Effect of Zinc on Microcystis aeruginosa UTEX LB 2385 and Its Toxin Production.

Cyanobacteria harmful algal blooms (CHABs) are primarily caused by man-made eutrophication and increasing climate-change conditions. The presence of heavy metal runoff in affected water systems may result in CHABs alteration to their ecological interactions. Certain CHABs produce by-products, such as microcystin (MC) cyanotoxins, that have detrimentally affected humans through contact via recreation activities within implicated water bodies, directly drinking contaminated water, ingesting biomagnified cyanotoxins in seafood, and/or contact through miscellaneous water treatment. Metallothionein

The efficiency of combined coagulant and ballast to remove harmful cyanobacterial blooms in a tropical shallow system.

We tested the hypothesis that a combination of coagulant and ballast could be efficient for removal of positively buoyant harmful cyanobacteria in shallow tropical waterbodies, and will not promote the release of cyanotoxins. This laboratory study examined the efficacy of coagulants [polyaluminium chloride (PAC) and chitosan (made of shrimp shells)] alone, and combined with ballast (lanthanum modified bentonite, red soil or gravel) to remove the natural populations of cyanobacteria collected from a shallow eutrophic urban reservoir with alternating blooms of Cylindrospermopsis and Microcystis.

Simultaneous Determination of Multiclass Cyanotoxins in Aquatic Products, Vegetables and Algal Dietary Supplements Using Dispersive Solid-Phase Extraction (dSPE)-UHPLC-MS/MS.

Cyanotoxins, prevalent in eutrophic aquatic ecosystems, pose significant health risks via contaminated food, yet analytical methods for detecting multiple toxin classes in foodstuffs remain limited. In the current study, a method based on dispersive solid-phase extraction (dSPE) coupled with ultra-high-performance liquid chromatography-tandem mass spectrometry (UHPLC-MS/MS) was developed to simultaneously determine four common classes of cyanotoxins (i.e., microcystins, cylindrospermopsins, nodularins, and anatoxins) in aquatic products, vegetables and algal dietary supplements. Microbial Fuel Cell Biosensor with Capillary Carbon Source Delivery for Real-Time Toxicity Detection.

A microbial fuel cell (MFC) biosensor with an anode as a sensing element is often unreliable at low or significantly fluctuating organic matter concentrations. To remove this limitation, this work demonstrates capillary action-aided carbon source delivery to an anode-sensing MFC biosensor for use in carbon-depleted environments, e.g., potable water. First, different carbon source delivery configurations using several thread types, silk, nylon, cotton, and polyester, are evaluated. Silk thread was determined to be the most suitable material for passive delivery of a 40 g L-1 acetate solution. T

Monitoring Cyanobacterial Blooms during the COVID-19 Pandemic in Campania, Italy: The Case of Lake Avernus.

Cyanobacteria are ubiquitous photosynthetic microorganisms considered as important contributors to the formation of Earth's atmosphere and to the process of nitrogen fixation. However, they are also frequently associated with toxic blooms, named cyanobacterial harmful algal blooms (cyanoHABs). This paper reports on an unusual out-of-season cyanoHAB and its dynamics during the COVID-19 pandemic, in Lake Avernus, South Italy. Fast detection strategy (FDS) was used to assess this phenomenon, through the integration of satellite imagery and biomolecular investigation of the environmental samples.

Fluorescent/colorimetric dual-mode for detecting of MC-LR using bidirectional RCA coupled with CdTe QDs.

A fluorescent/colorimetric dual-mode biosensor was designed using CdTe QDs and CRISPR/Cas for the efficient and ultrasensitive detection of microcystin-leucine-arginine (MC-LR). The biosensor mainly activates the trans-cleavage activity of Cas12a through nucleic acid amplification technology, such as bidirectional rolling circle amplification (B-RCA), to perform signal conversion, release Ag+ from the cleaving hairpin, quench QD fluorescence, and perform signal presentation. The biosensor can perform fluorometric and colorimetric detection, enabling rapid field assays. It exhibits enhanced sel

Carcinogenic aspects of protein phosphatase 1 and 2A inhibitors.

Okadaic acid is functionally a potent tumor promoter working through inhibition of protein phosphatases 1 and 2A (PP1 and PP2A), resulting in sustained phosphorylation of proteins in cells. The mechanism of tumor promotion with okadaic acid is thus completely different from that of the classic tumor promoter phorbol ester. Other potent inhibitors of PP1 and PP2A - such as dinophysistoxin-1, calyculins A-H, microcystin-LR and its derivatives, and nodularin - were isolated from marine organisms, and their structural features including the crystal structure of the PP1-inhibitor complex, tumor pro

Cyanotoxins induce hepatic inflammation and disrupt lipid metabolism in Sinocyclocheilus grahami.

Cyanobacterial blooms release a variety of secondary metabolites, severely impacting the survival of fish species. The multi-layered analytical approach (histology + biochemistry + transcriptomics + lipidomics) was used to investigate hepatic injury in Sinocyclocheilus grahami following a 96-hour exposure to Microcystis aeruginosa exudates (MaE) and their constituent, phytosphingosine (PHS; at 2.9 and 145 ng/mL). The results showed histopathological hepatic damage across treatments, with a significantly decreased hepatosomatic index (HSI) in the high-PHS-exposure group. Total bilirubin were sig

Studies of the liposolubility and the ecotoxicity of MC-LR degradation by-products using computational molecular modeling and in-vivo tests with Chlorella vulgaris and Daphnia magna.

Computational molecular modelling, mass spectrometry and in-vivo tests with Chlorella vulgaris (C. vulgaris) and Daphnia magna (D. magna) were used to investigate the liposolubility and ecotoxicity of MC-LR degradation by-products generated after oxidation by OH• radicals in Fenton process. Exposure of MC-LR (5 µg.L-1) to the most severe oxidation conditions (Fe2+ 20 mM and H2O2 60 mM) resulted in a reduction in the toxin concentration of 96% (0.16 µg.L-1), however, with the formation of many by-products. The by-product of m/z 445 was the most resistant to degradation and retained a toxic stru

Epidemiology of primary liver cancer in Serbia and possible connection with cyanobacterial blooms.

Today, the occurrence of harmful cyanobacterial blooms is a common phenomenon and a potential global health problem. Cyanobacteria can produce metabolites highly toxic to humans. More than 80% of reservoirs used for water supply in Central Serbia have bloomed over the past 80 years. A 10-year epidemiological study showed a significant increase in the incidence of primary liver cancer (PLC) in the regions where water from the blooming reservoirs was used for human consumption. At the same time, no correlation was found between the incidence of PLC and other risk factors, such as cirrhosis and h

Impact of toxic cyanobacterial blooms on Eurasian perch (Perca fluviatilis): experimental study and in situ observations in a peri-alpine lake.

Due to the importance of young-of-the-year (YOY) perch in the peri-alpine regions where they are consumed, the microcystin (MC) contamination of YOY perch was analysed both in field (Lake Bourget, France) and experimentally using force-feeding protocols with pure MCs. In-situ, schools of YOY perch present in the epilimnion of the lake were never found in direct contact with the P. rubescens blooms that were present in the metalimnion. However, MCs were detected in the muscles and liver of the fish and were thus assumed to reach YOY perch through dietary routes, particularly via the consumption

Cyanobacteria Microcystis aeruginosa Contributes to the Severity of Fish Diseases: A Study on Spring Viraemia of Carp.

Fish are exposed to numerous stressors in the environment including pollution, bacterial and viral agents, and toxic substances. Our study with common carps leveraged an integrated approach (i.e., histology, biochemical and hematological measurements, and analytical chemistry) to understand how cyanobacteria interfere with the impact of a model viral agent, Carp sprivivirus (SVCV), on fish. In addition to the specific effects of a single stressor (SVCV or cyanobacteria), the combination of both stressors worsens markers related to the immune system and liver health. Solely combined exposure re

Blue-green algae toxicosis in cattle.

Twenty-four of 175 heifers died after ingesting water from a stock pond containing blue-green algae (genus Microcystis) in southern Colorado. Affected cattle were found dead or had signs of nervousness, and were recumbent, weak, anorectic, and hypersensitive to noise when first examined. All cattle died within 3 days after the onset of signs. At necropsy, the rumen contained blue-green algae, and the liver was larger than normal, friable, and dark red. The most important histologic lesion was hepatocyte degeneration and necrosis. Intraperitoneal administration of lyophilized cell material from

First Confirmed Case of Canine Mortality Due to Dihydroanatoxin-a in Central Texas, USA.

The frequency of dogs becoming ill or dying from accidental exposure to cyanotoxins, produced by cyanobacteria, is increasing throughout the United States. In January and February of 2021, two dogs died and five dogs became ill after swimming in Lake Travis, central Texas, USA; one deceased dog (C1) was subjected to pathological testing. Algal materials, sediment samples, zebra mussel viscera, periphyton from shells, as well as fluids and tissues from the digestive tract of C1 were investigated for the following cyanotoxins: anatoxin-a, homoanatoxin-a, dihydroanatoxin-a (dhATX), cylindrospermo

Tonsillar microbiota in children with PFAPA (periodic fever, aphthous stomatitis, pharyngitis, and adenitis) syndrome.

Periodic fever, aphthous stomatitis, pharyngitis, and adenitis (PFAPA) is a childhood febrile syndrome of unknown origin that is often cured with tonsillectomy. We aimed to compare the bacterial microbiota of the tonsils removed from PFAPA patients with those of controls. We used next-generation sequencing technology to investigate the bacterial microbiota of the tonsils of 30 PFAPA patients and 24 controls. We found significant differences in the presence and relative abundance of many bacteria between PFAPA cases and controls. For example, cyanobacteria, potential producers of microcystins a

Investigation of the Occurrence of Cyanotoxins in Lake Karaoun (Lebanon) by Mass Spectrometry, Bioassays and Molecular Methods.

Lake Karaoun is the largest artificial lake in Lebanon and serves multiple purposes. Recently, intensive cyanobacterial blooms have been reported in the lake, raising safety and aesthetic concerns related to the presence of cyanotoxins and cyanobacterial taste and odor (T&O) compounds, respectively. Here, we communicate for the first time results from a recent investigation by LC-MS/MS covering multiple cyanotoxins (microcystins (MCs), anatoxin-a, cylindrospermopsin, nodularin) in water and fish collected between 2019 and 2020. Eleven MCs were identified reaching concentrations of 211 and 199

Tissue metallothionein response in the Japanese quail associated with exposure to cyanobacterial biomass, lead and the Newcastle disease virus.

OBJECTIVES: Here we tested the hypothesis that multiple toxic and infectious stressors combine in their adverse effects to produce higher tissue responses of metallothioneins (MTs) in birds. METHODS: We used Japanese quails as a model avian species. The study is based on data obtained from single and combined exposures of Japanese quails to cyanobacterial biomass containing microcystins, lead and a live Newcastle disease vaccination virus. Eight groups of 5 birds were exposed to single, double and triple combinations of these stressors and compared with controls. Birds were euthanized after t

Spatial and temporal variation of suspect screening derived cyanobacterial secondary metabolite mixtures during harmful algal bloom events in shallow agroecosystem lakes.

Freshwater harmful algal blooms (HABs) pose significant health risks to communities through drinking water sources and contact recreation. Cyanobacteria that drive HABs produce a complex array of secondary metabolites, notably cyanotoxins. The spatiotemporal variability of cyanometabolite mixtures during HAB events remains poorly understood, particularly in agroecosystems, where heavy nutrient loading can alter normal lake dynamics. Therefore, we applied high-resolution mass spectrometry-based suspect screening to characterize cyanometabolite mixture dynamics during active HAB events in two Ea
