Molecular approaches to the study of myiasis-causing larvae.

Among arthropod diseases affecting animals, larval infections - myiases - of domestic and wild animals have been considered important since ancient times. Besides the significant economic losses to livestock worldwide, myiasis-causing larvae have attracted the attention of scientists because some parasitise humans and are of interest in forensic entomology. In the past two decades, the biology, epidemiology, immunology, immunodiagnosis and control methods of myiasis-causing larvae have been focused on and more recently the number of molecular studies have also begun to increase. The 'new techn

Systematic evaluation of rDNA ITS2 as a DNA barcode for molecular identification of cyclorrhapha flies in China.

Morphological identification of medically important flies is often hindered by incomplete or damaged specimens, cryptic species, and morphological conservatism among closely related species. In this study, common fly species were collected from 15 cities across 10 provinces in China to evaluate rDNA internal transcribed spacer 2 (ITS2) as a potential DNA barcode for molecular identification. A total of 1252 fly specimens were collected, of which 933 were morphologically classified into 19 species, 15 genera, 9 families, and 3 superfamilies; 319 remained unidentified due to the loss of key morp

Antennal transcriptome analysis reveals sensory receptors potentially associated with host detection in the livestock pest Lucilia cuprina.

BACKGROUND: Lucilia cuprina (Wiedemann, 1830) (Diptera: Calliphoridae) is the main causative agent of flystrike of sheep in Australia and New Zealand. Female flies lay eggs in an open wound or natural orifice, and the developing larvae eat the host's tissues, a condition called myiasis. To improve our understanding of host-seeking behavior, we quantified gene expression in male and female antennae based on their behavior. METHODS: A spatial olfactometer was used to evaluate the olfactory response of L. cuprina mated males and gravid females to fresh or rotting beef. Antennal RNA-Seq analysis

How deadly sarcophagid fly larvae are for anurans? New interactions and review to Neotropical region.

Myiasis is defined as the infestation of living or necrotic tissues of vertebrates by dipteran larvae. In amphibians, it can be caused by fly larvae belonging to families Calliphoridae, Chloropidae, Muscidae, and Sarcophagidae. In this study, we report new records and infestation intensity of myiasis in anurans from two localities of the Atlantic Forest in southern Brazil, and also present a list with myiasis caused by flesh fly Sarcophagidae species in anurans of the Neotropical region. For this, we surveyed anuran communities during two breeding seasons, examined for myiasis signs, and when

Arthropods affecting the human eye.

Ocular infestations by arthropods consist in the parasitization of the human eye, either directly (e.g., some insect larvae causing ophthalmomyiasis) or via arthropods feeding on lachrymal/conjunctival secretions (e.g., some eye-seeking insects, which also act as vectors of eye pathogens). In addition, demodicosis and phthiriasis may also cause eye discomfort in humans. Ophthalmomyiasis by larvae of the families Oestridae, Calliphoridae and Sarcophagidae, are frequent causative agents of human ocular infestations. Over the last decades, the extensive use of macrocyclic lactones in cattle has r

Nuisance flies around a landfill: patterns of abundance and distribution.

Some members of the public living near landfills perceive the sites as a source of nuisance flies and their associated diseases. Both these factors contribute to frequent complaints to Environmental Health Officers about landfill operations. Fly monitoring using sticky targets was done at a working landfill site in the West Midlands in order to identify the key species of flies present at the site, and to establish whether these flies were similar to those occurring in the surrounding area. Targets were positioned in concentric rings, radiating away from the working face into the surrounding r

Traumatic myiasis by Wohlfahrtia magnifica in sheep flocks from southeastern Spain: prevalence and risk factors.

Wohlfahrtia magnifica (Diptera: Sarcophagidae) is an obligate myiasis-causing fly of livestock that is widely distributed throughout the Mediterranean basin. A total of 73 683 sheep from 122 flocks in Albacete Province (southeastern [SE] Spain) were examined to detect active traumatic myiasis. The influence of several individual and flock factors on the prevalence was also assessed. The overall flock prevalence of traumatic myiasis was 95.9%, with an individual prevalence of 7.1%. Wohlfahrtia magnifica was identified in all of the positive animals except one that was infested by Lucilia serica

Spatial and temporal habitat partitioning by calliphorid blowflies.

Calliphorid blowflies perform an essential ecosystem service in the consumption, recycling and dispersion of carrion nutrients and are considered amongst the most important functional groups in an ecosystem. Some species are of economic importance as facultative agents of livestock myiasis. The interspecific ecological differences that facilitate coexistence within the blowfly community are not fully understood. The aim of this work was to quantify differences in habitat use by calliphorid species. Thirty traps were distributed among three habitats at two sites in southwest England for collect

Genetic approaches for studying myiasis-causing flies: molecular markers and mitochondrial genomics.

"Myiasis-causing flies" is a generic term that includes species from numerous dipteran families, mainly Calliphoridae and Oestridae, of which blowflies, screwworm flies and botflies are among the most important. This group of flies is characterized by the ability of their larvae to develop in animal flesh. When the host is a live vertebrate, such parasitism by dipterous larvae is known as primary myiasis. Myiasis-causing flies can be classified as saprophagous (free-living species), facultative or obligate parasites. Many of these flies are of great medical and veterinary importance in Brazil

New World screwworm (Cochliomyia hominivorax) myiasis in feral swine of Uruguay: One Health and transboundary disease implications.

BACKGROUND: Feral swine (Sus scrofa) are highly invasive and threaten animal and human health in the Americas. The screwworm (Cochliomyia hominivorax) is listed by the World Organization for Animal Health as a notifiable infestation because myiasis cases affect livestock, wildlife, and humans in endemic areas, and outbreaks can have major socioeconomic consequences in regions where the screwworm has been eradicated. However, a knowledge gap exists on screwworm infestation of feral swine in South America, where the screwworm is endemic. Here, we report screwworm infestation of feral swine harve

Development and validation of a simulation model for blowfly strike of sheep.

A comprehensive simulation model for sheep blowfly strike due to Lucilia sericata (Meigen) (Diptera: Calliphoridae), which builds on previously published versions but also incorporates important new empirical data, is used to explain patterns of lamb and ewe strike recorded on 370 farms in south-west, south-east and central England and Wales. The model is able to explain a significant percentage of the variance in lamb strike incidence in all four regions, and ewe strike in three of the four regions. The model is able to predict the start of seasonal blowfly strike within one week in three of

Molecular basis of resistance to organophosphate insecticides in the New World screw-worm fly.

BACKGROUND: The emergence of insecticide resistance is a fast-paced example of the evolutionary process of natural selection. In this study, we investigated the molecular basis of resistance in the myiasis-causing fly Cochliomyia hominivorax (Diptera: Calliphoridae) to dimethyl-organophosphate (OP) insecticides. METHODS: By sequencing the RNA from surviving larvae treated with dimethyl-OP (resistant condition) and non-treated larvae (control condition), we identified genes displaying condition-specific polymorphisms, as well as those differentially expressed. RESULTS: Both analyses revealed

Confocal Laser Scanning Microscopy as a Method for Identifying Variation in Puparial Morphology and Establishing Characters for Taxonomic Determination.

Calliphoridae, or blow flies, are of much ecological and practical importance given their roles in decompositional ecology, medical and veterinary myiasis, and forensic entomology. As ephemeral and rapidly developing species, adults are frequently not present for identification, but puparia (the remaining outer integument of the third instar larvae) are frequently found. These heavily sclerotized remains are stable in the environment but they are of a conservative character. Historically, scanning electron microscopy (SEM) has been used for characterization, a technique which is not only time-

Overcoming doramectin failure: field efficacy of pour-on 5% fluralaner and an experimental 1% oral formulation for the prevention of Cochliomyia hominivorax-associated umbilical myiasis in newborn calves.

BACKGROUND: Myiasis caused by Cochliomyia hominivorax larvae is a reemerging challenge in the Americas due to the recent reappearance of this species in areas where it had been eradicated and reports of doramectin inefficacy, despite its recognition as the most effective avermectin for prevention in cattle. As an alternative, 5% fluralaner administered via pour-on has shown 100% efficacy in the treatment and prevention of myiasis. Therefore, this study aimed to evaluate the efficacy of three antiparasitic treatments, namely 1% doramectin administered subcutaneously, 5% fluralaner pour-on, and

Molecular epidemiology of the Old World screwworm fly (OWSF) in Iraq, and the genetic structure of various OWSF populations worldwide.

Despite being endemic in Iraq, no reports have been published in the past 10 years to update the molecular epidemiology of the Old World screwworm fly (OWSF), Chrysomya bezziana, in this country. In the present study, 130 sheep from 10 Iraqi governorates were found infected with C. bezziana larvae, whose identities were PCR-confirmed based on the cytochrome b (Cytb) gene, and 23 isolates from various tested governorates were successfully sequenced. Although most isolates (n = 20) belonged to the common haplotype circulating in Iraq, two new haplotypes were detected. Significant changes in OWSF
