Ultraviolet radiation reshapes the transcriptomic landscape of human skin aging: Insights from a multi-age comparative study.

BACKGROUND: Skin aging arises from both intrinsic processes and extrinsic factors, with ultraviolet (UV) radiation being the primary extrinsic cause of photoaging. However, the molecular mechanisms that differentiate these processes across the human lifespan remain incompletely characterized. OBJECTIVE: This study aimed to comprehensively compare the dynamic transcriptomic profiles of photoaged (neck, high UV exposure) and intrinsically aged (chest, low UV exposure) skin across three age groups (young, middle-aged, elderly), and to integrate these findings with biophysical skin measurements.

Current Evidence on the Use of Biomarkers for the Diagnosis of Acute Mesenteric Ischemia: A Narrative Review.

Acute mesenteric ischemia (AMI) is a life-threatening vascular emergency associated with high morbidity and mortality, largely because diagnosis is frequently delayed by non-specific clinical manifestations. Although computed tomography angiography (CTA) remains the diagnostic gold standard, considerable research has focused on identifying laboratory biomarkers that may facilitate earlier recognition and improve clinical decision-making. This narrative review summarizes the current evidence regarding routinely available and emerging laboratory biomarkers for the diagnosis of AMI. A comprehensi

Selective Thyroid Hormone Receptor-Beta (TRβ) Agonists: New Perspectives for the Treatment of Metabolic and Neurodegenerative Disorders.

Thyroid hormones (THs) elicit significant effects on numerous physiological processes, such as growth, development, and metabolism. A lack of thyroid hormones is not compatible with normal health. Most THs effects are mediated by two different thyroid hormone receptor (TR) isoforms, namely TRα and TRβ, with the TRβ isoform known to be responsible for the main beneficial effects of TH on liver. In brain, despite the crucial role of TRα isoform in neuronal development, TRβ has been proposed to play a role in the remyelination processes. Consequently, over the past two decades, much effort has be

UQCR10 and TNNT1: novel biomarkers for sarcopenia identified through integrated transcriptomic analysis and machine learning.

BACKGROUND: Sarcopenia, the age-related loss of muscle mass and function, significantly impacts health outcomes in older adults. Despite its prevalence, molecular diagnostics and targeted therapies remain limited due to incomplete understanding of its pathophysiology. METHODS: We analyzed the GSE226151 dataset comparing skeletal muscle transcriptomes from sarcopenic (n = 20) and healthy adults (n = 20) using differential expression analysis and weighted gene co-expression network analysis (WGCNA). Key genes were identified through the intersection of differentially expressed genes and hub gen

Dose-dependent effects of methylene blue on hemodynamics, cytokines, oxidative stress, and organ dysfunction in a rat model of CLP-induced sepsis: An experimental study.

BACKGROUND: Septic shock involves excessive nitric oxide (NO) production and overactivation of the NO/sGC/cGMP pathway, leading to severe vasodilation and microvascular dysfunction. Methylene blue (MB), an inhibitor of this pathway, has been proposed as a catecholamine-sparing adjunct, yet optimal therapeutic dosing remains unclear. This study investigated the dose-dependent effects of MB in a rat model of polymicrobial sepsis. METHODS: Fifty female Wistar rats were randomized into control, CLP-induced sepsis, or CLP+MB groups (10, 50, or 100 mg/kg, i.v.). Sepsis was induced via cecal ligatio

Integrated biomarker landscape for the early detection and management of calcific aortic valve disease.

BACKGROUND: Calcific aortic valve disease (CAVD) is the predominant valvular pathology in older adults, advancing from aortic sclerosis to life-threatening stenosis. Without effective medical therapies, intervention mainly relies on timely valve replacement, although silent myocardial and valvular damage may progress before symptoms arise. Early, non-invasive detection of disease activity is a crucial unmet need. AIMS: To review circulating and mechanistic biomarkers reflecting the core pathogenic pathways of CAVD and asses their potential for early detection and patient-specific risk stratif

In-depth cerebrovascular lipidomics profiling for discovering novel biomarkers and mechanisms in moyamoya and intracranial atherosclerotic disease.

BACKGROUND: Despite considerable research efforts, the precise etiology and underlying pathways contributing to moyamoya disease (MMD) remain poorly understood. Moreover, the overlapping vascular pathologies shared between MMD and intracranial atherosclerotic disease (ICAD) pose challenges in clinical differentiation, even with gold-standard cerebral angiography. An in-depth exploration of lipidomic alterations in cerebral intracranial MMD vessels could offer valuable insights into the pathogenesis of MMD-related mechanisms, encompassing MMD and ICAD, and unveil novel biomarkers and potential

Gut microbiota-based biomarkers for precision subtype classification and mechanistic understanding of biliary and hyperlipidemic acute pancreatitis.

BACKGROUND: Acute pancreatitis (AP) is an inflammatory disorder with distinct etiological subtypes, yet the role of gut microbiota in disease pathogenesis remains poorly understood. We hypothesized that biliary acute pancreatitis (BAP) and hyperlipidemic acute pancreatitis (HLAP) exhibit etiology-specific gut microbiota signatures that correlate with disease severity and metabolic dysfunction. METHODS: We conducted a cross-sectional study in which stool samples were collected from 20 BAP patients, 20 HLAP patients, and 20 healthy controls (HC) for 16S rRNA gene sequencing to compare gut micro

N-thiadiazole-4-hydroxy-2-quinolone-3-carboxamides bearing heteroaromatic rings as novel antibacterial agents: Design, synthesis, biological evaluation and target identification.

Due to the occurrence of antibiotic resistance, bacterial infectious diseases have become a serious threat to public health. To overcome antibiotic resistance, novel antibiotics are urgently needed. N-thiadiazole-4-hydroxy-2-quinolone-3-carboxamides are a potential new class of antibacterial agents, as one of its derivatives was identified as an antibacterial agent against S. aureus. However, no potency-directed structural optimization has been performed. In this study, we designed and synthesized 37 derivatives, and evaluated their antibacterial activity against S. aureus ATCC29213, which led

A Single Intravenous Dose of Indole-producing Attenuated Brucella in NOD Mice: Assessment of Type 1 Diabetes Onset and Islet Immune Remodeling.

Type 1 diabetes (T1D) results from autoimmune destruction of pancreatic β cells, and current therapies fail to restore durable immune tolerance. Metabolically engineered bacteria producing immunomodulatory metabolites represent a promising single-dose therapeutic strategy. Here, this paper presents a comprehensive protocol in which a single intravenous administration of an indole-producing attenuated Brucella melitensis strain BmΔvjbR::tnaA is used to assess effects on the onset of autoimmune diabetes in prediabetic female NOD/ShiLtJ mice. The protocol begins with preparing BmΔvjbR::tnaA from

Re-evaluating Cardiovascular Risk: A Narrative Review Challenging the Cholesterol Hypothesis and Identifying Modern Dietary Drivers.

Between 1920 and 1950, cardiovascular disease (CVD) underwent a profound epidemiological shift, rising from a relatively rare and infrequently diagnosed condition to become the leading cause of death in industrialized nations. This epidemic coincided with a series of changes in the food supply, including the expanded use of refined carbohydrates, industrial seed and vegetable oils, and trans fatty acids. In response, the "Diet-Heart Hypothesis" emerged, dominated by Ancel Keys' lipid theory, which focused scientific and public health attention on saturated fat and cholesterol as the primary ca

Lifestyle and mood correlates of cardiometabolic risk in people with serious mental illness on second-generation antipsychotic medications.

INTRODUCTION: Cardiovascular morbidity and mortality are high in people with serious mental illness (SMI). This problem is mediated, at least in part, by metabolic side effects of second-generation antipsychotics (SGAs) and by unhealthy lifestyle behaviors. We asked whether oral glucose tolerance testing (oGTT) or hemoglobin A1c (HbA1c) is superior in identifying people with SMI at high cardiometabolic risk and whether this risk is shaped by mood, cognition, or lifestyle habits. METHODS: We evaluated 40 patients with schizophrenia, schizoaffective, or bipolar disorder receiving SGAs by oGTT,

Vitamin D and Hypophosphatemia in Patients with Anorexia Nervosa and Avoidant/Restrictive Food Intake Disorder: A Case Control Study.

BACKGROUND: Refeeding hypophosphatemia (RH) is a common complication of nutritional restoration in malnourished individuals, yet clear risk stratification remains elusive. Individuals with anorexia nervosa (AN) and avoidant/restrictive food intake disorder (ARFID) may be deficient in vitamin D, an important component of dietary phosphorus absorption in the gut. The relationship between vitamin D and RH in AN and ARFID is unknown. Therefore, the aims of this of this study were to 1) describe the prevalence of low serum 25-hydroxy vitamin D levels and RH in AN and ARFID 2) report associations be
