Transcriptomic Approaches in the Zebrafish Model for Tuberculosis-Insights Into Host- and Pathogen-specific Determinants of the Innate Immune Response.

Mycobacterium marinum infection in zebrafish has become a well-established model of tuberculosis. Both embryonic and adult zebrafish infection studies have contributed to our knowledge of the development and function of tuberculous granulomas, which are typical of mycobacterial pathogenesis. In this review we discuss how transcriptome profiling studies have helped to characterize this infection process. We illustrate this using new RNA sequencing (RNA-Seq) data that reveals three main phases in the host response to M. marinum during the early stages of granuloma development in zebrafish embryo

Development of species-specific primers for identification of Biomphalaria arabica, the intermediate host of Schistosoma mansoni in Saudi Arabia.

Schistosoma mansoni is mediated through the intermediate host Biomphalaria arabica which lives in Saudi Arabia. Molecular characterization and identification of this intermediate host are important for epidemiological studies of schistosomiasis. The present work aimed to determine the molecular variations among the populations of B. arabica found in Southern part of Saudi Arabia, and to develop species-specific primers for identification of these snails as a first step in the development of multiplex PCR for simultaneously identifying the snails and diagnosing its infections in a single step.

Serogroup-specific interaction of Neisseria meningitidis capsular polysaccharide with host cell microtubules and effects on tubulin polymerization.

We have previously shown that during late stages of the infectious process, serogroup B meningococci (MenB) are able to escape the phagosome of in vitro-infected human epithelial cells. They then multiply in the cytosolic environment and spread intracellularly and to surrounding cells by exploiting the microtubule cytoskeleton, as suggested by results of infections in the presence of microtubule inhibitors and evidence of nanotubes connecting neighboring cells. In this study, by using microtubule binding assays with purified microtubule asters and bundles and microtubule bundles synthesized in

Identification of a novel tetrapeptide structure of the Mycobacterium avium glycopeptidolipid that functions as a specific target for the host antibody response.

Mycobacterium avium complex (MAC) is a group of non-tuberculous mycobacteria that cause tuberculosis-like diseases in humans. Unlike Mycobacterium tuberculosis, MAC expresses high levels of glycopeptidolipids (GPLs) containing a well-defined tetrapeptide-amino alcohol core, composed of D-phenylalanine, D-allo-threonine, D-alanine, and L-alaninol, that is modified with a fatty acid and sugar residues. Surprisingly, however, a careful scrutiny of the mass spectrum of MAC GPLs revealed the presence of ions that could not readily accountable for the known GPL structure. The magnitude of the ions w

Host-specific symbiotic requirement of BdeAB, a RegR-controlled RND-type efflux system in Bradyrhizobium japonicum.

Multidrug efflux systems not only cause resistance against antibiotics and toxic compounds but also mediate successful host colonization by certain plant-associated bacteria. The genome of the nitrogen-fixing soybean symbiont Bradyrhizobium japonicum encodes 24 members of the family of resistance/nodulation/cell division (RND) multidrug efflux systems, of which BdeAB is genetically controlled by the RegSR two-component regulatory system. Phylogenetic analysis of the membrane components of these 24 RND-type transporters revealed that BdeB is more closely related to functionally characterized or

Influence of host genetic variation on rubella-specific T cell cytokine responses following rubella vaccination.

The variability of immune response modulated by immune response gene polymorphisms is a significant factor in the protective effect of vaccines. We studied the association between cellular (cytokine) immunity and HLA genes among 738 schoolchildren (396 males and 342 females) between the ages of 11 and 19 years, who received two doses of rubella vaccine (Merck). Cytokine secretion levels in response to rubella virus stimulation were determined in PBMC cultures by ELISA. Cell supernatants were assayed for Th1 (IFN-gamma, IL-2, and IL-12p40), Th2 (IL-4, IL-5, and IL-10), and innate/proinflammator

Identification of cognate host targets and specific ubiquitylation sites on the Salmonella SPI-1 effector SopB/SigD.

Salmonella enterica is a bacterial pathogen responsible for enteritis and typhoid fever. Virulence is linked to two Salmonella pathogenicity islands (SPI-1 and SPI-2) on the bacterial chromosome, each of which encodes a type III secretion system. While both the SPI-1 and SPI-2 systems secrete an array of effectors into the host, relatively few host proteins have been identified as targets for their effects. Here we use stable isotope labeling with amino acids in cell culture (SILAC) and quantitative mass spectrometry-based proteomics to identify the host targets of the SPI-1 effector, SopB/Sig

Evaluation of a CD25-specific immunotoxin for prevention of graft-versus-host disease after unrelated marrow transplantation.

Donor T cells activated by recipient alloantigens cause graft-versus-host disease (GVHD) after hematopoietic cell transplantation. Activated T cells express CD25, among other components of the interleukin-2 receptor. We conducted a phase I/II study to determine whether administration of CD25-specific antibody conjugated to ricin toxin A could reduce the risk of grade III or IV GVHD after marrow transplantation from HLA-matched unrelated donors. All patients received methotrexate and cyclosporine after the transplantation. The immunotoxin was given to 36 patients for 4 consecutive days beginnin

Further evidence of host species-specific variation in antigens of Pneumocystis carinii using the polymerase chain reaction.

To further elucidate the extent of variation among Pneumocystis carinii obtained from different mammalian hosts, polymerase chain reaction (PCR) analysis of the genes encoding two antigens of P. carinii was done. Using primers based on the ferret P. carinii glycoprotein (gp)A gene and the rat P. carinii 45- to 55-kDa antigen gene, amplification was attempted with DNA isolated from P. carinii-infected ferret, rat, mouse, and human lungs. For both genes, amplification was successful only with P. carinii DNA isolated from the same host species from which the P. carinii gene was originally isolate

Analysis of Coxiella burnetii isolates in cell culture and the expression of parasite-specific antigens on the host membrane surface.

Coxiella burnetii isolates may be classified into several groups based on plasmid character. These groups may also be correlated with disease syndrome--chronic or short-term acute. L929 mouse fibroblast cells were exposed, independently, to two members of the three major C. burnetii groups, and their growth/morphological characteristics analysed by light and electron microscopy, including High Voltage Electron Microscopy. The fates of the isolates were followed. Two acute isolates [Nine Mile (RSA 493) and Henzerling (RSA 331), QpH1-type plasmids] and two chronic isolates (S Q217 and L Q216, pl

Transcription during the development of bacteriophage phi 29: production of host- and phi 29-specific ribonucleic acid.

The synthesis of ribonucleic acid (RNA) during development of the virulent Bacillus subtilis bacteriophage phi29 has been analyzed. Transcription of host deoxyribonucleic acid (DNA) continues at the preinfection rate throughout the latent period of viral growth. RNA-DNA hybridization was used to show that host messenger RNA synthesis continues late into the phage lytic cycle. Amino acid-labeling experiments show that this RNA is continuously used to produce protein. Ribosomal RNA production is not inhibited by phage infection. Small quantities of phage-specific RNA first appear between min 6 a

The influence of food competition and host specificity on the transmission of Triaenophorus crassus (Cestoda) and Cystidicola farionis (Nematoda) to Coregonus lavaretus and Coregonus albula (Pisces:Coregonidae) in Finland.

As a superior competitor for planktonic food, vendace (Coregonus albula), when abundant, is expected to displace whitefish (Coregonus lavaretus) from feeding on plankton and to force it to rely more on benthic food. The predicted result would be a reduced abundance of the copepod-transmitted cestode Triaenophorus crassus in whitefish, but an increase in the abundance of the nematode Cystidicola farionis transmitted via benthic amphipods. We studied the occurrence of both parasites in whitefish during 1991-1996 in three interconnected areas at Lake Saimaa, Finland, where the densities of the ve

Host-Specific Circulation and Molecular Diversity of Babesia, Hepatozoon and Bartonella Species Among Wild Carnivores in Northwestern Italy.

Vector-borne diseases (VBDs) represent a concern for animal and public health, and wildlife surveillance is valuable for their monitoring. The occurrence, molecular diversity and host distribution of tick-borne protozoa (TBP: Babesia spp., Theileria spp. and Hepatozoon spp.) and Bartonella spp. were investigated in wild carnivores from three regions of northwestern Italy. Spleen samples from 690 wild carnivores were screened for TBP and/or Bartonella spp. by PCR followed by sequencing. Among the 503 carnivores screened for TBP, 197 animals (39.2%) tested positive for at least one targeted prot

Mapping of Phenotype Specific Host-Microbiome Protein-Protein Interaction Networks in Colorectal Cancer Using Deep Learning.

Colorectal cancer (CRC) pathogenesis is driven by complex protein-protein interactions (PPIs) between the host and the gut microbiome, yet these molecular dialogs remain largely unmapped. This study utilizes a Deep Learning framework, enhanced by protein structure embeddings, to predict approximately 8.9 billion interspecies PPIs from clinical metagenomic data. The model achieved high accuracy with an AUROC of 0.9960, identifying a high-confidence interactome representing roughly 16% of evaluated protein pairs. Phenotype-specific analysis revealed that while microbial hubs shift-transitioning

Genomic Component Interactions Shape Host-Specific Systemic Infection in Tomato Leaf Curl New Delhi Virus.

Tomato leaf curl New Delhi virus (ToLCNDV) is a bipartite begomovirus with a broad host range and marked geographic variation in host adaptation. Whereas Asian isolates readily infect solanaceous crops, Mediterranean isolates are mainly adapted to cucurbits and infect tomato poorly. The molecular basis of this divergence remains unclear. Here, we compared Indian (ToLCNDV-IN) and Spanish (ToLCNDV-ES) isolates using reciprocal pseudo-recombinants to infect Solanum lycopersicum and Nicotiana benthamiana. Both DNA-A components replicated in tomato leaves, indicating that the inability of ToLCNDV-E

The Farooq, not the Lerner, grading system predicts disease-specific and overall survival in duodenal graft-versus-host disease.

Histopathological evaluation of graft-versus-host disease (GVHD) is crucial in its diagnosis. In the upper gastrointestinal tract, the duodenum has been reported as the most affected site. However, studies characterizing the features of duodenal GVHD (dGVHD) are sparse and no study to date have compared the utility of the traditionally use Lerner grading system and the recently proposed Farooq grading system. 130 cases were included with a median age of 40 years (range: 0.8-73) and 50.8% of the patients were men. The median follow-up time was 32 months (range: 0-185), 83.1% of patients receive

Zone-specific hepatocytes orchestrate the early onset of host immune defenses during Staphylococcus aureus bloodstream infection.

BACKGROUND: Bloodstream infections (BSI) are life-threatening conditions initiated by pathogens entering the circulation, with Staphylococcus aureus among the most lethal causative agents. The liver is the primary site for the initial detection and clearance of blood-borne pathogens. Emerging evidence indicates that the hepatic immune response to pathological insults is influenced by liver zonation, exhibiting both spatial and cellular specialization. Because direct experimental evidence linking liver zonation to the immune response to BSI is currently lacking, this study aimed to characterize

A comprehensive review on microbial production of vanillin: metabolic engineering strategies and pathway-specific yields across microbial hosts.

Vanillin is one of the chief flavors used in the food, beverage, fragrance, and medicinal industries. It is a phenolic aldehyde with aldehyde, ethers, and hydroxyl as functional groups and is chemically known as 4-hydroxy-3-methoxybenzaldehyde, obtained from species like Vanilla planifolia, Vanilla tahitensis, and Vanilla pompona. The Food and Drug Administration (FDA) and the Flavor and Extract Manufacturers Association (FEMA) have recommended vanillin as a safe flavor additive for consumption. The international vanillin market has experienced noteworthy growth in the past few years, recordin

Pathogen-specific host response in critically ill patients with blood stream infections: a nested case-control study.

BACKGROUND: Knowledge of the contribution of the pathogen to the heterogeneity of the host response to infection is limited. We aimed to compare the host response in critically ill patients with a bloodstream infection (BSI). METHODS: RNA profiles were determined in blood obtained between one day before and after a positive blood culture. Differential expression and pathway analyses were performed on independent patients' samples by RNA sequencing (discovery) or microarray (validation). Additional patients were included for the discovery and validation of transcriptome classifiers of pathogen

Modeling Mycobacterium tuberculosis pathogenesis in lung epithelial organoids reveals strain-specific host responses and intercellular crosstalk.

Emerging studies have identified alveolar epithelial cells as a conducive niche for Mycobacterium tuberculosis (Mtb) replication and spread during early infection. However, the host-pathogen interactions and intercellular crosstalk within the lung epithelial microenvironment remain inadequately understood. Here, we developed a lung epithelial organoid coculture model and exposed it to the virulent H37Rv strain or the avirulent Bacillus Calmette-Guérin (BCG) strain to investigate tuberculosis (TB) pathogenesis. Transcriptomic analyses revealed that Mtb infection markedly alters cell death patte
