Parkinson's disease (PD) is a neurodegenerative disease that primarily affects motor function. Dopaminergic cell death occurs progressively in the PD brain and autophagic dysfunction is critically implicated in the neuronal death. Therefore, modulating impaired autophagic flux is a promising strategy to treat dopaminergic neurodegeneration, addressing the unmet needs in PD. This study aimed to investigate efficacy and the underlying mechanism of Paeonia lactiflora extract (PLE) focusing on autophagic dysfunction in 6-hydroxydopamine (6-OHDA)-induced PD models in vitro and in vivo for the first

Paeoniflorin (PF) is a key active ingredient with anti-inflammatory and antioxidant properties extracted from the root of Paeonia lactiflora. Non-alcoholic fatty liver disease (NAFLD), recently referred to as metabolic dysfunction-related fatty liver disease (MAFLD), is the leading cause of chronic liver disease worldwide. However, the potential mechanisms and targets of paeoniflorin's anti-inflammatory and antioxidant therapy for MAFLD remain to be thoroughly investigated. Thus, in cellular experiments, we added free fatty acids (P/O) to AML-12 cells and cultured them for 24 h. In animal expe

Imatinib mesylate (IM) is a first-line therapy for chronic myeloid leukemia (CML) and exhibits good therapeutic effects, but not in all patients with CML owing to drug resistance. Our previous study showed that Cyr61 plays a key role in IM resistance in CML cells. Paeoniflorin (PF) is a bioactive compound isolated from the traditional Chinese medicine Paeonia lactiflora Pall that displays anticancer activity. Little is, however, known regarding the role of PF in IM-resistant CML cells. This study aimed to evaluate whether PF could decrease Cyr61 production and improve IM-resistant CML cell sen

BACKGROUND: In China, Rhizoma atractylodis macrocephalae-Paeonia lactiflora Pall (Biazhu-Baishao, BZBS) is a classic herb pair used to treat intestinal stress syndrome, ulcerative colitis and other diseases. However, the mechanism of BZBS in the treatment of functional constipation (FC) has been little studied and remains unclear. In this study, a behavioral investigation, colon tissue morphology, enzyme-linked immunosorbent assay (Elisa) and intestinal microflora analysis have been used to illuminate the potential mechanism of the effects of BZBS on FC in a rat model.

The search for new antibiotics against multidrug-resistant (MDR), Gram-negative bacteria is crucial with respect to filling the antibiotics development pipeline, which is subject to a critical shortage of novel molecules. Screening of natural products is a promising approach for identifying antimicrobial compounds hosting a higher degree of novelty. Here, we report the isolation and characterization of four galloylglucoses active against different MDR strains of Escherichia coli and Klebsiella pneumoniae. A crude acetone extract was prepared from Paeonia officinalis Linnaeus leaves, and bioaut

Baicalin (BA), a flavonoid with diverse bioactivities, faces clinical application challenges due to its poor solubility, low bioavailability, and limited stability. This study reports on the development of BA-loaded peony seed polypeptide nanoparticles (BA-PSP NPs) using a pH-driven method to overcome these limitations. These nanoparticles (NPs) were spherical, 88.20 ± 2.19 nm in size, with a zeta potential of -35 mV, demonstrating excellent stability under thermal, pH, salt ion, and storage conditions. Physicochemical assays confirmed that BA was encapsulated in an amorphous state within the

Typical adverse effects of Shakuyaku-kanzo-To (SKT), an herbal medicine containing licorice, is a licorice-induced pseudoaldosteronism with hypokalemia and hypertension. The risk factors for this side effect are still unclear. To identify the risk factors, we surveyed 37 cases of licorice-induced pseudoaldosteronism in the literature and serum potassium levels in our patients receiving SKT and Shosaiko-To (SST), which contain 6 g and 1.5 g of licorice in the daily dose, respectively. In the case report survey, pseudoaldosteronism developed a median 35 (range 2-231) days after the administratio

Ulcerative colitis (UC) is a recurrent inflammatory bowel disease characterized by mucosal inflammation. Recently, the incidence rate of UC increases year by year, and patients diagnosed with UC usually have the poor quality of life. The search for effective treatments of UC remains a crucial research priority. Since traditional Chinese medicine (TCM) is an important treatment for UC, the components of these TCMs were analyzed. Our data indicated that Mairin was the common core component of TCMs with UC therapeutic effects, including Licorice, Paeoniae Radix Alba and Aucklandiae Radix. Recentl

Inflammatory bowel disease (IBD) is a chronic immune-mediated disorder characterized by intestinal inflammation, with frequent progression to complications including intestinal stenosis, obstruction, and perforation. The sustained inflammatory burden and gastrointestinal bleeding in IBD culminate in systemic iron depletion, predisposing patients to iron deficiency anemia (IDA). Here, we report a rationally engineered nanozyme, Radix Paeoniae Alba polysaccharide‑iron complexes (PPFeCs), designed to integrate superoxide dismutase-/catalase-mimicking antioxidase activities with bioavailable iron

Hepatic ischemia/reperfusion (I/R) injury is a major cause of high morbidity and mortality after liver resection, transplantation, and hemorrhagic shock. Paeoniflorin (PF), the main substance of glucosides in Radix Paeoniae Alba, has been widely used to treat various hepatic inflammatory diseases including I/R injury. However, the underlying mechanisms of PF on hepatic I/R injury remain further investigated. In this study, the liver I/R model was performed by clamping the portal vein and hepatic artery with an atraumatic clamp for 90 min followed by 6 hr reperfusion. PF (100 mg/kg) was given t

BACKGROUND/OBJECTIVES: Botanical supplements are increasingly investigated for their potential to address women's health concerns. Compounds that modulate progesterone receptor (PR) signaling may help manage gynecologic disorders such as endometriosis, uterine hyperplasia, and preterm birth. Because PR ligands often cross-react with the glucocorticoid receptor (GR), this study examined two botanical compounds, paeoniflorin from Paeonia lactiflora (white peony root) and isoorientin from Vitex agnus-castus (chasteberry), that were identified as modulators of GR or PR signaling.
METHODS: Lucifer
