Initiation of metformin in early pregnancy results in fetal bioaccumulation, growth restriction, and renal dysmorphology in a primate model.

BACKGROUND: In recent years, pragmatic metformin use in pregnancy has stretched to include prediabetes mellitus, type 2 diabetes mellitus, gestational diabetes mellitus, and (most recently) preeclampsia. However, with its expanded use, concerns of unintended harm have been raised. OBJECTIVE: This study developed an experimental primate model and applied ultrahigh performance liquid chromatography coupled to triple-quadrupole mass spectrometry for direct quantitation of maternal and fetal tissue metformin levels with detailed fetal biometry and histopathology. STUDY DESIGN: Within 30 days of

Stabilities of Three Key Biological Trisulfides with Implications for Their Roles in the Release of Hydrogen Sulfide and Bioaccumulation of Sulfane Sulfur.

Trisulfides and higher polysulfides are important in the body due to their function as key reservoirs of sulfane sulfur and their rapid reactions to release persulfides. Recent work has shown that persulfides act as powerful antioxidants and release hydrogen sulfide, an emerging gasotransmitter with numerous therapeutic effects. Despite the important role of polysulfides, there is a lack of understanding of their stabilities in aqueous systems. To investigate the reactivity of trisulfides and polysulfides, three key biologically important trisulfides were synthesized from cysteine, glutathione

Combining of C, N and specific Hg stable isotopes to track bioaccumulation of monomethylmercury in coastal and freshwater seafood.

Human exposure to monomethylmercury (MMHg) through seafood consumption is a global concern. This study investigates the potential sources and processes of MMHg in seafood of coastal and freshwater areas through combing of δ13C, δ15N, and specific Hg (including MMHg and inorganic Hg (IHg)) isotopes. The results showed that δ13C and δ15N values exhibit different patterns in coastal and freshwater species. Δ199HgMMHg/δ202HgMMHg values suggested that coastal and freshwater seafood undergo similar aqueous MMHg photodegradation processes. The Δ199HgMMHg values could distinguish that, coastal fish ab

Seagrass and green macroalgae Halimeda as biomonitoring tools for metal contamination in Chuuk, Micronesia: Pollution assessment and bioaccumulation.

In this study, we evaluated metal accumulation in different species and tissues of seagrasses and green macroalgae Halimeda and assessed metal pollution levels in Chuuk, Micronesia. In seagrass, the concentrations of Ni, Cu, Zn, Cd, Pb, and Hg were higher in leaves than in roots, whereas Cr and As concentrations were higher in roots. Halimeda had higher concentrations of Ni than of the other metals, and the mean Ni concentration was approximately 2.1 times higher in Halimeda than in seagrass leaves. The concentrations of Cr, As, Cu, Pb, and Hg in Halimeda were similar to those in seagrasses, w

Bioaccumulation of mercury and transcriptional responses in tusk (Brosme brosme), a deep-water fish from a Norwegian fjord.

High concentrations of mercury (Hg) have been documented in deep-water fish species from some Norwegian fjords. In this study, tusk (Brosme brosme) was sampled from four locations in the innermost parts of Sognefjorden in Western Norway. Total Hg and methylmercury (MeHg) levels were measured in liver tissue. To search for potential sublethal effects of Hg, we characterized the hepatic transcriptome in tusk with high and low levels of Hg bioaccumulation using global transcriptomics analysis (RNA-seq). The results showed that there was a significant correlation between fish weight and accumulate

Detail investigation of toxicity, bioaccumulation, and translocation of Cd-based quantum dots and Cd salt in white mustard.

In this study, a model crop plant white mustard (Sinapis alba L.) was treated with an aqueous dispersion of silica-coated CdTe quantum dots (CdTe/SiO2 QDs) in a 72-h short-term toxicity test. The toxicity was established via measurements of (i) the root length and (ii) the chlorophyll fluorescence. These results were compared to two other sources of cadmium, free Cd ions (CdCl2) and prime un-shell nanoparticles CdTe QDs. Tested compounds were applied in concentrations representing 20 and 200 μM Cd. The uptake and translocation of Cd were investigated using inductively coupled plasma optical em

Mercury bioaccumulation in zooplankton and its relationship with eutrophication in the waters in the karst region of Guizhou Province, Southwest China.

Zooplankton play an important role in the transfer of mercury (Hg) from the lower to upper trophic positions in the food chain. In this study, total mercury (THg) and methylmercury (MeHg) levels were measured in three size fractions of zooplankton collected from three reservoirs (Hongfeng, Baihua, and Aha Reservoir) and one wetland in karst areas to understand mercury accumulation in zooplankton from alkaline environments. The results showed that the alkaline waters had lower zooplankton MeHg levels (0.1 to 66.8 ng g-1) than most of the acidic waters reported. However, the zooplankton THg leve

Bioaccumulation, cellular and molecular effects in adult zebrafish after exposure to cadmium sulphide nanoparticles and to ionic cadmium.

Few works have addressed the effects provoked by the exposure to cadmium containing nanoparticles (NPs) on adult zebrafish (Danio rerio). We studied the effects of CdS NPs (5 nm) or ionic cadmium (10 μg Cd/L) after 3 and 21 d of exposure and at 6 months post-exposure (mpe). Acute toxicity was recorded after exposure to both forms of cadmium. Significant cadmium accumulation was measured in the whole fish after both treatments and autometallography showed a higher accumulation of metal in the intestine than that in the liver. Histopathological alterations, such as inflammation in gills and vacu

Bioaccumulation and distribution of cadmium by Burkholderia cepacia GYP1 under oligotrophic condition and mechanism analysis at proteome level.

Bacteria have been applied for the bioremediation of cadmium-contaminated environment. Less is known about the bioaccumulation of high concentration of Cd over time under the oligotrophic environment. Burkholderia cepacia GYP1, which was isolated from multiple heavy metal contaminated farmland, was studied for its bioaccumulation mechanism of Cd under oligotrophic condition. GYP1 possessed highly accumulation capacity for cadmium reaching 116 mg Cd/g biomass (dry weight). ATR-FTIR, electron microscopy, flow cytometry along with subcellular fraction demonstrated that the uptake and distribution

Distribution and bioaccumulation of POPs and mercury in the Ga-Selati River (South Africa) and the rivers Gudbrandsdalslågen and Rena (Norway).

Biomagnification of Hg and persistent organic pollutants (POPs: polychlorinated biphenyls (PCBs), organochlorine pesticides (OCPs), polybrominated diphenyl ethers (PBDEs)) in aquatic food chains can lead to high pollutant concentrations in top predators, including humans. Despite this threat to human health, research concerning bioaccumulation is still underrepresented in the southern hemisphere and in (sub)arctic and (sub)tropical areas, emphasizing the need for research in these locations. In this study, samples of water, sediment and aquatic biota were analyzed to determine concentrations o

Mercury and methylmercury bioaccumulation by polychaete worms is governed by both feeding ecology and mercury bioavailability in coastal mudflats.

Polychaete worms are abundant in many mudflats but their importance to coastal food web Hg biomagnification is not known. We sampled sediments and polychaete worms from mudflats in the Bay of Fundy to investigate the bioaccumulation of mercury (Hg) and methylmercury (MeHg) in the coastal invertebrate food web. Hg concentrations in the sediments were low (<20 μg kg(-1)). Labile Hg (methanol/KOH sediment extraction) in surface sediments (0-1 cm) was positively correlated with Hg bioaccumulation by surface sediment-ingesting polychaetes but, surprisingly, there was a negative correlation between

Combined effect of copper and cadmium on heavy metal ion bioaccumulation and antioxidant enzymes induction in Chlorella vulgaris.

The relationships between metal uptake and antioxidant enzyme activities or a response to membrane lipid peroxidation (i.e., malondialdehyde production) in Chlorella vulgaris exposed to Cu and Cd compounds singly and in combination were investigated. The results showed that bioaccumulation of a single metal was influenced by the presence of the other metal. The activities of superoxide dismutase and peroxidase increased to more than fivefold of the control after exposure to Cu(1.5 μM) alone or to Cu(1.5 μM) with Cd mixtures. Malondialdehyde levels in C. vulgaris also increased to approximately

Bioaccumulation of As, Cd, Cr, Hg(II), and MeHg in killifish (Fundulus heteroclitus) from amphipod and worm prey.

Elevated metal levels in fish are a concern for the fish themselves, their predators, and possibly humans who consume contaminated seafood. Metal bioaccumulation models often rely on assimilation efficiencies (AEs) of ingested metals and loss rate constants after dietary exposure (k(ef)s). These models can be used to better understand processes regulating metal accumulation and can be used to make site-specific predictions of metal concentrations in animal tissues. Fish often consume a varied diet, and prey choice can influence these two parameters. We investigated the trophic transfer of As,

Intra- and inter-specific variability in total and methylmercury bioaccumulation by eight marine fish species from the Azores.

We relate fish biological and ecological characteristics to total and organic mercury concentrations to determine whether accumulation is influenced by trophic level, Hg concentration in the diet, and vertical distribution. Levels of total mercury and methylmercury were determined in the muscle tissue of eight species of fish: Pagellus acarne, Trachurus picturatus, Phycis phycis, P. blennoides, Polyprion americanus, Conger conger, Lepidopus caudatus and Mora moro, caught in the Azores. All such fishes are commercially valuable and were selected to include species from a wide range of vertical

Bioaccumulation and translocation of rare earth elements in Medicago sativa L. grown in coal fly ash-amended substrates.

A greenhouse pot experiment investigated the bioaccumulation and translocation of Cerium (Ce), Erbium (Er), Europium (Eu), Neodymium (Nd), and Praesodymium (Pr) in Medicago sativa L. grown in a substrate amended with 0, 20, 30, and 40% coal fly ash (CFA, w/w). Bioaccumulation factors (BAF) were highest in the unamended control, indicating that CFA loading reduced uptake efficiency consistent with pH-driven REE immobilization. Shoot REE concentrations exceeded root concentrations for most elements at later growth stages, with Eu dominating accumulation, peaking at 304.44 µg g-1 (shoot + root) u

Assessing the accumulation of microplastics in earthworms (Eisenia fetida) using traditional bioaccumulation modeling and synchrotron-based microcomputed tomography.

Microplastics (MPs) are recognized as a ubiquitous contaminant in terrestrial ecosystems. Earthworms, which play key roles in soil structure, nutrient cycling, and ecosystem function, are important bioindicators for assessing MP exposure and fate. However, uncertainties remain regarding MP toxicokinetics in earthworms, including ingestion dynamics, retention, and the potential for translocation across the gut epithelium. This study investigated the movement and fate of MPs in Eisenia fetida by quantifying uptake and elimination across five common MP morphologies (films, fibers, foams, fragment

Copper-nickel interaction effects on bioaccumulation and physiological responses of the acid-tolerant microalga Trebouxiophyceae sp. MA-1.

Copper (Cu) and nickel (Ni) coexist widely in industrial and municipal wastewater, especially acid mine drainage (AMD). However, research concerning the toxic effects and ecological risks of combined Cu/Ni stress on microalgae is scant, and a systematic comparison of microalgal responses between single and co-exposure conditions has yet to be systematically conducted. An indigenous acid-tolerant AMD microalga, Trebouxiophyceae sp. MA-1, was isolated and examined for its Cu and Ni tolerance and removal in this study. The algal growth was inhibited more severely by Cu than Ni. Co-exposure caused

Microplastic co-exposure increases PFAS bioaccumulation without universally amplifying toxicity in aquatic organisms: A multi-endpoint meta-analysis.

Aquatic ecosystems are important sinks for microplastics (MPs) and per- and polyfluoroalkyl substances (PFASs), while their combined toxic effects on aquatic organisms vary across biological endpoints. This quantitative meta-analysis was performed based on 810 datasets from 29 laboratory studies, to explore MPs-mediated changes in PFASs toxicity and identify key influencing factors. MPs-PFASs co-exposure significantly increased PFASs bioaccumulation, indicating enhanced particle-mediated transport, co-ingestion, or altered internal retention. However, this increase of bioaccumulation did not t

Survival, antioxidant enzyme activities, and bioaccumulation response in freshwater snail Radix plicatula (W. H. Benson, 1842) to arsenic.

Arsenic contamination threatens freshwater aquatic ecosystems; however, integrated studies of bioaccumulation and toxicological mechanisms in gastropods remain limited. This study assessed acute and chronic toxicity in Radix plicatula by integrating survival, antioxidant enzyme activity, and tissue bioaccumulation measurements. We exposed snails to three arsenic concentrations for 21 days. Survival declined dose- and time-dependently from 98.3% (controls) to 83.3% (high concentrations) by day 21. Antioxidant enzymes showed biphasic responses, catalase (CAT) peaked on day 7 and then declined, w

When nanoplastics (NPs) meet algae: Heteroaggregates exacerbate bioaccumulation, immunotoxicity, and microbial dysbiosis in the green mussel (Perna viridis).

Heteroaggregates (HAs) formed by nanoplastics (NPs) and microalgae occur ubiquitously in natural aquatic systems. However, their influence on the toxicokinetics and biological effects of NPs in marine mussels remains largely unknown. Here, the green mussels (Perna viridis) were exposed to NPs and HAs at their environmentally relevant concentrations for 21 d, followed by a 7-d depuration phase. The effects on toxicokinetics, immunological responses, and microbiota of digestive gland were evaluated. The results showed that HAs increased the uptake rate constant in digestive gland by 5.5-fold and
