An Experimental Study of Incremental Buckling-Resistant Inchworm-Type Insertion of Microwire Neural Electrodes.

Chronically implanting microelectrodes for high-resolution action potential recording is critical for understanding the brain. The smallest and most flexible electrodes, most suitable for chronic recordings, are also the most difficult to insert due to buckling against the thin but hard-to-penetrate brain meninges. To address such implantation challenges without introducing further damage to the brain, this paper presents our design and prototype of an inchworm-type insertion device that conducts a grip-feed-release incremental motion for planar microelectrode insertion. To optimize the operat

In vitro and preclinical assessment of drug interactions between fluoroquinolones and a nonsteroidal antiinflammatory drug predicting risk of seizure.

Seizures are a common reason for drugs to fail during development, but they are difficult to predict preclinically. Enoxacin, a fluoroquinolone, rarely causes convulsions as a monotherapy, but convulsions have been seen after combination therapy with fenbufen, a nonsteroidal antiinflammatory drug. The interaction between the drugs is thought to result from inhibition of GABAA receptors, which are not their primary targets. Here, we show that, among 15 marketed fluoroquinolones and 1 active metabolite, 6, including enoxacin, inhibited γ-aminobutyric acid (GABA)-evoked depolarization in cells ex

Visual fields in patients with posterior GPi pallidotomy.

The objective of this study was to describe the incidence and types of visual field defects after posterior globus pallidus internus (GPi) pallidotomy for Parkinson's disease. The creation of the pallidotomy lesion carries a risk of damaging neighboring structures such as the optic tract. The reported frequency of visual field defects in patients after pallidotomy varies from 0 to 40%. Goldmann visual field testing was performed on 40 patients who underwent microelectrode-guided posterior GPi pallidotomy. The optic tract was identified during the procedure by listening during microelectrode re

VLSI circuits for bidirectional interface to peripheral and visceral nerves.

This paper presents an architecture for sensing nerve signals and delivering functional electrical stimulation to peripheral and visceral nerves. The design is based on the very large scale integration (VLSI) technology and amenable to interface to microelectrodes and building a fully implantable system. The proposed stimulator was tested on the vagus nerve and is under further evaluation and testing of various visceral nerves and their functional effects on the innervated organs.

Sub-second striatal dopamine release measured by in vivo voltammetry.

High speed cyclic voltammetry was used to measure electrically stimulated striatal dopamine release in vivo with 25 ms time resolution. Dopamine release after 1 s stimulations was readily detectable and could be manipulated by drugs known to influence dopaminergic neurones. Using data-averaging dopamine release could be detected following stimulus trains as short as 100 ms. The use of carbon fibre microelectrodes gave a spatial resolution better than 5 micron.

Network Analysis Pipeline for Neuronal 2D and 3D Organoid Multielectrode Recordings.

Multielectrode recordings from 2D or 3D cultures are commonly used to assess neuronal activity including action potential firing and burst rates. Yet, multielectrode recordings, which carry parallel time series, can also be used to infer functional connectivity and to compare network features from neuronal networks within neuronal 2D cultures or 3D organoids. This chapter provides a protocol for using the microelectrode array network analysis pipeline (MEA-NAP). With MEA-NAP, users with limited computational experience can easily evaluate network activity in their microelectrode array (MEA) re

Dithiobis(succinimidyl propionate) modified gold microarray electrode based electrochemical immunosensor for ultrasensitive detection of cortisol.

Gold microelectrode arrays functionalized with dithiobis(succinimidyl propionate) self-assembled monolayer (SAM) have been used to fabricate an ultrasensitive, disposable, electrochemical cortisol immunosensor. Cortisol specific monoclonal antibody (C-Mab) was covalently immobilized on the surface of gold microelectrode array and the sensors were exposed to solutions with different cortisol concentration. After C-Mab binding, unreacted active groups of DTSP were blocked using ethanol amine (EA) and label-free electrochemical impedance (EIS) technique was used to determine cortisol concentratio

[Development of conductometric biosensor based on alkaline phosphatase for determining concentration of cadmium ions].

The paper describes a novel conductometric biosensor sensitive to cadmium ions based on alkaline phosphatase immobilized on gold planar microelectrodes used as transducers. Assays have been carried out with paranitrophenyl phosphate as substrate for the immobilized enzyme. Various parameters such as reticulation time, along with pH, ionic strength and buffer concentration of the measuring solution were studied. The optimized biosensor was stable, reproducible and it exhibited a detection limit of 4.45 microM for cadmium ions.

Safety profile of intracranial electrode implantation for video-EEG recordings in drug-resistant focal epilepsy.

Invasive electroencephalography recordings with depth or subdural electrodes are necessary to identify the ictogenic area in some drug-resistant focal epilepsies. We aimed to analyze the safety profile of intracranial electrode implantation in a tertiary center and the factors associated with its complications. We retrospectively examined complications in 163 intracranial procedures performed in adult patients. Implantation methods included oblique depth stereotactic approach (n = 128) and medial-temporal depth stereotactic approach in combination with subdural strip placement (n = 35). 1201 d

Patterning cells on optically transparent indium tin oxide electrodes.

The ability to exercise precise spatial and temporal control over cell-surface interactions is an important prerequisite to the assembly of multi-cellular constructs serving as in vitro mimics of native tissues. In this study, photolithography and wet etching techniques were used to fabricate individually addressable indium tin oxide (ITO) electrodes on glass substrates. The glass substrates containing ITO microelectrodes were modified with poly(ethylene glycol) (PEG) silane to make them protein and cell resistive. Presence of insulating PEG molecules on the electrode surface was verified by c

Luminal acid increases apical cell membrane resistance in isolated Necturus antral mucosa.

BACKGROUND & AIMS: The gastric mucosa must have efficient protective mechanisms to maintain physiological intracellular pH. The aim of this study was to investigate the effect of low luminal pH on apical membrane permeability. METHODS: Chambered Necturus antral mucosa was perfused with Ringer's/95% O2-5% CO2 at pH 7.25. The mucosal side was exposed to pH 4.0-2.0 with four microelectrodes placed in surface cells. Two-dimensional cable analysis was used to measure apical, basolateral, and shunt resistances. In some experiments, liquid sensor pH or Na(+)-selective microelectrodes were used. RES

A Triboelectric Nanogenerator-Based Electromechanical Synchronized Sensing Probe for Simultaneous Detection of Cardiac Physiological Activities.

Cardiovascular diseases are recognized as the leading global cause of mortality and disability, where elucidating electromechanical coupling is critical for early diagnosis and treatment. However, current technologies have been limited by the lack of synchronous acquisition of cardiac electrical and mechanical signals, restricting the quantitative evaluation of severe clinical complications such as electromechanical dissociation. In this work, an electromechanical synchronized sensing probe was developed by integrating a triboelectric nanogenerator (TENG)-based pressure sensor with a microelec

Electrical stimulation of primary olfactory nerve induces two types of variations in the extracellular potassium activity within the glomerulus of the rat olfactory bulb in vivo.

Primary olfactory nerve stimulation induces a two-step increase in extracellular potassium, as recorded by K+-sensitive microelectrodes inserted into single glomeruli of the rat olfactory bulb. The second rise in K+ (K-peak) occurs in a graded, activity-dependent manner at the basal pole of the glomerular neuropile. This phenomenon appears to be directly related to the rate of extracellular K+ accumulation and is accompanied by a substantial depression of the mitral cell firing rate.

Silk-enabled conformal intraventricular interfaces for minimally invasive neural recordings.

Flexible neural interfaces capable of monitoring subcortical neuronal activity facilitate the study of deep brain neural circuits and their interactions with the cortex. However, there exists a paucity of translational tools for interfacing subcortical nuclei surfaces within the intraventricular cerebrospinal fluid. Here, we developed a flexible and conformal intraventricular interface (IVI) featuring a deformable microelectrode array paired with a silk scaffold. The IVI can be minimally invasively implanted into the lateral ventricles with the assistance of commonly used clinical catheters, s

High-resolution mapping of infraslow cortical brain activity enabled by graphene microtransistors.

Recording infraslow brain signals (<0.1 Hz) with microelectrodes is severely hampered by current microelectrode materials, primarily due to limitations resulting from voltage drift and high electrode impedance. Hence, most recording systems include high-pass filters that solve saturation issues but come hand in hand with loss of physiological and pathological information. In this work, we use flexible epicortical and intracortical arrays of graphene solution-gated field-effect transistors (gSGFETs) to map cortical spreading depression in rats and demonstrate that gSGFETs are able to record, wi

Bringing cellular clarity to the cortical component of ALS with a high-density multi-electrode array system.

There is selective vulnerability in diseases, requiring the understanding of cell-type specific aspects of neurodegeneration with cellular resolution. Single-cell electrophysiology enables direct investigation of neuronal excitability, ion-channel dynamics, and synaptic transmission with high temporal precision, which is crucial in understanding neuronal circuitries and how they are affected in diseases. Microelectrode arrays (MEAs) have emerged as powerful platforms enabling long-term, parallel, and non-invasive extracellular measurements. The advent of complementary-metal-oxide-semiconductor

Multimodal monitoring of human cortical organoids implanted in mice reveal functional connection with visual cortex.

Human cortical organoids, three-dimensional neuronal cultures, are emerging as powerful tools to study brain development and dysfunction. However, whether organoids can functionally connect to a sensory network in vivo has yet to be demonstrated. Here, we combine transparent microelectrode arrays and two-photon imaging for longitudinal, multimodal monitoring of human cortical organoids transplanted into the retrosplenial cortex of adult mice. Two-photon imaging shows vascularization of the transplanted organoid. Visual stimuli evoke electrophysiological responses in the organoid, matching the

Dramatically Amplified Thoracic Sympathetic Postganglionic Excitability and Integrative Capacity Revealed with Whole-Cell Patch-Clamp Recordings.

Thoracic paravertebral sympathetic postganglionic neurons (tSPNs) comprise the final integrative output of the distributed sympathetic nervous system controlling vascular and thermoregulatory systems. Considered a non-integrating relay, what little is known of tSPN intrinsic excitability has been determined by sharp microelectrodes with presumed impalement injury. We thus undertook the first electrophysiological characterization of tSPN cellular properties using whole-cell recordings and coupled results with a conductance-based model to explore the principles governing their excitability in ad
