The ciliary transitional zone and nephrocystins.

Loss of cilia and ciliary protein causes various abnormalities (called ciliopathy), including situs inversus, renal cystic diseases, polydactyly and dysgenesis of the nervous system. Renal cystic diseases are the most frequently observed symptoms in ciliopathies. Cilia are microtubule-based organelles with the following regions: a ciliary tip, shaft, transitional zone and basal body/mother centriole. Joubert syndrome (JBTS), Meckel Gruber syndrome (MKS) and Nephronophthisis (NPHP) are overlapping syndromes. Recent studies show that JBST and MKS responsible gene products are localized in the tr

[On the ontogenetic origin of human beings: the scientific solution].

Every living being is the result of a genome-environment interaction. Neither human oocytes nor spermatozoids have human functional genomes, but the zygote that they constitute may have a human functional genome and other functional genomes such as those of the hydatidiform mole, polyploids, and non-human living beings. When the zygotic human functional genome is integrated and activated, the biotic humanity is acquired. This may occur when the paternal chromatin decondenses; the nuclear environment and envelope of both nuclei are changed to constitute pronuclei; the replacement of sperm prota

Paternal effects on cell division in the human preimplantation embryo.

Cell divisions in the human preimplantation embryo can be compromised by deficiencies in sperm nuclear genome or sperm-derived developmentally relevant cytoplasmic factors, oocyte activating substance and centriole. Sperm nuclear deficiencies are usually not detected before the 8-cell stage of embryo development, when a major expression of sperm-derived genes has begun. Sperm cytoplasmic deficiencies can be detected as early as the 1-cell zygote and then throughout the preimplantation development. The terms 'late paternal effect' and 'early paternal effect' have been suggested to denote these

Homozygous mutations in DZIP1 can induce asthenoteratospermia with severe MMAF.

BACKGROUND: Asthenoteratospermia, one of the most common causes for male infertility, often presents with defective sperm heads and/or flagella. Multiple morphological abnormalities of the sperm flagella (MMAF) is one of the common clinical manifestations of asthenoteratospermia. Variants in several genes including DNAH1, CEP135, CATSPER2 and SUN5 are involved in the genetic pathogenesis of asthenoteratospermia. However, more than half of the asthenoteratospermia cases cannot be explained by the known pathogenic genes. METHODS AND RESULTS: Two asthenoteratospermia-affected men with severe MMA

Development of Polyspermic Rice Zygotes.

Fertilization is a general feature of eukaryotic uni- and multicellular organisms to restore a diploid genome from female and male gamete haploid genomes. In most animals and fucoid algae, polyspermy block occurs at the plasmogamy step. Because the polyspermy barrier in animals and in fucoid algae is incomplete, polyspermic zygotes are generated by multiple fertilization events. However, these polyspermic zygotes with extra centrioles from multiple sperms show aberrant nuclear and cell division. In angiosperms, polyspermy block functions in the egg cell and the central cell to promote faithful

Taenia solium: germinal cell precursors in tapeworms grown in hamster intestine.

BACKGROUND: In a previous study, it was shown that growth of evaginated metacestodes occurs in the germinative tissue of the neck by duplication of somatic stem cells. In these specimens, it was not possible to find the mitotic figures required to demonstrate duplication of germ cell lines. METHODS: Taenia solium strobilae were collected from the intestinal lumen of outbred hamsters infected orally with 10 metacestodes dissected from naturally infected pigs. Animals were anesthetized 1-10 days postinfection, the small intestine excised, submerged in PBS, and cut open longitudinally. Live Taen

Rational design of CZL-S092: A novel indazole-based PLK4 inhibitor targeting neuroblastoma through virtual screening and fragment-based drug design strategies.

Polo like kinase 4 (PLK4) is a critical member of the polo-like kinase family that works as a master regulator of centriole duplication and mitotic progression. Overexpression of PLK4 has been documented in various cancers and PLK4 inhibitors are regarded as a potential strategy for cancer treatment. In this study, we identified indazole derivative 8 as a hit compound through a virtual screening approach. Subsequent fragment-based rational drug design strategy was used to modify the chemical structure of compound 8 and yielded 34 indazole derivatives targeting PLK4. Among them, compound 34b (C

Biallelic TEDC1 variants cause a new syndrome with severe growth impairment and endocrine complications.

We encountered two affected male patients born to non-consanguineous parents, who presented with prenatal-onset severe growth impairment, primary microcephaly, developmental delay, adrenal insufficiency, congenital glaucoma, delayed bone aging, craniosynostosis, congenital tracheal stenosis, and primary hypogonadism. By exome sequencing, we identified compound heterozygous TEDC1 variants (NM_001134877.1 c.[104-5C>G];[787delG] p.[?];[(Ala263LeufsTer29)] in both affected siblings. We confirmed that the splice site variant, c.104-5C>G, leads to no TEDC1 protein production via nonsense-mediated mR

CCDC189 depletion leads to oligo-astheno-teratozoospermia and male infertility in mice†.

In male reproductive system, proteins containing the coiled-coil domain (CCDC) are predominantly expressed in specific regions including the testis, epididymis, seminal vesicle, and prostate. They play a vital role in centriole formation, sperm motility and flagellar development in male gametes. Despite being highly expressed in the testis, the exact physiological function of the coiled-coil domain-containing 189 (Ccdc189) gene remain largely unclear. Our research provides a comprehensive and detailed investigation into the localization of CCDC189 protein within the testis seminiferous tubules

Identification of Pathogenic Mutations in Primary Microcephaly- (MCPH-) Related Three Genes CENPJ, CASK, and MCPH1 in Consanguineous Pakistani Families.

Microcephaly (MCPH) is a developmental anomaly of the brain known by reduced cerebral cortex and underdeveloped intellectual disability without additional clinical symptoms. It is a genetically and clinically heterogenous disorder. Twenty-five genes (involved in spindle positioning, Wnt signaling, centriole biogenesis, DNA repair, microtubule dynamics, cell cycle checkpoints, and transcriptional regulation) causing MCPH have been identified so far. Pakistani population has contributed in the identification of many MCPH genes. WES of three large consanguineous families revealed three pathogenic

A novel function of Huntingtin in the cilium and retinal ciliopathy in Huntington's disease mice.

Huntington's disease (HD) is a neurodegenerative disorder caused by the toxic expansion of polyglutamine in the Huntingtin (HTT) protein. The pathomechanism is complex and not fully understood. Increasing evidence indicates that the loss of normal protein function also contributes to the pathogenesis, pointing out the importance of understanding the physiological roles of HTT. We provide evidence for a novel function of HTT in the cilium. HTT localizes in diverse types of cilia--including 9 + 0 non-motile sensory cilia of neurons and 9 + 2 motile multicilia of trachea and ependymal cells--whic

A novel apical midpiece defect in the spermatozoa of a bull without an apparent decrease in motility and fertility. A case study.

Despite some limitations as predictors of fertility, evaluation of sperm morphology and progressive motility is the commonest method to assess viability of frozen/thawed semen. In this article we describe by light and transmission electron microscopy a novel midpiece structural defect observed in 24-36% of frozen/thawed sperm cells from a Charolais bull, used in artificial insemination programs without any apparent ill effect to the fertility. After thawing, the sperm progressive motility ranged from 65 to 80% and the pregnancy rate for all artificial inseminations performed (43%) did not diff

The Meckel-Gruber Syndrome proteins MKS1 and meckelin interact and are required for primary cilium formation.

Meckel-Gruber syndrome (MKS) is an autosomal recessive lethal malformation syndrome characterized by renal cystic dysplasia, central nervous system malformations (typically, posterior occipital encephalocele), and hepatic developmental defects. Two MKS genes, MKS1 and MKS3, have been identified recently. The present study describes the cellular, sub-cellular and functional characterization of the novel proteins, MKS1 and meckelin, encoded by these genes. In situ hybridization studies for MKS3 in early human embryos showed transcript localizations in agreement with the tissue phenotype of MKS p

Characterization of antinuclear autoantibodies present in the serum from nonobese diabetic (NOD) mice.

The nonobese diabetic (NOD) mouse develops insulin-dependent type 1 diabetes in response to autoantibodies and T-cell attack directed against pancreatic islet cell antigens. Sera obtained from nondiabetic and diabetic female mice demonstrated a 1.4-fold increase in IgG levels when compared to BALB/c control animals. Nondiabetic and diabetic male mice had a 2.1- and 3-fold increase, respectively, in serum IgG levels over that of control mice. Seven of 11 non-diabetic and 7/10 diabetic sera from female NOD mice contained antibody to cytoplasmic or nuclear components of HEp-2 cells. Cytoplasmic s

The making of abnormal spermatozoa: cellular and molecular mechanisms underlying pathological spermiogenesis.

Fertilization in mammals occurs via a series of well-defined events in the secluded environment of the female reproductive tract. The mode of selection of the fertilizing spermatozoon nevertheless remains unknown. As has become evident during in vitro fertilization by sperm microinjection into the oocyte, abnormal spermatozoa can successfully fertilize oocytes. Under these extreme conditions, post-fertilization events, early embryonic development and implantation are significantly compromised indicating that the contribution of spermatozoa extends beyond sperm penetration. Cep70 and Cep131 contribute to ciliogenesis in zebrafish embryos.

BACKGROUND: The centrosome is the cell's microtubule organising centre, an organelle with important roles in cell division, migration and polarity. However, cells can divide and flies can, for a large part of development, develop without them. Many centrosome proteins have been identified but the roles of most are still poorly understood. The centrioles of the centrosome are similar to the basal bodies of cilia, hair-like extensions of many cells that have important roles in cell signalling and development. In a number of human diseases, such Bardet-Biedl syndrome, centrosome/cilium proteins a

Big1 is a cell-cycle regulator linking cell size to basal body number in Tetrahymena thermophila.

Cell-size control is necessary for the optimum function and fitness of cells. In many eukaryotes, cell size is controlled through the initiation of cell division once a size threshold is achieved.1 Cells sense their size through the relationship of cell-size-dependent and independent factors, though the mechanistic details are known for few eukaryotes.2,3 An additional dimension of cell-size control is the subcellular localization and control of cell-cycle regulators. The cell cortex is a known site of cell-cycle regulator accumulation that maintains an established cell size based on surface a

Long-lasting arrest of murine polycystic kidney disease with CDK inhibitor roscovitine.

Polycystic kidney diseases (PKDs) are primarily characterized by the growth of fluid-filled cysts in renal tubules leading to end-stage renal disease. Mutations in the PKD1 or PKD2 genes lead to autosomal dominant PKD (ADPKD), a slowly developing adult form. Autosomal recessive polycystic kidney disease results from mutations in the PKHD1 gene, affects newborn infants and progresses very rapidly. No effective treatment is currently available for PKD. A previously unrecognized site of subcellular localization was recently discovered for all proteins known to be disrupted in PKD: primary cilia.

Ultrastructure of the pineal body of the common vampire bat, Desmodus rotundus.

The type AB pineal body of the common vampire bat, Desmodus rotundus, was recessed and lobulated, was extensively vascularized and intimately related to great veins, and was unassociated with the epithalamic region. The habenular and the posterior commissures coursed anteriorly and were unassociated with the pineal. The saccular suprapineal recess of the third ventricle extended dorsally juxtaposed to the pineal body. These anatomical features are likely to make pinealectomies in the vampire more difficult to manage. The pineal parenchyma consisted of light pinealocytes surrounded by canalicul

CEP170 as a novel molecular link between centrosomal function and cerebral cortical development.

BACKGROUND: The centrosome is a critical regulator of cortical development, orchestrating microtubule dynamics, cell cycle progression, and neuronal migration. Disruptions in centrosome-associated proteins have been associated with a range of neurodevelopmental disorders. CEP170, a microtubule-binding protein localized to the subdistal appendages (SDA) of centrioles, has been implicated in centrosome function, yet its role in corticogenesis remains poorly defined. METHODS: We analyzed CEP170 expression in mouse cortex using western blotting, qPCR, scRNA-seq, and spatial transcriptomics, and e
