Sex specific modulation of LC neuronal activity in a binge drinking mouse model.

Binge drinking substantially elevates the risk of developing alcohol use disorder, with pronounced sex-specific differences observed. However, there is a limited understanding of the underlying sex differences in the neural mechanisms that underpin binge drinking. The present study utilized the Drinking in the Dark (DID) paradigm, in combination with electrophysiology and chemogenetics, to reveal the critical role of the locus coeruleus (LC) in regulating sexually dimorphic binge-like drinking behavior. Electrophysiological analysis revealed that after three weeks of DID modeling, female mice

Pituitary adenylate cyclase-activating polypeptide (PACAP)overexpression in the paraventricular nucleus of the thalamus alters motivated and affective behavior in female rats.

BACKGROUND: Pituitary adenylate cyclase-activating polypeptide (PACAP) has been found to be involved in a wide range of motivated and affective behaviors. While the PACAP-38 isoform is more densely expressed than PACAP-27 in most of the brain, PACAP-27 is more highly expressed in the rodent paraventricular nucleus of the thalamus (PVT), where females also have greater expression than males. Notably, the role of PACAP-27 expression in cells of the PVT has not been explored. METHODS: Adult, female Long-Evans rats were injected in the PVT with an adeno-associated virus (AAV) to increase expressi

Development and validation of a behavioral observation measure for the syndrome of psychosis, intermittent hyponatremia, and polydipsia.

A behavioral observation scale (Virginia Polydipsia Scale; VPS) for monitoring drinking patterns was developed and its reliability tested during 25 hours of tandem ratings among six patients with the syndrome of psychosis, intermittent hyponatremia, and polydipsia (PIPS). These ratings were compared to those collected from a control group of six psychiatric inpatients who were similarly observed for 25 hours. The scale was subsequently used to assess day-long drinking in a single PIPS patient. Results demonstrated that the VPS can be reliably administered by trained raters and that it clearly

Integration of impulsivity and positive mood to predict risky behavior: development and validation of a measure of positive urgency.

In 3 studies, the authors developed and began to validate a measure of the propensity to act rashly in response to positive affective states (positive urgency). In Study 1, they developed a content-valid 14-item scale, showed that the measure was unidimensional, and showed that positive urgency was distinct from impulsivity-like constructs identified in 2 models of impulsive behavior. In Study 2, they showed that positive urgency explained variance in risky behavior not explained by measures of other impulsivity-like constructs, differentially explained positive mood-based risky behavior, diff

Identifying Sensitive Periods for Alcohol Use: The Roles of Timing and Chronicity of Child Physical Abuse.

BACKGROUND: Childhood maltreatment, particularly physical abuse (PA), increases the risk of alcohol use during young adulthood. Although prior research underscores the importance of examining the roles of PA-event characteristics such as timing of and chronicity of PA in initiating and maintaining alcohol use, few studies have explored the risk of developing alcohol use based on the timing and chronicity of PA. METHODS: Using a community sample of 300 young adults (ages 18 to 25), this study examined how variations in timing and chronicity of PA relate to 4 distinct drinking behaviors includi

Assessing sexual motives for drinking alcohol among HIV-positive men who have sex with men.

Individuals who drink alcohol for the explicit motive of facilitating or enhancing sex may be more likely to engage in risky sexual behavior, including having sex under the influence of alcohol. However, efforts to assess sexual motives for drinking (SMDs) have been very limited to date. We examined the psychometric properties of a 5-item measure of SMDs in a sample of HIV-positive heavy drinking men who have sex with men. Findings provided excellent support for the scale's internal consistency and concurrent validity with a well-established measure of sexual alcohol expectancies (SAEs). Good

Excessive alcohol consumption and drinking expectations among clients in methadone maintenance.

Excessive alcohol consumption and related problems are common among clients in methadone maintenance treatment (MMT), yet relatively little is known about the psychological and social determinants of alcohol-related attitudes and behaviors during treatment. This study reports on the prevalence of alcohol dependence, patterns of alcohol consumption and preliminary findings about clients' beliefs that they will change their drinking behavior in the future. Data were gathered from personal interviews with 66 clients attending a MMT program in South London (some 80.5% of the eligible caseload). Fo

Event-Level Examinations of Impaired Control, Protective Behavioral Strategies, and Alcohol-Related Consequences.

OBJECTIVE: Impaired control over alcohol use is a well-established, prospective risk factor for alcohol use disorder. However, research is mixed regarding the risk conveyed by event-level impaired control behaviors. While most of this research evaluates unplanned drinking episodes, a continuous estimate, such as the number of unintended drinks consumed, appears to be an important but overlooked consideration. Moreover, there is limited understanding of protective factors that may buffer the effects of impaired control at the daily level. Thus, the current study tested the association between u

Associations of alcohol drinking with incident dementia: a prospective study from the UK Biobank.

Relationship between alcohol drinking and incident dementia remained uncertain. This study used UK Biobank cohort data to investigate the association between alcohol drinking and dementia risk, and potential effect modifications by cardiovascular disease (CVD) risk, APOE4 gene, and sex. We excluded infrequent drinkers and participants with baseline dementia or dementia within two years of follow-up. Drinking status was defined as non-drinking, low-moderate and heavy drinking (by weekly alcohol units). Drinking behaviors included drinking with meals and drinking type. Primary outcome was all-ca

Wild Chimpanzees (Pan troglodytes schweinfurthii) Use Tools to Access Out of Reach Water.

The use of tools to drink water is well-documented in wild chimpanzees, but the specific function of this behavior is unclear. Here we use a large data set of drinking behaviors spanning 14 years of observation from the Kanyawara community of chimpanzees living in Kibale National Park, Uganda, to test two possible functions of leaf-sponges and other drinking tools. On the one hand, chimpanzees may use tools to access water that is hard to reach, which predicts that chimpanzees will preferentially use tools to drink at tree holes and crevices compared to all other locations. Conversely, chimpan

An evaluation of relations among antisocial behavior, psychopathic traits, and alcohol problems in incarcerated men.

BACKGROUND: The association between antisocial behavior and alcohol problems is well established, but the role that psychopathic traits and other factors such as drinking patterns and motives and family history play in this relationship has not been subjected to much empirical scrutiny. METHODS: A large sample (n=329) of male Federal Correctional Institution inmates participated in structured interviews designed to permit diagnosis of antisocial personality disorder and to assess facets of psychopathic personality as defined by the Hare Psychopathy Checklist-Revised. Behavioral effects of bombesin administration in rats.

Bombesin (BBS, 0.1-4.0 micrograms) administered to the lateral cerebral ventricle (IVT) of rats decreased food intake and feeding behavior. Grooming behavior increased and resting behavior decreased as doses greater than or equal to 0.01 microgram. IVT BBS (4.0 micrograms) caused greater suppression of food-deprivation-induced food intake and feeding behavior than the same dose and volume administered intraperitoneally (IP). After IVT BBS, rats displayed more grooming and less resting than normal, but after IP BBS, rats displayed a normal frequency of grooming and more resting. IVT BBS (greate

Lateral hypothalamus CRFR1 regulation of chronic binge drinking: divergence along anterior-posterior axis.

Binge alcohol drinking increases the risk of developing an alcohol use disorder (AUD) and comorbid psychopathology. The lateral hypothalamus (LH) is a brain structure that integrates cognitive and sensory information to tightly regulate motivated behavior, including binge drinking. Importantly, LH function is vulnerable to modulation by the pro-stress neuropeptide corticotropin-releasing factor (CRF), and acute antagonism of CRF receptor 1 (CRFR1) in the LH blunts binge drinking. However, the role of LH CRFR1 in chronic binge drinking is unknown. We used genetically targeted knockdown (KD) of

Is social drinking during pregnancy harmless? There is reason to think not.

This paper reviews the scientific evidence bearing on the safety of social drinking in pregnancy (that is, patterns or levels of alcohol intake that cannot be diagnosed as alcohol abuse or alcohol dependence). Human studies have shown an increased incidence of spontaneous abortion, greater risk for a growth-retarded infant, measurable changes in newborn behavior, differences in development at age eight months, and differences in behavior at age four years, related to patterns of social drinking during pregnancy. Animal studies and in vitro studies of human umbilical vessels add to doubts about

Anticipating problem alcohol use developmentally from childhood into middle adulthood: what have we learned?

This commentary reviews and comments on six major longitudinal studies from the United States, Great Britain and Finland, that test predictive models of drinking and problem drinking behavior across a developmental span of one to two generations. The large Ns, in two instances involving population samples, and the broad and study-overlapping variable domains make this collection of studies unique and of special interest vis-à-vis the issue of cross-study replicability of findings. Significant cross-study commonalities are noted, involving the strong cross-study replicability of an undercontrol

Identifying the Intersection of Alcohol, Adherence and Sex in HIV Positive Men on ART Treatment in India Using an Adapted Timeline Followback Procedure.

People living with HIV (PLHIV) on anti-retroviral treatment (ART) who drink are less adherent and more likely to engage in unprotected sex but the connections among these events are correlational. Using an adapted Timeline Follow-Back (A-TLFB) procedure, this paper examines the day by day interface of alcohol, medication adherence and sex to provide a fine grained understanding of how multiple behavioral risks coincide in time and space, explores concordance/discordance of measures with survey data and identifies potential recall bias. Data are drawn from a survey of behavior, knowledge and at

A Novel Mouse Home Cage Lickometer System Reveals Sex- and Housing-Based Influences on Alcohol Drinking.

Alcohol use disorder (AUD) is a significant global health issue. Despite historically higher rates among men, AUD prevalence and negative alcohol-related outcomes in women are rising. Loneliness in humans has been associated with increased alcohol use, and traditional rodent drinking models involve single housing, presenting challenges for studying social enrichment. We developed LIQ PARTI (Lick Instance Quantifier with Poly-Animal RFID Tracking Integration), an open-source tool to examine home cage continuous access two-bottle choice drinking behavior in a group-housed setting, investigating

Youth Problem Drinking: The Role of Parental and Familial Relationships.

BACKGROUND: Alcohol use continues to be an important global public health problem and adolescence seems to be a decisive period of time in the development of drinking patterns into adulthood. While most studies concentrate on frequency and amount of alcohol, fewer studies address "problem drinking." Gathering information on youth's alcohol-related behavioral consequences is especially important. OBJECTIVES: Current research focuses on gathering information on the background of problem drinking behavior with special attention to parental/familial relationships. METHODS: The survey was conduct

Drinking motives for self and others predict alcohol use and consequences among college women: the moderating effects of PTSD.

Although drinking motives have been shown to influence drinking behavior among women with trauma histories and PTSD, no known research has examined the influence of drinking motives on alcohol use and alcohol-related consequences for women with PTSD as compared to women with a trauma history but no PTSD and women with no trauma history. Therefore, the present study sought to examine the associations between drinking motives women held for themselves as well as their perception of the drinking motives of others and their own alcohol use and consequences, and whether this was moderated by a hist

Mild or borderline intellectual disability as a risk for alcohol consumption in adolescents - A matched-pair study.

BACKGROUND: Studies that investigate the association between mild or borderline intellectual disability (MBID) and alcohol use in adolescents have not examined whether MBID is an independent risk factor for drinking. AIM: It is important to examine whether MBID is a risk factor for alcohol consumption by controlling concomitant factors in a matched-pair design. METHOD: Overall, 329 students from two schools for children with MBID self-reported their drinking behavior via questionnaires, and 329 students from regular schools were matched to this group by gender, age, family composition, and p
