The inherent bioinertness of zirconia (ZrO2) hinders its early bone integration, presenting a significant obstacle to its widespread use in dental implant technologies. Addressing this, we developed a surface coating leveraging the synergistic effects of catecholamine and coordination chemistry inspired by the mussel byssus cuticle. This coating, named PDPA@Sr, is enriched with strontium ions and amine groups, resulting from a simple immersion of polydopamine (PD)-coated ZrO2 in an alkaline strontium chloride and poly(allylamine) (PA) solution. Compared to conventional mussel-inspired PD coati

In vivo histone deacetylase (HDAC) inhibition by vorinostat under clinically acceptable dosing is limited by its poor pharmacokinetics properties. A new type of nontoxic pH-responsive delivery system has been synthesized by ring-opening metathesis polymerization, allowing for the selective distribution of vorinostat in mesothelioma tumors in vivo and subsequent histone reacetylation. The delivery system is synthesized by generic click chemistry, possesses native stealth properties for passive tumor targeting, and does not need additional chemistry for cellular internalization. Although vorinos

The tetrazine ligation, an inverse electron-demand Diels-Alder reaction between tetrazine and a dienophile such as norbornene (Nb) or trans-cyclooctene (TCO), has characteristics that make it a potentially ideal chemistry for the preparation of nanoparticle bioconjugates. Although this click chemistry has been used for this purpose, relatively little is known about how to optimize nanoparticle surface chemistry and tagging with reactive functional groups to maximize the chances of successful conjugation. Here, we addressed this open question with quantum dots (QDs) by preparing and testing a p

Sulphur fluoride exchange (SuFEx) is a category of click chemistry that enables covalent linking of modular units through sulphur connective hubs. Here, we reported an efficient synthesis and in situ screening method for building a library of sulphonamides on the picomolar scale by SuFEx reaction between a sulphonyl fluoride (RSO2F) core and primary or secondary amines. This biocompatible SuFEx reaction would allow us to rapidly synthesise sulphonamide molecules, and evaluate their ChE inhibitory activity. Compound T14-A24 was identified as a reversible, competitive, and selective AChE inhibit

In this contribution, Fe3O4 magnetic nanoparticles (MNPs) have been functionalized with a tetraphosphonate cavitand receptor (Tiiii), capable of complexing N-monomethylated species with high selectivity, and polyethylene glycol (PEG) via click-chemistry. The grafting process is based on MNP pre-functionalization with a bifunctional phosphonic linker, 10-undecynylphosphonic acid, anchored on an iron surface through the phosphonic group. The Tiiii cavitand and the PEG modified with azide moieties have then been bonded to the resulting alkyne-functionalized MNPs through a "click" reaction. Each r

Ultrasound-responsive chemical scaffolds capable of operating under clinically relevant conditions remain rare, limiting the development of programmable ultrasound-controlled molecular systems. Here, we report the discovery of a stable yet ultrasound-responsive arylurea-dihydrotetrazine (ArU-DHTz) scaffold that undergoes efficient activation under 1 MHz therapeutic ultrasound without requiring exogenous sonosensitizers. Structure-responsiveness studies revealed that activation arises from a unique combination of a monosubstituted arylurea motif and an appropriately tuned DHTz core, while mecha

Proximity labeling has revolutionized the study of dynamic subcellular proteomes by enabling the capture of transient protein interactions within living cells, yet the application of existing platforms to hard-to-transfect primary cells remains challenging. Here, we leverage a bioorthogonal proximity labeling platform based on the copper-dependent tyrosinase BmTyr to profile subcellular proteomes in primary T cells. This system catalyzes the subcellular incorporation of an alkyne-phenol probe, enabling subsequent click-compatible conjugation to versatile azide-bearing tags for fluorescence ima

The use of gold nanoclusters in biomedical applications has been steadily increasing in recent years. However, water solubility is a key factor for these applications, and water-soluble gold nanoclusters are often difficult to isolate and susceptible to exchange or oxidation in vivo. Herein, we report the isolation of N-heterocyclic carbene (NHC)-protected atomically precise gold nanoclusters functionalized with triethylene glycol monomethyl ether groups. These clusters are highly luminescent and water soluble and are shown to be stable in biological media. Importantly, the core structure, sta

Cellulose nanocrystals (CNCs) are promising nanomaterials for biomedical applications owing to their biocompatibility, low toxicity, and versatile surface chemistry. However, efficient surface modification of CNCs with hydrophobic moieties and functional polymers remains challenging under mild aqueous conditions. Herein, we present an aqueous-phase "grafting-to" strategy that enables orthogonal conjugation of meso-tetra(4-carboxyphenyl)porphine (TCPP) and polyethylene glycol (PEG)-based polymers using a heterobifunctional linker. Aldehyde-functionalized CNCs were modified with dibenzocycloocty

Translation inhibitors have therapeutic potential against Candida species. Here, we present a protocol to measure translation inhibition in Candida spp. We describe steps for employing an alkynylated methionine analog, L-homopropargylglycine (HPG), that becomes incorporated into newly synthesized proteins. We then detail procedures to perform a click reaction of the alkyne with a fluorescent azide, which is visualized using fluorescence microscopy and quantified by flow cytometry. For complete details on the use and execution of this protocol, please refer to Puumala et al.,1 Fu et al.,2 and I

We report here a Cu-catalyzed azide-alkyne-thiol reaction forming thiotriazoles as the major byproduct under widely used bio-orthogonal protein labeling "click" conditions. The development of Cu(I)-catalyzed azide-alkyne cycloaddition (CuAAC) had a tremendous impact on many biological discoveries. However, the considered chemoselectivity of CuAAC is hampered by the high reactivity of cysteine free thiols, yielding thiotriazole protein conjugates. The reaction byproducts generate false-positive protein hits in functional proteomic studies. The reported detail investigation of conjugates between

In this study, we have successfully synthesized bis (cholesterol-dibenzo-18-crown-6-ether)-pillar[5]arene compound 1 through a click reaction, which could spontaneously insert into lipid bilayers to form ion channel due to the membrane anchor cholesterol group and show significant transport activity of K+ superior to Na+, with a permeability ratio of K+/Na+ equal to 4.58. Compound 1 two crown ether modules act as selective filters similar to natural K+ channel, which are determined to 1 : 2 binding stoichiometry to K+ by Job's plot and NMR titration. This structurally unambiguously unimolecule

In this work, we report the use of bioorthogonal chemistry, specifically the strain-promoted click azide-alkyne cycloaddition (SPAAC) for the covalent attachment of magnetic nanoparticles (MNPs) on living cell membranes. Four types of MNPs were prepared, functionalized with two different stabilizing/passivation agents (a polyethylene glycol derivative and a glucopyranoside derivative, respectively) and two types of strained alkynes with different reactivities: a cyclooctyne (CO) derivative and a dibenzocyclooctyne (DBCO) derivative. The MNPs were extensively characterized in terms of physicoch

Interrogating biological systems is often limited by access to biological probes. The emergence of "click chemistry" has revolutionized bioconjugate chemistry by providing facile reaction conditions amenable to both biologic molecules and small molecule probes such as fluorophores, toxins, or therapeutics. One particularly popular version is the copper-catalyzed azide-alkyne cycloaddition (AAC) reaction, which has spawned new alternatives such as the strain-promoted azide-alkyne cycloaddition reaction, among others. This focused review highlights practical approaches to AAC reactions for the s

Poly(aminoester) (PAE) dendrimers are attractive materials for biomedical applications owing to their biodegradability and precise structure, effective synthesis and favourable safety profile. We report here a click chemistry-enabled divergent-convergent strategy for the efficient synthesis of amphiphilic PAE dendrimers. These dendrimers exhibit negligible cytotoxicity, generation-dependent degradation behaviour, and tunable self-assembly controlled by the hydrophobic chain length. This study offers an accessible synthesis of PAE dendrimers, advancing their use in future biomedical application

Modified peptides are an important class of drugs due to their high specificity and affinity in particular to pharmacological targets that are not druggable by conventional small molecules. Methods that allow the efficient diversification of peptides with non-canonical modifications are therefore highly sought after. One possible strategy for accessing broadly diversified peptides is on-resin late-stage modification, whereby a functional site-typically an amino acid side chain or the N-terminus-is usually singly modified after peptide chain assembly, while the other functionalities remain prot

mRNA is a new medical modality as vaccine and raises hopes to become a protein replacement agent. Despite notable advances in manufacturing, stabilization and modification, its short half-life remains a major limitation. Circular RNA bears potential to solve this issue, as degradation of mRNA proceeds primarily from the 5' and 3' ends. However, circular RNAs normally lack a 5' cap and are less efficiently translated. We present a strategy to circularize mRNA covalently via click chemistry by modifying the 5' cap and the 3' poly(A) tail with bioorthogonal functional groups. The resulting Cyclic

Proteolysis-targeting chimeras (PROTACs) have become a popular therapeutic strategy, and the development of multi-functional PROTACs has added complexity to their synthetic process. Although click reactions have been widely applied to prepare highly functionalized biomolecules, most of them are limited to two-component reactions, restricting the creation of more complex structures. Here, we developed a convenient multi-component assembly strategy via strain-promoted alkyne-nitrone cycloaddition (SPANC), which can be extended to a 3-component reaction when combined with nitrone formation. We synthesized two green-light activatable 5'-caps for oligonucleotides based on the BODIPY and coumarin scaffold. Both bear an alkyne functionality allowing their use in numerous biological applications. They were successfully incorporated in oligonucleotides via solid-phase synthesis. Copper-catalyzed alkyne-azide cycloaddition (CuAAC) using a bisazide photo-tether gave cyclic oligonucleotides that could be relinearized by activation with green light and were shown to exhibit high stability against exonucleases. Chemical ligation as another example for bioconjugation yielded oligonucleotides

In the study of multivalent interactions at interfaces, as occur for example at cell membranes, the density of the ligands or receptors displayed at the interface plays a pivotal role, affecting both the overall binding affinities and the valencies involved in the interactions. In order to control the ligand density at the interface, several approaches have been developed, and they concern the functionalization of a wide range of materials. Here, different methods employed in the modification of surfaces with controlled densities of ligands are being reviewed. Examples of such methods encompas
