Blood meal sources and bacterial microbiome diversity in wild-caught tsetse flies.

Tsetse flies are the vectors of African trypanosomiasis affecting 36 sub-Saharan countries. Both wild and domestic animals play a crucial role in maintaining the disease-causing parasites (trypanosomes). Thus, the identification of animal reservoirs of trypanosomes is vital for the effective control of African trypanosomiasis. Additionally, the biotic and abiotic factors that drive gut microbiome diversity in tsetse flies are primarily unresolved, especially under natural, field conditions. In this study, we present a comprehensive DNA metabarcoding approach for individual tsetse fly analysis

Characterization of the bacterial communities of life stages of free living lone star ticks (Amblyomma americanum).

The lone star tick (Amblyomma americanum) is an abundant and aggressive biter of humans, domestic animals, and wildlife in the southeastern-central USA and an important vector of several known and suspected zoonotic bacterial pathogens. However, the biological drivers of bacterial community variation in this tick are still poorly defined. Knowing the community context in which tick-borne bacterial pathogens exist and evolve is required to fully understand the ecology and immunobiology of the ticks and to design effective public health and veterinary interventions. We performed a metagenomic su

Prevalence and subtype analysis of Blastocystis in healthy Indian individuals.

There is a growing interest in subtype (ST) analysis of the intestinal parasite Blastocystis due to its extensive genetic diversity that might reflect differences in pathogenicity. Although essential for reference, few studies are available on Blastocystis in healthy individuals. Moreover, molecular epidemiology data on Blastocystis in India still remain to emerge. In the present study we identified the prevalence and ST distribution of Blastocystis in healthy Indian individuals. A total of 220 stool samples were obtained; four of 100 samples from 100 adults were chosen randomly for constructi

Stability and accuracy assessment of identification of traditional Chinese materia medica using DNA barcoding: a case study on Flos Lonicerae Japonicae.

DNA barcoding is a novel molecular identification method that aids in identifying traditional Chinese materia medica using traditional identification techniques. However, further study is needed to assess the stability and accuracy of DNA barcoding. Flos Lonicerae Japonicae, a typical medicinal flower, is widely used in China, Korea, and other Southeast Asian countries. However, Flos Lonicerae Japonicae and its closely related species have been misused and traded at varying for a wide range of prices. Therefore, Flos Lonicerae Japonicae must be accurately identified. In this study, the ITS2 an

Forensic Species Identification: A Case Involving Trafficked Fish Maws and Shark Fins.

Illegal wildlife trade and other forms of wildlife crime pose significant threats to global biodiversity. Accurate species identification of wildlife products is crucial for effectively combating illegal wildlife trade. In recent years, forensic species identification using molecular methods has gained increasing attention. Here, we report a case involving 1777 trafficked fish maws and 18,170 trafficked shark fins. We employed DNA barcoding for forensic species identification and a large-sample inference strategy for population estimation. A total of 325 samples (197 fish maws and 128 shark fi

DNA Barcoding-Enabled Tracking of Lipid Nanoparticles: Drug-Loading-Dependent Biodistribution and Tumor Microenvironment Targeting.

Lipid nanoparticles (LNPs) are versatile drug delivery systems, yet the impact of drug loading (DL) on their biodistribution and cellular uptake remains poorly understood. Optimizing drug loading is crucial for enhancing therapeutic efficacy and safety, as higher loading allows for lower LNP doses, reducing overall nanomaterial burden. Addressing this knowledge gap is essential for advancing LNP-based cancer therapies. This study integrates DNA barcoding technology with LNPs to evaluate their in vivo delivery behaviors under varying drug loadings. Using a sequential nanoprecipitation method, D

Urban environmental drivers of eukaryotic microbiota and parasite prevalence in domestic pigeon faeces: a metabarcoding-based public health risk assessment in Seoul, South Korea.

Feral pigeons in Seoul, South Korea, pose a significant public health risk due to their potential to spread zoonotic pathogens. However, the diversity of their eukaryotic microbiota, particularly parasitic organisms, remains underexplored with urban environmental factors. We aimed to characterize the eukaryotic microbiota in pigeon faeces and assess how urban factors such as housing density, proximity to water sources and parks and the presence of homeless individuals influence parasite diversity. Faecal samples were collected from pigeons in various regions of Seoul and adjacent cities. Metab

An integrative approach to DNA barcoding, geometric morphometrics, and machine learning for field identification of Culex mosquitoes (Diptera: Culicidae), with implications for vector-borne disease surveillance.

Culex mosquitoes are of considerable medical and veterinary importance, acting as vectors of arboviruses such as Japanese encephalitis, Rift Valley fever, and West Nile virus, as well as the filarial parasite Wuchereria bancrofti. Accurate identification of Culex species, however, remains challenging due to their close morphological similarity, frequent damage to field-collected specimens, and the limited availability of trained taxonomists. To address these challenges, this study employed an integrative framework combining DNA barcoding, wing geometric morphometrics (GM), and Random Forest (R

Interspecific and sexual shape variation in the filariasis vectors Mansonia dives and Ma. bonneae.

In the South of Thailand, six Mansonia species are recorded as filariasis vectors, among which Ma. bonneae and Ma. dives. These two species are distributed in the same breeding place, mainly the swamp forest, but appear to be of problematic identification using traditional morphological characters. Because of the risk of wrong identification during epidemiological or biological studies, complementary techniques are needed to distinguish the two species. We used on the same field collected specimens both genetic (DNA barcoding) and phenetic (geometric morphometrics) techniques. Both methods con

Comparative Assessment of Environmental DNA and Bulk-Sample Metabarcoding in Biosecurity Surveillance for Detecting Biting Midges (Ceratopogonidae).

Biting midges, Culicoides spp. (Diptera: Ceratopogonidae), are significant vectors capable of transmitting arboviruses, such as bluetongue virus, to livestock. New Zealand is free of Culicoides, and a national surveillance programme is in place for the early detection of an incursion. Traditionally, insect trap samples from the surveillance programme are analyzed using morphology-based diagnostics under microscopes, which is time-consuming and relies on specialized taxonomic expertise. Here, we assessed the effectiveness of DNA metabarcoding using insect bulk samples and environmental DNA (eDN

DIVERSITY, HOST RANGE, AND SPECIFICITY OF ECHINOSTOMES (FAMILY ECHINOSTOMATIDAE; LOOSS, 1899) IN THE MIDWESTERN UNITED STATES.

Trematodes in the family Echinostomatidae (Looss, 1899) are globally distributed parasites of both humans and wildlife. In North America, echinostomes are nearly ubiquitous in freshwater systems, where they commonly parasitize snails as their first intermediate hosts, aquatic hosts such as snails or tadpoles as second intermediate hosts, and waterfowl and semi-aquatic mammals as their definitive hosts. Our study expands on sampling efforts in the midwestern United States and contributes novel ND1, ITS2, and 28S sequences from 12 lineages of echinostomes in the genera Echinostoma, Echinoparyphi

Morphological and molecular phylogeny of Clinostomum sp. (Digenea: Clinostomidae) metacercariae, using DNA barcode from a South American freshwater fish.

Here, we present a comprehensive morphological and molecular phylogenetic analysis of Clinostomum sp. (Digenea: Clinostomidae) metacercariae parasitizing two freshwater fish species from Southeast Brazil: Serrasalmus spilopleura (piranha) and Callichthys callichthys (tambuatá). The morphological examination revealed distinct characteristics of metacercariae in each host. Using the cytochrome c oxidase I (COI) gene barcode region, we obtained DNA sequences that allowed for accurate phylogenetic placement. Phylogenetic analyses revealed that Clinostomum sp. HM41 (metacercariae), isolated from S.

Prevalence, risk factors, and species diversity of strongylid nematodes in domesticated Thai horses: insights from ITS-2 rDNA metabarcoding.

Strongylid nematodes represent a major health and performance concern for equids globally. However, the epidemiology of strongylid infections in horse populations remains largely unexplored in Thailand. This study investigated the prevalence of strongylid parasites and the associated risk factors in domesticated horses in Thailand. Additionally, the study utilized ITS-2 rDNA metabarcoding to characterize the diversity and co-occurrence patterns of strongylid species. Of the 408 horses examined, 50.98% tested positive for strongyle infection, with an average intensity of 445.67 ± 639.58 eggs. N

Detection of Bartonella sp. in ticks and their small mammal hosts in mangrove forests of Peninsular Malaysia.

Bacteria of the genus Bartonella have been known as emerging zoonotic pathogens for several human diseases including cat scratch disease, Carrion's disease and trench fever. Numerous species of small mammals have been reported to play a role as a suitable reservoir to many pathogenic Bartonella. These infections are thought to be transmitted through blood-feeding arthropod vectors such as ticks, fleas and lice. The purpose of this study is to detect the presence of Bartonella species from tick samples collected from small mammals in mangrove forests of Peninsular Malaysia. Herein, 38 individua

Identification of Indian crocodile species through DNA barcodes.

The biodiversity of India includes three crocodile species, Crocodylus palustris, Crocodylus porosus, and Gavialis gangeticus, whose status is threatened due to bushmeat crisis and illegal hunting. The crocodilian conservation management requires novel techniques to help forensic analysts to reveal species identity. DNA barcoding is a species identification technique, where a partial cytochrome c oxidase subunit 1 gene is used as a marker for species identification. Herein, the DNA barcoding technique is evaluated for three Indian crocodiles by analyzing an approximately 750-bp barcode region.

Morphological and molecular evidence uncovers hidden species diversity in the leatherleaf slug genus Valiguna (Systellommatophora, Veronicellidae) from Thailand.

The poorly studied leatherleaf slug genus Valiguna in Thailand was carefully investigated. Members of this genus are phenotypically similar, making their identification very challenging. This study clarifies the taxonomic status of all Valiguna species in Thailand by combining morphological and anatomical studies with DNA barcoding. Monophyly of all Valiguna species was confirmed by analysis of the mitochondrial COI data and that all Valiguna species have the acropleurocaulis type of penis. Currently, three Valiguna species are recognised: V.siamensis, V.semicerina Mitchueachart & Panha, sp. n

Designation of a neotype and redescription of Hesione reticulata von Marenzeller, 1879 from Japan (Annelida, Hesionidae).

The hesionid polychaete Hesione reticulata von Marenzeller, 1879 was described from Enoshima Island, Japan and has been recorded also from the Red Sea. Depending on researchers, it has been regarded as either a distinct species or synonymous with older established ones. The type specimen has been lost. In order to clarify its taxonomic status, Hesione reticulata is herein redescribed, illustrated, and a neotype is proposed based on recent material collected near the type locality. The diagnostic features include the presence of several dorsal, discontinuous longitudinal bands, interrupted by p

Dissecting the factors shaping fish skin microbiomes in a heterogeneous inland water system.

BACKGROUND: Fish skin microbiomes are rarely studied in inland water systems, in spite of their importance for fish health and ecology. This is mainly because fish species distribution often covaries with other biotic and abiotic factors, complicating the study design. We tackled this issue in the northern part of the Jordan River system, in which a few fish species geographically overlap, across steep gradients of water temperature and salinity. RESULTS: Using 16S rRNA metabarcoding, we studied the water properties that shape the skin bacterial communities, and their interaction with fish ta

Arbuscular mycorrhizal fungi promote coexistence and niche divergence of sympatric palm species on a remote oceanic island.

Microbes can have profound effects on their hosts, driving natural selection, promoting speciation and determining species distributions. However, soil-dwelling microbes are rarely investigated as drivers of evolutionary change in plants. We used metabarcoding and experimental manipulation of soil microbiomes to investigate the impact of soil and root microbes in a well-known case of sympatric speciation, the Howea palms of Lord Howe Island (Australia). Whereas H. forsteriana can grow on both calcareous and volcanic soils, H. belmoreana is restricted to, but more successful on, volcanic soil,

Morphological and molecular characterization of Neotropic Lymnaeidae (Gastropoda: Lymnaeoidea), vectors of fasciolosis.

Lymnaeidae play a crucial role in the transmission of fasciolosis, a disease of medical and veterinary importance. In the Neotropic, a region where fasciolosis is emergent, eight Lymnaeidae species are currently considered valid. However, our knowledge of the diversity of this taxon is hindered by the fact that lymnaeids exhibit extremely homogeneous anatomical traits. Because most species are difficult to identify using classic taxonomy, it is difficult to establish an epidemiological risk map of fasciolosis in the Neotropic. In this paper, we contribute to our understanding of the diversity
