Rapid GC-MS confirmation of amphetamines in urine by extractive acylation.

Amphetamine and related derivatives are widely abused central- and psychostimulants. Detection of certain derivatives, such as methcathinone, by commonly available immunoassay screening techniques is insufficient. Multi-analyte confirmations for amphetamine type stimulants are therefore required, but traditional gas chromatography-mass spectrometry methods necessitate lengthy analytical procedures with prolonged sample turn-around times. A validated rapid GC-MS assay for urinary confirmation of amphetamine, methamphetamine, methcathinone, ephedrine, norephedrine, methylenedioxyamphetamine, met

Naltrexone for the treatment of amphetamine dependence: a randomized, placebo-controlled trial.

OBJECTIVE: Currently there is no approved pharmacotherapy for amphetamine dependence. Recent human laboratory studies have demonstrated that naltrexone modulates some of the reinforcing effects of amphetamine. The aim of this study was to investigate the efficacy of naltrexone in comparison with placebo in reducing relapse to amphetamine use in amphetamine-dependent patients. METHOD: Eighty patients who met DSM-IV criteria for amphetamine dependence were randomized to 12 weeks of double-blind naltrexone (50 mg) or placebo treatment. Patients visited the clinic twice weekly to receive medicati

Effects of D-amphetamine on defensive behaviors related to fear and anxiety.

In rodents, the administration of amphetamine has been associated with increased locomotor activity and stereotypy, and an emerging body of evidence suggests that it also enhances anxiety-like behavior in a number of animal models. Ethoexperimental analyses have outlined an array of defensive behaviors to threat that are responsive to anxiolytic, panicolytic-like and panicogenic agents, suggesting that the characterization of amphetamine effects on defense may provide further insights into the emotionality consequences of this drug. In Experiment 1, intraperitoneal administration of amphetamin

Oxazepam does not modulate the behavioral effects of d-amphetamine in humans.

Benzodiazepines, which are gamma-aminobutyric acid-A (GABA(A)) receptor positive modulators, can block the behavioral effects of psychomotor stimulants. In the present study, the ability of oxazepam, which may have less abuse potential compared to some other benzodiazepines, to attenuate the discriminative-stimulus, subject-rated and psychomotor performance effects of d-amphetamine in humans was determined. Six healthy participants (2 female, 4 male) learned to discriminate 15 mg oral d-amphetamine. After acquiring the discrimination (i.e., > or = 80% correct responding on 4 consecutive days),

Role of nitrergic system in behavioral and neurotoxic effects of amphetamine analogs.

Several amphetamine analogs are potent psychostimulants and major drugs of abuse. In animal models, the psychomotor and reinforcing effects of amphetamine, methamphetamine (METH), 3,4-methylenedioxymethamphetamine (MDMA; Ecstasy), and methylphenidate (MPD; Ritalin) are thought to be dependent on increased extracellular levels of dopamine (DA) in mesocorticolimbic and mesostriatal pathways. However, amphetamine analogs that increase primarily serotonergic transmission, such as p-chloroamphetamine (PCA) and fenfluramine (FEN), have no potential for abuse. High doses of METH, MDMA, PCA, and FEN p

The Neuropsychopharmacology and Toxicology of 3,4-methylenedioxy-N-ethyl-amphetamine (MDEA).

This paper reviews the pharmacology and toxicology of 3,4-methylenedioxy-N-ethylamphetamine (MDEA, "eve"). MDEA is a ring-substituted amphetamine (RSA) like MDMA, its well known N-methyl analog. Both have become very popular substances of abuse in the techno- and house-music scene. They can evoke psychomotor stimulation, mild alterations of perception, sensations of closeness and a positive emotional state as well as sympathomimetic physical effects. At present, the name "ecstasy" is no longer used only for MDMA, but for the whole group of RSAs (MDA, MDMA, MDEA and MBDB) as they are chemically

Striatal dopamine sensitization to D-amphetamine in periadolescent but not in adult rats.

The neurobiological and behavioral facets of adolescence have been poorly investigated in relation to the vulnerability to psychostimulants. Periadolescent (33-43 days) and adult (>70 days) Sprague-Dawley rats underwent a 3-day treatment history with D-amphetamine (AMPH) at 0, 2, or 10 mg/kg (once a day). After a short 5-day-long withdrawal interval, freely moving animals were challenged with a 2-mg/kg AMPH dose and their behavior as well as in vivo intrastriatum dopamine (DA) release in the CNS were assessed. Microdialysis data indicated that AMPH-history periadolescent rats showed a prominen

Distinct accumbal subareas are involved in place conditioning of amphetamine and cocaine.

In considering the heterogeneous function of the nucleus accumbens (NAC), the present work evaluated the conditioned place preference (CPP) after local infusion of d-amphetamine (AMP; 10, 15 microg/side) or cocaine (COC; 50, 100 microg/side) into two subareas of NAC, core and shell. A regular two-compartment CPP apparatus was used to test the place conditioning effects after 6 pairings of drug in one compartment and 6 pairings of vehicle in the other one. Significant CPP was observed with either AMP infused in the core area or COC infused into the shell area. Neither AMP in shell nor COC in co

Conditional control by midazolam and amphetamine in a rapid appetitive discrimination procedure.

The present two experiments examined conditional control by midazolam and amphetamine cues in an appetitive discrimination procedure. In each of two experiments, male Wistar rats were subjected to a small number of two types of training sessions in a conditioning box. During one type of session, a stimulus was consistently followed by food in a magazine, whereas during the other type of session, this very same stimulus was followed by nothing. Groups of rats were injected with either midazolam (0.1 mg/kg, s.c.) or amphetamine (0.5 mg/kg, s.c.), prior to each food-reinforced session, and with s

Changes in the structure of the agonistic behavior of mice produced by D-amphetamine.

The effects of three acute doses of D-amphetamine (0.25, 1.5 and 3 mg/kg) were studied in a model of isolation-induced aggression in male mice. An ethopharmacological analysis of the encounters was carried out, which studied the frequency, total and mean duration of different behavioral categories, including the temporal distribution of attacks and the duration of inter-attack intervals. The results show a reduction in the total and mean duration of the Attack category and an increase in motor activity manifested by longer durations, both total and mean, of Non Social Exploration and shorter I

Repeated binge exposures to amphetamine and methamphetamine: behavioral and neurochemical characterization.

Stimulant psychosis and addiction are most commonly associated with repeated, high-dose binges or runs, typically preceded by a more intermittent pattern of stimulant abuse. We previously reported that rats exposed to an escalating dose-run pattern of amphetamine administration exhibited changes in their behavioral response profile that differed both qualitatively and quantitatively from the response to either acute or intermittent daily treatment. To determine the generality of these effects and characterize further the nature of the behavioral and neurochemical changes of this treatment, rat

Role of behavioral and pharmacological variables in the loss of tolerance to amphetamine hypophagia.

Tolerance to amphetamine-induced hypophagia is lost when drug injections are withdrawn for 4 weeks while milk tests are continued (Wolgin and Hughes 1996). The purpose of this study was to determine whether the loss of tolerance is a function of drug withdrawal per se. Rats made tolerant to amphetamine (2 mg/kg, IP) were assigned to one of three groups. During the next 4 weeks (phase), one group continued to receive amphetamine injections prior to daily milk tests (Before group), one group received drug injections after the milk tests (After group), and one group received injections of saline

Stereotyped behavior: effects of d-amphetamine and methylphenidate in the young rat.

The proclivity of d-amphetamine and methylphenidate to induce perseverative motoric and vocal side effects detracts from the clinical efficacy of these stimulants in the treatment of Attention-Deficit Hyperactivity Disorder (ADHD). In an attempt to develop a model for these deleterious treatment effects, this study explored the behavioral influences exerted by d-amphetamine and methylphenidate in the young laboratory rat. This experiment revealed that doses of these stimulants that typically induce stereotypy provoke diverging behavioral profiles: while animals given 5 mg/kg d-amphetamine exhi

Effects of d-amphetamine on temporal distributions of schedule-induced polydipsia.

Food-deprived rats were divided into four groups according to the equal interval and time durations of a multiple fixed-interval, fixed-time schedule (15, 30, 60, and 120 s). Fixed-time components were signaled by a tone and lever withdrawal. d-Amphetamine (0.25-4.0 mg/kg) produced similar dose-dependent reductions in the drinking and licking induced by fixed-interval and fixed-time schedules. These dose-dependent decrements were a function of the interfood interval length. More licks occurred early in the interfood intervals with doses of d-amphetamine. Dose-dependent shifts to the left were

State-dependency of conditioning and extinction of an appetitive response with amphetamine and midazolam.

Rats received a single conditioning session in which unsignaled food pellets were delivered to the food magazine of a training box, followed by a single extinction session without pellet presentations. Groups of rats differed in the drug state induced prior to conditioning and extinction according to a 2 x 2 design using d-amphetamine sulphate (AMP, 0.5 mg/kg, S.C.) and midazolam (MID, 0.1 mg/kg, S.C.). Subsequently, all rats received nonreinforced test sessions under AMP and MID. The dependent measure was the frequency of magazine visits during selected portions of each session. Rats that rec

Effect of sensitization of stereotypy on the acquisition and retention of tolerance to amphetamine hypophagia.

The purpose of this study was to determine whether prior sensitization of stereotypy interferes with the development and retention of tolerance to amphetamine-induced hypophagia. Rats were given intermittent injections of either amphetamine (2.5 mg/kg) to induce sensitization of stereotypy, or saline. Subgroups from each group then received daily injections of either amphetamine (2 mg/kg) or saline and access to milk for 30 min. Both sensitized and nonsensitized groups became tolerant to drug-induced hypophagia at about the same rate and to about the same extent. Such tolerance was accompanied

Naloxone reduces the neurochemical and behavioral effects of amphetamine but not those of cocaine.

The specific opioid receptor antagonist naloxone modifies the effects of amphetamine in a wide variety of behavioral paradigms. Naloxone also attenuates the amphetamine-induced increase in extracellular dopamine in the brain of rats. Therefore, these experiments were designed to replicate the neurochemical and behavioral interactions between naloxone and amphetamine, and to extend these observations to interactions between naloxone and cocaine. Microdialysis was performed on adult male rats of Sprague-Dawley descent. Rats were pretreated with a subcutaneous injection of 5.0 mg/kg naloxone or v

Effects of naloxone on the behaviors evoked by amphetamine and apomorphine in adult cats.

1. This work was undertaken in order to study whether the opioid system is involved in the modulation of the behaviors induced by two agonists of the dopaminergic system, amphetamine and apomorphine in adult cats. 2. Naloxone, an antagonist of the mu, delta and kappa opioid receptors was administered to twelve female mongrel cats; 0.5, 1.0 and 2.0 mg/kg s.c. were injected in order to analyse its own effect of naloxone. This drug produced NREMs behavior and accordingly the cat showed an overall decrease of its activities. 3. Amphetamine (2.5 mg/kg s.c.) and apomorphine (2.0 mg/kg s.c.) were inj

Environment-specific reinstatement of amphetamine-mediated hyperdipsia by morphine and (-)-norpseudoephedrine.

In a study designed to determine whether environmental and pharmacological stimuli have the ability to take control of amphetamine-mediated hyperdipsia, rats were injected with d,l-amphetamine (AMPH; 4 mg/kg, IP) alone or in combination with (-)-norpseudoephedrine (NPE; 10 mg/kg, IP) and then returned to the home cage or transferred to a distinct environment (test cage). Water intake was measured hourly for 3 h, in the absence of food. AMPH treatment lasted for 10 days, followed by a 6-day extinction phase during which AMPH, but not NPE, injections were discontinued. Subsequently, all animals

Effect of chlorpromazine on mouse ambulatory activity sensitization caused by (+)-amphetamine.

The development of sensitization to the ambulation-increasing effect of (+)-amphetamine (2.5 mg kg-1) was found to be dose-dependently inhibited when 1 or 2 mg kg-1 chlorpromazine was administered concomitantly, and the sensitization to (+)-amphetamine was almost completely suppressed when co-administered with 4 mg kg-1 chlorpromazine. Following a challenge dose of 2.5 mg kg-1 (+)-amphetamine, mice pretreated with (+)-amphetamine alone or with (+)-amphetamine plus 1 or 2 mg kg-1 chlorpromazine showed similar marked enhancement of the sensitization. However, mice that had been given (+)-ampheta
