The aging interactome: From cellular dysregulation to therapeutic frontiers in age-related diseases.

Biological aging is a complex, multifaceted process characterized by the progressive erosion of cellular homeostasis, driven by intersecting pathways of inflammaging, immune senescence, mitochondrial dysfunction, and genomic instability. This review delineates current knowledge on the interconnected mechanisms underlying aging-related diseases, including atherosclerosis, obesity, sarcopenia, neurodegenerative disorders, rheumatoid arthritis, and cancer. We further discuss the critical role of contributing factors such as microbiome dysbiosis, sex differences, and exosome-mediated communication

Leveraging Autonomous Biological Oscillators for Directed Evolution of Switchable Proteins.

Engineering proteins that can switch between states is of strong interest to biotechnology. These include multi-state transcription factors, membrane receptors, kinases, and synthetic molecular logic gates. While traditional directed evolution approaches excel at optimizing steady-state protein functionalities, they require substantial adaptation to treat transitions in states of proteins. This chapter describes how the emerging regulatory logic of autonomous biological oscillators can be used to impose time-varying selection pressure for evolving switchable proteins. We first review the chall

[Update on Gastric Pacemaking: Relevance for Gastroparesis].

Gastroparesis is a disease in which gastric emptying is significantly decreased in the absence of a mechanical obstruction of the gastric outlet. In addition to conservative or pharmacological treatment options, gastric electrical stimulation (GES) is a possible alternative therapy for refractory courses of the disease. However, the available data from randomised controlled trials on efficacy do not permit an unqualified recommendation. The aim of this review is to introduce the clinical picture of gastroparesis, including the underlying pathophysiology, and to classify the therapeutic role of

Time's arrow flies like a bird: two paradoxes for avian circadian biology.

Biological timekeeping in birds is a fundamental feature of avian physiology, behavior and ecology. The physiological basis for avian circadian rhythmicity has pointed to a multi-oscillator system of mutually coupled pacemakers in the pineal gland, eyes and hypothalamic suprachiasmatic nuclei (SCN). In passerines, the role of the pineal gland and its hormone melatonin is particularly important. More recent molecular biological studies have pointed to a highly conserved mechanism involving rhythmic transcription and translation of "clock genes". However, studies attempting to reconcile the phys

[Slow-wave fluctuations in craniosacral space: hemo-liquorodynamic conception of origin].

In the paper, the mechanism of forming of rhythmic slow-wave fluctuations in craniospinal cavity was investigated. In five young healthy persons, at rest and under voluntary respiration arrest test, the bioimpedansograms of head and lumbosacral part of vertebral column were synchronously registered as these recordings reflect the changes of relationships between blood/CSF volumes in cranial and lumbosacral regions, respectively. The recordings were subjected to frequency and spectral computer analysis (PC Macintosh G-4, Chart 5.2. software). The rapid (pulsatile) as well as slow and counter-di

Effects of working memory load on oscillatory power in human intracranial EEG.

Studies of working memory load effects on human EEG power have indicated divergent effects in different frequency bands. Although gamma power typically increases with load, the load dependency of the lower frequency theta and alpha bands is uncertain. We obtained intracranial electroencephalography measurements from 1453 electrode sites in 14 epilepsy patients performing a Sternberg task, in order to characterize the anatomical distribution of load-related changes across the frequency spectrum. Gamma power increases occurred throughout the brain, but were most common in the occipital lobe. In

Headache due to photosensitive magnesium depletion.

Clinical and paraclinical data (visual stress tests, electroencephalographic and cerebrovascular photic driving, visual evoked potentials) demonstrate that the concept of photosensitive headache is fully justified. The interictal hallmark of photosensitive cephalalgic patients is potentiation (or sensitization) instead of habituation. The aetiopathogenic mechanisms of photosensitive headache associate hypofunction of the biological clock and magnesium depletion. The new concept of headache due to photosensitive magnesium depletion seems justified. It appears logical to add the treatments of ma

Analysis of amplitude and frequency variations of essential and Parkinsonian tremors.

Variations in the amplitude and period of essential and Parkinsonian tremors were studied. It was found that the variations in frequency (or period) were mostly similar to the white noise, with the standard deviation typically less than 10% of the mean, whereas the variations in amplitude were much larger, with standard deviations greater than 30% of the mean, and so could not be effectively smoothed by running means. It is conjectured that variations in frequency reflect the stable nature of the neural network that generates the rhythmicity responsible for the tremor. The variations in amplit

About-weekly variations in nocturia.

The aim of this study was to assess components of variation in nocturia and to determine any putative geomagnetic influence. A 54-year old man with benign prostatic hyperplasia had recorded for about 4 years the number of times he awoke each night to urinate. The data have been reanalyzed for chronomics, the mapping of time structures (chronomes), involving the computation of least squares spectra of the urinary record and of environmental variables recorded during the same 4-year span. In addition to the previously reported monthly variation, other periodicities have been documented, includin

Magnesium maintains the length of the circadian period in Arabidopsis.

The circadian clock coordinates the physiological responses of a biological system to day and night rhythms through complex loops of transcriptional/translational regulation. It can respond to external stimuli and adjust generated circadian oscillations accordingly to maintain an endogenous period close to 24 h. However, the interaction between nutritional status and circadian rhythms in plants is poorly understood. Magnesium (Mg) is essential for numerous biological processes in plants, and its homeostasis is crucial to maintain optimal development and growth. Magnesium deficiency in young Ar

Circadian Rhythms in Acute Respiratory Distress Syndrome: Molecular Mechanisms and Therapeutic Implications.

Acute respiratory distress syndrome (ARDS) is a high-mortality condition lacking targeted treatments. Emerging evidence indicates that circadian rhythm disruption is a key factor in the development of ARDS. Core clock proteins control essential processes, including alveolar-capillary barrier function, inflammation, and tissue repair. The intensive care unit (ICU) environment and underlying illness create double hits that impair biological clocks, leading to a cycle of excessive inflammation and organ damage. This review highlights the central role of circadian rhythms in ARDS. Despite strong p

Effect of monochromatic light on the temporal expression of N-acetyltransferase in chick pineal gland.

The avian pineal gland is an independent molecular oscillator that receives external light information that regulates the synthesis and secretion of melatonin. Arylalkylamine N-acetyltransferase plays an important role in the pineal gland by controlling the rhythmic production of melatonin. Previous study showed that monochromatic light influences the secretion of melatonin, which is regulated by the molecular circadian clock genes in chick pineal gland. This study was designed to investigate the effect of monochromatic light on the circadian rhythm of levels of cAanat, clock protein (CLOCK an

Associations of epigenetic age acceleration with motor impairment: Evidence from the Parkinson's Progression Markers Initiative cohort.

Motor decline is a hallmark of Parkinson's disease (PD) and biological aging, yet the specific relationship between systemic biological aging and neuromotor function remains under-characterized. This study leveraged longitudinal phenotypic and whole-blood DNA methylation data from the Parkinson's Progression Markers Initiative (PPMI) to evaluate associations between seven epigenetic aging measures, spanning first-generation, second-generation (risk-optimized), rate-based, and next-generation deep-learning clocks, and motor impairment. Motor function was assessed using the Movement Disorder Soc

Circadian mechanisms in heart failure: Cell-specific regulation and chrono-medicine.

Emerging evidence indicates that circadian rhythms profoundly influence the pathophysiology of heart failure (HF). The circadian clock system governs myocardial functions via synchronized actions from multiple cell types involving cardiomyocytes and non-cardiomyocytes. This review summarizes state-of-the-art advances on the cardiovascular circadian system and provides a perspective on time-of-day effects in the pathophysiology of HF, including energy metabolism, tissue inflammation and fibrosis. In particular, we focus on cell-specific circadian regulation and synthesize how circadian clocks a

Circadian rhythms in haematological malignancies: therapeutic potential and personalised interventions.

The circadian clock, a fundamental cellular mechanism, regulates the rhythmic expression of numerous genes and biological processes across various organs. Disruptions in this system, driven by genetic or environmental factors, have been reported to be involved in cancer progression. This review explores the role of the circadian clock in cancer hallmarks and its impact on cellular homeostasis within haematological malignancies. Drawing on findings from in vitro, in vivo, and clinical trials, this review highlights the potential of clock genes as diagnostic and prognostic biomarkers, and as the

Genetic dissection of drug effects in clinical practice: CLOCK gene and clozapine-induced diurnal sleepiness.

Psychotic patients treated with clozapine often experience persistent daytime sleepiness. This is a frequent side effect of clozapine that may reduce patient compliance. We hypothesized that clozapine might interfere with the circadian rhythms regulated by the biological clock. In 171 patients with major psychosis, we investigated the association between hypersomnolence during clozapine therapy and a CLOCK gene polymorphism (3111 T/C substitution). Forty-six patients showed persistent daytime sleepiness and were classified as "sleepy". "Sleepy" patients were significantly more likely to have a

Circadian (De)regulation in Head and Neck Squamous Cell Carcinoma.

Head and neck cancer encompass different malignancies that develop in and around the throat, larynx, nose, sinuses and mouth. Most head and neck cancers are squamous cell carcinomas (HNSCC) that arise in the flat squamous cells that makeup the thin layer of tissue on the surface of anatomical structures in the head and neck. Each year, HNSCC is diagnosed in more than 600,000 people worldwide, with about 50,000 new cases. HNSCC is considered extremely curable if detected early. But the problem remains in treatment of inoperable cases, residues or late stages. Circadian rhythm regulation has a b

[Practical Pharmaceutical Researches Utilizing Clock Genes].

The biological clock system regulates gene transcription in approximately 24 h cycles, creating circadian rhythms with respect to drug pharmacokinetics and pharmacological efficacy. My research focuses on clinical applications of clock systems and the elucidation of pathophysiological mechanisms involving clock genes. In the present study, I aimed to identify the optimal timing of methylprednisolone (mPSL) administration to pediatric patients undergoing liver transplantation. A randomized clinical trial of 60 such patients demonstrated that evening administration (20:00) was associated with si

The adrenal gland circadian clock exhibits a distinct phase advance in spontaneously hypertensive rats.

The circadian clock influences a multitude of cellular and biological processes, including blood pressure control. Spontaneously hypertensive rats (SHR) exhibit aberrant circadian rhythms affecting cardiovascular parameters, and they also have abnormal clock gene expression profiles in several organs. Given the important role of the adrenal gland in orchestrating circadian oscillations, we investigated the adrenal gland circadian clock in SHR and control Wistar-Kyoto rats maintained under a 12-hour light-dark cycle. Adrenal glands, livers, and serum samples were collected every 4 h and mRNA wa

Circadian Clock Genes Modulate Immune, Cell Cycle and Apoptosis in the Diagnosis and Prognosis of Pan-Renal Cell Carcinoma.

Background: Pan-renal cell carcinoma (pan-RCC) is mainly divided into renal clear cell carcinoma (KIRC), renal papillary cell carcinoma (KIRP), and chromophobe cell carcinoma (KICH). Pan-RCC is a common malignant neoplasm with a high incidence and poor prognosis. Several studies have demonstrated a close association between cancer development and circadian rhythms; however, the clinical significance and molecular mechanism of the clock gene remain unclear in pan-RCC. Methods: In this study, we systematically characterized the alterations of 15 well-known clock genes of three types of kidney ca
