Strategic Control of Drug-Resistant HIV: Multi-Strain Modeling with Diagnosis, Adherence, and Treatment Switching.

A central challenge in Human Immunodeficiency Virus (HIV) public health policy lies in determining whether to universally expand treatment access, despite the risk of sub-optimal adherence and consequent drug resistance, or to adopt a more strategic allocation of resources that balances treatment coverage with adherence support. This dilemma is further complicated by the need for timely switching to second-line therapy, which is critical for managing treatment failure but imposes additional burdens on limited healthcare resources. In this study, we develop and analyze a compartmental model of

Global dynamics of an age-structured cholera model with saturation incidence and multiple transmission pathways.

Cholera is an acute diarrheal disease caused by the bacterium Vibrio cholerae. With the consideration of the transmission mechanism and heterogeneity of population, an age-structured cholera epidemic model is proposed, involving saturation incidence rates that describe direct and indirect transmission pathways and all class-ages with the susceptible age of susceptible individuals, infection age of infected individuals and biological age of Vibrio cholerae. The focus is to investigate the global dynamics of the model by using the basic reproduction number R 0 . After establishing the well-pose

Epidemiological Consequences of Individual Centrality on Wild Chimpanzees.

Disease outbreaks are one of the key threats to great apes and other wildlife. Because the spread of some pathogens (e.g., respiratory viruses, sexually transmitted diseases, ectoparasites) are mediated by social interactions, there is a growing interest in understanding how social networks predict the chain of pathogen transmission. In this study, we built a party network from wild chimpanzees (Pan troglodytes), and used agent-based modeling to test: (i) whether individual attributes (sex, age) predict individual centrality (i.e., whether it is more or less socially connected); (ii) whether i

BF.7: a new Omicron subvariant characterized by rapid transmission.

OBJECTIVES: BF.7 (BA.5.2.1.7) is a novel sublineage of Omicron BA.5, whose clinical characteristics are not yet established. METHODS: From 28 September 2022 to 3 October 2022, the first 421 patients with BF.7 were assessed in Hohhot China and the clinical data were extracted and analysed. The basic reproduction number (R0) was estimated using a statistical model calculation method. RESULTS: The R0 value was determined to be 13.79 (95% confidence interval: 12.44-15.24). The mean age was 33.43 ± 18.78 years. Asymptomatic, mild, moderate, severe, and critical patients accounted for 12.35% (52/4

On the benefits of flattening the curve: A perspective.

The many variations on a graphic illustrating the impact of non-pharmaceutical measures to mitigate pandemic influenza that have appeared in recent news reports about COVID-19 suggest a need to better explain the mechanism by which social distancing reduces the spread of infectious diseases. And some reports understate one benefit of reducing the frequency or proximity of interpersonal encounters, a reduction in the total number of infections. In hopes that understanding will increase compliance, we describe how social distancing (a) reduces the peak incidence of infections, (b) delays the occ

Vaccination of dogs in an African city interrupts rabies transmission and reduces human exposure.

Despite the existence of effective rabies vaccines for dogs, dog-transmitted human rabies persists and has reemerged in Africa. Two consecutive dog vaccination campaigns took place in Chad in 2012 and 2013 (coverage of 71% in both years) in the capital city of N'Djaména, as previously published. We developed a deterministic model of dog-human rabies transmission fitted to weekly incidence data of rabid dogs and exposed human cases in N'Djaména. Our analysis showed that the effective reproductive number, that is, the number of new dogs infected by a rabid dog, fell to below one through November

A Bayesian inferential approach to quantify the transmission intensity of disease outbreak.

BACKGROUND: Emergence of infectious diseases like influenza pandemic (H1N1) 2009 has become great concern, which posed new challenges to the health authorities worldwide. To control these diseases various studies have been developed in the field of mathematical modelling, which is useful tool for understanding the epidemiological dynamics and their dependence on social mixing patterns. METHOD: We have used Bayesian approach to quantify the disease outbreak through key epidemiological parameter basic reproduction number (R0), using effective contacts, defined as sum of the product of incidence

A multigroup model for a heterosexually transmitted disease.

A multigroup model is considered for a disease transmitted heterosexually in which there is a core group that has higher sexual activity than other groups. In order to develop control strategies to eradicate the disease, type reproduction numbers are determined. A graph-theoretic interpretation of type reproduction numbers for a star network with a core group at the center is established. Using these general formulas for the star network, a two-group model consisting of a core and a non-core group is considered. If disease can persist in the core group in isolation, then the amount of increase

A novel mathematical model of seasonality in Oropouche virus transmission in Amazonas, Brazil.

Oropouche virus (OROV) is a vector-borne arbovirus that has caused more than 500,000 infections across South America, Central America, and the Caribbean. Its primary vector, the midge species Culicoides paraensis, acquires the virus from wild reservoir hosts and transmits it to humans during blood feeding. As outbreaks within the Amazon basin continue to occur and confirmed cases steadily increase, a proper understanding of the interplay between climatic variables, vector behavior, and OROV transmission is needed to develop effective public health interventions. In this paper, we develop a nov

Dynamics of infectious disease transmission in heterogeneous networks considering population interaction mechanism.

In public health emergencies, infectious disease transmission often varies significantly across populations. To investigate population heterogeneity and interaction mechanisms in cross-population transmission dynamics, we develop a community-based SIR (susceptible-infectious-removed) compartmental model applicable to multiple populations. This model is grounded in a heterogeneous network that incorporates both intra-population structural differences and inter-population coupling. Considering the structural characteristics and interactions between the service-providing and the served population

A Kermack-McKendrick type epidemic model with double threshold phenomenon (and a possible application to Covid-19).

The suggestion by K.L. Cooke (1967) that infected individuals become infective if they are exposed often enough for a natural disease resistance to be overcome is built into a Kermack-McKendrick type epidemic model with post-latency age. Both the case that the resistance may be the same for all hosts and the case that it is distributed among the host population are considered. In addition to the familiar threshold behavior of the final size of the epidemic with respect to a basic reproductive number, an Allee effect is generated for the final cumulative force of infection by the final cumulati

A multi-strain sequential super-infection model for dengue fever with antibody dependent enhancement.

Dengue fever is a vector-borne disease that is transmitted by the aedes aegypti mosquito. The disease is caused by four different viral serotypes which are DEN-1, DEN-2, DEN-3, and DEN-4. Of these serotypes, DEN-1 and DEN-2 are the most prevalent, and their co-circulation presents complex disease dynamics due to the antibody-dependent enhancement (ADE) phenomenon as well as the cross-immunity interaction of these viral serotypes. In this study, we formulate a novel multi-strain sequential super-infection model that captures the primary and secondary infection dynamics with temporary cross immu

Estimated transmission dynamics of SARS-CoV-2 variants from wastewater are unbiased and robust to differential shedding.

The COVID-19 pandemic has accelerated the development and adoption of wastewater-based epidemiology. Wastewater samples can provide genomic information for detecting and assessing the spread of SARS-CoV-2 variants in communities and for estimating important epidemiological parameters such as the selection advantage of a viral variant. However, despite demonstrated successes, epidemiological data derived from wastewater suffers from potential biases. Of particular concern are shedding profiles, which can affect the relationship between true viral incidence and viral loads in wastewater. Changes

Advanced ANN-LMB modeling of hepatitis B transmission across sexual networks and its disability burden.

Hepatitis B virus (HBV) remains a major global health concern, with sexual transmission being a key driver among adults. This study develops a gender-stratified compartmental model of HBV spread that integrates long-term disability through gender-specific parameters. A key contribution is the integration of mechanistic modeling with artificial neural networks (ANNs), enabling efficient emulation of the model's nonlinear dynamics. Numerical solutions from the classical Runge-Kutta 4th-order (RK4) method were used as training data for an ANN optimized with the Levenberg-Marquardt algorithm (ANN-

Assessing the impact of the Wolbachia-based control of malaria.

Malaria remains a significant infectious disease globally, causing hundreds of thousands of deaths each year. Traditional control methods, such as disease surveillance and mosquito control, along with the development of malaria vaccines, have made strides in reducing the disease's impact, but new control methods are urgently needed. Wolbachia is a natural bacterium that can infect mosquitoes and reduce their ability to transmit diseases. While initially used to control dengue fever, recent research explored its potential for malaria control. In this study, we develop and analyze a novel mathem

Modeling different infectious phases of hepatitis B with generalized saturated incidence: An analysis and control.

Hepatitis B is one of the global health issues caused by the hepatitis B virus (HBV), producing 1.1 million deaths yearly. The acute and chronic phases of HBV are significant because worldwide, approximately 250 million people are infected by chronic hepatitis B. The chronic stage is a long-term, persistent infection that can cause liver damage and increase the risk of liver cancer. In the case of multiple phases of infection, a generalized saturated incidence rate model is more reasonable than a simply saturated incidence because it captures the complex dynamics of the different infection pha

A Modeling Study on the Effect of Interstate Mobility Restrictions on the SARS-CoV-2 Pandemic.

Mobility is a crucial element in comprehending the possible expansion of the transmission chain in an epidemic. In the initial phases, strategies for containing cases can be directly linked to population mobility restrictions, especially when only non-pharmaceutical measures are available. During the pandemic of COVID-19 in Brazil, mobility limitation measures were strongly opposed by a large portion of the population. Hypothetically, if the population had supported such measures, the sharp rise in the number of cases could have been suppressed. In this context, computational modeling offers s

Real-time nowcasting and forecasting of COVID-19 dynamics in England: the first wave.

England has been heavily affected by the SARS-CoV-2 pandemic, with severe 'lockdown' mitigation measures now gradually being lifted. The real-time pandemic monitoring presented here has contributed to the evidence informing this pandemic management throughout the first wave. Estimates on the 10 May showed lockdown had reduced transmission by 75%, the reproduction number falling from 2.6 to 0.61. This regionally varying impact was largest in London with a reduction of 81% (95% credible interval: 77-84%). Reproduction numbers have since then slowly increased, and on 19 June the probability of th

Mathematical modelling for scarlet fever with direct and indirect infections.

Scarlet fever is an acute respiratory infectious disease and the incidence rate is increasing from 2011 throughout the world. In this paper, the mathematical models are proposed, which incorporate both direct transmissions and indirect transmissions of scarlet fever. The threshold conditions for disease invasion are obtained in terms of the basic reproduction number. The peak value, final size and epidemic time in a seasonal prevalence are investigated numerically. Furthermore, the effects of seasonal fluctuations on disease outbreak are also studied on the basis of real data in China.
