the other NFI family members given its unique ability to drive both tumor suppressive and oncogenic programs. Thus, the ultimate impact of deregulated NFIB signaling is cancer-specific and strongly influenced by an intricate network of upstream regulators and downstream effectors. Deciphering the events that drive NFIB's paradoxical roles within these networks will enable us to

Targeting PIN1 exerts potent antitumor activity in pancreatic ductal carcinoma via inhibiting tumor metastasis.

The human prolyl isomerase PIN1, best known for its association with carcinogenesis, has recently been indicated in the disease of pancreatic ductal adenocarcinoma (PDAC). However, the functions of PIN1 and the feasibility of targeting PIN1 in PDAC remain elusive. For this purpose, we examined the expression of PIN1 in cancer, related paracarcinoma and metastatic cancer tissues by immunohistochemistry and analyzed the associations with the pathogenesis of PDAC in 173 patients. The functional roles of PIN1 in PDAC were explored in vitro and in vivo using both genetic and chemical PIN1 inhibitio

CAF-derived PTGDS drives pancreatic cancer neuroendocrine differentiation and chemoresistance via PAQR9-MAPK.

The tumor microenvironment, particularly cancer-associated fibroblasts (CAFs), critically influences pancreatic ductal adenocarcinoma (PDAC) progression. Neuroendocrine differentiation (NED) is associated with aggressive tumor behavior and therapy resistance in multiple malignancies, yet its regulation within the PDAC environment is poorly characterized. Through integrated analysis of clinical specimens and transcriptomic datasets, we identified significant enrichment of NED markers correlating with poor patient outcomes. Single-cell RNA sequencing further identified Glycoprotein prostaglandin

DNMT3A Cooperates with YAP/TAZ to Drive Gallbladder Cancer Metastasis.

Gallbladder cancer (GBC) is an extremely lethal malignancy with aggressive behaviors, including liver or distant metastasis; however, the underlying mechanisms driving the metastasis of GBC remain poorly understood. In this study, it is found that DNA methyltransferase DNMT3A is highly expressed in GBC tumor tissues compared to matched adjacent normal tissues. Clinicopathological analysis shows that DNMT3A is positively correlated with liver metastasis and poor overall survival outcomes in patients with GBC. Functional analysis confirms that DNMT3A promotes the metastasis of GBC cells in a man

A Multiscale Map of the Stem Cell State in Pancreatic Adenocarcinoma.

Drug resistance and relapse remain key challenges in pancreatic cancer. Here, we have used RNA sequencing (RNA-seq), chromatin immunoprecipitation (ChIP)-seq, and genome-wide CRISPR analysis to map the molecular dependencies of pancreatic cancer stem cells, highly therapy-resistant cells that preferentially drive tumorigenesis and progression. This integrated genomic approach revealed an unexpected utilization of immuno-regulatory signals by pancreatic cancer epithelial cells. In particular, the nuclear hormone receptor retinoic-acid-receptor-related orphan receptor gamma (RORγ), known to driv
