Radiosensitivity enhancement of human thyroid carcinoma cells by the inhibitors of histone deacetylase sodium butyrate and valproic acid.

Radiotherapy is one of the leading treatments for clinical cancer therapy. External beam radiotherapy has been proposed as an adjuvant treatment for patients bearing differentiated thyroid cancer refractory to conventional therapy. Our purpose was to study the combined effect of HDAC inhibitors (HDACi) and ionizing irradiation in thyroid cancer cell lines (Nthy-ori 3-1, WRO, TPC-1 and 8505c). HDACi radiosensitized thyroid cancer cells as evidenced by the reduction of survival fraction, whereas they had no effect in the normal cells. HDACi enhanced radiation-induced cell death in WRO cells. Gam

Evaluating Individual Radiosensitivity for the Prediction of Acute Toxicities of Chemoradiotherapy in Esophageal Cancer Patients.

In this work, individual radiosensitivity was evaluated using DNA damage response and chromosomal aberrations (CAs) in peripheral blood lymphocytes (PBLs) for the prediction of acute toxicities of chemoradiotherapy (CRT) in esophageal cancer patients. Eighteen patients with esophageal cancer were enrolled in this prospective study. Prescribed doses were 60 Gy in 11 patients and 50 Gy in seven patients. Patients received 2 Gy radiotherapy five days a week. PBLs were obtained during treatment just before and 15 min after 2 Gy radiation therapy on the days when the cumulative dose reached 2, 20,

MicroRNA-9 suppresses the sensitivity of CNE2 cells to ultraviolet radiation.

MicroRNA (miR)-9 has been demonstrated to regulate the radiosensitivity of tumor cells. In the present study, the mechanism by which miR-9 modulates the sensitivity of nasopharyngeal carcinoma (NPC) cells to ultraviolet (UV) radiation was investigated. The results demonstrated that exposure of NPC cells to UV light resulted in a significant increase in the expression of miR-9, and that CNE2 cells overexpressing miR-9 exhibited reduced levels of DNA damage and increased levels of total glutathione upon UV exposure. Accordingly, the inhibition of the expression of miR-9 promoted UV-induced DNA d

Variation of radiation-sensitivity of neural stem and progenitor cell populations within the developing mouse brain.

PURPOSE: We investigated the DNA damage response (DDR) of fetal neural stem and progenitor cells (NSPC), since exposure to ionizing radiation can severely impair the brain development. MATERIAL AND METHODS: We compared apoptosis induction in the dorsal telencephalon and the lateral ganglionic eminences (LGE) of mouse embryos after an in utero irradiation. We used two thymidine analogs, together with the physical position of nuclei within brain structures, to determine the fate of irradiated NSPC. RESULTS: NSPC did not activate an apparent protein 21(p21)- dependent G1/S checkpoint within the

Caffeine-induced radiosensitization is independent of nonhomologous end joining of DNA double-strand breaks.

After exposure to ionizing radiation, proliferating cells actively slow down progression through the cell cycle through the activation of checkpoints to provide time for repair. Two major complementary DNA double-strand break (DSB) repair pathways exist in mammalian cells, homologous recombination repair (HRR) and nonhomologous end joining (NHEJ). The relationship between checkpoint activation and these two types of DNA DSB repair pathways is not clear. Caffeine, as a nonspecific inhibitor of ATM and ATR, abolishes multi-checkpoint responses and sensitizes cells to radiation-induced killing. H

Phase 1a Evaluation of LP-184 in Recurrent Glioblastoma: Safety, Pharmacokinetics, and Translational Optimization of CNS Exposure.

PURPOSE: Limited central nervous system (CNS) bioavailability and pharmacodynamics are obstacles to effective systemic therapies for glioblastoma (GBM). One strategy to overcome these challenges is drug combinations enhancing CNS penetration and/or tumor chemosensitivity. LP-184 (zirdafulven), a synthetic acylfulvene class alkylator, induces DNA damage and inhibits glioblastoma cell viability in pre-clinical models. LP-184 is a prodrug converted to active metabolites by intracellular oxidoreductase prostaglandin reductase 1 (PTGR1) that is over-expressed in >70% of glioblastoma. DNA damage ind

Phase 1 clinical trial of the ataxia telangiectasia and Rad3-related inhibitor berzosertib with irinotecan in patients with advanced solid tumors (ETCTN 9938).

BACKGROUND: Ataxia telangiectasia and Rad3-related (ATR) inhibition with berzosertib potentiates the efficacy of irinotecan in preclinical models. The authors hypothesize that this combination is well tolerated, modulates the DNA damage response to irinotecan, and is associated with clinical activity in advanced solid tumors. METHODS: In this phase 1 study (NCT02595931), berzosertib 60-270 mg/m2 was administered with irinotecan 180 mg/m2 every 2 weeks in a 4-week cycle. The primary end point was determination of the maximum tolerated dose and recommended phase 2 dose (RP2D). Antitumor activit

Long-term exposure to the ethanol-derived metabolite acetaldehyde elevates structural genomic alterations but not base substitutions.

Acetaldehyde is the primary metabolite of ethanol, and routes of exposure include endogenous sources, food and cigarette smoke. To explore whether the mutagenic effect of acetaldehyde is responsible for the carcinogenicity of ethanol, we use whole genome sequencing on four human cell lines subjected to long-term, physiologically relevant, analytically validated acetaldehyde treatments. Unexpectedly, the treatments do not induce increased base substitution and short insertion/deletion mutagenesis, nor the appearance of the alcohol-associated cancer mutation signature SBS16. In contrast, we obse

Combination of PARPi and topoisomerase I inhibitors is highly effective in olaparib-resistant ovarian cancer.

High-grade serous ovarian carcinoma (HGSOC) often shows an initial response to poly (ADP-ribose) polymerase inhibitors (PARPi) such as olaparib, particularly in tumors with defects in homologous recombination (HR), but resistance to these drugs is a major clinical challenge. To address this, we screened over 1400 FDA-approved compounds to find agents that could restore olaparib sensitivity in resistant ovarian cancer cells. DNA topoisomerase I inhibitors (TOP1i) emerged as the most effective partner. The combination of TOP1 inhibitors with PARPi showed strong synergistic anticancer effects acr

Identification of a damaged-DNA binding domain of the XPA protein.

The XPA (xeroderma pigmentosum group A) protein is a zinc metalloprotein consisting of 273 amino acids which binds preferentially to UV- or chemical carcinogen-damaged DNA, suggesting that it is involved in the recognition of several types of DNA damage during nucleotide excision repair processes. Here we identify a DNA binding domain of the XPA protein. The region of the XPA protein responsible for preferential binding to DNA damaged by UV or cis-diammine-dichloroplatinum(II) (cisplatin) is contained within a truncated derivative of the XPA protein, MF122, consisting of 122 amino acids and co

Dehydration-based preservation of cat cumulus-oocyte complexes is not improved by membrane-permeable trehalose.

INTRODUCTION: Dry-preservation of gametes offers a promising alternative to cryobanking by potentially storing cells at ambient temperatures; however, successful preservation of whole oocytes remains limited by dehydration-induced cellular damages. This study evaluated whether intracellular delivery of trehalose via a membrane-permeable derivative, trehalose hexaacetate (6-O-Ac-Tre), could enhance dehydration tolerance of domestic cat cumulus-oocyte complexes (COCs), a relevant model for mammalian fertility preservation. METHODS: Cat COCs were incubated with 0, 3, 10, or 30 mM of 6-O-Ac-Tre t

Arginine Methyltransferase PRMT8 Provides Cellular Stress Tolerance in Aging Motoneurons.

Aging contributes to cellular stress and neurodegeneration. Our understanding is limited regarding the tissue-restricted mechanisms providing protection in postmitotic cells throughout life. Here, we show that spinal cord motoneurons exhibit a high abundance of asymmetric dimethyl arginines (ADMAs) and the presence of this posttranslational modification provides protection against environmental stress. We identify protein arginine methyltransferase 8 (PRMT8) as a tissue-restricted enzyme responsible for proper ADMA level in postmitotic neurons. Male PRMT8 knock-out mice display decreased muscl

Inhibition of Translesion DNA Synthesis Sensitizes BRCA-Deficient Ovarian Cancer to PARP Inhibitors.

Poly(ADP-ribose) polymerase inhibitors (PARPi) exploit synthetic lethality to treat BRCA1/2-mutated ovarian cancer, yet clinical responses are often incomplete and resistance frequently emerges. PARPi exert cytotoxic effects by inhibiting single-strand break repair and trapping PARP on DNA, which impedes replication fork progression and generates lethal double-strand breaks. However, tumor cells can mitigate these effects through activation of alternative DNA repair mechanisms. Here, we investigated the role of translesion synthesis (TLS), a DNA damage tolerance pathway in which specialized po

Exogenous N-Acetyl-L-cysteine Improves Soybean Saline-Alkali Tolerance by Enhancing the hGSH Pathway to Alleviate Oxidative Damage.

Soil salinization and alkalization severely threaten global agricultural sustainability. Although N-acetylcysteine (NAC) is well known as an antioxidant in animal research, its effects on plant salt-alkali tolerance and the underlying mechanisms remain unclear. This study investigated the regulatory effects of foliar-sprayed NAC on two soybean (Glycine max) cultivars-salt-tolerant HF50 and salt-sensitive HN95-under saline-alkali stress. Exogenous NAC alleviated seedling growth inhibition by supplying cysteine, which elevated glutamate-cysteine ligase (GCL) activity and promoted homoglutathione

Recombinant FSH Improves Sperm DNA Damage in Male Infertility: A Phase II Clinical Trial.

UNLABELLED: Background and objectives: Male infertility is a global health dilemma and Follicle-Stimulating Hormone (FSH) administration has shown promising results. Several studies showed that infertile men with normal semen parameters have low levels of DNA damage while infertile men with abnormal semen parameters have more damage at the DNA level. Sperm DNA damage may affect the reproductive outcome and has been associated with failure in the achievement of competent embryos and pregnancy fulfillment. The aim of this study was to evaluate whether the administration of recombinant FSH (Gonal

DNA repair responses in human skin cells.

Sunlight and some environmental chemical agents produce lesions in the DNA of human skin cells that if unrepaired may interfere with normal functioning of these cells. The most serious outcome of such interactions may be malignancy. It is therefore important to develop an understanding of mechanisms by which the lesions may be repaired or tolerated without deleterious consequences. Our models for the molecular processing of damaged DNA have been derived largely from the study of bacterial systems. Some similarities but significant differences are revealed when human cell responses are tested a

HslO ameliorates arrested ΔrecA polA cell growth and reduces DNA damage and oxidative stress responses.

Chromosome damage combined with defective recombinase activity has been widely considered to render cells inviable, owing to deficient double-strand break repair. However, temperature-sensitive recAts polA cells grow well upon induction of DNA damage and supplementation with catalase at restrictive temperatures. These treatments reduce intracellular reactive oxygen species (ROS) levels, which suggests that recAts polA cells are susceptible to ROS, but not chronic chromosome damage. Therefore, we investigated whether polA cells can tolerate a complete lack of recombinase function. We introduced

Rho GTPases in cancer resistance: mechanisms, vulnerabilities, and therapeutic opportunities.

Intrinsic and adaptive resistance to therapy remain major barriers to effective cancer treatment. Diverse resistance mechanisms, including epithelial-mesenchymal transition, enhanced tolerance to DNA damage, impaired cell death pathways, metabolic reprogramming, and cues from the tumour microenvironment, are increasingly recognised as being tightly integrated with Rho GTPase signalling networks. Accumulating evidence positions these pathways as central regulators of therapeutic resistance across multiple cancer types. In this review, we synthesise recent experimental findings linking Rho GTPas

Pathological modulation of genome maintenance by cancer/testes antigens (CTAs).

The Cancer Testis Antigens (CTAs) are a group of germ cell proteins that are absent from normal somatic cells yet aberrantly expressed in many cancer cells. When mis-expressed in cancer cells, many CTAs promote tumorigenic characteristics including genome instability, DNA damage tolerance and therapy resistance. Here we highlight some of the CTAs for which their roles in genome maintenance in cancer cells are well established. We consider three broad CTA categories: (1) Melanoma Antigens (MAGEs) (2) Mitotic CTAs and (3) CTAs with roles in meiotic homologous recombination. Many cancer cells rel

Exogenous arginine induced metabolic modulation and biomass enhancement in Nannochloropsis oceanica under high CO2 stress.

Given that excessive CO2 induces reactive oxygen species (ROS) which inhibit photosynthesis and microalgal survival, this study explores how exogenous arginine enhances stress resilience and biomass yield of Nannochloropsis oceanica under high CO2 stress. Arginine promoted NO synthesis, enhancing antioxidant enzyme activities to combat ROS damage. Transcriptomic analysis revealed that key enzymes in the Calvin cycle (PRK, GAPDH, FBPase and SBPase) and TCA cycle (CS, IDH, SCS and MDH) were significantly up-regulated, supporting carbon fixation and energy metabolism. Arginine also alleviated cel
