Nanomechanical properties of lipid bilayer: asymmetric modulation of lateral pressure and surface tension due to protein insertion in one leaflet of a bilayer.

The lipid membranes of living cells form an integral part of biological systems, and the mechanical properties of these membranes play an important role in biophysical investigations. One interesting problem to be evaluated is the effect of protein insertion in one leaflet of a bilayer on the physical properties of lipid membrane. In the present study, an all atom (fine-grained) molecular dynamics simulation is used to investigate the binding of cytotoxin A3 (CTX A3), a cytotoxin from snake venom, to a phosphatidylcholine lipid bilayer. Then, a 5-microsecond [corrected] coarse-grained molecula

Brain imaging, genetics and emotion.

This paper reviews the published evidence on genetically driven variation in neurotransmitter function and brain circuits involved in emotion. Several studies point to a role of the serotonin transporter promoter polymorphism in amygdala activation during emotion perception. We also discuss other polymorphisms (e.g. the COMT val158met polymorphism, tryptophan hydroxylase-2 -703 G/T) and putative effects on affective processing in cortical and limbic regions. A different line of research concerns studies with genetic disorders. Although at a less fine-grained level, studies with individuals wit

Dose Variations Using an X-Ray Cabinet to Establish in vitro Dose-Response Curves for Biological Dosimetry Assays.

In biological dosimetry, dose-response curves are essential for reliable retrospective dose estimation of individual exposure in case of a radiation accident. Therefore, blood samples are irradiated in vitro and evaluated based on the applied assay. Accurate physical dosimetry of the irradiation performance is a critical part of the experimental procedure and is influenced by the experimental setup, especially when X-ray cabinets are used. The aim of this study was to investigate variations and pitfalls associated with the experimental setups used to establish calibration curves in biological

Follicular lymphoma: are we ready for a risk-adapted approach?

Follicular lymphoma is the most common indolent non-Hodgkin lymphoma in the Western hemisphere. The natural history of FL appears to have been favorably impacted by the introduction of rituximab after randomized clinical trials demonstrated that the addition of rituximab to standard chemotherapy induction has improved the overall survival. Yet, the disease is biologically and clinically heterogeneous with wide variations in outcomes for individual patients. The ability to accurately risk-stratify patients and then tailor therapy to the individual is an area of ongoing research. Historically, t

Chemical Profile and Biological Effects of an Herbal Mixture for the Development of an Oil-in-Water Cream.

Three individual hydroalcoholic extracts derived from Hamamelis virginiana leaves, Krameria lappacea root, Salix alba bark, and the resulting herbal mixture (HM) were assessed for the phytochemical profile as well as for antibacterial and cytotoxic potential. The chemical composition of the individual extracts and of their mixture was analyzed by chromatographical (LC-MS) and spectrophotometrical methods. The antimicrobial properties were evaluated by using the agar-well diffusion and the broth microdilution assays, whereas the potential cytotoxicity was investigated on human keratinocyte cell

Structural and chemical heterogeneity of Proterozoic organic microfossils of the ca. 1 Ga old Angmaat Formation, Baffin Island, Canada.

Organic microfossils in Meso- and Neoproterozoic rocks are of key importance to track the emergence and evolution of eukaryotic life. An increasing number of studies combine Raman spectroscopy with synchrotron-based methods to characterize these microfossils. A recurring observation is that Raman spectra of organic microfossils show negligible variation on a sample scale and that variation between different samples can be explained by differences in thermal maturation or in the biologic origin of organic precursor material. There is a paucity of work, however, that explores the extent to which

Deep Learning in Infant Functional Neuroimaging: Challenges, Advances, and Future Directions.

Infancy is a critical developmental window characterized by rapid functional brain reorganization, during which large-scale networks emerge, individualized connectome signatures continue to form, and early deviations may shape long-term cognitive and clinical outcomes. Functional MRI (fMRI) offers an opportunity to study these processes in vivo, yet extracting developmentally meaningful information from it remains challenging due to comparatively short scan duration, structured motion artifacts, variable scan states, and rapid brain maturation. Amid these challenges, deep learning has expanded

Forensic DNA phenotyping: Prediction of eye and hair colour and allelic frequency estimation in the Italian population for the development of a reference dataset.

In recent years, Forensic DNA Phenotyping (FDP) has emerged as an innovative "DNA intelligence approach" aimed at predicting externally visible individual characteristics from biological traces collected at crime scenes or from biological samples of unidentified corpses or skeletal remains. This study aims to analyse the allele frequency distribution of genetic markers involved in the prediction of eye and hair colour in an Italian population sample, to enhance the existing European dataset. A large cohort of Italian individuals with at least three generations of ancestry was recruited, ensuri

Proliferating cell nuclear antigen (PCNA) expression in patients with breast carcinoma in comparison to patients with benign breast diseases.

The proliferating cell nuclear antigen (PCNA) was studied by immunohistochemistry on FNA specimens of 20 breast (ductal) carcinomas, 20 fibroadenomas and 20 atypical fibrocystic lesions of the breast. Sixteen cases of breast carcinomas, eight fibroadenomas but no atypical fibrocystic lesions were found to display nuclear PCNA staining. A significant difference was found, between PCNA index of breast carcinomas (mean PCNA index 56%) and fibroadenomas (mean PCNA 21, 25%). This suggests that breast carcinomas display a higher proliferation index than fibroadenomas and fibrocystic disease. Further

Pharmacogenetics in Response to Biological Agents in Inflammatory Bowel Disease: A Systematic Review.

Inflammatory bowel diseases (IBDs) are chronic inflammatory disorders influenced by microbial, environmental, genetic, and immune factors. The introduction of biological agents has transformed IBD therapy, improving symptoms, reducing complications, and enhancing patients' quality of life. However, approximately 30% of patients exhibit primary non-response, and 50% experience a loss of response over time. Genetic and non-genetic factors contribute to variability in treatment outcomes. This systematic review aims to thoroughly analyze and assess existing studies exploring the relationships betw

Inter-individual variations of human mercury exposure biomarkers: a cross-sectional assessment.

BACKGROUND: Biomarkers for mercury (Hg) exposure have frequently been used to assess exposure and risk in various groups of the general population. We have evaluated the most frequently used biomarkers and the physiology on which they are based, to explore the inter-individual variations and their suitability for exposure assessment. METHODS: Concentrations of total Hg (THg), inorganic Hg (IHg) and organic Hg (OHg, assumed to be methylmercury; MeHg) were determined in whole blood, red blood cells, plasma, hair and urine from Swedish men and women. An automated multiple injection cold vapour a

A novel PRRX1 loss-of-function variation contributing to familial atrial fibrillation and congenital patent ductus arteriosus.

Atrial fibrillation (AF) represents the most common type of sustained cardiac arrhythmia in humans and confers a significantly increased risk for thromboembolic stroke, congestive heart failure and premature death. Aggregating evidence emphasizes the predominant genetic defects underpinning AF and an increasing number of deleterious variations in more than 50 genes have been involved in the pathogenesis of AF. Nevertheless, the genetic basis underlying AF remains incompletely understood. In the current research, by whole-exome sequencing and Sanger sequencing analysis in a family with autosoma

Are Alterations in DNA Methylation Related to CKD Development?

The modifications in genomic DNA methylation are involved in the regulation of normal and pathological cellular processes. The epigenetic regulation stimulates biological plasticity as an adaptive response to variations in environmental factors. The role of epigenetic changes is vital for the development of some diseases, including atherogenesis, cancers, and chronic kidney disease (CKD). The results of studies presented in this review have suggested that altered DNA methylation can modulate the expression of pro-inflammatory and pro-fibrotic genes, as well those essential for kidney developme

Management of High-Risk Non-Muscle-Invasive Bladder Cancer.

High-risk non-muscle-invasive bladder cancer represents a biologically heterogeneous disease with wide variation in recurrence and progression risk, creating persistent challenges in diagnosis, risk stratification, and management. Current guidelines rely on well-described clinicopathologic features to define risk; however, these factors alone often incompletely predict individual patient outcomes and efforts to refine prognostication using molecular profiling and advanced imaging techniques such as multiparametric MRI are under active investigation. Complete transurethral resection (TUR), incl

Risk factors for dementia in people with down syndrome: issues in assessment and diagnosis.

It has been clearly established that there is an increased incidence of early onset dementia of the Alzheimer type (DAT) in people who have Down syndrome. There are variations in the age of onset of the clinical signs of DAT, which may be accounted for by different risk factors. In this review we examined the evidence that different biological and psychological factors may influence the risk for DAT. Limitations in design of early studies, the need for consistent diagnostic criteria for DAT in individuals with Down syndrome, and the lack of adequate psychometric tools to detect cognitive chang

The Scoop on Honey Badger Poop: Validation of a Non-Invasive Method for Assessing Physiological Responses in Captivity.

Captive honey badgers (Mellivora capensis) are routinely exposed to environmental and husbandry-related stressors that differ markedly from their natural conditions; however, physiological welfare assessments in this species remain limited. This study aimed to validate a non-invasive approach to evaluate stress-related fecal glucocorticoid metabolite (fGCM) concentrations and individual responses to visitors and other environmental stressors in two zoo-housed individuals. Five enzyme immunoassays (EIAs) were assessed for suitability using routine veterinary examinations as a presumed biologica

Correlation of damage score in PTOA with changes in stress on cartilage in an ovine model.

OBJECTIVE: There is a high risk of developing osteoarthritis (OA) following traumatic injury to the knee. Severe ligament injuries can disrupt the integrity of the multicomponent knee at both biological and biomechanical levels. We hypothesize changes in cartilages stresses could lead to tissue damage and development of OA. DESIGN: The in-vivo gait kinematics of the stifle (knee) joint of four adult female ovine subjects were recorded prior to and at ten-and-twenty weeks following partial ACL-MCL transection. The subjects were sacrificed and the experimental joint from each subject was mounte

Neural and cognitive characteristics of extraordinary altruists.

Altruistic behavior improves the welfare of another individual while reducing the altruist's welfare. Humans' tendency to engage in altruistic behaviors is unevenly distributed across the population, and individual variation in altruistic tendencies may be genetically mediated. Although neural endophenotypes of heightened or extreme antisocial behavior tendencies have been identified in, for example, studies of psychopaths, little is known about the neural mechanisms that support heightened or extreme prosocial or altruistic tendencies. In this study, we used structural and functional magnetic

Quantitative Genetic Analysis of Baboon Facial Variation Challenges a Classic Papionin Trait.

The Linnaean family of monkeys that includes baboons, macaques, and mandrills (among others) is notably characterized by large, extended muzzles that vary in lateral nasal dorsum profile. In paleontological studies, this variation is referred to as the anteorbital drop (AOD). Here, we first devise a method to quantify this phylogenetic character. We then employ quantitative genetic analyses using a MCMCglmm model of facial variation in a captive, pedigreed baboon colony (n = 934), including admixed individuals derived from at least two Papio species, from the Southwest National Primate Researc

Bridging language phenotypes, neural dynamics and gene regulation in autism.

Autism Spectrum Disorders (ASDs) affect approximately 1% of the global population, with prevalence rising over recent decades. Although ASDs are heritable (~70%), translating genetic findings into effective therapeutics remains challenging due to the vast number of implicated risk genes (>700) and the limited influence of individual genes on disease expression. Their collective impact and convergent expression patterns of small-effect genes provide insights into ASD-related neurobiological mechanisms. Current interventions remain symptom-focused and do not address underlying neurodevelopmental
