[Histone deacetylase inhibitors as therapeutic agents for polyglutamine disorders].

During the past few years, gene expression studies have shown that the perturbation of transcription frequently results in neuronal dysfunction in polyglutamine (PolyQ) diseases such as Huntington's disease (HD). Histone deacetylases (HDACs) act as repressors of transcription through interaction with co-repressor complexes, leading to chromatin remodelling. Aberrant interaction between PolyQ proteins and regulators of transcription could be one mechanism by which transcriptional dysregulation occurs. Here, the authors discuss the possible mechanism of transcriptional dysfunction in PolyQ disea

Histone deacetylase: a potential therapeutic target for fibrotic disorders.

Histone deacetylases (HDACs) are enzymes that balance the acetylation activities of histone acetyltransferases on chromatin remodeling and play essential roles in regulating gene transcription. In the past several years, the role of HDACs in cancer initiation and progression, as well as the therapeutic effects of HDAC inhibitors in various types of cancer, has been well studied. Recent studies indicated that HDAC activity is also associated with the development and progression of some chronic diseases characterized by fibrosis, including chronic kidney disease, cardiac hypertrophy, and idiopat

Histone deacetylase inhibitors and neurodegenerative disorders: holding the promise.

Neurodegenerative disorders (NDs) such as Huntington's disease, Alzheimer's disease, Parkinson disease, amyotrophic lateral sclerosis, spinal muscular atrophy, Friedreich's ataxia, and others are multi-factorial illnesses, in which many pathways (still poorly understood) act serially and in parallel to give a determined pathologic phenotype. Thus, presently there are no effective cures for these diseases. Some phenotypic as well as mechanistic features, common to the most of NDs, can be linked to epigenetic defects, that can lead to alteration of acetylation homeostasis and impairment of the h

Histone deacetylase inhibitors: possible implications for neurodegenerative disorders.

During the past six years numerous studies identified histone deacetylase (HDAC) inhibitors as candidate drugs for the treatment of neurodegenerative disorders. Two major neuroprotective mechanisms of HDAC inhibitors have been identified, namely the transcriptional activation of disease-modifying genes and the correction of perturbations in histone acetylation homeostasis, which have been shown to be intimately involved in the neurodegenerative pathomechanisms of Huntington's, Parkinson's and Kennedy disease, amyotropic lateral sclerosis, Rubinstein-Taybi syndrome as well as stroke. Histone modifications in Rett syndrome lymphocytes: a preliminary evaluation.

Most cases of Rett syndrome (RTT) are associated with mutations in the coding region of the transcriptional regulator MeCP2. This gene appears to repress gene expression through chromatin conformational changes secondary to histone modifications, mainly histone deacetylation of core histones H3 and H4. There is limited and contradictory information about histone modifications in RTT tissues. The present study intended to provide a preliminary characterization of histone acetylation (AcH3, AcH4) and methylation (MeH3) in RTT, with emphasis on non-selected peripheral cells and molecular-neurolog

Structure of the GCN5 histone acetyltransferase bound to a bisubstrate inhibitor.

Histone acetyltransferases (HATs) use acetyl CoA to acetylate target lysine residues within histones and other transcription factors, such as the p53 tumor suppressor, to promote gene activation. HAT enzymes fall into subfamilies with divergence in sequence and substrate preference. Several HAT proteins have been implicated in human cancer. We have previously reported on the preparation of peptide-CoA conjugate inhibitors with distinct specificities for the p300/CBP [cAMP response element binding protein (CREB)-binding protein] or GCN5 HAT subfamilies. Here we report on the crystal structure o

TGIF2 interacts with histone deacetylase 1 and represses transcription.

TG-interacting factor (TGIF) is a transcriptional repressor, which represses transcription by binding directly to DNA or interacts with transforming growth factor beta (TGF beta)-activated Smads, thereby repressing TGF beta-responsive gene expression. Mutation of TGIF in humans causes holoprosencephaly, a severe genetic disorder affecting craniofacial development. Searching human expressed sequence tag data bases revealed the presence of clones encoding a TGIF-related protein (TGIF2), which contains two regions of high sequence identity with TGIF. Here we show that, like TGIF, TGIF2 recruits h

Histone H1(o) expression in the developing cat retina.

Histone H1(o) is a subtype of the non-core H1 histones located in the linker region of DNA between nucleosome cores and postulated to be involved in the regulation of gene expression. Studies in both the mouse retina and rat brain have correlated the terminal differentiation of cell types in these tissues to the expression of H1(o)a The expression of H1(o) in mouse retina occurs after light exposure suggesting that light may trigger the expression of H1(o). The aims of the present research were to: (1) describe the relationship of the appearance of H1(o) protein immunoreactivity to the formati

Binding of the von Willebrand factor A1 domain to histone.

Activation of the von Willebrand Factor (vWF) A1 domain is a critical factor in regulating the interaction of vWF with its platelet membrane receptor, the glycoprotein (GP) Ib-IX-V complex. This activation controls vWF-dependent platelet adhesion at high shear. The vWF-GP Ib-IX-V interaction is induced in vivo by exposure of platelet-rich plasma to high shear force, or by association of vWF with one or more unidentified components of the subendothelial matrix. In vitro, soluble vWF is activated to bind to platelets by nonphysiological modulators, such as the bacterial glycopeptide, ristocetin,

The phosphorylation region of lysine-rich histone in dividing cells.

N-Bromosuccinimide cleavage of in vivo 32P-labelled lysine-rich histone isolated from rapidly dividing cells has been studied. N-Bromosuccinimide cleaves F1-histone into two fragments, a small N-terminal piece and a larger C-terminal portion. The phosphate-induced microheterogeneity and associated radioactivity which has been linked to cell replication, is found in the carboxyterminal fragment, No phosphorous is found associated with the amino-terminal fragment when histone phosphorylation is associated with cell division. The specific tryptic phosphopeptides obtained from in vivo labelled F1

Functional Characterization of Breast Cancer-Associated Histone Mutations.

Cancer-related somatic histone mutants are present in 4% of diverse tumor types and display a similar genomic prevalence to other somatic mutations in cancer, but they are not well characterized. We curated and characterized 17 of the most frequently recurring missense mutations in histone H2A, H2B, and H3 genes in breast cancers occurring at glutamate (E), aspartate (D), serine (S), and arginine (R) residues. Many of these mutants co-occur with PIK3CA mutations in breast cancers. We expressed these breast cancer-associated histone mutants in MCF-7 and MDA-MB-468 breast cancer cells, or MCF-10

KDM6 Histone Demethylases Tune Pancreatic Cancer Plasticity.

Aberrant epigenetic regulation is a central driver of tumorigenesis, and the reversibility of epigenetic modifications underscores their potential as therapeutic targets. Epigenetic factors often function in a highly context-dependent manner, necessitating rigorous study of their dynamic and complex roles. Here, we uncovered an intricate regulation of tumor development and plasticity by the KDM6 family of H3K27me3 histone demethylases in pancreatic ductal adenocarcinoma (PDAC), a disease with sustained high mortality and limited treatment options. Discovery of first histone deacetylase 10 fluorescent probe.

Histone deacetylase 10 (HDAC10) is an understudied member of the HDAC enzyme family, with its biological functions just beginning to be elucidated. Recent studies showed that HDAC10 is involved in key cellular processes and may contribute to disease pathology. However, no selective fluorescent probes for HDAC10 have been reported. This study presents the first series of HADC10-targeted fluorescent probes based on naphthalimide scaffold, in which probe 4 shows the most promising characteristics. It displays moderate inhibitory potency against HDAC10 (IC50 = 60.2 nM) and features a distinct pH-r

Histone Modifications in Cardiovascular Disease: Mechanisms and Therapeutic Opportunities.

Cardiovascular diseases remain the leading cause of death worldwide. Maladaptive transcriptional programs drive the fibrosis, hypertrophy, and vascular inflammation that characterize these pathologies. Histone posttranslational modifications regulate these programs by remodeling chromatin accessibility and transcriptional output in cardiomyocytes, vascular cells, and immune cells. These modifications include methylation, acetylation, and metabolite-derived acylations. While the enzymatic machinery of classical histone marks is increasingly well defined, the cell-type-specific integration of th

Mechanistic insights into acetylated histone recognition by the CECR2 bromodomain.

Chromatin remodeling regulator CECR2 (cat eye syndrome chromosome region candidate 2) plays critical roles in neurulation, spermatogenesis, DNA damage response, and cancer metastasis, yet the structural determinants underlying its recognition of acetylated histones remain poorly understood. Here, we characterize the histone-binding specificity and structural determinants of the CECR2 bromodomain using isothermal titration calorimetry, biochemical assays, and structure-guided computational modeling. The CECR2 bromodomain recognizes multiple monoacetylated histone H4 peptides, including H4K5ac,

Histone modification cross talk between a host and pathogen.

Bacterial pathogens modulate host cell physiology by secreting effector proteins that rewire host signaling pathways. A subset of these effectors directly modify host chromatin to reprogram gene expression and promote infection. While these enzymes are thought to function autonomously, the extent to which the host epigenetic landscape regulates their activity remains largely unknown. RomA and its homolog LegAs4 are Set domain-containing lysine methyltransferases from Legionella pneumophila that methylate histone H3 at lysine 14 (H3K14) to suppress host immune responses and enhance intracellula

Age-dependent role of histone deacetylase Sirt7 on haematopoiesis.

Sirt7 is a member of the sirtuin family of proteins, which are NAD+-dependent deacetylases and ADP-ribosyltransferases. It is involved in a wide range of cellular processes. To study the specific role of Sirt7 in hematopoiesis during aging, the gene was specifically inactivated in hematopoietic stem cells (HSC). Vav1 promoter mediated expression of CRE recombinase in floxed Sirt7 mice resulted in specific inactivation of Sirt7 in the hematopoietic stem and progenitor cells. Young mice exhibited a normal peripheral blood count and no detectable hematological aberrancies. Peripheral blood of 19-

Histone Lactylation Links Glycolysis to Ferroptosis in Diabetic Cataract.

AIMS: This study aimed to delineate a novel mechanistic axis linking hyperglycemia-driven glycolytic reprogramming to ferroptotic death in lens epithelial cells (LECs) and to determine its therapeutic significance in diabetic cataract (DC). Specifically, we sought to define the integration of metabolic, epigenetic (histone lactylation), and post-translational (fucosylation) pathways in DC pathogenesis. RESULTS: Under hyperglycemic conditions, LECs exhibited robust glycolytic activation and lactate accumulation. This metabolic shift drove selective histone H3K18 lactylation at the promoter of

Histone decrotonylation plays a distinct role in HIV latency.

The role of epigenetic regulation in HIV latency remains incompletely understood. We show that histone deacetylase 3 (HDAC3) inhibits trans-activator of transcription (Tat)-mediated HIV transcription through histone decrotonylation (HDCR), independent of deacetylase activity. Chemical biology approaches identified selective HDCR inhibitors (HDCRis) that reverse HIV latency with minimal impact on other histone acylations. Although HDAC2, HDAC3, and HDAC8 exhibit HDCR activity, genetic and chemical studies reveal that the HDCRi citarinostat is selective for HDAC3 and HDAC8. Molecular docking sug
