Midostaurin response in AML is shaped by a progenitor-like cell state selectively targeted by SMAC mimetics.

FLT3-mutated acute myeloid leukemia (AML) remains difficult to treat due to frequent resistance to FLT3 inhibitors like midostaurin. In this study, we observed a progenitor-like CD38+CD45RA+ leukemic cell population that may be associated with midostaurin resistance. Midostaurin-resistant cells display disrupted membrane architecture and a shift in signaling from STAT5 to PI3K/AKT, favoring survival over apoptosis. Functional drug testing was consistent with clinical response to midostaurin, and together with multi-omic profiling, including single-cell and proteomic analyses, indicated the pre

Immunization with mosaic nanoparticles reveals that antigen avidity shapes the clonal hierarchy of the B cell response.

Vaccination with real-world complex antigens often elicits a hierarchical immune response to different epitopes. This immunodominance is a major obstacle in vaccine design because nonprotective epitopes often divert the immune response away from protective epitopes. We posited that B cell epitope avidity (i.e., total multivalent binding strength) is a key regulator of immunodominance hierarchy by modulating both germinal center (GC) seeding and B cell competitive fitness in GCs. To test these hypotheses, we used two sets of mosaic nanoparticle immunogens to precisely modulate epitope avidity b

Design-dependent shear and liquid media transport characteristics in microfluidic U-shaped cell traps.

Single-cell analysis has become central to fields ranging from cancer biology to drug screening. To exploit its potential, tools that can isolate individual cells and maintain them under controlled conditions are needed. Microfluidic platforms are an ideal fit for this, offering precise control over microenvironments. In these systems, cell behavior is shaped by parameters such as fluid shear stress (the mechanical force exerted by flow) and the transport of soluble factors (nutrients and signaling molecules). However, both are set by flow velocity, so they cannot be tuned independently. Here,

ShapeSpaceExplorer: Analysis of morphological transitions in migrating cells using similarity-based shape space mapping.

Here we describe the development of ShapeSpaceExplorer, an interactive software for the extraction, visualisation and analysis of complex 2D shape series. We also demonstrate its application to the analysis of cell morphology changes during cell migration. Cell migration is essential for many physiological and pathological processes. Intracellular force generation and transmission of these forces to extracellular structures or neighbouring cells drives cell migration. The emergent property of the processes driving cell migration is a change in cell shape. We describe a machine learning approac

Direct and capacitive electrical stimulation shapes neural progenitor cell survival and orientation on conductive scaffolds.

Electrical stimulation (ES) of neural progenitor cells (NPCs) on conductive biomaterials offers a promising strategy to enhance regenerative therapies for neurological disorders. In this study, we designed and evaluated two ES platforms, direct and capacitively coupled, using silver nanowire-integrated collagen scaffolds to investigate their effects on NPC orientation and viability. Direct stimulation promoted NPC alignment along the electric field, suggesting enhanced guidance potential for neural repair. In contrast, the capacitively coupled system did not alter cell morphology but improved

Myeloid Cell States in Influenza-Associated Pulmonary Aspergillosis Are Shaped by Iron Overload and Metabolic Reprogramming.

Virus-associated pulmonary aspergillosis is a life-threatening secondary infection that substantially increases morbidity and mortality in critically ill patients with respiratory virus infections. Influenza A virus (IAV) and SARS-CoV2 are known to disrupt pulmonary homeostasis, the mechanisms by which these perturbations render the host susceptibility to Aspergillus fumigatus (Af) remain incompletely understood. Here, we integrate an established murine model of influenza-associated pulmonary aspergillosis (IAPA) with single-cell RNA sequencing (scRNA-seq) to define the myeloid cell dysfunctio

Regnase-1 in cDC1 controls T cell priming and shapes the dynamics of experimental autoimmune encephalomyelitis.

INTRODUCTION: Multiple sclerosis is a chronic inflammatory demyelinating disease of the central nervous system (CNS), in which various immune cells contribute to disease progression, yet the role of dendritic cells (DCs) remains incompletely understood. The RNA-binding protein Regnase-1 plays an important role in regulating immune cell function, but its function in DCs, particularly conventional type 1 DCs (cDC1), has not been defined. METHODS: We investigated the role of Regnase-1 in cDC1 and its impact on experimental autoimmune encephalomyelitis (EAE) on Regnase-1 fl/+ Xcr1 -Cre+ and contr

Mitochondrial DNA transfer between malignant cells and T lymphocytes shapes the cancer-immunity dialogue.

Nonmutated mitochondrial DNA (mtDNA) from T lymphocytes can be incorporated into cancer cells bearing mutated mtDNA to repair their bioenergetic deficiency. However, a recent paper by Ikeda et al. indicates that mutated mtDNA from malignant cells can also be transferred into tumor-infiltrating T lymphocytes to subvert their function in cancer immunosurveillance.

Inflammatory conditions shape phenotypic and functional characteristics of lung-resident memory T cells in mice.

Lung tissue-resident memory T cells (TRM) are critical for the local control of respiratory tract infections caused by influenza A viruses (IAV). Here we compare TRM populations induced by intranasal adenoviral vector vaccines encoding hemagglutinin and nucleoprotein (NP) with those induced by an H1N1 infection in BALB/c mice. While vaccine-induced TRM express high levels of CD103 and persist longer in the lung parenchyma, short-lived, H1N1-induced TRM have a transcriptome associated with higher cytotoxic potential and distinct transcriptional profile as shown by single-cell RNA sequencing. Th17 cells/IL-17A shape Pasteurella multocida serotype A infection in murine and rabbit models.

Pasteurella multocida (Pm) is an important zoonotic respiratory pathogen, especially serotype A, which can cause death in various hosts, including humans, farm animals, and wildlife. However, the exact mechanisms by which hosts mobilize the immune system against Pasteurella multocida serotype A (PmA) in mammals remain largely unexplored. In the present study, we found that the control of lung infection is critical for PmA infection. Furthermore, unbiased RNA sequencing analysis and histopathological studies revealed that T-cell associated immunity was not fully activated during PmA infection,

Ehmt2 inactivation in pancreatic epithelial cells shapes the transcriptional landscape and inflammation response of the whole pancreas.

Introduction: The Euchromatic Histone Methyl Transferase Protein 2 (EHMT2), also known as G9a, deposits transcriptionally repressive chromatin marks that play pivotal roles in the maturation and homeostasis of multiple organs. Recently, we have shown that Ehmt2 inactivation in the mouse pancreas alters growth and immune gene expression networks, antagonizing Kras-mediated pancreatic cancer initiation and promotion. Here, we elucidate the essential role of Ehmt2 in maintaining a transcriptional landscape that protects organs from inflammation. Methods: Comparative RNA-seq studies between normal

Oxygen Exposure and Tolerance Shapes the Cell Wall-Associated Lipids of the Skin Commensal Cutibacterium acnes.

Cutibacterium acnes is one of the most abundant bacteria on the skin. Being exposed to oxygen and oxic stress, the secretion of the bacterial antioxidant protein RoxP ensures an endogenous antioxidant system for the preservation of skin health. To investigate the impact of the antioxidant RoxP on oxidation of the bacteria, wildtype and an isogenic roxp mutant were cultured in anaerobic and oxic conditions. The carbonylated status of proteins were recorded, as were the most significant modifications in a relative intensity of free fatty acids (FFA) and lipids containing fatty acids (FA), such a

TIAR and FMRP shape pro-survival nascent proteome of leukemia cells in the bone marrow microenvironment.

Chronic myeloid leukemia (CML) cells circulate between blood and bone marrow niche, representing different microenvironments. We studied the role of the two RNA-binding proteins, T-cell-restricted intracellular antigen (TIAR), and the fragile X mental retardation protein (FMRP) in the regulation of protein translation in CML cells residing in settings mimicking peripheral blood microenvironment (PBM) and bone marrow microenvironment (BMM). The outcomes showed how conditions shaped the translation process through TIAR and FMRP activity, considering its relevance in therapy resistance. The QuaNC

The regional distribution of resident immune cells shapes distinct immunological environments along the murine epididymis.

The epididymis functions as transition zone for post-testicular sperm maturation and storage and faces contrasting immunological challenges, i.e. tolerance towards spermatozoa vs. reactivity against pathogens. Thus, normal organ function and integrity relies heavily on a tightly controlled immune balance. Previous studies described inflammation-associated tissue damage solely in the distal regions (corpus, cauda), but not in the proximal regions (initial segment, caput). To understand the observed region-specific immunity along the epididymal duct, we have used an acute bacterial epididymitis

NK cells with tissue-resident traits shape response to immunotherapy by inducing adaptive antitumor immunity.

T cell-directed cancer immunotherapy often fails to generate lasting tumor control. Harnessing additional effectors of the immune response against tumors may strengthen the clinical benefit of immunotherapies. Here, we demonstrate that therapeutic targeting of the interferon-γ (IFN-γ)-interleukin-12 (IL-12) pathway relies on the ability of a population of natural killer (NK) cells with tissue-resident traits to orchestrate an antitumor microenvironment. In particular, we used an engineered adenoviral platform as a tool for intratumoral IL-12 immunotherapy (AdV5-IL-12) to generate adaptive anti

MHC-II alleles shape the CDR3 repertoires of conventional and regulatory naïve CD4+ T cells.

T cell maturation and activation depend upon T cell receptor (TCR) interactions with a wide variety of antigenic peptides displayed in a given major histocompatibility complex (MHC) context. Complementarity-determining region 3 (CDR3) is the most variable part of the TCRα and -β chains, which govern interactions with peptide-MHC complexes. However, it remains unclear how the CDR3 landscape is shaped by individual MHC context during thymic selection of naïve T cells. We established two mouse strains carrying distinct allelic variants of H2-A and analyzed thymic and peripheral production and TCR

Shape-engineered titanium dioxide nanoparticles (TiO2-NPs): cytotoxicity and genotoxicity in bronchial epithelial cells.

The aim of this study was to evaluate cytotoxicity (WST-1 assay), LDH release (LDH assay) and genotoxicity (Comet assay) of three engineered TiO2-NPs with different shapes (bipyramids, rods, platelets) in comparison with two commercial TiO2-NPs (P25, food grade). After NPs characterization (SEM/T-SEM and DLS), biological effects of NPs were assessed on BEAS-2B cells in presence/absence of light. The cellular uptake of NPs was analyzed using Raman spectroscopy. The cytotoxic effects were mostly slight. After light exposure, the largest cytotoxicity (WST-1 assay) was observed for rods; P25, bipy

Dendritic cell factor 1 deletion leads to developmental defects in mushroom-shaped dendritic spines.

Dendritic spines are divided into four subtypes, namely, Mushroom, Stubby, Thin, and Branched. The mushroom-shaped spines are related to learning and memory. Previous studies have shown that the dendritic cell factor 1 (Dcf1, a transmembrane protein) affects the memory process and regulates the development of dendritic spines by inhibiting the expression of lipocalin 2 (Lcn2, a member of the family containing over 20 small secreted proteins). However, the exact subtype of dendritic spines that are specifically affected by Dcf1 remains unknown. Here, we identified that deletion of Dcf1 leads to

NK cells shape pancreatic and oral tumor microenvironments; role in inhibition of tumor growth and metastasis.

We have recently shown that natural killer (NK) cells select and differentiate cancer stem cells (CSCs)/undifferentiated tumors via secreted and membrane bound IFN-gamma (IFN-γ) and TNF-alpha (TNF-α), preventing tumor growth and inducing remodeling of the tumor microenvironment. Since many conventional therapeutic strategies, including chemotherapy and radiotherapy remain fairly unsuccessful in treating CSCs/poorly differentiated tumors, there has been an increasing interest in NK cell-targeted immunotherapy for the treatment of aggressive tumors. In our recent studies, we used humanized-BLT (
