Purification, characterization and three-dimensional structure prediction of multicopper oxidase Laccases from Trichoderma lixii FLU1 and Talaromyces pinophilus FLU12.

Broad-spectrum biocatalysts enzymes, Laccases, have been implicated in the complete degradation of harmful pollutants into less-toxic compounds. In this study, two extracellularly produced Laccases were purified to homogeneity from two different Ascomycetes spp. Trichoderma lixii FLU1 (TlFLU1) and Talaromyces pinophilus FLU12 (TpFLU12). The purified enzymes are monomeric units, with a molecular mass of 44 kDa and 68.7 kDa for TlFLU1 and TpFLU12, respectively, on SDS-PAGE and zymogram. It reveals distinct properties beyond classic protein absorption at 270-280 nm, with TlFLU1's peak at 270 nm a

Enhanced lactic acid production from P2O5-pretreated biomass by domesticated Pediococcus pentosaceus without detoxification.

Expensive cellulase and complex detoxification procedures increase the cost of biomass lactic acid fermentation. Therefore, it is of great significance to develop a robust method to ferment lactic acid using biomass by avoiding cellulase and detoxification. This study demonstrates the advantage of combining mechanocatalytic P2O5 pre-treatment and strain domestication. We show that an enzyme-free mechanocatalytic saccharification process by combining mix-milling of P2O5 with biomass and successive hydrolysis produces a fermentable hydrolysate with much less inhibitory compounds than the hydroly

Zingerone Alleviates Morphine Tolerance and Dependence in Mice by Reducing Oxidative Stress-Mediated NLRP3 Inflammasome Activation.

Morphine (MPH) is widely used for pain management; however, long-term MPH therapy results in antinociceptive tolerance and physical dependence, limiting its clinical use. Zingerone (ZIN) is a natural phenolic compound with neuroprotective effects. We investigated the effects of single and repeated doses of ZIN on MPH-induced tolerance, dependence, and underlying biochemical mechanisms. After a dose-response experiment, tolerance was developed to MPH (10 mg/kg, i.p.) for seven days. In the single-dose study, ZIN was administered on day seven. In the repeated-dose study, ZIN was administered for

Gigantol Ameliorates Metabolic Dysfunction-Associated Steatohepatitis by Promoting TFEB-Mediated Lipophagy and Fatty Acid Oxidation.

Gigantol, a naturally active molecule derived from the edible plant Dendrobium, has been demonstrated to have hepatoprotective potential, but its effect in the treatment of metabolic dysfunction-associated steatohepatitis (MASH) remains unclear. Here, we found that gigantol treatment greatly corrected dyslipidemia, hepatic dysfunction, and hepatic histological changes that arrested MASH occurrence in high-fat and high-fructose-diet (HFFD)-fed mice. Mechanistically, gigantol promoted the nuclear translocation of the transcription factor EB (TFEB) to regulate the key factors of lipophagy and fat

Zingerone modulates the circulating steroids hormone levels and testicular steroidogenic markers expression in mice: An in vivo and in silico study.

Zingerone (4-(4-hydroxy-3-methoxyphenyl)-2-butanone) is a phytochemical with potent anti-inflammatory and antioxidant properties. It has been shown that zingerone protects testis under pathological conditions by its antioxidant and anti-inflammatory effects. However, there is no direct evidence that zingerone specifically affects the testicular steroidogenesis in normal conditions. Therefore, in this study, we have investigated the effects of zingerone on testicular steroidogenesis by using in vivo and silico approaches. The mice were divided into four groups: Control, 10, 25, and 50 mg/kg dos

Zingerone ameliorates sodium arsenite-induced cardiotoxicity in rats by suppressing oxidative stress and inflammation via Nrf2 /GCLM\GCLC signaling pathways.

Arsenic toxicity is a serious threat to human health, transmitted through many factors in the environment, especially water and contaminated food. Epidemiologic studies have reported that arsenite increases mortality and morbidity by causing cardiac damage, but the mechanism of action on cardiotoxicity remains to be elucidated. Zingerone (ZNG) obtained from ginger root is a monomer with pharmacological effects such as antioxidant, anti-inflammatory, and anticancer. This study was conducted to investigate the protective potential of zingerone against sodium arsenite-induced cardiac damage in ra

Microbial-derived metabolites coordinate growth regulation and stress tolerance in Phalaenopsis orchids.

Microbial metabolites regulate plant growth and stress responses, yet their roles in crassulacean acid metabolism (CAM) plants remain insufficiently characterized. Here, we investigated the effects of metabolites from Bacillus licheniformis strain BlCP6, identified by 16S rDNA sequencing and whole-genome analysis, on growth and stress responses in Phalaenopsis orchids. Genome sequencing and metabolite profiling revealed that siderophores represent major bioactive compounds with direct plant growth-promoting potential, whereas osmoprotectants and extracellular lytic enzymes may contribute indir

Metal oxides-based catalytic pyrolysis of lignocellulosic biomasses for bio-oil development: a comparative assessment on process optimization and characterization.

This study investigated the conventional fixed-bed catalytic pyrolysis using metal oxides (CaO, Fe₂O₃, and TiO₂) to enhance bio-oil yield from three lignocellulosic biomasses: safflower press cake (SPC), coconut press cake (CPC), and coconut shell (CS). The primary characterization, using ultimate, proximate, and compositional analyses, as well as TGA-DTG-DSC, was carried out in the experiment. SPC biomass had a high hemicellulose content (50.8 ± 0.11%), which is suited for bio-oil applications. An initial non-catalytic pyrolysis experiment was conducted at 550 °C with a heating rate of 25 °C/

Physiological responses of plants and mites to salicylic acid improve the efficacy of spirodiclofen for controlling Tetranychus urticae (Acari: Tetranychidae) on greenhouse tomatoes.

Salicylic acid (SA) is a signaling molecule that can induce plant resistance to certain herbivores. Although the role of jasmonic acid in mediating mite-tomato plant interactions has been well studied, the role of salicylic acid has not. This study examined how the application of exogenous SA, via its effects on tomato plant physiology, alters the activity of mite digestive enzymes, mite energy reserves, and mite susceptibility to spirodiclofen. Enzymatic activity-including superoxide dismutase, ascorbate peroxidase, guaiacol peroxidase, polyphenol oxidase, and phenylalanine ammonia-lyase-alon

Signaling Molecule Hydrogen Sulfide Improves Seed Germination and Seedling Growth of Maize (Zea mays L.) Under High Temperature by Inducing Antioxidant System and Osmolyte Biosynthesis.

Hydrogen sulfide (H2S) is a novel type signaling molecule in plants. Seed germination is a key stage of life cycle of plants, which is vulnerable to environmental stress including high temperature. However, under high temperature stress, whether pre-soaking of maize seeds with NaHS (a H2S donor) could improve seed germination and seedling growth and the possible mechanisms are not completely clear. In this study, maize seeds pre-soaked with NaHS enhanced germination percentage, sprout length, root length, and fresh weight compared with the control without NaHS treatment, illustrating that H2S

FZD7 regulates BMSCs-mediated protection of CML cells.

BACKGROUND: Phthalates are man-made chemical compounds with numerous applications especially known for their use as plasticizers. They have weak bonding to the polymeric matrix or products in which they are used. Owing to this reason, they are readily released into the environment which makes them ubiquitous. The agricultural soils are also reported to be polluted with phthalates up to a considerable extent which causes adverse effects on flora and fauna. A few studies have been conducted on phthalate-induced phytotoxicity, which has revealed that phthalates affect the quality and yield of edi
