Autologous peripheral blood stem cell transplantation in tumor-stage mycosis fungoides: predictors of disease-free survival.

Nine patients with mycosis fungoides (age range 27-67) underwent autologous peripheral blood stem cell transplantation (PBSCT). All patients had tumor-stage disease, and four had lymph node involvement. Eight patients exhibited a peripheral blood T cell clone using PCR/SSCP analysis of the TCR gamma gene, six prior to harvest and two at the time of harvest. Mobilization of CD34+ stem cells was achieved with etoposide and G-CSF. Harvested cells were positively selected for CD34. After negative selection for CD4 and CD8, only two samples became PCR negative. Conditioning prior to reinfusion of s

Laboratory Evaluation and Pathological Workup of Neoplastic Monocytosis - Chronic Myelomonocytic Leukemia and Beyond.

PURPOSE OF REVIEW: Monocytosis is a distinct but non-specific manifestation of various physiologic and pathologic conditions. Among hematopoietic stem cell neoplasms, depending on the criteria used for disease classification, monocytosis may be a consistent and integral component of diseases such as chronic myelomonocytic leukemia or acute myeloid leukemia with monocytic differentiation, or it may represent an inconsistent finding that often provides a clue to the underlying genetic changes driving the neoplasm. The purpose of this review is to provide the readers with a laboratory-based appro

Targeting Sphingosine Kinase Isoforms Effectively Reduces Growth and Survival of Neoplastic Mast Cells With D816V-KIT.

Mastocytosis is a disorder resulting from an abnormal mast cell (MC) accumulation in tissues that is often associated with the D816V mutation in KIT, the tyrosine kinase receptor for stem cell factor. Therapies available to treat aggressive presentations of mastocytosis are limited, thus exploration of novel pharmacological targets that reduce MC burden is desirable. Since increased generation of the lipid mediator sphingosine-1-phosphate (S1P) by sphingosine kinase (SPHK) has been linked to oncogenesis, we studied the involvement of the two SPHK isoforms (SPHK1 and SPHK2) in the regulation of

[Significance of mitotic spindle checkpoint genes in leukemia].

Leukemia is a clonal proliferative disorder of the multipotent hematopoietic stem cells that leads to abnormal cell growth and (or) differentiation. The hallmark of the disease is the presence of oncogene expression in bone marrow or peripheral blood as a result of some chromosome translocations. The development of leukemia with transfer to disease progression presents a multistage process implicating series of molecular changes leading to chromosomal instability and aneuploidy. The most possible of these changes include the followings: appearance of additional chromosome translocations, activ

Normal and malignant human myeloid progenitors differ in their sensitivity to hyperthermia.

The effect of in vitro hyperthermia on normal human bone marrow granulocyte-macrophage progenitor cells (granulocyte-macrophage colony-forming units, CFU-GM) was compared to its effect on clonogenic acute nonlymphocytic leukemic (ANLL) cells. Mononuclear normal bone marrow cells, blasts from patients with ANLL, and HL-60 cells were incubated at room temperature (control) and at 42 degrees-44 degrees C for 0-120 min prior to assay in methylcellulose. The heat sensitivity of the leukemic cells was significantly greater than that of normal bone marrow progenitors. Two-h exposure to 43 degrees C,

Head and Neck Solitary Fibrous Tumor Presenting as Salivary Gland Tumor: Two Case Reports and Review of the Literature.

Solitary fibrous tumors (SFTs) are soft tumors (mesenchymal origin) that most likely develop from adult mesenchymal stem cells. SFTs are not common in the head and neck region, and the characteristics of tumors in this location are unclear. The present study describes the clinicopathological findings of 2 cases of SFTs arising in the parotid gland and buccal space, presenting as salivary gland tumors. The first case is a 76-year-old man presenting with a painless tumor on the right parotid gland who subsequently underwent partial superficial parotidectomy. According to the results of histopath

Therapeutic potential of MicroRNAs in targeting breast cancer stem cells: A systematic review.

BACKGROUND: A distinct subpopulation of tumour cells, known as breast cancer stem cells (BCSCs), plays a critical role in driving poor therapeutic outcomes due to its high proliferative capacity, metastatic potential and resistance to treatment. MicroRNAs (miRNAs) have emerged as a promising focus of research owing to their stability and ability to modulate tumour biology. However, the role of miRNAs in regulating BCSC characteristics remains insufficiently understood. This systematic review aims to synthesise evidence from in vitro and in vivo studies to evaluate the potential of miRNA-based

Mast cell hyperplasia: role of cytokines.

Mast cell hyperplasia is found in different pathologies such as chronic inflammatory processes, fibrotic disorders, wound healing or neoplastic tissue transformation. The functional significance of the accumulation of mast cells in these processes is largely unknown. It is now established that bone marrow-derived mast cell progenitors circulate in peripheral blood and subsequently migrate into the tissue where they undergo final maturation under the influence of local microenvironmental factors. Cytokines are of particular importance for mast cell recruitment, development, and function. Stem c

The dynamics of the metastatic niche: Lung alveolar type 2 cell reprogramming and cancer cell stemness.

The complex cellular interactions supporting metastasis formation remain incompletely understood. In this issue of Developmental Cell, Rodrigues et al. describe a positive feedback loop within the lung metastatic niche, whereby disseminated cancer cells promote the multilineage potential of alveolar type 2 cells, favoring cancer cell stemness and metastasis initiation.

Solitary Primary Intracranial Extramedullary Plasmacytoma With Lymph Node Metastasis.

Multiple myeloma is a neoplastic disease of plasma cells. Plasma cell disorders can present as a single lesion (solitary plasmacytoma) or as multiple lesions (multiple myeloma). Solitary plasmacytomas can occur in bone (plasmacytoma of bone) or in soft tissues (extramedullary plasmacytoma), and both can serve as a precursor lesion to multiple myeloma. These lesions may occur anywhere, however, intracranial presentations are rare. Here, we present a rare case of solitary intracranial extramedullary plasmacytoma in a patient complaining of headaches and vision changes. Despite radiation treatmen

Systemic mastocytosis with associated myeloproliferative neoplasm with t(8;19)(p12;q13.1) and abnormality of FGFR1: report of a unique case.

Systemic mastocytosis is a neoplastic proliferation of mast cells that frequently presents with associated clonal hematological non-mast cell lineage disease. Myeloid and lymphoid neoplasms with abnormalities of the FGFR1 gene are a heterogenous group of rare and aggressive hematopoietic stem cell disorders. About a dozen of chromosome changes involving the FGFR1 gene, presenting as myeloid or lymphoid neoplasms, have been described in the literature. To date, only 2 cases of myeloid and lymphoid neoplasms with abnormalities of the FGFR1 gene have been reported in association with systemic mas

Targeting mitotic kinesin KIF20A: A differentiation-based therapeutic strategy for glioblastoma stem/progenitor cells.

Glioblastoma multiforme (GBM) remains refractory to current treatment modalities. Differentiation-based approaches, which force cancer stem/progenitor cells to exit the cell cycle and adopt terminal fates, offer an alternative therapeutic strategy. Cell fate regulators operating during stem/progenitor cell divisions integrate proliferative and anti-proliferative cues and represent particularly attractive points of intervention. Here, we investigated the therapeutic potential of targeting mitotic kinesin KIF20A in GBM stem/progenitor cells. KIF20A is a crucial component of cytokinetic machinery

Chronic Neutrophilic Leukemia in a Patient With Multiple Myeloma.

High-dose chemotherapy used for autologous stem cell transplantation (ASCT) is often associated with the proliferation of cells with neoplastic capabilities. As a result, expansion of one particular cell line, i.e., clonal hematopoiesis (CH), can occur. The frequency of CH in multiple myeloma (MM) has been reported as ~21.6%. Although neutrophilia in MM could be a leukemoid reaction, one should not forget hematologic neoplasms such as chronic neutrophilic leukemia (CNL) - which is characterized by CSF3R mutation, and absence of cytopenia and/or significant morphologic dyspoiesis. In this case

Transcription phenotype of circulating tumor cells in non-metastatic breast cancer.

The presented research is relevant, as breast cancer is the most commonly diagnosed cancer in the female population worldwide, with the exception of skin cancer. The aim of this article is to study the transcription phenotype of circulating tumor cells in non-metastatic breast cancer. The transcriptional phenotype of circulating tumor cells (CTCs) was studied using real-time polymerase chain reaction (PCR). Three-year OS was 79.2, and 90.8 without the expression with p Log-Rank = 0.04. Independent prognostic factors for the recurrence of disease include the presence of CTCs expressing BIRC5 ge

Intravascular Lymphoma: A Unique Pattern Underlying a Protean Disease.

Intravascular lymphoma (IVL) is a rare, aggressive subtype of non-Hodgkin lymphoma (NHL) characterized by the selective proliferation of neoplastic lymphoid cells within small and medium-sized blood vessels, most frequently of B-cell origin (IVLBCL). Its protean clinical presentation, lack of pathognomonic findings, and absence of tumor masses or lymphadenopathies often lead to diagnostic delays and poor outcomes. IVLBCL can manifest in classic, hemophagocytic syndrome-associated (HPS), or cutaneous variants, with extremely variable organ involvement including the central nervous system (CNS),

The CD34 epitope is expressed in neoplastic and malformative lesions associated with chronic, focal epilepsies.

The etiology and pathogenesis of complex focal lesions associated with chronic, intractable epilepsy are largely unknown. Some data indicate that malformative changes of the central nervous system may precede the development of gangliogliomas and other epilepsy-associated neoplasms. In the present immunhistochemical study, we have examined epilepsy-associated lesions for CD34, a stem cell marker transiently expressed during early neurulation. Surprisingly, most tissue samples from patients with chronic epilepsy (n = 262) revealed neural cells immunoreactive for CD34. Prominent immunoreactivity

Advances in the research of osteosarcoma stem cells and its related genes.

Osteosarcoma is a malignant tumor that often occurs in adolescents. There is an urgent need for new treatment options for osteosarcoma due to its poor prognosis after metastasis. Cancer stem cell (CSC) theory states that CSCs represent a small proportion of cancer cells. These CSC have self-renewal ability and are closely associated with cancer growth and metastasis as well as chemotherapy resistance. Similarly, osteosarcoma stem cells (OSCs) play an important role in the growth, metastasis, and chemotherapy resistance of osteosarcoma cells. Targeting OSCs may represent a future treatment of o

HSPC-like blasts in acute lymphoblastic leukemia: biology and therapeutic opportunities.

Stem cell-like blasts have been associated with hierarchical tumor-initiating potential and poor outcomes in myeloid leukemias. Previous studies using primary samples of acute lymphoblastic leukemia (ALL) have identified blasts that immunophenotypically and transcriptomically resemble hematopoietic stem and progenitor cells (HSPCs), but failed to consistently demonstrate hierarchical tumor-initiating potential in xenograft models. Recent multi-omic profiling of lymphoblastic and mixed-phenotype leukemias has improved our understanding of the phenotypes of HSPC-like blasts and their association
