Cold plasma triggered cell death with a curcumin and capecitabine loaded magnetic nanocluster-based multifunctional system on the MCF-7 cell line: a smart therapy platform.

The development of smart, selective, and multifunctional nanotherapeutics is crucial for advancing next-generation cancer treatments. In this study, superparamagnetic iron oxide nanoclusters (SPIONCs) were coated with mesoporous silica, functionalized with folic acid (FA), and co-loaded with curcumin (CUR) and capecitabine (CAPE) to create a novel nanocarrier system. To enhance cellular internalisation, magnetophoresis was applied before exposure of the cells to cold atmospheric plasma (CAP). The resulting FA-conjugated, CUR and CAPE-loaded nanoclusters were evaluated in vitro in MCF-7 breast

The extracts from centaurea species abolished the cytotoxic effects of doxorubicin on breast cancer cell line - MCF-7 and bortezomib on prostate cancer cell line- PC3.

The use of plant extracts by cancer patients during chemotherapy poses potential risks, as they may reduce the effectiveness of treatment or interact negatively with chemotherapeutic drugs. There is a lack of comprehensive studies evaluating the effects of various Centaurea spp. plant extracts on chemotherapy outcomes, highlighting the need for caution and medical supervision. Therefore, the aim of this study was to evaluate the effects of five Centaurea spp. extracts in concentrations of 125 μg/ml and 250 μg/ml on the cytotoxicity induced by doxorubicin (DOX, 1 μM) in MCF-7 breast cancer cell

Alterations in transforming growth factor-alpha and -beta production and cell responsiveness during the progression of MCF-7 human breast cancer cells to estrogen-autonomous growth.

Hormonal management of breast cancer is confounded by an almost inevitable progression of cell growth from a steroid-regulated to a steroid-autonomous state. We have experimentally induced this progression in the estrogen growth-responsive MCF-7 human breast cancer cell line by long-term culture in the absence of steroids. After an initial period (10-12 weeks) of slowed growth in response to steroid deprivation, rapid, steroid-independent growth rates were consistently established. In these cells, which contained 3-fold elevated, functional estrogen receptor levels (as determined by induction

Endophytic Xylaria sp. from Syzygium myrtifolium as a novel source of cytochalasin D with antimalarial activity against Plasmodium falciparum and anticancer activity against MCF-7 cells.

Endophytic fungi associated with Syzygium myrtifolium have significant potential as rapid sources of bioactive compounds for drug discovery. This study aimed to identify antimalarial and anticancer compounds from an endophytic fungal isolate Xylaria sp., associated with S. myrtifolium. The Xylaria sp. SmPdDh-4 isolate was identified using morphological characterisation and phylogenetic analysis. Metabolite profiling was performed using LC-HRMS and further confirmed by NMR spectroscopy, identifying cytochalasin D as the major compound, which was also confirmed by X-ray diffraction examination.

Estrogen-related mechanisms underlying bisphenol A-Induced proliferation and migration in MCF-7 breast cancer cells: An integrated network toxicology, molecular docking, and in vitro study.

Bisphenol A (BPA) is an environmental endocrine-disrupting chemical widely present in plastics and food packaging. Owing to its estrogen-like activity, BPA may increase the risk of estrogen-related tumors, including breast cancer, by interfering with endocrine function; however, the underlying molecular mechanisms remain unclear. To systematically evaluate the potential effects of BPA on estrogen-related tumors, including breast, ovarian, and endometrial cancers, this study integrated network toxicology, molecular docking, and in vitro validation. Bioinformatics approaches were first used to p

Novel benzenesulfonamide-aroylhydrazone conjugates as carbonic anhydrase inhibitors that induce MAPK/ERK-mediated cell cycle arrest and mitochondrial-associated apoptosis in MCF-7 breast cancer cells.

A series of novel 4-alkylthio-2-chloro-5-[(2-arylmethylidene)hydrazinecarbonyl]benzenesulfonamide derivatives 3-22 were synthesized and evaluated for their inhibitory activity against human carbonic anhydrase isozymes hCA I, hCA II, hCA IX, and hCA XII. These compounds showed varying degrees of activity against the studied isoenzymes. However, the importance of substituent choice in designing potent carbonic anhydrase inhibitors is highlighted by the strong inhibition profiles of compounds 3 and 10 against hCA IX and the low average KI values for compounds 9 and 10 (134 nM and 77 nM, respectiv

Unraveling the cytotoxicity and cellular uptake of low, medium and high molecular weight polyethylene glycol polymers in MCF-7 cells by green UPLC-MS/MS methods.

Unraveling the cytotoxicity and cellular uptake of low, medium and high molecular weight polyethylene glycol (PEG) in cells is important for evaluation of therapeutic efficacy and toxicity of PEGylated drug delivery systems. In this study, cellular uptake of PEG600, PEG2K, PEG4K and PEG10K in MCF-7 cells was first studied by an UPLC-MS/MS assay coupled with collision induced dissociation (CID) in source technique. The CID of PEG in source with high values of declustering potentials generates numerous PEG-related product ions. These PEG-related fragment ions can be further broken into specific

Optimized semisolid self-nanoemulsifying system based on glyceryl behenate: A potential nanoplatform for enhancing antitumor activity of raloxifene hydrochloride in MCF-7 human breast cancer cells.

Raloxifene hydrochloride (RLX) is a selective estrogen receptor modulator used for treatment and protection against postmenopausal osteoporosis. The drug has been used for protection against breast cancer and more recently, for management of the disease by virtue of its estrogen antagonist action. However, the drug has reduced bioavailability related to low water solubility and first pass metabolism. To surmount these pitfalls, this study aimed at developing and optimizing RLX-loaded semisolid self-nanoemulsifying system (SSNES) with minimized globule size to improve the drug solubility, tumor

Inhibitory effect of silver nanoparticles on proliferation of estrogen-dependent MCF-7/BUS human breast cancer cells induced by butyl paraben or di-n-butyl phthalate.

In this study the effect of silver nanoparticles (AgNPs) on proliferation of estrogen receptor (ER)-positive human breast cancer MCF-7/BUS cells was assessed by means of in vitro assay. The cells were exposed in the absence of estrogens to AgNPs alone or in combination with aluminum chloride (AlCl3), butyl paraben (BPB) and di-n-butyl phthalate (DBPh). The results revealed that AgNPs at the non-cytotoxic concentrations (up to 2μg/mL) and AlCl3 (up to 500μM) did not induce proliferation of MCF-7/BUS cells whereas BPB and DBPh showed strong estrogenic activity with the highest effect at 16μM and

Induction of cytochrome P450 1A1 in MCF-7 human breast cancer cells by 4-chlorobiphenyl (PCB3) and the effects of its hydroxylated metabolites on cellular apoptosis.

Several studies suggest an involvement of PCBs in breast cancer formation, but the results are ambiguous and the mechanisms not clear. We propose that local activation of cytochrome P450 enzymes, CYP1A1 and CYP1B1 by PCB3, may generate active metabolites which affect apoptosis and thereby promote mammary carcinogenesis. To test this hypothesis MCF-7 human breast cancer cells were exposed to 300 nM PCB3 and its hydroxylated metabolites, 4OH-PCB and 3,4diOH-PCB3. The enzyme activity for CYP1A1 was assayed using the EROD assay, and CYP1A1 and CYP1B1 protein expression by western blotting. PCB3 in

Aldo-keto reductase 1C3 expression in MCF-7 cells reveals roles in steroid hormone and prostaglandin metabolism that may explain its over-expression in breast cancer.

Aldo-keto reductase (AKR) 1C3 (type 5 17beta-hydroxysteroid dehydrogenase and prostaglandin F synthase), may stimulate proliferation via steroid hormone and prostaglandin (PG) metabolism in the breast. Purified recombinant AKR1C3 reduces PGD(2) to 9alpha,11beta-PGF(2), Delta(4)-androstenedione to testosterone, progesterone to 20alpha-hydroxyprogesterone, and to a lesser extent, estrone to 17beta-estradiol. We established MCF-7 cells that stably express AKR1C3 (MCF-7-AKR1C3 cells) to model its over-expression in breast cancer. AKR1C3 expression increased steroid conversion by MCF-7 cells, leadi

Evaluation of potential implication of membrane estrogen binding sites on ERE-dependent transcriptional activity and intracellular estrogen receptor-alpha regulation in MCF-7 breast cancer cells.

The potential involvement of membrane estrogen binding sites in the induction of ERE-dependent transcriptional activity as well as in the regulation of intracellular estrogen receptor alpha (ER-alpha) level under estradiol (E2) stimulation was investigated. Our approach relied upon the use of two DCC-treated E2-BSA (bovine serum albumin) solutions (E2-6-BSA and E2-17-BSA). The absence of detectable free E2 in these solutions was established. Both E2-BSA conjugates led to a transient dose-dependent stimulation of the expression of ERE-luciferase (LUC) reporter gene in MVLN cells (MCF-7 cells st

Plasma membrane receptors for the cancer-regulating progesterone metabolites, 5alpha-pregnane-3,20-dione and 3alpha-hydroxy-4-pregnen-20-one in MCF-7 breast cancer cells.

Recent observations indicate that the progesterone metabolite, 5alpha-pregnane-3,20-dione (5alphaP), which is produced at higher levels in tumorous breast tissue, promotes cell proliferation and detachment, whereas 3alpha-hydroxy-4-pregnen-20-one (3alphaHP), which is produced at higher levels in nontumorous breast tissue, suppresses proliferation and detachment of MCF-7 breast cancer cells. The objective of the current study was to determine the presence and characteristics of binding sites for these endogenous putative cancer-regulating steroid hormones. Radiolabeled 5alphaP and 3alphaHP were

Identification of new ER-targeted anticancer hits active against MCF-7 breast cancer cells in the marine fungal secondary metabolite library using computative and experimental approaches.

The identification of novel estrogen receptor (ER)-targeted anticancer compounds from marine fungal secondary metabolites was performed using a combined computational and experimental approach. A library of 461 marine fungal metabolites was analyzed using chemical space visualization, structural similarity assessment using Tanimoto indices, and target-prediction tools to identify potential ER ligands. Scaffold analysis of predicted hits suggested decalin/bicyclic terpenoid (6-6), tricyclic 6-6-6 terpenoid, bicyclic 5-6 system and several polycyclic system chemotypes as novel for ER ligands. Th

T-type voltage-activated calcium channel Cav3.1, but not Cav3.2, is involved in the inhibition of proliferation and apoptosis in MCF-7 human breast cancer cells.

T-type voltage-gated Ca2+ channels have unique electrophysiological properties, suitable for generating Ca2+ oscillations and waves and thus controlling the proliferation of various tumor cells. In the present study, we investigated the role of Cav3.1, a candidate tumor suppressor gene, in neoplastic processes, and compared the differences between Cav3.1 with Cav3.2 channels. While the overexpression of a full-length Cav3.1 clone suppressed cell proliferation, the knockdown of the Cav3.1 gene by siRNA, or treatment with ProTx-I, a relatively selective inhibitor for Cav3.1, promoted the cell pr

Comparative effects of 2,3,7,8-tetrachlorodibenzo-p-dioxin on MCF-7, RL95-2, and LNCaP cells: role of target steroid hormones in cellular responsiveness to CYP1A1 induction.

A study was conducted to investigate whether target hormones affect 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD)-inducible gene expression, using as an experimental model system three human cancer cell lines, breast (MCF-7), uterine (RL95-2), and prostate (LNCaP). Exposure to TCDD induced the CYP1A1 gene in all three cell lines. MCF-7 and RL95-2 cells showed more than 15- and 10-fold induction of EROD (7-ethoxyresorufin O-deethylase) activity, respectively, compared with the less responsive LNCaP cells. Surprisingly, however, TCDD-induced reporter gene activity driven by a single XRE element was

Aptamer based strategy for cytosensing and evaluation of HER-3 on the surface of MCF-7 cells by using the signal amplification of nucleic acid-functionalized nanocrystals.

The electrochemical detection of cell lines of MCF-7 (human breast cancer) has been reported, using magnetic beads for the separation tool and high-affinity DNA aptamers for signal recognition. The high specificity was obtained by using the magnetic beads and aptamers, and the good sensitivity was realized with the signal amplification of DNA capped CdS or PbS nanocrystals. The ASV (anodic stripping voltammetry) technology was employed for the detection of cadmic cation and lead ions, for electrochemical assay of the amount of the target cells and biomarkers on the membrane of target cells, re
